AIM: In order to study the relationship between mitochondrial deficiency and Alzheimer's disease(AD), we used sodium azide, a specific inhibitor of cytochrome C oxidase (COX), to develop a cell model of mitochondrial complex IV deficiency and investigated the impairment of microtubules and microtubule-associated proteins. METHODS: Primary cultured hippocampal neurons of hewborn rats were exposed to sodium azidethen cell viability was measured by MTT method; cell morphology, immunofluorecence-stained cellular microtubules and microtubule-associated proteins were observed by confocal microscopy. RESULTS: Primary cultured hippocampal neurons were exposed to 8-128 mmol/L sodium azide for 3-24 h, MTT absorbance decreased dose-and time-dependently. Exposed to 64 mmol/L sodium azide for 6 h, the processes of cells retracted, synapses disappeared, axons were shortened under contrast microscope. Meanwhile, microtubles were disassembled and became disorderly, the expression of microtubule-associated proteins were also reduced especially in the processes observed by confocal microscopy. CONCLUSION: Sodium azide inhibits the assembly and polymerization of tubulin in microtubules which may be reduced by low expression of microtubule-associated proteins in nerve cells. The damage of axons induced by microtubule collapse further blocks the intercellular signal transduction and intracellular material transportation which are important causes in cell death.  相似文献

AIM: To investigate the effects of Ginkgo biloba extract 761 (Egb761) on synaptophysin (SYN) expression in hippocampus of vascular dementia (VD) rats.METHODS: VD rat models, established by repeatedly cerebral ischemia/reperfusion, were randomly divided into two groups: model group and EGb761 treated group (both n=30), and another 30 condition-matched rats were selected as the sham-operated controls. Spatial learning and memory abilities of rats were assessed by Morris water maze (MWM) task, and SYN expression in hippocampal formation of rats in different groups was detected by immunohistochemical technique and image analysis.RESULTS: The MWM escape latency (EL) in model group was highly longer than that in the sham-operated group (P<0.01), while the EL of EGb761-treated group was significantly shorter than that in model group, but still longer than that in the sham-operated group at 1 m, 2 m and 4 m after VD modeling operation (P<0.05 or P<0.01). Immunohistochemical analysis showed that the SYN immunoreactive expression in hippocampal formation in model group greatly decreased and mean optical density of SYN expression highly increased compared with both sham-operated group and EGb761-treated group at three time points (P<0.01).CONCLUSION: EGb761 can increase the expression of SYN in hippocampus, which may be one of important mechanisms of EGb761 in improving learning and memory dysfunction of VD rats.  相似文献

AIM:To investigate the effects of hypobaric hypoxic exposure on the morphological changes of dendritic spines and the expression of filamin-A in the neurons of mouse hippocampal CA1 region. METHODS:C57BL/6 male mice (6~8-week-old) were divided into normoxia 7 d group, normoxia 14 d group, hypobaric hypoxia 7 d group and hypobaric hypoxia 14 d group. The mice in hypobaric hypoxia exposure groups were placed in a hypobaric chamber with hypobaric hypoxia exposure to simulate the plateau at an altitude of 6 000 m. Golgi staining assay was used to observe the branch number of dendrites, and the length and density of basal and apical dendritic spines in the hippocampal CA1 region. The protein expression of filamin-A in the hippocampus of the mice was determined by Western blot. The protein expression and distribution of filamin-A in the hippocampal CA1 region were detected by immunofluorescence staining. RESULTS:Compared with normoxia exposure group, no significant difference of the number of dendritic branches in the hippocampal CA1 region after hypobaric hypoxia exposure was observed. However, the length of basal spines and apical spines was increased significantly (P<0.05), and the density of basal spines and apical spines was significantly reduced after hypobaric hypoxia exposure (P<0.01). The results of Western blot showed that the protein expression of filamin-A in the hippocampus of the mice after hypobaric hypoxia exposure was lower than that in normoxia exposure group (P<0.01 or P<0.05). Immunofluorescence staining showed that the filamin-A protein was expressed in the mouse hippocampal CA1 region, and the expression level after hypobaric hypoxia exposure was lower than that in normoxia group. CONCLUSION:Chronic hypobaric hypoxia exposure affects the protein expression level of filamin-A in the mouse hippocampal CA1 region, thus leading to the morphological changes of dendritic spines in the hippocampal CA1 region.  相似文献

