2011年4月,从海南工厂化养殖的患病方斑东风螺体内分离得到3株优势菌,经感染实验确定菌株DFL11-01为该暴发性疾病的致病菌,其对方斑东风螺注射感染的LD50为2.6×106CFU/g。采用常规形态和生理生化特征,对菌株DFL11-01进行鉴定,并以16SrRNA基因序列同源性分析法对所有3株分离菌进行同源性分析,结果表明,3株细菌均为哈氏弧菌Vibrio harveyi。药敏试验结果表明,菌株DFL11-01对阿洛西林、利福平等15种药物耐药;对头孢他啶、四环素等3种药物中介敏感;对氨苄西林、恩诺沙星、头孢三嗪、哌拉西林、左旋氧氟沙星等5种药物敏感。  相似文献

用室内实验生态学方法研究不同规格和温度对方斑东风螺(Babyloniaareolata)耗氧率(O)和排氨率(N)的影响。实验所用方斑东风螺规格为壳高(3 147±0 115)~(6 862±0 237)cm,软体部分干重(2 0145±0 12)~(11 0636±0 36)g。结果表明:(1)个体大小、温度以及二者的综合效应均对其耗氧率和排氨率有极显著的影响(P<0 01)。在实验温度范围内(13~33℃),不同规格的方斑东风螺耗氧率的总平均值为1 413mg/(h·g),排氨率的总平均值为217 05μg/(h·g)。(2)在实验条件下,方斑东风螺的软体部分干重(W)与耗氧量(R)之间的回归关系符合幂函数方程R=aWb,其中a的取值范围为0 9984~3 0128,平均值是1 7894;b值的范围为0 7692~0 8985,平均值为0 8430。在13~28℃温度范围内,不同规格的方斑东风螺的耗氧率随着温度的升高而增加,当温度升高到33℃时,耗氧率反而下降。(3)实验温度下,W与排氨量(A)也可用幂函数方程A=cWd表示,c值的范围为277 4091~405 3933,平均值为326 6857;d值的范围是0 4519~0 8864,平均值是0 6683。随着温度的升高方斑东风螺的排氨率呈上升趋势。(4)实验证实,高温将进一步提高方斑东风螺蛋白质的代谢水平,耗氧率和排氨率与温度(T)、W的二元线性回归方程分别为:O=-0 521-0 038W+0 050T、N=-152 640-10 581W  相似文献

在(25.8±1.7)℃条件下,测定了方斑东风螺(5.25±0.53)g在不同时间(7、15、25和40d)饥饿处理后再投饵30d过程中的生长参数、基本营养成分及组织RNA/DNA比值的变化。饥饿状态下,螺体水分含量逐渐上升,第15天时显著高于对照组;脂肪与糖原含量均下降,并分别于饥饿15d、25d时显著低于对照组(P<0.05);而蛋白质含量在不同处理组间无显著性差异(P>0.05);足肌与肝胰脏中RNA/DNA比值均随饥饿时间延长而逐渐降低。恢复生长后,除饥饿40d组含水量显著高于对照组外(P<0.05),该组其余营养成分及其它各组相应指标均恢复至或接近对照组;RNA/DNA比值除在饥饿40d组肝胰脏中仍较低外均接近或显著高于对照组。各组幼螺摄食率(FR)均高于或显著高于对照组(P<0.05),体重增量、食物转化率(FCE)在前3处理组及特殊生长率(SGR)在饥饿7d与25d组均与对照组间无显著差异(P>0.05),而饥饿15d组的SGR显著高于对照组(P<0.05);饥饿40d组尽管FR显著提高,但体重增量、FCE及SGR均显著低于对照组(P<0.05)。结果表明,方斑东风螺幼螺饥饿时主要消耗脂肪与糖原...  相似文献

为研究泥东风螺线粒体基因组遗传进化和遗传多样性,采用PCR扩增和直接测序技术,对泥东风螺线粒体基因组全序列及3个野生群体线粒体基因测序,并用生物软件进行分析。研究结果表明,泥东风螺线粒体基因组上的tRNA-Met、tRNA-Tyr、tRNA-Cys、tRNA-Trp、tRNA-GLn、tRNA-Gly、tRNA-Glu和tRNA-Thr位于环形线粒体L链上;18种腹足纲动物线粒体全长基因相似性为70.1%~92.9%,种间的12SRNA和16SRNA相似性与线粒体基因组全长相似性相当;12SRNA的391~491bp区域和16SRNA的1~106bp和361~481bp区域为高度变异区,而16SRNA的1191~1317bp区域为高度保守区;以F5引物扩增测序3个野生群体,共检测到由40个多态位点组成的26个单倍型,野生群体单倍型多样性为(0.406±0.112)~(0.532±0.113),野生群体核苷酸多样性为(0.00094±0.00124)~(0.00111±0.00123);北海野生群体与连江和长乐野生群体之间的遗距离分别为0.00094和0.00099。其结果可以为泥东风螺遗传进化、人工增殖和种质资源保护评估提供参考。  相似文献

