Summary Two linseed flax (Linum usitatissimum) lines transformed with a mutant Arabidopsis ALS gene conferring resistance to sulfonylurea herbicides were tested in a replicated, randomized field test against its nontransformed commercial cultivar parent (cv. NorLin) in normal soil and in soil containing the commonly used sulfonylurea herbicides chlorsulfuron (Glean®) or metsulfuron methyl (Ally®). There were no significant differences between the transgenic lines and the parent for any agronomic trait measured in untreated soil, indicating that there is no detrimental effect of T-DNA or foreign gene expression. Similarly, there were no significant differences for performance of the transgenic lines between the untreated and the herbicide treated soils, indicating that the transferred gene does confer a field level of tolerance to the flax. Successful identification of a desirable segregant depends partly on the parents chosen to make the crosses. This experiment was conducted to compare performance and genetic variability for seed yield, yield components, agronomic traits and harvest index of lines derived from low- and high-yielding flax (Linum usitatissimum L.) crosses, and to identify important yield components for flax seed yield improvement. The lines chosen as high yielding parents for this experiment were‘Linott’and 'Summit; and the low yielding parents were‘Grant’and Ci2395. The high-yielding lines produced significantly more seeds per boll and had a higher harvest index than the low-yielding lines. Evaluation of 161 F2:6 lines from four crosses among these lines showed that the greatest genetic variability, highest cross average, and highest F6 line seed yield occurred in the low x high cross,‘Grant’x‘Linott’. All low X high crosses exhibited higher genetic variances for seed yield than the high x high crosses. The high x high cross 'Summit’x‘Linott’had low genetic variance for seed yield. Number of bolls per area was determined by linear regression, path coefficient analysis, and stepwise multiple regression analysis to be the most important component of seed yield. This study showed that hybridization of low and high-yielding flax lines may be useful to increase genetic variability and obtain high-yielding flax lines.  相似文献

A pot experiment was conducted at University of Wales Aberystwyth, UK, Frongach field station to evaluate the effects of sowing dates on the dry matter partitioning of linseed. Root, stem and capsule (boll) weight increased progressively as sowing was delayed up to third sowing (19 April) but showed a gradual decrease when sowing was further delayed. The contribution of each of the three parts to total weight did not respond to sowing date in any consistent manner. The total dry matter per plant followed a similar pattern to the dry matter of plant parts, which increased with sowing up to third sowing (19 April) and then decreased when sowing was further delayed. Dry matter accumulation per day degree > 4 °C also increased as sowing was delayed with a peak at third sowing (19 April). Dry matter accumulation per day degree is concluded to be directly related to the total day degrees received during the growth period.  相似文献

Flax engineering to yield increased resistance to pathogens is the goal of this study. Since carotenoids act as antioxidants it is thus postulated that the accumulation of a higher quantity of these compounds in the transgenic plants might improve their resistance to pathogen infection.Our approach was based on the generation of transgenic flax overproducing carotene and analysis of its susceptibility to Fusarium infection. For transformation bacterial gene – crtB was used. As expected, transgenic plants showed increased resistance against pathogen infection.The impact of carotenoids on plant resistance to infection was verified by generation and analysis of transgenic flax with decreased content of carotene. The transgenic plants were obtained by suppression of endogenous flax gene coding for lycopene β-cyclase. Plant analysis revealed decrease in carotene content, however, an unexpected increase in resistance against Fusarium infection was detected. Further analysis of metabolites in the plants revealed that an increase in accumulation of other terpenoids and tocopherols, squalene and menthol were among them. Thus, it is suggested that repression of carotene synthesis results in the redirecting of substrates to other branches of isoprenoids synthesis.We conclude that a general level of antioxidants rather than the presence of any particular compound is the most important factor in resistance of the flax plant to pathogen infection.  相似文献

Research was conducted at the Macdonald Campus of McGill University (Québec, Canada) at three sites in 1997 and one site in 1998 to determine the effects and interactions of seeding depth (0, 1, 2, 4 or 6 cm) and seedbed preparation (i.e. soil rolling): none, rolling before or rolling after seeding on fibre flax (cv. Ariane) establishment, growth and yield. Seedbed preparation had little impact on the parameters measured while seeding depth had a variable effect on plant density, plant height, stem diameter and retted straw yield. Seeding depths of 1–4 cm provided consistently good establishment, growth and yield results. In 1997, there was an interaction between seeding depth and seedbed preparation on plant height, branching ratio and retted straw yield, although results were generally variable and tended to be site‐specific. In 1998, there was an interaction between seeding depth and seedbed preparation on plant height and stem diameter prior to harvest, with the results varying for all seeding depth‐seedbed preparation treatment combinations except for the 2‐cm depth treatment. Rolling of the seedbed before seeding on lighter soils and at a depth of 2 cm on most soils can improve establishment, growth and yields of fibre flax under eastern Canadian growing conditions.  相似文献

亚麻是一种重要的韧皮纤维作物, 木质素对亚麻纤维的性能和品质具有较大影响。以亚麻(Linum usitatissimum L.)茎秆表皮细胞的mRNA为模板, 利用木质素合成途径中关键酶基因的同源基因保守序列设计简并引物, 通过RT-PCR扩增, 电泳获得13个特异带。分别将这些DNA片段连接到T载体后转化大肠杆菌, 从重组质粒转化菌分别挑取5~8个单菌落测定插入片段序列, 得到关键酶基因新的片段序列8个。生物信息学分析结果表明, 这些新片段分别属于3个基因家族。其中, 2个CCoAOMT基因片段(GenBank登录号为EF214740, EF214741), 3个4CL基因片段(GenBank登录号为EF214737, EF214738, EF214739), 3个F5H基因片段(GenBank登录号为EF214745, EF214746, EF214747), 推测亚麻中这几个木质素代谢关键调控酶基因存在多基因家族。新获得的基因片段序列为进一步克隆全序列和了解调控亚麻木质素的生物合成奠定了基础。  相似文献

分离和克隆了亚麻纤维素合成酶基因(CesA)及其类蛋白D基因家族(CSLD)的部分序列,并对其进行生物信息学分析。纤维素合成酶类蛋白(CSL)作为一种重要的膜蛋白与纤维素合成酶具有相似的蛋白结构,都含有D,D,D,QXXRW保守区。利用其它物种中CesA基因的保守序列设计简并引物,采用RT-PCR方法扩增基因片段,经多次重复测序,获得了6个CesA基因片段和5个CSL基因片段。应用ClustalW和NCBI数据库对获得的基因片段进行序列分析,它们分别属于CesA基因家族和CSLD基因家族,6个CesA基因片段之间核酸和蛋白的相似性分别在67%~88%和71%~98%之间,5个CSLD基因片段之间核酸和蛋白的相似性分别在68%~83%和76%~94%之间。推测LusiCesA1与LusiCesA3,LusiCesA4与LusiCesA5及LusiCSLD1与LusiCSLD2的亲缘关系较近。  相似文献

以胡麻品种陇亚10号为对照,在景泰县对11个胡麻新品系进行了引种比较试验,结果表明,在露地条播栽培条件下,籽粒产量以新品系97047-13最高,达2 642.18 kg/hm~2,比对照品种陇亚10号增产122.61%;新品系2005×94301-14、陇亚10号(CK)含油率最高,均为39.72%;油产量也以新品系97047-13最高,达1 043.66 kg/hm~2,比对照品种陇亚10号增产121.40%。初步认为胡麻新品系97047-13适宜在景泰县露地种植。  相似文献
