IntroductionCystic echinococcosis (CE) is a chronic zoonotic disease caused by the larval stage of Echinococcus granulosus (E. granulosus), which affects domestic and wild carnivores as the definitive host and ungulates as intermediate hosts. In intermediate hosts, both Th1 and Th2 cells are involved in the immune responses to an echinoccocal infection. This study aimed to investigate production of IL-4, IL-10, and IFN-γ cytokines in peripheral blood mononuclear cells (PBMCs) of CE patients before and after surgical treatment.MethodsTo evaluate cytokine production in response to E. granulosus antigens, we investigated IL-4, IL-10, and IFN-γ production in PBMCs of 20 CE patients in response to hydatid cyst fluid antigen (HCF-Ag) before and after surgical treatment using ELISA.ResultsThe mean IL-4 production from HCF-Ag stimulated PBMCs was significantly decreased (p < 0.05), while IFN-γ was significantly increased in HCF-Ag stimulated PBMCs in patients after surgery (p = 0.005).Furthermore, our results showed that there is no significant difference between IL-10 production in patients before and after treatment (p = 0.562).ConclusionsOur data Indicated production of IL-4 in cultured PBMCs of CE patients stimulated with HCF-Ag was decreased significantly. While, production of IFN-γ was increased significantly in responses to HCF Ag after surgery. We concluded that the evaluation of IL-4 and IFN-γ in HCF-Ag stimulated PBMCs of CE patients should be considered as a useful marker in the follow up of patients with cystic echinococcosis.  相似文献

ABSTRACT

1. This study quantified xylanase-induced changes in soluble monosaccharides, xylooligosaccharides (XOS) and volatile fatty acid (VFA) contents of the different sections of the gastrointestinal tract (GIT) and whether these were related to altered bird performance.

2. An in vitro digestion of the wheat-based diet was carried out with the xylanase (Econase XT at 16,000BXU/kg diet) to compare the in vitro and in vivo generation of these XOS and monosaccharides. For the in vivo study, 80 male Ross 508 b roiler chicks were split into two groups fed a wheat-based diet with or without Econase XT (16,000BXU/kg diet) for 21 days.

3. There were no effects of Econase XT inclusion on growth performance characteristics, likely a result of the high-quality wheat diet, the corresponding high performance of the control group (FCR average of 1.45 in controls) and the relatively young age of the birds (from four to 26 days of age).

4. Econase XT supplementation increased the xylotetraose (X4) content in the colon (P = 0.046, enzyme x GIT section interaction) and the xylose contents in the colon and caeca (P < 0.001, enzyme x GIT section interaction).

5. The trend for increased acetate production in the caeca of Econase XT treated birds (P = 0.062) suggested that the XOS generated were subsequently fermented in the caeca, potentially impacting upon the types of microbiota present.

The present study suggested that wheat arabinoxylan degradation was enhanced by xylanase supplementation, which may have increased the production of beneficial volatile fatty acids (VFA) in the caeca, and thereby potentially modulated the caecal microbiome, but without affecting bird performance at this early age.  相似文献

为研究可食性酪蛋白膜对食品贮藏过程中油脂氧化和抗氧化能力的影响,以草莓、马铃薯、鸡胸肉为材料,研究不同可食性酪蛋白膜处理对其蛋白质含量、可溶性糖含量、抗坏血酸含量、过氧化值、TBA值和正己醛浓度的影响。结果表明,草莓涂膜组的蛋白质、可溶性糖、抗坏血酸含量最高,脂溶性涂膜组次之,水溶性涂膜组稍高于对照组;马铃薯涂膜组比对照组的氧化程度低;鸡胸肉涂膜与未涂膜样品感官明显不同,抗氧化作用明显。由此可知,酪蛋白涂膜液优于涂膜对食品的保鲜效果,脂溶性涂膜效果优于水溶性涂膜,涂膜食品保鲜技术具有较佳的抗氧化能力和抑菌效果,在一定程度上可延长货架期。  相似文献

为了研究雏鸡在正常发育过程中免疫器官内B淋巴细胞的变化规律,以不同日龄的SPF鸡为研究对象,用流式细胞术检测鸡脾脏、胸腺及法氏囊内IgA+、IgG+及IgM+B淋巴细胞含量的变化情况。结果显示,在脾脏中,4日龄时B淋巴细胞含量比较多,7日龄时B淋巴细胞含量下降,随后,7至21日龄细胞含量逐渐升高;胸腺内则是在4日龄时IgM+B淋巴细胞含量相对较高,而IgA+、IgG+B淋巴细胞几乎没有;在法氏囊中,IgA+、IgG+及IgM+B淋巴细胞含量在4至14日龄时逐渐升高,并且IgA+和IgG+细胞含量在21日龄时基本趋于稳定,但是IgM+B细胞含量则在21日龄时有所下降。试验结果表明,雏鸡出壳后各免疫器官B淋巴细胞始终以IgM+细胞含量最多。  相似文献

