Effects of melatonin,p-chlorophenylalanine,and α-methylparatyrosine on plasma gonadotropin level and ovarian activity in the catfish,Heteropneustes fossilis: A study correlating changes in hypothalamic monoamines

The effects of (ip, 10 injections over 20 days) of melatonin (75 g 100 g^–1 BW), the serotonin (5-HT)-synthesis blocker, para-cholorophenylalanine (p-CPA, 10 mg 100g^{–1 BW}) and the catecholamine-synthesis blocker, -methylparatyrosine (-MPT, 10 mg 100 g^–1 BW) on gonadotropin (GTH) secretion and ovarian activity were studied in ^{Heteropneustes fossilis} during late preparatory to early prespawning (April–May). The treatments resulted in significant reductions of plasma GTH and estradiol-17 levels, the gonadosomatic index, frequency distribution of vitellogenic and postvitellogenic oocytes, and ovarian and serum ³²p-labelled alkali-labile phosphoprotein (a marker of vitellogenic activity). Most of the oocytes were nonvitellogenic or had undergone atretic changes. The hepatic ³²-phosphoprotein content increased significantly over the saline control value. The effects were similar and pronounced in the p-CPA and melatonin-treated groups but were moderate in the -MPT-treated group. Hypothalamic 5-HT content and turnover were significantly inhibited in the p-CPA and melatonin-treated groups but the content and turnover of catecholamines were not. The -MPT treatment decreased significantly the content and turnover of dopamine (DA), noradrenaline (NA), and adrenaline (A) but did not influence the 5-HT content or turnover. These results suggest that 5-HT, NA and A are stimulatory to GTH secretion and that melatonin may act on the serotonergic system to inhibit the pituitary-gonadal axis.to whom correspondence should be addressed.A part of the results was presented at the International Workshop on Pineal gland: Its molecular signals and published as an abstract in Neuroendocrinol. 共轭亚油酸（CLA）是含有共轭双键的亚油酸家族的总称,在这个家族中至少有28个同分异构体,它们主要存在于反刍动物的脂肪中.其中含量最多,生物学功能最为广泛的c9t11 CLA,占整个CLA总量的80％～90％.其生物学功能包括抗肿瘤细胞生长、改善心血管系统功能、增强机体免疫力以及预防糖尿病等.其次为t1oc12 CLA,占CLA总量的3％～5％,其主要生物学功能为降低脂肪酸的合成,抑制脂肪细胞的分化与生长等.给反刍动物饲喂具有不同脂肪酸含量特征的油菜籽、长链不饱和脂肪酸等,或对体内硬酯酰辅酶A去饱和酶的活性进行调节,均可提高反刍动物组织中的CLA含量,从而生产出具有促进人体健康的功能性畜产品.未来的研究应该阐明共轭亚油酸中的共轭结构是如何发挥其生物学功能的,在其他营养物质中是否也存在具有生物学功能的结构形式.  相似文献

动物的肠道完整性在营养、代谢和机体内环境稳定中起着非常重要的作用,是机体抵御内源性和外源性有害物质的第一道屏障.肠上皮细胞的完整性是保障肠道功能的生理基础,肠上皮受损会影响营养物质的吸收,从而降低生产性能和影响动物的健康.N-乙酰半胱氨酸(NAC)是L-半胱氨酸与还原型谷胱甘肽(GSH)的前体物质,NAC的生理功能和应用效果在很大程度上与维持细胞内的GSH水平有关.饲粮中添加NAC可以缓解脂多糖应激引起的肠黏膜损伤、乙酸刺激引起的结肠炎及改善小肠吸收功能,这些与NAC促进表皮生长因子的生成、抗黏膜细胞凋亡和促进紧密连接蛋白表达有关.本文就NAC对猪肠道功能的保护作用相关研究作一综述.  相似文献   

水稻OsNHO1基因cDNA序列分离及表达分析

OsNHO1是一种甘油激酶,受多种因素的诱导,在植物先天免疫反应中起着重要的作用。笔者采用同源序列法,根据已报道的拟南芥NHO1基因序列结合水稻基因组测序结果,筛选水稻OsNHO1基因,采用RT-PCR获得水稻OsNHO1基因全长cDNA(1 590 bp,529AA),并通过半定量RT-PCR方法分析OsNHO1基因在SA,PXO99刺激下的表达模式,结果显示OsNHO1基因的表达受SA,PXO99的诱导,为进一步研究OsNHO1的具体功能和作用机理奠定了基础。  相似文献   

Effect of microRNA-132 transfection on lipopolysaccharide-induced inflammation in rat alveolar macrophages

AIM: To investigate the effect of microRNA-132 (miR-132) transfection on the lipopolysaccharide (LPS)-induced inflammation in rat alveolar macrophages. METHODS: The rat alveolar macrophage NR8383 cultured without pyrogen in vitrowere divided into blank control group, negative control group and transfected group. The cells in the 3 groups were transfected with phosphate buffer solution (PBS), Lipofectamine 2000 and synthesized miR-132 mimic respectively. The cell proliferation was detected by Cell Counting Kit-8 (CCK-8) assay. Real-time PCR was used to detect the expression of miR-132 in the cells. After NR8383 cells were stimulated with LPS for 6 h, the NF-κB DNA-binding activity was measured by electrophoretic mobility shift assay (EMSA). The expression of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in NR8383 cells was assayed by Western blotting.RESULTS: After transfection, the expression of miR-132 was significantly higher than that in blank control group and negative control group. The growth of NR8383 cells in transfected group was significantly inhibited compared with blank control group and negative control group (P<0.05). After the cells were stimulated with LPS, the productions of NF-κB, TNF-α and IL-6 in transfected NR8383 cells were decreased compared with blank control group and negative control group (P<0.05).CONCLUSION: Transfection of alveolar macrophages with miR-132 significantly suppresses the cell growth, and inhibits inflammatory responses induced by LPS.  相似文献

Serum neopterin levels in female dogs with malignant mammary tumours

In this study, we have determined serum neopterin levels in female dogs with primary malignant mammary tumours. The study involved 50 female dogs which had a malignant mammary tumours removed surgically (32 animals with carcinoma, 12 animals with sarcoma and 6 animals with carcinosarcoma) and 10 clinically healthy female dogs. Serum neopterin levels were determined using a commercial ELISA kit. The mean neopterin levels were lower in the malignant tumour groups than in healthy animals but differences were statistically significant only in carcinoma and sarcoma groups. The decrease of neopterin levels in animals with malignant mammary tumours may suggest their decreased cellular immunity. Moreover, it might indicate that decreased activity of cellular mechanisms of the anti‐neoplastic response is one of the factors associated with the development and course of malignant mammary tumours in female dogs; however, further studies are necessary.  相似文献   

Congenital Peripheral Vestibular Syndrome in a Domestic Ferret (Mustela putorius furo)

A 3-month-old intact female ferret (Mustela putorius furo) was presented with a 2-month history of ataxia. On physical examination, the ferret had difficulty standing upright. During the neurologic examination, the patient had a left head tilt and positional strabismus, circled to the left, and was ataxic. Results of the complete blood count were consistent with a mild normocytic normochromic anemia. Initial treatment was supportive. Magnetic resonance imaging was performed and revealed an asymmetry of the inner ears. A brainstem auditory evoked response test was also performed. History, clinical signs, and diagnostic test results indicated that the ferret was suffering from congenital peripheral vestibular syndrome and left-sided deafness. Congenital disease should be considered in the differential diagnosis of young ferrets with peripheral vestibular syndrome. Supportive care and physiotherapy can improve balance and motor function, leading to an acceptable quality of life.  相似文献
