The wild species of Sesamum have long been recognized as an important source of many valuable pest and disease resistance genes and other novel traits such as male sterility and plant type for the cultivated taxon. Utilization of these species in crop improvement is marginal because of their inherent low crossability with the cultigen. An understanding of the biological nature of incompatibility systems that impede hybridization offers tools for successful introgressions into cultivated sesame. The objective of this investigation was to observe pollen germination and pollen tube growth in cross pollinated S. indicum pistils as an indicator of pre-zygotic barriers operating in wide crosses involving S. indicum. Three wild species: S. alatum, S. radiatum and S. laciniatum were used as pollen donors in hybridization with S. indicum. In the cross S. indicum × S. alatum 85.23% of pollen grains germinated and pollen tubes reached ovaries within 8 h after pollination. In other two crosses, S. indicum × S. laciniatum and S. indicum × S. radiatum the level of incompatibility was high as evidenced by a reduced pollen germination and slow rate of pollen tube growth. Measures to circumvent incompatibility in these crosses to produce interspecific hybrids are discussed.  相似文献

BACKGROUND: Gene transfer from weeds to crops could produce weedy individuals that might impact upon the evolutionary dynamics of weedy populations, the persistence of escaped genes in agroecosystems and approaches to weed management and containment of transgenic crops. The present aim was to quantify the gene flowrate from weedy red rice to cultivated rice, and evaluate the morphology, phenology and fecundity of resulting hybrids. Field experiments were conducted at Stuttgart and Rohwer, Arkansas, USA. Twelve red rice accessions and an imazethapyr‐resistant rice (Imi‐R; Clearfield?) were used. RESULTS: Hybrids between Imi‐R rice × red rice were 138–150 cm tall and flowered 1–5 days later than the rice parent, regardless of the red rice parent. Hybrids produced 20–50% more seed than the rice parent, but had equivalent seed production to the majority of red rice parents. Seeds of all hybrids were red, pubescent and dehisced at maturity. For the majority of hybrids, seed germination was higher than that of the red rice parent. The gene flowrate from red rice to rice was 0.01–0.2% and differed by red rice biotype. The hybrids had higher fecundity and potential competitive ability than the rice parent, and in some cases also the red rice parent. CONCLUSIONS: Red rice plants are vectors of gene flow back to cultivated rice and other weedy populations. The progeny of red rice hybrids from cultivated rice mother plants have higher chances of persistence than those from red rice mother plants. The pharmacokinetics and urinary excretion following single intramuscular administration of levofloxacin at a dose of 4 mg/kg was investigated in seven male cross bred calves. Appreciable plasma concentration of levofloxacin (0.38 ± 0.06 µg/ml) was detected at 1 min after injection and the peak plasma level of 3.07 ± 0.08 µg/ml was observed at 1 h. The drug level above MIC90 in plasma was detected up to 12 h after administration. Rapid absorption of the drug was also evident by the high value of the absorption rate constant (2.14 ± 0.24 /h). The overall systemic bioavailability of levofloxacin, after intramuscular administration, was 56.6 ± 12.4%. The high value of AUC (7.66 ± 0.72 mg . h/ml) reflected the vast area of body covered by drug concentration. Extensive distribution of the drug into various body fluids and tissues was noted by the high value of Vdarea (1.02 ± 0.05 l/kg). The high ratio of AUC/MIC (76.6 ± 7.25) obtained in this study indicated excellent clinical and bacteriological efficacy of levofloxacin in calves. The elimination half-life and MRT were 3.67 ± 0.4 h and 5.57 ± 0.51 h, respectively. The total body clearance (ClB) was 204.9 ± 22.6 ml/kg/h. On the basis of the pharmacokinetic parameters, a suitable intramuscular dosage regimen for levofloxacin in calves would be 1.5 mg/kg repeated at 12 h intervals.  相似文献

Near-isogenic lines (NILs) constitute valuable tools in genetic investigations and plant breeding programs. Conventional methods for developing these are time consuming and tedious. An innovative method for identifying NILs is proposed and validated. The method involves computation of simple correlation coefficients of all possible pairs of genotypes within a mapping population using molecular marker data, and phenotypic characterization of those pairs with very high positive correlation. The pairs having both genomic and phenotypic similarity except for a single trait are considered as NILs. This strategy was tested with a doubled haploid mapping population involving CT9993 and IR62266. This population was saturated with 315 markers and comprised 154 lines. The pairs showing very high correlation coefficients (0.70–0.97) and differing for less than 10% of the markers were considered as Genotypically Closely Related Pairs (GCRPs). Graphical genotyping was employed to visualize the genome of the closely related lines. A total of 39 such pairs were subjected to rigorous evaluation for root and shoot morphological traits in two contrasting moisture regimes. Four GCRPs under well-watered condition and ten GCRPs under low moisture stress condition are statistically significant for a single phenotypic trait and are considered as NILs for their respective traits and would be the valuable materials for genetic studies. Mapped QTLs and candidate genes were employed to explain the probable cause of phenotypic difference in NILs.  相似文献