AIM: To investigate the effect of chronic intermittent hypoxia on AMP-activated protein kinase(AMPK) pathway in the brain of young rats. METHODS: Part one: SD mice(3~4 weeks old) were randomly divided into 4 groups(n=8): simulated air control group for 2 weeks(2AC), chronic intermittent hypoxia group for 2 weeks(2IH), simulated air control group for 4 weeks(4AC) and chronic intermittent hypoxia group for 4 weeks(4IH). Part two: SD mice(3~4 weeks old) were randomly divided into 2 groups(n=8): chronic intermittent hypoxia group for 4 weeks(4IH) and chronic intermittent hypoxia group treated with AMPK inhibitor for 4 weeks(4IHI). After modeling, the eight-arm maze test was performed. TUNEL method was used to detect the neuronal apoptosis in the hippocampal and prefrontal cortical tissues. The mRNA expression of adenosine A2a receptor was examined by RT-qPCR, and the protein levels of phosphorylated AMPK(p-AMPK) and mammalian target of rapamycin(p-mTOR) were determined by Western blot. RESULTS: Compared with control group, the numbers of reference memory error(RME), working memory error(WME) and total error(TE) in 2IH group and 4IH group significantly increased(P<0.01). Compared with 2IH group, the numbers of errors in 4IH group also increased significantly(P<0.01). Compared with 4IH group, the values in 4IHI group significantly decreased. Compared with control group, the neuronal apoptosis of hippocampus and prefrontal cortex in 2IH group and 4IH group increased, and that in 4IH group was more evident(P<0.05). In 4IHI group, the neuronal apoptosis decreased. The mRNA expression of adenosine A2a receptor in the hippocampal and cortical tissues in 2IH group and 4IH group was higher than that in control group. The protein level of p-AMPK was higher, and p-mTOR was lower in 2IH group and 4IH group, and those in 4IH group were more evident(P<0.05). Compared with 4IH group, the protein level of p-AMPK was lower, and p-mTOR was higher in 4IHI group. CONCLUSION: Chronic intermittent hypoxia induces neuronal apoptosis, resulting in impairment of learning and memory in a time-dependent manner by upregulating adenosine A2a receptor, activating AMPK activity, and inhibiting mTOR phosphorylation in rats.  相似文献

为评价复合营养型强化剂对灰海马(Hippocampus erectus)幼体氨基酸组成模式和营养品质的影响,用0.045 mL/L(A)、0.090 mL/L(B)和0.180 mL/L(C)3个浓度的50DE-微囊强化的卤虫投喂灰海马幼体。结果显示,灰海马氨基酸总量(TAA)的平均值为51.10%(质量分数,干样);灰海马含有17种氨基酸(AA),其中8种人体必需氨基酸总量(TEAA)占TAA比例的平均值为31.00%,4种呈味氨基酸总量(TDAA)占TAA的比例超过45.00%(质量分数,干样)。限制性氨基酸为异亮氨酸(Ile)和亮氨酸(Leu)。必需氨基酸指数(EAAI)的平均值为36.97。随着强化剂浓度升高,卤虫和灰海马体内部分AA、TAA、TDAA和非必需氨基酸(TNEAA)的含量及大多数单个必需氨基酸含量与必需氨基酸总量比值(A/E)均与强化剂浓度呈反比(P<0.05)。但是,卤虫和灰海马的TEAA/TAA和TEAA/TNEAA的值却随强化剂浓度升高而上升(P<0.05)。同时,从氨基酸评分(AAS)、化学评分(CS)和EAAI的评分模式可以看出,B、C两组灰海马的评分总体显著高于A组(P<0.05),但B、C两组间大部分氨基酸评分差异不显著(P>0.05)。结果说明,过高浓度的强化剂无法显著提高氨基酸的相对含量,但却能在一定程度上改善氨基酸的组成模式。  相似文献

