Induction of cell DNA fragmentation by treatment of recombinant human Tumor Necrosis Factor alpha (rhTNF alpha) was examined by using mouse L929 cells derived from mouse fibroblast cells. The amount of DNA fragments derived from rhTNF alpha-treated cells, detected by alkaline elution technique, was smaller than that derived from X-irradiated cells. The rhTNF alpha caused the DNA fragmentation depending on its incubation time and concentration. The DNA damage caused by rhTNF alpha treatment correlated with its cytotoxicity. This result suggested that the DNA fragmentation is one of causes of cell death. The treatment with proteinase K of DNA obtained from rhTNF alpha-treated cells did not increase the amount of DNA fragmentation, which indicates that rhTNF alpha causes DNA-fragmentation but not DNA-protein cross-linking.  相似文献

The decay risk of airborne wood-decay fungi in the same volume of air was investigated by using an air sampler over the course of a year at three different sampling sites. Japanese cedar disks measuring 7.8 cm in diameter and about 3 mm in thickness, and with a moisture content of about 100 % were placed in a “BIOSAMP” air sampler and then exposed to 1000 l of air in the northern, central, and southwest parts of Japan. The exposed disks were incubated for 20 weeks in a damp container maintained at 26 ± 2 °C and degraded by fungi trapped on the disks. The decay risk was calculated from the mass loss during incubation, and the factors affecting the said risk were explored. The results showed that sampling sites apparently do not affect decay risk, even though the Scheffer’s climate indexes of the sites were quite different. The relation between the sampling month and decay risk reveals that decay risk remains virtually the same year-round. Relative humidity on a sampling day is one of the key factors affecting decay risk in sampling conducted at the central or southwest site. In contrast, no weather factors influenced decay risk at the northern sampling site.  相似文献

To study the effect of ionizing radiation on thyroid glands, 66 Japanese Black cattle residing in the restricted area of the Fukushima Daiichi Nuclear Power Plant in 2013–2017 were pathologically examined. There were no clinical symptoms of thyroid disease in these cattle. Three cases of goiter and seven of atrophy were found in two among the four farms examined. Cases of goiter exhibited normal morphological structure without mass or nodule formation in thyroid glands. Cellular atypia or capsular invasion of the follicular epithelium was absent. The estimated integrated dose of external radiation in goiter cases ranged from maximum 797 mSv to minimum 24 mSv. All lobules in the seven atrophic thyroid glands were affected, but pathological findings, such as inflammatory cell infiltration or stromal fibrosis, were not observed. The estimated integrated dose of external radiation in atrophic thyroids ranged from maximum 589 mSv to minimum 8 mSv. Immunohistochemical analysis of anti‐nitroguanosine and the TUNEL method in goiter and atrophic thyroid glands did not reveal any positive findings. The present study indicates that there was no significant relationship between a radiation effect and pathological findings in any thyroid glands.  相似文献

White blood cells, especially lymphocytes, are susceptible to radiation exposure. In the present study, red blood cell, total white blood cell, and lymphocyte counts were repeatedly measured in cattle living on three farms located in the “difficult‐to‐return zone” of the Fukushima nuclear accident, and compared with two control groups from unaffected areas. Blood cell counts differed significantly between the two control groups, although almost all the values fell within the normal range. The blood cell counts of the cattle in the “difficult‐to‐return zone” varied across sampling times even on the same farms, being sometimes higher or lower than either of the two control groups. However, neither a statistically significant decrease in blood cell counts nor an increase in the rate of cattle with extremely low blood cell counts was observed overall. The estimated cumulative exposure dose for the cattle on the most contaminated farm was within a range of 500–1000 mSv, exceeding the threshold for the lymphopenia. Because of the low dose rate on these farms, potential radiation damages would have been repaired and have not accumulated enough to cause deterministic effects.  相似文献

We investigated the environmental factors in Japan, including meteorological conditions, on the fertility of a European cattle breed, Holstein–Friesian, by examining conception rates in different regions. First artificial insemination and associated conception details were recorded for 69,952 Holstein female cattle. In general, meteorological conditions vary considerably according to latitude in Japanese islands. Conception rates were higher in the Northern (above 37°N) than the Southern (below 37°N) region (61.3% vs. 53.3%). All the factors analyzed in the statistical model, including insemination year, region, month, AI technician, service sire and interaction between region and month, had significant effects on the conception rate. In the Southern region, conception rates were lower in the summer months (average temperature, 27.8 °C, and maximum temperature, 32.3 °C). However, this seasonal decline was not observed in the Northern region (average temperature, 23.7 °C, and maximum temperature, 28.4 °C). Regression analysis of conception rate in relation to temperature showed highly significant negative regression coefficients (− 0.66 to − 0.63% per °C; p < 0.001) in the Southern region, but non-significant coefficients in the Northern region. Humidity had a minimal effect on conception rate in both regions. Our data collectively suggest that 37°N latitude is a threshold that affects Holstein–Friesian conception rates in Japan.  相似文献

