The gonadotropin (GTH)-stimulated testicular androgen secretionin vitro and the ultrastructure of Leydig and Sertoli cells was studied during the pubertal development in male African catfish. Testicular weight increased from less than 1 mg in the ninth week of age to nearly 600 mg in the 28th week. Immature testes (stage I: spermatogonia) were highly sensitive to GTH and secreted very high amounts of androgens per mg of tissue. The secretion per mg tissue decreased gradually in stages II (spermatogonia and spermatocytes) and III (spermatogonia, spermatocytes, and spermatids), but precipitously in stage IV (all germ cell stages, including spermatozoa). However, due to the testicular weight gain, the total androgen output per pair of testes increased slightly in stage III and strongly in stage IV. The sensitivity to GTH decreased with the appearance of haploid germ cells in stage III. Leydig cells but not Sertoli cells showed the ultrastructural characteristics of steroid producing cells. Leydig cell morphology did not change in stages I–III, while in stage IV, more smooth endoplasmic reticulum was present. The ultrastructural characteristics of Sertoli cells did not change prominently. Thus, spermatogonial multiplication and spermatocyte formation takes place when the testicular steroidogenic system is highly active and responsive to GTH; whereas the differentiation of haploid germ cells is accompanied by a reduced responsiveness to GTH and by the secretion of several-fold lower androgen amounts per mg of tissue.  相似文献

An experiment was conducted to induce ovulation in Asian catfish, Clarias batrachus, by a single injection of SGnRHa (d ‐Arg6, Trp7, Leu8, Pro9, Net) in combination with domperidone. The effects of latency periods, 11, 14, 17, 20 and 23 h, and doses of inducing agent, 10 μg SGnRHa+5 mg domperidone, 20 μg SGnRHa+10 mg domperidone, 30 μg SGnRHa+15 mg domperidone and 40 μg SGnRHa+20 mg domperidone kg?1 body weight, were studied on the total egg output, stripping response, fertilization, hatching and normal larval production. The highest (P<0.05) number of eggs were stripped at 23 h of post injection of 20 μg SGnRHa+10 mg domperidone kg?1 female body weight. The highest (P<0.05) stripping response was observed when the females were stripped at 20 and 23 h latency, at all dose levels of the inducing agent. The eggs stripped at 11 h latency did not fertilize, and hence did not hatch irrespective of administration of any dose levels of the inducing agent. The fertilization and hatching per cent of eggs had significantly increased (P<0.05) with increase in latency period to 14–23 h at a dose of 20 μg SGnRHa+10 mg domperidone. The latency period of 14–17 h, and dose of 20 μg SGnRHa+10 mg domperidone and 30 μg SGnRHa+15 mg domperidone kg?1 of female, was found to be suitable to obtain best spawning performance, and good‐quality egg and larval production in C. batrachus.  相似文献

Clarias ganepinus from wild population, which was purchased from fish mongers in Ado-Ekiti, Nigeria, were examined for possible deformities in their body. Eighty individuals were collected. Various types of deformities observed were recorded and later photographed. The present study reported for the first time the occurrence of morphological deformities in C. gariepinus natural ecosystem in Ado-Ekiti, Nigeria. The rivers, where the fish were collected, were found to have been exposed to persistent human activities. The possible causes of the deformities may be pollution from human activities or other factors such as poor nutrition, heredity, diseases. The etiology of the deformities in Clarias under this condition is recommended to be studied in detail.  相似文献

Abstract

Effects of salinity on embryonic development and growth of African catfish, Clarias gariepinus, eggs and larvae were studied. Eggs were incubated at 27-29°C in 2,4,6,8, and 10 ppt sodium chloride. Rate of embryonic development was delayed in all salt solutions by 15, 15,28 and 30 minutes, in 2,4,6, and 8 ppt sodium chloride, respectively, when compared with the control group (0% salt); total mortality occurred at 12 hours after gastrula stage in the 10 ppt concentration. Percentage hatching was 45.1,47.7, 59.5,49.2, and 26.6% while percentage deformity was 10.4, 16.1, 52.0, 28.6, and 71.6% in 0, 2, 4, 6, 8, and 10 ppt salt treatments, respectively. There were significant differences (P <0.05) in the hatching percentage and in deformity percentage between 4, 6, and 8 ppt. Rate of yolk absorption was significantly faster in the control and 2 ppt salt treatments, but slower in 4, 6, and 8 ppt. Rate of increase in length was slower with increasing salinity. The optimum salinity for African catfish eggs and was between 0-2 ppt and acceptable up to 6 ppt. The results suggest that increasing salinity delayed hatching and development of African catfish eggs and larvae, respectively, as well as increased the deformity of the larvae.  相似文献

