以豇豆胰蛋白酶抑制剂(CpTI)转基因水稻自交品系和其衍生的花培品系为实验材料,通过PCR扩增和CpTI表达蛋白活性的测定,研究了CpTI基因的遗传稳定性和表达特性。结果表明CpTI基因在转基因自交后代可以稳定遗传,而在部分花培品系中发生丢失。同时CpTI基因的表达表现出一定的时空特性,同一转基因品系中叶片CpTI蛋白的表达活性通常高于茎杆,而不同发育时期的CpTI蛋白的表达活性以分蘖期最高。除花培品系DH401外,转基因品系叶片和茎杆的蛋白酶抑制剂的活性随着水稻的生长发育呈下降的趋势。  相似文献

Summary An efficient transformation system for Chinese cabbage cotyledon explants was developed using Agrobacterium tumefaciens strains LBA4404 harbouring the plasmid pMOG 411 and the plasmid pBinΩSCK respectively. Various factors affecting the transformation efficiency and subsequent regeneration were identified. The age of seedlings, growth conditions and status of Agrobacterium suspension, preculture of explants, cocultivation time, ratio between Agrobacterium and explants, acetosyringone and concentration of kanamycin had a significant influence on transformation frequency and plant regeneration. The presences of the antibacterial peptide gene and the cowpea trypsin inhibitor gene in those selected shoots in kanamycin medium were confirmed by PCR, Southern blotting and Northern blotting, The frequency of regeneration from cotyledons was analyzed in eight parental lines of Chinese cabbage and five lines were suitable for obtaining transformed plants, among which Qingmaye C and Qingmaye D were more responsive than other lines.  相似文献

Summary Genetic changes induced by grafting were observed in soybean storage proteins, involving the 11S acidic subunit composition and Kunitz trypsin inhibitor. Changes from Gly 1 Gly 1to Gly 1 gly 1involved the 11S acidic subunit, and those from Ti 0Ti0 to Ti 3Ti0 involved the KTI in the first progeny (G1S1) in the scion (Gly 1 Gly 1 Ti 0Ti0, var. Kinzu) grafted onto var. Raiden (gly 1 gly 1 Ti 3 Ti 3). The progenies G1S2 and G2S1 from the seeds that had been changed segregated for both proteins. However, the segregation ratio in the progenies were different from those of the F2's from the sexual crosses between Kinzu and Raiden used for grafting. These findings show that the changes (changed genes) were transmitted to the progenies in an unstable manner.  相似文献

Five tannin-related compounds – gallic acid, flavogallonic acid dilactone, valoneic acid dilactone, gallagyldilactone, ellagic acid – were isolated from the heartwood of Shorea laeviforia, and the inhibitory activity of each against rat liver 5-reductase was evaluated. Valoneic acid dilactone and gallagyldilactone exhibited positive inhibitory activity, but gallic acid and ellagic acid did not. Flavogallonic acid dilactone stimulated 5-reductase activity, even though this compound is structurally similar to valoneic acid dilactone. The kinetic study of valoneic acid dilactone and gallagyldilactone indicated that the inhibitory behavior of 5-reductase were not competitive against the substrate (testosterone) and were partially competitive against the cofactor (NADPH). Additionally, double inhibition analysis of valoneic acid dilactone and NADP+ showed synergetic inhibition. These results suggested that neither valoneic acid dilactone nor gallagyldilactone can affect the binding of testosterone but that either compound could interact with an enzyme–NADP+ complex to inhibit 5-reductase.  相似文献

The inhibitory effects of 17 samples prepared from Thailand plants on 5-reductase activity were examined. The acetone extract of leaves ofArtocarpus altilis showed potent 5-reductase inhibitory activity. Fractionation guided by 5-reductase inhibition led to the isolation of 2-geranyl-2,3,4,4-tetrahydroxydihydrochalcone from the acetone extract of leaves ofA. altilis. This compound showed more potent inhibitory effect (IC50=38M) than-linolenic acid known as a naturally occurring potent inhibitor.  相似文献

The putative transgenic hybrid triploid poplars [(P. tomentosa × P. bolleana) × P. tomentosa] with CpTI gene have been outplanted in test field for 2 years. Although the authors’ previous studies have proved that they are highly resistant to 3 species of poplar-threatening insect pests and contain high content of CpTI protein in foliage, incorporation status of foreign CpTI gene in poplar genome is uncertain. In this present study, the incorporation of foreign CpTI gene in genome of 5 transgenic poplars was confirmed by PCR and Southern blotting analysis. DNA amplification showed that there were clear DNA bands of about 450bp specific to CpTI gene in transgenic lanes, while no corresponding band in non-transgenic lane was observed. Correspondingly, clear DNA hybridization signals and no signal were exhibited on film for DNA Southern blotting analysis in transgenic lanes and non-transgenic lane, respectively, which further confirmed the stable integration of foreign CpTI gene in genome of 2-year-old transgenic poplar.  相似文献

采用分级盐析和Sephadex G-50的方法分离纯化无种皮南瓜籽胰蛋白酶抑制剂,用BAPNA法进行抑制活性的测定,用SDS-PAGE法测定蛋白质的分子量,用不同的温度处理方法对它的热稳定性进行研究,并对抑制剂的Lys和Cys进行了化学修饰。结果表明在饱和度为60%的硫酸铵作用下,能够沉淀出活性较强的胰蛋白酶抑制剂,其分子量为29 512 Da,并且具有一定热稳定性,在80℃恒温处理1 h仍保持一定的抑制活性。证明了无种皮南瓜籽胰蛋白酶抑制剂是一种分子量较小的,以Lys为活性中心的具有一定热稳定性的蛋白质。  相似文献

本研究将抑制农杆菌生长的两种抑菌剂头孢霉素和特泯丁,以及3种筛选剂潮霉素B、卡那霉素和PPT除草剂,以不同浓度添加到香蕉的3种受体系统中,研究它们对香蕉组织生长与分化的影响。实验结果表明:不同抑菌剂和筛选剂对香蕉再生体系的影响程度不同,不同香蕉再生体系对相同抑菌剂和筛选剂的敏感性也不同,因此不同香蕉再生体系需要选择不同的抑菌剂与筛选剂的最佳的配比和组合配比。3种筛选剂在香蕉的3种再生体系中的筛选能力顺序都为:PPT〉潮霉素B〉卡那霉素。  相似文献