  • 1. The implications of shrinkage associated with desiccation and ethanol preservation for seahorses (genus Hippocampus) were investigated using Hippocampus guttulatus (European long‐snouted seahorse) as a model. Specifically, this research addressed the implications of preservation for taxonomy and life history studies and the application of minimum size limits (MSL) for managing seahorse trade.
  • 2. In 2004, the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) listed all seahorse species on its Appendix II, and recommended a 10 cm MSL as an interim means of ensuring sustainable international trade. Inconsistencies in seahorse measurement methods and repeatability posed challenges for applying the MSL. Moreover, the shrinking effect of desiccation on body length observed in other fish was assumed to be negligible for seahorses because of their high degree of ossification.
  • 3. Changes in seahorse sizes were measured following immersion in ethanol and desiccation. H. guttulatus shrank on average by 0.1–2.3% when preserved in ethanol and 3.0–6.4% when dried, depending on the trait measured. Similar trends were observed in a sample of H. kuda (yellow seahorse). Specimen posture during drying, and measurement methods also influenced estimates of size.
  • 4. Based on the shrinkage observed, 14–44% of captured seahorses that are dried could shrink to below the recommended MSL, even if all seahorses were longer than the MSL at capture. This demonstrates that small changes in body lengths can have significant implications for trade of species managed with size limits.
  • 5. Recommendations are to (1) standardize seahorse measurement methods, (2) consider the effects of preservation and measurement technique on body lengths, and apply appropriate corrections in comparative studies and when developing fisheries management strategies, and (3) adjust size limits at the point of capture to ensure retained seahorses comply with the CITES recommended MSL.

为了解灰海马的细胞遗传学特征,便于今后开展灰海马的种质评价与鉴定、规模化人工繁育、亲缘关系研究及人工选育等工作,实验以雌雄灰海马的背鳍为材料,采用秋水仙素浸泡和常规热滴片法制备染色体标本。借助Photoshop图像软件,将同源染色体配对、拼贴,做出染色体核型图。采用Image J软件的自定义曲线测量功能,以着丝粒的中心位置为起点,顺着染色体弯曲的形态,到染色体臂末端为终点,测量线段长度,得出图片中染色体的臂长,根据相同放大倍数下标尺的测量值,换算出染色体的实际臂长。根据臂比值,将染色体进行配对、分类后,得出灰海马的染色体核型公式。结果显示,灰海马的背鳍组织可作为其染色体制备的理想材料,实验选用的染色体制备方法能获得图像清晰、形态良好的细胞分裂相。此法简单、效果好,解决了海龙科鱼类染色体制备的难题;灰海马具有22对染色体,二倍体染色体数目2n=44,雄鱼的染色体核型公式为2n=2sm+20st+22t,雌鱼的染色体核型公式为2n=1m+2sm+20st+21t,雌鱼存在性染色体异型的现象,因此灰海马的性染色体为ZW/ZZ型。  相似文献

  • 1. The fish family Syngnathidae (seahorses, pipefish, pipehorses and seadragons) is fully protected in New South Wales, Australia, but in some countries certain species are threatened by unsustainable collecting, capture as incidental bycatch, and habitat degradation.
  • 2. Within Sydney Harbour, two species of seahorses (Hippocampus abdominalis and Hippocampus whitei) have been found to colonize artificial structures such as jetty pylons and protective netted swimming enclosures. These protective nets are subject to fouling from epibiotic growth (algae, ascidians, bryozoans, etc.) and rubbish, which causes the nets to collapse from the additional weight. Local authorities employ diving contractors on an ad hoc basis to remove the epibiota from nets.
  • 3. Surveys showed a significant decline in the numbers of both seahorse species at one site following the replacement of a net, and recovery of the H. whitei population took more than 15 months.
  • 4. A manipulative experiment tested the importance of epibiotic growth for seahorses. H. whitei, tagged with individual marks, were allocated to sections of a net that had undergone different cleaning procedures. Seahorse size, position on the net and total population abundance were recorded every 2 weeks over a 3 month period. It was demonstrated that seahorses have a significant positive association with epibiotic growth and proximity to the sea floor. Seahorse populations also showed seasonal variation in abundance with increased numbers on the net during the breeding season (spring–summer).
  • 5. This project has led to the development of best practice net cleaning procedures for local authorities in Sydney Harbour to manage growth on the nets while minimizing impacts on seahorse populations. Copyright © 2009 John Wiley & Sons, Ltd.
