Lizard fish (Saurida elongata) muscle protein was hydrolyzed using neutral protease to produce protein hydrolysate (LFPH), and the hydrolysis conditions were investigated using response-surface methodology. The optimum conditions for producing peptides with the highest angiotensin-I converting enzyme (ACE)-inhibitory activity were the following: enzyme-to-substrate ratio of 10,000 U/g, temperature of 48 °C, pH 7.0, and hydrolysis time of 2 h. Under these conditions, the ACE-inhibitory activity of LFPH and the degree of hydrolysis were 84% and 24%, respectively. A novel ACE-inhibitory peptide was isolated from LFPH using ultrafiltration, Sephadex G-15, and high-performance liquid chromatography. The amino acid sequence of the ACE-inhibitory peptide was identified as Ser-Pro-Arg-Cys-Arg (SPRCR), and its IC50 was 41 ± 1 µM. 相似文献

湿地是自然界生物多样性最丰富的生态景观和人类最重要的生存环境之一。近来,随着社会和经济的发展,对湿地资源的过度开发及不合理利用,造成湿地生态系统结构和功能遭到严重干扰及破坏,极大制约社会经济可持续发展,因此,对湿地保护与恢复显得尤为重要。本研究以安徽省级自然保护区沱湖湿地为研究对象,通过访谈、调查、TWINSPAN分类和DCA排序等方法,对沱湖湿地的环境状况、植物多样性和群落学特征进行分析,结果表明沱湖湿地生态环境退化明显,水生群落组成简单,主要由菹草组成,珍稀濒危植物有野莲、野大豆和野菱3种。DCA二维排序较好地验证了TWINSPAN的分类结果,其纵、横坐标都反应群丛所处生境的梯度变化。结合沱湖水域和水生生态系统的特点,分别从政策法规、技术方案、保护策略、建设模式方面提出生态重建的具体建议。 相似文献

在野外试验基础上,利用水盐平衡原理对河套灌盐荒地水盐运移规律进行研究。结果表明,盐荒地作物生育期输入盐分27.929kg/hm2,秋浇期流失盐分26.882kg/hm2,盐分积聚1.047kg/hm2,是输入盐分的3.749%。盐荒地具有一定的积盐能力。研究结果可为灌区节水灌溉的实施和土壤盐渍化的防治提供基础理论依据。 相似文献

采用超声波辅助提取枇杷花中的总黄酮,研究超声波功率、乙醇浓度、提取温度、提取时间、液料比及提取次数对总黄酮提取得率的影响。根据单因素试验,选择在超声波功率80 W、一次浸提条件下优化提取工艺;采用Box-Behnken中心组合试验设计原理,设计4因素3水平试验,以响应面分析法优化乙醇浓度、提取温度、提取时间及液料比4个因素对总黄酮提取得率的影响。结果表明:当超声波功率为80 W时,枇杷花中总黄酮超声波辅助提取的优化工艺参数为:乙醇浓度47.33%、提取温度58.42 ℃、提取时间12.01 min、液料比(mL ∶ g)45.56 ∶ 1,在此条件下枇杷花总黄酮一次浸提的提取得率可达10.48%。 相似文献

Brown seaweeds produce useful bioactive substances with high cosmetic and pharmacological values due to the presence of antioxidant derivatives, mainly phlorotannins (PHT), which are of particular interest. A central composite rotatable design (CCRD) with three variables (extraction time, dry material-to-solvent ratio and ethanol concentration) and two responses was performed to optimize the microwave-assisted extraction (MAE) of phlorotannins from Cystoseira sedoides using response surface methodology (RSM). Results showed that the independent variables significantly affected both phlorotannin content and the scavenging capacity. The optimum operating conditions were extraction time, 101.74 sec; dry material-to-solvent ratio, 1:10 g/mL; and ethanol extraction, 50%. Under these conditions, the predicted values of PHT content and radical scavenging activity-IC50 were close to the observed values and were 383.887 µg PGE/g Dm and 18.353 µg/mL, respectively. Characterization of the phlorotannin-rich fraction was conducted by high-performance liquid chromatography and Fourier transform infrared spectroscopy. The evaluation of the anticancer activity against MCF-7 breast cancer cell line showed a potent activity to trigger apoptotic death in more than a half of the MCF-7 cells, with an estimated IC50 value of 78 μg/mL. ABSTRACTExtrusion cooking plays an important role in the production of puffed snacks, which enjoy wide acceptance all over the world and are potential vehicles for increasing the consumption of n-3 fatty acids. Response surface methodology (RSM) with central composite design was adopted to investigate the effects of four independent variables (fish powder, fish oil, moisture, and screw speed) at five levels on expansion ratio, breaking strength index, hardness, color, and thiobarbituric acid reactive substances (TBARS). A second order polynomial model was used to study the main, interactive, and quadratic effects of the independent variables and to explain the relationship between the responses and independent variables. Moisture was found to be the most significant factor (p < 0.001) influencing expansion ratio, breaking strength index, and color. Inclusion of fish powder and fish oil were found to be negatively associated with the expansion characteristics, resulting in decreased lipid oxidation due to less exposed surface area of the extrudates to air. Further, the absence of linear effect of fish powder and fish oil on TBARS demonstrated the possibility to incorporate higher levels of these ingredients to produce oxidatively stable extruded snacks rich in fish proteins. 相似文献