[目的]从抗炎方面探讨贵阳中医二附院院内制剂“复方肿节风雾化剂”在对咽炎治疗上的作用机制.[方法]采用小鼠耳二甲苯致炎法、角叉莱胶所致炎症肿胀度等法制作炎症模型观察其抗炎作用;而体外抑菌实验则用其对致咽炎的5种常见病菌的抑菌作用进行评价;同时抽取使用不同剂量复方肿节风雾化剂的咽炎试验组及空白组大鼠的血液通过分析,观察血常规的变化及其白细胞介素-1 (IL-1)的水平.[结果]复方肿节风雾化剂高、中、低3个不同浓度均能减轻在二甲苯所诱发的小鼠耳肿胀、角叉莱胶所致大鼠足趾肿胀的炎症肿胀度,但抗炎作用随着浓度的降低而降低.在体外试验中,复方肿节风雾化剂对肺炎链球菌、溶血链球菌、白喉杆菌、金黄色葡萄球菌及伤寒杆菌的最低抑菌浓度分别是76、105、38、65、30 mg/ml.复方肿节风雾化剂降低了咽炎模型大鼠血浆中的白细胞数、中性白细胞和淋巴细胞含量;阳性对照组及复方肿节风雾化剂高、中、低3个不同浓度组IL-1水平均低于空白对照组.[结论]复方肿节风雾化剂可能通过抗炎、抑菌、降低IL-1水平,达到治疗咽炎的作用.该研究为复方肿节风雾化剂的临床应用奠定基础.  相似文献

鸡脂肪组织TCF21基因启动子区DNA甲基化与其表达的关系   总被引:1,自引:1,他引:0  
旨在研究鸡脂肪组织中TCF21基因启动子区DNA甲基化水平与其表达的关系。以东北农业大学高、低腹脂双向选择品系(简称高、低脂系)第24世代7周龄肉鸡为试验材料,利用RT-qPCR检测高、低脂系肉鸡腹部脂肪组织中TCF21基因的mRNA表达水平;利用生物信息学和双荧光素酶报告系统分析TCF21基因启动子的结构与功能;利用Sequenom MassARRAY飞行质谱检测高、低脂系肉鸡腹部脂肪组织中TCF21基因启动子区CpG位点的甲基化水平;利用CpG甲基转移酶处理TCF21启动子报告基因质粒,分析DNA甲基化对TCF21基因启动子活性的影响。结果显示,高脂系肉鸡腹部脂肪组织中TCF21基因的mRNA表达水平极显著高于低脂系(P<0.001);TCF21基因的启动子区存在40个CpG位点,且在启动子的近端和远端均有分布,但不存在CpG岛;将TCF21基因的启动子划分为5个功能区域,分别为R1区域(-2 000~-1 500 bp)、R2区域(-1 500~-1 000 bp)、R3区域(-1 000~-500 bp)、R4区域(-500~-200 bp)和Core区域(-200~-100 bp);高脂系R2、R3和R2+R3区域的DNA甲基化水平显著或极显著高于低脂系(P<0.05或P<0.001);R2、R3、R2+R3区域的DNA甲基化水平与TCF21基因mRNA表达水平呈显著正相关(R2区域:r=0.438,P<0.05;R3区域:r=0.371,P<0.05;R2+R3区域:r=0.489,P<0.05);R2区域的DNA甲基化显著抑制其转录活性(P<0.05)。综上所述,TCF21基因在高、低脂系肉鸡腹部脂肪组织中的表达水平主要与其启动子R2区域的DNA甲基化水平有关。  相似文献

为研究热应激条件下,鸡SFRP1和PDK4两个与代谢相关的基因在鸡肝脏组织中的差异表达,选取12只60日龄体重相近的黄羽肉母鸡,分为正常对照组(CL)、热应激反应组(HS)和热应激反应后的降温组(HF)3个处理组,并对这两个基因在三组肝脏组织中的表达水平进行荧光定量表达分析。结果表明,在热应激状态下,SFRP1和PDK4基因在肝脏中的表达均上调,且PDK4在对照组和热应激组间差异显著(P<0.05)。本研究结果还表明,当热应激反应后温度重新降至(25±1)℃时,SFRP1和PDK4在肝脏中的相对表达量均有不同程度的恢复,进一步说明这两个基因在热应激中发挥的作用。本研究为从分子机制分析热应激提供了相应的参考。  相似文献