Background

To investigate plant hybrid sterility, we studied interspecific hybrids of two cultivated rice species, Asian rice (Oryza sativa) and African rice (O. glaberrima). Male gametes of these hybrids display complete sterility owing to a dozen of hybrid sterility loci, termed HS loci, but this complicated genetic system remains poorly understood.

Results

Microspores from these interspecific hybrids form sterile pollen but are viable at the immature stage. Application of the anther culture (AC) method caused these immature microspores to induce callus. The segregation distortion of 11 among 13 known HS loci was assessed in the callus population. Using many individual calli, fine mapping of the HS loci was attempted based on heterozygotes produced from chromosome segment substitution lines (CSSLs). Transmission ratio distortion (TRD) from microspores was detected at 6 of 11 HS loci in the callus population. The fine mapping of S1 and S19 loci using CSSLs revealed precise distances of markers from the positions of HS loci exhibiting excessive TRD.

Conclusions

We demonstrated that AC to generate callus populations derived from immature microspores is a useful methodology for genetic study. The callus population facilitated detection of TRD at multiple HS loci and dramatically shortened the process for mapping hybrid sterility genes.

The cytotoxic effects of canine NK cells on CL-1 target cells were examined by scanning electron microscopy (SEM). NK cell mediated cytotoxicity on CL-1 target cells was detected by 51Cr release assay. SEM showed that a canine NK cell extended projections to the CL-1 target cell. Furthermore, the surface of CL-1 target cells changed a mesh-like structure. Therefore, the cytotoxic effects of canine NK cells on CL-1 target cells were morphologically demonstrated.  相似文献

We evaluated the suitability of fish waste diet (FWD) for culturing the minute rotifer Proales similis through the observation of their population growth and particle size selective feeding. A total of five treatments either with Nannochloropsis oculata or FWD (0.75 and 0.50 g/L) or the combination of N. oculata and FWD were set up in triplicates. P. similis were cultured in diluted natural seawater (8 g/L salinity) and 26 ± 1°C with the diet treatments being applied randomly. We daily monitored the rotifer density, the number of bacteria and the water quality in all the cultures. The population density of P. similis increased exponentially in all treatments, while the mean growth in FWD 0.75 g/L was significantly higher (p < .05) than that in the control. Addition of N. oculata to FWD resulted to lowered P. similis population growth. Bacterial colony count was high in FWD and introduction of P. similis to the diet decreased their density. The estimated bacteria ingestion rates were generally in the range of 6.03 × 102–1.24 × 104 bacteria/rotifer/hr and there was a positive linear relationship between bacterivory and rotifer population growth. We also observed a shift in the particle size distribution with a reduction in the frequency and concentration of small‐sized particles (<2.5 μm) at day 6. These results accentuate the potential of fish‐processing waste as diet for culturing P. similis which feeds on bacteria and small particles (≤2.5 μm) that are by‐products of degradation of this diet.  相似文献

Drought-sensitive bryophytes are especially vulnerable to edge effects caused by forest fragmentation. Because of increased forest fragmentation, these bryophyte species are declining and are in need of conservation. Considering that a field survey including all bryophyte species is very time consuming, methods that make identification less difficult should be tested for their usefulness in conservation. This paper describes an alternative survey method that utilizes the correlation between bryophyte life-forms and microclimates for evaluating drought-sensitive bryophytes. This survey method was examined using epiphytic bryophyte flora on tree trunks in 27 fragmented forests of Kyoto city, in the western part of Japan. The usefulness of life-forms for this evaluation was discussed based on the correlation of life-forms with species richness and microclimates. The results indicated that, while life-from richness was considered to reflect the heterogeneity of moisture availability and light intensity in bryophyte habitats, a certain life-form category (for example, fans and dendroids) seemed to correlate with drought sensitivity of bryophytes. Considering this correlation, an alternative survey method was proposed that utilizes the richness of hygrophilous life-forms. This survey method seems to be more cost-effective than a traditional all-species survey, and provides an important step toward the conservation of drought-sensitive species.  相似文献