胡子鲇脑型芳香化酶基因全长cDNA克隆及表达   总被引:1,自引:0,他引:1  
采用RT-PCR和RACE法,克隆了胡子鲇(Clarias fuscus)脑型芳香化酶基因Cyp19a1b,应用荧光实时定量PCR检测其在前脑、下丘脑、脑垂体、肝、精巢和卵巢6种组织,以及性腺分化前后(出膜后12~30 d)全鱼中的mRNA表达。结果表明,Cyp19a1b cDNA全长2 347 bp,5′端非编码区219 bp,3′端[不包括poly(A)]596 bp,开放阅读框(ORF)1 503 bp,编码500个氨基酸,推测其编码蛋白质分子量为56.388 kD。序列分析及分子系统进化树结果表明,胡子鲇Cyp19a1b氨基酸序列与非洲鲇(Clarias gariepinus)Cyp19a1b同源性最高,达95.6%;与南方大口鲇(Silurusmeridionalis)、黄颡鱼(Pelteobagrus fulvidraco)、斑马鱼(Danio rerio)、斑点叉尾(Ictalurus punctatus)、赤点石斑鱼(Epinephelus akaara)、鲤(Cyprinus carpio)、鲫(Carassius auratus)及稀有鲫(Gobiocypris rarus)Cyp19a1b同源性达75%以上,但与上述鱼类的性腺型芳香化酶基因(Cyp19a1a)同源性低于62%,表明其为脑型芳香化酶,同源性分析结果与根据传统形态学和生化特征分类的结果相一致。荧光实时定量PCR结果显示,Cyp19a1b在胡子鲇上述组织中均有表达,其中下丘脑中表达量最高,肝和精巢最低,在脑部的表达存在明显的性别差异(P<0.05)。此外,在胡子鲇性腺分化前(出膜后12 d)Cyp19a1b即开始表达,但分化前后表达量无显著性差异(P>0.05)。结果暗示Cyp19a1b可能不是引起胡子鲇性腺分化的直接因素,但其可能通过"下丘脑垂体性腺轴"对性腺分化过程产生间接影响。  相似文献

A preliminary feeding trial was conducted to evaluate the nutritive value of adult variegated grasshopper (Zonocerus variegatus) meal (VGM) as a replacement for fish meal in the diets of Clarias gariepinus fingerlings. Proximate analysis result showed that VGM had 61.50% crude protein, 6.87% crude lipid, 27.33 NFE and 9.35% chitin. Five isonitrogenous (30%) and isoenergetic (18.42 MJ kg?1) diets were formulated in which VGM progressively replaced fishmeal at five inclusion levels (0%, 25%, 50%, 75% and 100%). The best performance in terms of % weight gain was observed in the group of fish fed ED1 (25% inclusion level) and was not significantly different (P>0.05) from the groups fed the control diet. Weight gain and specific growth rate were significantly lower (P<0.05) in the group fed ED4 (100%VGM). Feed conversion ratio and protein efficiency ratio were numerically but not significantly different (P>0.05) among the different groups. The results indicate that apparent protein and lipid digestibility were high at all levels, although there were decreases with an increase in the inclusion levels. Carcass lipid decreased with an increase in the inclusion level of VGM. The VGM can replace fish meal up to 25% in the diets of C. gariepinus fingerlings without any adverse effect on growth and nutrient utilization.  相似文献

Two gonadotropins (GtH; Qa and Qb) were purified by gel filtration and ion exchange chromatography from the pituitaries of Indian walking catfish (Clarias batrachus). The presence of GtH during purification was assessed by in vitro oocyte maturation and in vivo steroidogenic activity, and their identities were determined by elution profiles, molecular weight, biological activities and yield. The molecular weights of Qa and Qb were 37 and 42 kDa, respectively, and composed of distinct subunits (Qa: 20 and 14 kDa and Qb: 26 and 18 kDa). Polyclonal antibodies raised against Qa immunostained Qa, Qb and pituitary GtH cells. A competitive Qa‐ELISA was developed whose sensitivity was 6.25 ng mL?1 (1.25 ng well?1) with intra‐ (3.5%) and inter‐ (12.4%) assay coefficients of variation. Displacement curves parallel to the standard were obtained with plasma and pituitary extracts of catfish, Qb and carp GtHII. The assay was validated by measuring the plasma Qa levels after LHRH treatment and in relation to ovarian growth in the female catfish during different reproductive phases. Based on the results, Qa and Qb corresponded to fish LH and FSH respectively. The findings will increase the knowledge of the mechanisms controlling fish reproduction and identification of sensitive phases in fish in captivity for hormonal manipulation.  相似文献

Enteric septicaemia of catfish (ESC) caused by Edwardsiella ictaluri is becoming an increasing problem in aquaculture and has been reported worldwide in a variety of fish species. This study reports ESC in hybrid catfish, Clarias macrocephalus (Günther) × Clarias gariepinus (Burchell), cultured in southern Thailand. The bacteria were identified as E. ictaluri by conventional and rapid identification systems, as well as by genetic and phylogenetic characterization. Analysis of 16S rRNA indicated 100% homology to the 16S rRNA sequence of several E. ictaluri strains in GenBank. Plasmid profiles demonstrated 4.0‐ and 5.6‐kb plasmids, compared with the 4.8‐ and 5.6‐kb plasmids in the US isolates, and representative genes of three of the four known pathogenicity islands of US isolates were present. Serologically, lipopolysaccharide (LPS) purified from the Thai isolates was not recognized by a monoclonal antibody against the LPS of US isolates. Fish experimentally infected with E. ictaluri showed 23–100% mortality within 14 days with a 168‐h LD50 of 6.92 × 107 CFU mL?1 by immersion and a 96‐h LD50 of 1.58 × 106 CFU fish?1 by intraperitoneal injection. Examination of tissue sections obtained from both naturally and experimentally infected fish indicated that infection of hybrid catfish with E. ictaluri produced lesions in several organs including liver, kidney, spleen, heart and brain. Histopathology findings included cellular necrosis, focal haemorrhage, infiltration of lymphocytes and multifocal granulomatous inflammation in the infected organs.  相似文献