卢氏鸡是我国唯一片羽型的可产绿壳蛋的地方鸡种,壳色有绿壳和褐壳2种颜色。本研究采集131只卢氏绿壳蛋鸡,45只卢氏褐壳蛋鸡及18只地方白壳蛋鸡的血液,提取基因组DNA,用微卫星AY493302标记探讨了这3种壳色的基因型差异。结果表明,微卫星AY493302在卢氏绿壳蛋鸡、卢氏褐壳蛋鸡以及地方白壳蛋鸡之间基因型分布差异极显著,基因型AA(225bp/225bp)在卢氏鸡中的频率为0.396 9,且只在卢氏绿壳蛋鸡中出现。其A基因频率在卢氏绿壳蛋鸡、卢氏褐壳蛋鸡和地方白壳蛋鸡中分别为0.698 5,0.500 0,0.500 0。进一步进行测序发现,卢氏绿壳蛋鸡AA基因型在19bp处碱基和217bp处碱基均为G,但卢氏绿壳蛋鸡AB基因型在19bp处碱基却为C,在217bp处碱基与绿壳蛋鸡AA基因型同为G。卢氏褐壳蛋鸡AB基因型和地方白壳蛋鸡的AB基因型则表现在19bp处碱基为C,217bp处碱基为T。AA基因型的序列与NCBI数据库中红色原鸡比较,达99%同源的序列有V00425.1、J02015.1和NW_003763484.1,均位于鸡的1号染色体上。该微卫星标记的群体杂合度、群体多态信息含量在卢氏绿壳蛋鸡中都低于卢氏褐壳蛋鸡和地方白壳蛋鸡。尽管在所研究的卢氏绿壳群体中只有39.7%的个体表现为纯合AA基因型,但卢氏褐壳蛋鸡和地方白壳蛋鸡中没有该基因型,故而认为微卫星AY493302的纯合AA基因型可以作为对卢氏绿壳蛋鸡进行分子标记辅助选择的基因型。  相似文献

Nutraceuticals have gained immense importance in poultry science recently considering the nutritional and beneficial health effects of their constituents. Besides providing nutritional requirements to birds, nutraceuticals have beneficial pharmacological effects, for example, they help in establishing normal physiological health status, prevent diseases and thereby improve production performance. Nutraceuticals include amino acids, vitamins, minerals, enzymes, etc. which are important for preventing oxidative stress, regulating the immune response and maintaining normal physiological, biochemical and homeostatic mechanisms. Nutraceuticals help in supplying nutrients in balanced amounts for supporting the optimal growth performance in modern poultry flocks, and as a dietary supplement can reduce the use of antibiotics. The application of antibiotic growth enhancers in poultry leads to the propagation of antibiotic-resistant microbes and drug residues; therefore, they have been restricted in many countries. Thus, there is a demand for natural feed additives that lead to the same growth enhancement without affecting the health. Nutraceuticals substances have an essential role in the development of the animals’ normal physiological functions and in protecting them against infectious diseases. In this review, the uses of amino acids, vitamins and minerals as well as their mode of action in growth promotion and elevation of immune system are discussed.  相似文献

An 8-year-old neutered male Yorkshire Terrier dog presented with head pressing, vestibular ataxia, neck tenderness, and no oculocephalic reflex. A demarcated lesion in the pons was identified on MRI. The patient was tentatively diagnosed with a glioma and was treated with hydroxyurea plus imatinib and prednisolone. After 30 days of therapeutic treatment, the patient showed a clear improvement in neurological signs, which lasted for 1117 days. On day 569 after the initiation of treatment, 18F-fluorodeoxyglucose (FDG)-positron emission tomography (PET) was performed with no significant findings on visual analysis. The average and maximal standardized uptake values (SUVs) were 1.92 and 2.29, respectively. The tumor-to-normal-tissue (T/N) ratio was 0.97. The first evidence of clinical deterioration was noticed on day 1147. On day 1155, 3,4-dihydroxy-6-[18F]-fluoro-l-phenylalanine (18F-FDOPA)-PET was performed. High uptake of 18F-FDOPA was observed in the intracranial lesion. The mean and maximal SUVs of the tumor were 1.59 and 2.29, respectively. The T/N ratio was 2.22. The patient was euthanized on day 1155 and histopathologic evaluations confirmed glioma (astrocytoma). This case shows that chemotherapy with hydroxyurea plus imatinib may be considered in the treatment of canine glioma. Furthermore, this is the first case describing the application of 18F-FDG and 18F-FDOPA in a dog with glioma.  相似文献