A growth trial was conducted to determine the effects of dietary carbohydrate level on growth performance, feed utilization and metabolism of juvenile spotted babylon. Six isonitrogenous and isoenergetic experimental diets (48% crude protein and 15 MJ kg− 1 diet) using wheat starch as the carbohydrate source, were formulated to contain six carbohydrate levels. Triplicate groups of 45 animals (initial average weight, 168.39 ± 0.69 mg) were stocked in 120-l tanks and fed to apparent satiation twice daily for 10 weeks. Growth performance and feed utilization were significantly affected by dietary carbohydrate level. Maximum weight gain and specific growth rate occurred at 20% dietary starch inclusion, survival and soft body to shell ratios were not significantly different among diets. There were significant differences in protein, lipid, moisture and glycogen content in soft body. Glycogen content in soft body was positively correlated with dietary starch level. The activities of glucose-6-phosphate dehydrogenase and fructose-1,6-bisphosphatase were significantly affected by dietary starch level, with both peaking in the 20% treatment; however, there were no significant differences in 6-phosphofructokinase activity in any treatment. Quadratic regression analysis of weight gain against dietary starch level indicated that the optimal dietary carbohydrate level for maximum weight gain of juvenile spotted babylon is 27.1% of dry diet.  相似文献

Growth and water quality for growing-out of juvenile spotted Babylon, Babylonia areolata, were determined at three water-exchange regimes of 7-, 15- and 30-day intervals in large-scale operation of earthen ponds over a 5-month period. Results showed that growth was significantly different among the water-exchange treatments (P < 0.05). The higher body weight gains and shell length increments were observed in snails held at water exchange of 7- and 15-day intervals when compared with those held at water exchange of 30-day intervals. At the end of the experiment, average body weight gains were 4.22, 3.73 and 2.77 g for snails held in water-exchange treatments of 7-, 15- and 30-day intervals, respectively, and 16.63, 15.04 and 13.78 mm for those of shell length increments, respectively, The average final survivals were 83.60, 80.90 and 74.20% for snails held in water-exchange treatments of 7-, 15- and 30-day intervals, respectively. Seawater temperature, salinity, pH, dissolved oxygen, nitrite–nitrogen and ammonia–nitrogen had gradually changed throughout the culture period for all water-exchange treatments. With one exception, the total alkalinity showed the greatest changes during the culture period for all water-exchange treatments ranging from 58.67–97.50 mg/l.  相似文献

Eight growth trials of juvenile B. areolata at different water depths (20 and 50 cm) and water flow durations (6, 12, 18, and 24 h) were conducted in a flow-through culture system over 5 months. One-way ANOVA performed on each growth data set showed that there were significant differences among water depth (WD) and water flow duration (WFD) treatments (P < 0.05). The highest weight gain (6.29 g /snail) was found in snails reared in the treatment of WD50 /WFD24, followed by those from WD50 /WFD18 (6.15 g/snail), WD50/WFD12 (5.90 g/snail), WD50 /WFD6 (5.83 g/snail), WD20 /WFD24 (5.86 g/snail), and WD20 /WFD18 (5.87 g /snail), while the lowest one was found in WD20 /WFD6 (5.52 g/snail) and WD20 /WFD12 (5.57 g/snail). The relative weight gain (RWG) and individual weight gain (IWG) of the snails showed similar trends as the weight gain. Final survival exceeded 95% for all treatments. Based on all criteria, the best growth performances of juvenile spotted babylon were found in the WD50 /WFD18 and WD50 /WFD24 treatments, followed by the treatments of WD50/WFD12, WD50 /WFD6, WD20 /WFD24, and WD20 /WFD18; the lowest growth was found in WD20 /WFD6 and WD20 /WFD12.  相似文献

对海南发生的方斑东风螺(Babylonia areolata)“急性死亡症”进行了病原分离纯化、人工感染试验,以及病原16SrDNA分析和生理生化鉴定。结果显示病原菌为哈维弧菌(Vibrioharveyi);药敏试验结果显示,该病原菌对常见的抗菌药物如恩诺沙星、复合磺胺等敏感,而对青霉素表现为耐药;组织病理结果显示鳃叶、腹足、肝胰腺、消化道、肾等器官的细胞出现变性、坏死、血细胞浸润等现象。  相似文献

为了解方斑东风螺养殖系统中细菌种群结构,研究和优化方斑东风螺地膜覆沙池养殖体系的管理模式。试验采用PCR-DGGE技术,对方斑东风螺地膜覆沙池养殖系统和潮线下养殖系统底质样品和水体样品中的细菌种群结构进行了分析。试验结果显示,以广东省湛江市东海岛海水为基础构建的方斑东风螺养殖系统内主要细菌种群包括变形菌门、绿弯菌门、蓝细菌门和放线菌门。其中地膜覆沙池养殖系统主要细菌种群包括γ-变形菌纲、α-变形菌纲、δ-变形菌纲、变形菌纲、厌氧绳菌纲、蓝细菌和放线菌等;潮线下养殖系统主要细菌种群包括γ-变形菌纲、α-变形菌纲、δ-变形菌纲、厌氧绳菌纲、蓝细菌,而变形菌纲和放线菌的分布较少。被检测的两个养殖系统中细菌种群均具有较高的多样性,UPGMA聚类分析结果表明,被检测的两个养殖系统底质样品S1和S2为一簇,种群结构相似度为54.5%;两个养殖系统的水体样品S3和S4为一簇,种群结构相似度为84.0%;试验结果说明,通过池塘水体更新和微生物种群监测等调控手段有效控制养殖系统微生物种群结构,能够使方斑东风螺地膜覆沙池养殖系统的微生物生态水平接近潮线下养殖系统。  相似文献
