AIM: To elucidate the effect of calcitonin gene-related peptide (CCRP) in the therapy of atherosclerosis.METHODS:Effect of CGRP on cell cycle kinetics of cultured vascular smooth muscle cells(HA-VSMC) was investigated by flow cytometry. The expression of cyclins D1 and E required for initiation of S phase were also studied by immunochemistry method. RESULT: CGRP was shown to arrest VSMC in the G0/G1 phase of cell cycle and reduced expression of cyclins D1 and E. CONCLUSION:CGRP inhibits proliferation of HA-VSMC by arresting cells in G1 phase via limiting accumulation of cyclin D1 and E. It might play a role in the therapy of atherosclerosis.  相似文献

AIM: To elucidate the in vivo mechanisms of the proliferation of vascular smooth muscle cells (VSMCS) in injuried arteries. METHODS: A VSMCS proliferative model was constructed by injury of rabbit iliac arteries with balloon catheters and a probe designed for rabbit platelet-derived growth factor B chain (PDGF-B ) mRNA was used to detect the expression of it by intimal VSMCS on the vascular cross sections using an in situ hybridization technique at the indicated times. The relation of this expression to the proliferation of VSMCS by their expression of proliferating cell nuclear antigen (PCNA) and vascular intimal areas were estimated. RESULTS: The expression of PDGF-B mRNA of intimal VSMCS was increased when calculating the intimal PDGF-B mRNA positive cells per millimetre area at ×400 magnification with average numbers of 31.93±14.64 in 1 week group, 26.50±9.25 in 2 weeks group and 24.85±13.65 in 4 weeks group. This was in accordance with the expression of PCNA by VSMCS and the increase of intimal areas. CONCLUSION: The local production of PDGF-B by VSMCS via an autocrine mechanism is responsible for the continuous proliferation of these cells and formation of neointima after the injury. The probe designed is very useful for detecting rabbit PDGF-B mRNA.  相似文献

AIM: To investigate the mechanism of vascular smooth muscle cells (VSMC) apoptosis after balloon injury. METHODS: VSMCs apoptosis, Bax protein and Bcl-2 protein was measured by terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) and immunohistochemical technique after balloon injury. RESULTS:VSMCs apoptosis occurred in vascular media at 3 days after balloon injury and reached a peak in media and intima where apoptosis was mainly present at 7 days, then decreased. At 28 days after balloon injury, only a few apoptosis cells were in intima. Irbesartan significantly increased VSMCs apoptosis (P<0.01). Expressions of Bax and Bcl-2 were significantly higher in vascular media at 3 days after balloon injury than sham(P<0.01). At 7 days, Bax expression reached a peak which was three times as that of sham, and then decreased. After balloon injury, Bcl-2 expression was generally increased and reached its peak at 28 days. The ratio of Bax to Bcl-2 (Bax/Bcl-2) also reached its peak at 7 days and was decreased under basal level at 28 days. Irbesartan increased Bax and the ratio of Bax to Bcl-2, while decreased Bcl-2. CONCLUSION: Bcl-2 and Bax contribute to the regulation of VSMC apoptosis after balloon injury.  相似文献

AIM: To observe the effect of thichosanthes injection on the expression of proliferating cell nuclear antigen (PCNA) of vascular smooth muscle cell (SMC). METHODS: The expression of PCNA of cultured rabbit aortic SMC was examined with LSAB immunohistochemical technique, and [3H]-thymidine( [3H]-TdR) incorporation data of SMC and the contents of superoxide dismutase (SOD), lipid peroxide (LPO), prostacyclin (PGI2) and cyclic AMP (cAMP) in medium were simultaneously determined. RESULTS: Thichosanthes injection has an effects of increasing SOD activity, decreasing LPO, elevating PGI2 and cAMP, reducing [3H]-TdR incorporation and expression of PCNA (all P<0.05,P<0.01). CONCLUSION: Thichosanthes could inhibit SMC proliferation.  相似文献

对我国肖山鸡、白耳鸡、狼山鸡、乌骨鸡、油鸡、白耳鸡(♂)×肖山鸡(♀)、肖山鸡(♂)×白耳鸡(♀)7个地方鸡种进行了血浆、肌肉磷脂含量的比较研究.结果表明:①血浆中磷脂含量由高到低依次为乌骨鸡、狼山鸡、白耳鸡、白耳鸡(♂)×肖山鸡(♀)、肖山鸡、肖山鸡(♂)×白耳鸡(♀)、油鸡,其中乌骨鸡显着高于其它鸡种(P<0.05),狼山鸡、白耳鸡显着高于肖山鸡、肖山鸡(♂)×白耳鸡(♀)、油鸡,白耳鸡(♂)×肖山鸡(♀)显着高于油鸡(P<0.05);②肌肉中磷脂含量由高到低依次为肖山鸡、乌骨鸡、狼山鸡、肖山鸡(♂)×白耳鸡(♀)、白耳鸡、白耳鸡(♂)×肖山鸡(♀)、油鸡,但鸡种间不存在显着性差异(P>0.05);③血浆、肌肉磷脂含量有公鸡高于母鸡的趋势,但差异不显着(P>0.05).血浆、肌肉磷脂含量及12周龄体重相关分析显示,血浆磷脂含量与肌肉磷脂含量呈极显着正相关(P<0.01),血浆磷脂含量与12周龄体重呈显着负相关(P<0.01),而肌肉磷脂含量与12周龄体重相关不显着(P>0.05).  相似文献
