携带有家蚕促前胸腺激素PTTH基因的重组转移载体pHcMU1PTTH(含有麻蝇毒素IA信号序列 )与美国白蛾核型多角体病毒 (HcNPV)基因组DNA共转染SPIM细胞 ,经 2轮空斑纯化 ,筛选出重组病毒HcNPVPTTH+ ,SDSPAGE和Westernblot分析表明 :在细胞培养上清中可检测到PTTH的存在 ,说明麻蝇毒素IA信号序列可被SPIM细胞识别。用抗PTTH的兔血清进行Westernblot分析可显示分子量为2 0ku左右的 2条带 ,但当培养液内加入衣霉素时 ,仅出现 1条分子量小于 1 8ku的信号带 ,推测 2条信号带的产生与糖基化修饰不一致有关 相似文献

The lesions of patch/plaque cutaneous T-cell lymphosarcoma (CTCL) are similar to many benign inflammatory dermatoses, both histopathologically and clinically. In humans, attempts to develop a simple, reliable immunophenotypic technique to differentiate between the malignant and benign dermatoses have been unsuccessful. The purpose of our study was to determine, using a proliferating cell nuclear antigen (PCNA)/CD3 double immunolabelling technique of paraffin-embedded sections, if the rate of T-cell proliferation in canine CTCL was greater than that of other diseases with non-neoplastic lymphocyte epitheliotropism. We selected cases of patch/plaque (PP) and tumour stage CTCL, erythema multiforme (EM) and cutaneous lupus erythematosus (CLE). We did not find a significant difference in the rate of T-cell proliferation in the epithelia nor the superficial dermis between PP-CTCL and EM. Whereas epithelial T-cell proliferation is significantly higher in PP-CTCL than CLE, it is not diagnostically useful because of overlap in the labelling indices. 相似文献

The aim of this study was to examine the effects of a digital filter on the detectability of a phantom lesion in a scintigram of the equine tarsus. Lateral images containing 50, 100, 150, 500, or 1000 kcounts were acquired. A created phantom lesion of 0 (normal), 10, 15, 20, or 30% increased intensity relative to local background was placed in the centrodistal tarsal joint area in the images of different count levels. Duplicate images were filtered with a Metz filter. The complete set of filtered and unfiltered images totaling 180 images was projected as slides to a group of 9 observers. The sensitivity in 50 and 100 kcount images was improved by applying the filter. Digital filtering increased the false positive fraction at all count levels but this effect was most pronounced in 500 and 1000 kcount images. Based on receiver operating characteristic analysis, filtering of a 50 kcount image improved image quality to that of a 150 kcount image. 相似文献

To study the effect of Hieracium pilosella L. invasion on the transformations of soil organic matter of New Zealand tussock grassland soils (Ustochrepts), plant material
and soils underneath Hieracium, the surrounding halo, and the adjacent herbfield (depleted tussock grassland) were examined for their chemical composition.
An attempt was made to reveal possible changes in chemical composition of the soil organic matter induced by H. pilosella invasion. Small differences were detected by solid-state 13C nuclear magnetic resonance (NMR) spectroscopy in the composition of the plant and soil materials from these zones. Most
of the differences in soil organic matter occurred due to differences in the amount and quality of plant-residue inputs. Comparable
amounts of phenolic C were detected in the solid-state 13C NMR spectra of H. pilosella and herbfield vegetation, while alkaline CuO oxidation yielded considerable lower lignin oxidation products for H. pilosella. A slightly higher proportion of these compounds in H. pilosella soil revealed an accumulation and a low degradation rate of lignin compounds under H. pilosella. The HCl hydrolysis and solid-state 15N NMR spectroscopy showed similar chemical compositions of the N fractions of the three different soils. The absence of 15N NMR signal intensity assignable to aniline derivatives or aromatic heterocyclic N indicates that the condensation of phenolic
compounds with N groups plays a minor role in N sequestration in these soils.