Elicitors of systemic acquired resistance are well known to reduce severity of several plant pathogenic diseases caused by fungi, bacteria and viruses. Their field applications for management of plant diseases are, however, limited because of yield penalties. Our studies on affect of Benzo (1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH), an elicitor of systemic acquired resistance, on chickpea blight caused by a fungal pathogen Didymella rabiei showed that multiple foliar applications of the chemical were effective in management of the disease under economic threshold levels. Multiple applications, however, affected chickpea grain yield adversely. The BTH induced yield penalties could be prevented by foliar spray schedule comprised of BTH and a contact fungicide mancozeb. One spray of BTH (50 ppm) followed by another of mancozeb (0.2%) was less effective (8.3% severity) than three sprays of BTH (4.2% severity) in blight control, however, this treatment enhanced grain yield significantly (1.241 t ha−1) over three sprays of BTH (0.922 t ha−1).  相似文献

Complexes of the type [M(apabh)Cl] and [M(Hapabh)(H2O)SO4], where M = Mn(II), Co(II), Ni(II), Cu(II) and Zn(II); Hapabh = acetone p-amino acetophenone benzoylhydrazone have been synthesized and characterized. Electronic spectra and μeff values suggest a square planar geometry for Co(II), Ni(II) and Cu(II) chloride complexes, whereas, octahedral geometry for the sulfato complexes. ESR data show isotropic spectra for [Cu(apabh)Cl] and axial spectra for [Cu(Hapabh)(H2O)SO4] and dx2y2 as the ground state for both Cu(II) complexes. The ligand acts as tridentate monobasic in all chloro complexes bonding through two >CN and a deprotonated enolate groups, whereas tridentate neutral in all sulfato complexes coordinating through two >CN and a >CO groups. Thermal analysis (TGA & DTA) of [Ni(apabh)Cl] complex shows a multi-step exothermic decomposition pattern. The complexes show a significant antifungal activity against Rizoctonia sp., Aspergillus sp. and Penicillium sp. and a considerably fair antibacterial activity against Pseudomonas sp. and Clostridium sp. The activity increases at higher concentration of the compound.  相似文献

The HYDRUS-2D model was experimentally verified for water and salinity distribution during the profile establishment stage (33?days) of almond under pulsed and continuous drip irrigation. The model simulated values of water content obtained at different lateral distances (0, 20, 40, 60, 100?cm) from a dripper at 10, 20, 30, 40, 50, 60, 70, 80, 90, 100, 120, 140 and 160?cm soil depths at different times (5, 12, 19, 26 and 33?days of profile establishment) were compared with neutron probe measured values under both irrigation scenarios. The model closely predicted water content distribution at all distances, times and soil depths as RMSE values ranged between 0.017 and 0.049. The measured mean soil water salinity (ECsw) at 25?cm from the dripper at 30, 60, 90 and 150?cm soil depth also matched well with the predicted values. A correlation of 0.97 in pulsed and 0.98 in continuous drip systems with measured values indicated the model closely predicted total salts in the root zone. Thus, HYDRUS-2D successfully simulated the change in soil water content and soil water salinity in both the wetting pattern and in the flow domain. The initial mean ECsw below the dripper in pulsed (5.25?dSm?1) and continuous (6.07?dSm?1) irrigations decreased to 1.31 and 1.36?dSm?1, respectively, showing a respective 75.1 and 77.6% decrease in the initial salinity. The power function [y?=?ax ?b ] best described the mathematical relationship between salt removal from the soil profile as a function of irrigation time under both irrigation scenarios. Contrary to other studies, higher leaching fraction (6.4–43.1%) was recorded in pulsed than continuous (1.1–35.1%) irrigation with the same amount of applied water which was brought about by the variation in initial soil water content and time of irrigation application. It was pertinent to note that a small (0.012) increase in mean antecedent water content (θ i ) brought about 8.25–9.06% increase in the leaching fraction during the profile establishment irrespective of the emitter geometry, discharge rate, and irrigation scenario. Under similar θ i , water applied at a higher discharge rate (3.876?Lh?1) has resulted in slightly higher leaching fraction than at a low discharge rate (1.91?Lh?1) under pulsing only owing to the variation in time of irrigation application. The influence of pulsing on soil water content, salinity distribution, and drainage flux vanished completely when irrigation was applied daily on the basis of crop evapotranspiration (ETc) with a suitable leaching fraction. Therefore, antecedent soil water content and scheduling or duration of water application play a significant role in the design of drip irrigation systems for light textured soils. These factors are the major driving force to move water and solutes within the soil profile and may influence the off-site impacts such as drainage flux and pollution of the groundwater.  相似文献
