北川白山羊是在特定生态条件下未经系统繁育而形成的优良地方山羊品种,在生长发育、繁殖和抗逆等性状方面具有突出优势。通过对122只北川白山羊个体基因组的扩增,结果从80条RAPD引物中选出12条多态引物,北川白山羊群体平均遗传相似率和遗传多样性指数分别为0.859 8±0.077 6和0.919 8,表明北川白山羊品种具有一定的遗传分化和较丰富的遗传多样性。在12条多态性引物中,SBS06同时对体质量(P=0.028)、体高(P=0.017)和体长(P=0.037)具有显著影响,SBS02对体质量(P=0.033)和体高(P=0.034)具有显著效应,所以推断影响体质量和体高性状的QTL基因座可能与RAPD标记SBS06和SBS02相连锁,可以应用于北川白山羊体质量和体高性状的标记辅助选择以及相应主效基因的进一步研究。 相似文献

Polymorphic mountain whitefish (Prosopium williamsoni) in a coastal riverscape: size class assemblages,distribution, and habitat associations 下载免费PDF全文
We compared the assemblage structure, spatial distributions, and habitat associations of mountain whitefish (Prosopium williamsoni) morphotypes and size classes. We hypothesised that morphotypes would have different spatial distributions and would be associated with different habitat features based on feeding behaviour and diet. Spatially continuous sampling was conducted over a broad extent (29 km) in the Calawah River, WA (USA). Whitefish were enumerated via snorkelling in three size classes: small (10–29 cm), medium (30–49 cm), and large (≥50 cm). We identified morphotypes based on head and snout morphology: a pinocchio form that had an elongated snout and a normal form with a blunted snout. Large size classes of both morphotypes were distributed downstream of small and medium size classes, and normal whitefish were distributed downstream of pinocchio whitefish. Ordination of whitefish assemblages with nonmetric multidimensional scaling revealed that normal whitefish size classes were associated with higher gradient and depth, whereas pinocchio whitefish size classes were positively associated with pool area, distance upstream, and depth. Reach‐scale generalised additive models indicated that normal whitefish relative density was associated with larger substrate size in downstream reaches (R2 = 0.64), and pinocchio whitefish were associated with greater stream depth in the reaches farther upstream (R2 = 0.87). These results suggest broad‐scale spatial segregation (1–10 km), particularly between larger and more phenotypically extreme individuals. Waxy (Wx) protein is a key enzyme for synthesis of amylose in endosperm. Amylose content in wheat grain influences the quality of end‐use products. Seven alleles have been described at the Wx‐D1 locus, but only two of them (Wx‐D1b, Wx‐D1e) were genotyped with codominant markers. The waxy wheat line K107Wx1 developed by treating ‘Kanto 107’ seeds with ethyl methanesulphonate carries the Wx‐D1d allele. However, no molecular basis supports this nomenclature. In the present study, DNA sequence analysis confirmed that a single nucleotide polymorphism in the sixth exon of Wx‐D1 changed tryptophan at position 301 into a termination codon. Based on this sequence variation, a PCR‐based KASP marker was developed to detect this point mutation using 68 BC8F1 plants and 297 BC8F2 lines derived from the cross ‘Ningmai 14’*9/K107Wx1. Combined with codominant markers for the Wx‐A1 and Wx‐B1 alleles, waxy and non‐waxy near‐isogenic lines were distinguished. 利用13对ISSR引物对28个栽培种花生品种进行PCR扩增,研究这些品种的DNA多态性及其亲缘关系。结果表明,有7对引物检测到明显的多态性,从28个花生品种中扩增出90条带,其中多态性带达到79条,多态性比率为87.8%,平均每个引物可扩增出12.86条带,11.29条为多态性条带。品种间的遗传相似系数值在0.411~0.800之间,平均为0.620。在UPGMA聚类分析简单匹配系数值为0.57处,可把28个花生品种分成3个品种群。由此表明,这些不同来源的花生品种具有较高的DNA多态性,亲缘关系不同。 相似文献

查尔酮的合成是类黄酮生物合成途径中的一个关键节点。从10个柑橘种质中克隆了查尔酮合成酶(chalcone synthase,CHS)基因,并对其不同时期的果实和叶片中的类黄酮含量进行了测定,分析了CHS基因的序列多态性与类黄酮含量之间的关系;同时通过qRT-PCR检测了CHS基因在不同种质之间以及同一种质的不同组织间的表达差异。结果表明,柑橘CHS基因的核苷酸序列高度保守,相似度达98%以上。聚类分析表明,CHS氨基酸序列的多态性有物种特异性,而且与类黄酮含量有一定的相关性。CHS基因的表达水平在不同种质、部位及生长发育时期有显著差异,这些差异与类黄酮的含量有显著的相关性,说明CHS基因对类黄酮的生物合成有明显影响。 相似文献

【目的】随着不同棉种序列数据库的逐步完善以及高通量测序技术的发展,棉花单核苷酸多态性(Single nucleotide polymorphism,SNP)标记开发可利用的公共数据资源逐步增加。【方法】本研究基于陆地棉祖先基因组的现代种亚洲棉表达序列标签(Expressed sequence tag,EST)数据库,利用CAP3对亚洲棉EST数据库进行拼接。拼接获得7 187个重叠群(Contig),再利用Quality SNP软件进行SNP位点分析。【结果】在807条含有4条以上EST序列的Contig中查找到2 690个SNP位点。通过筛选次要等位基因频率大于30%的位点,获得953个可靠度较高的候选SNP,通过电子筛选,最终获得可用于陆地棉分析的SNP 149个,利用位点特异性聚合酶链式反应以及酶切扩增多态序列验证了EST-SNP的准确性。【结论】本研究证实基于亚洲棉EST数据库挖掘用于陆地棉研究的EST-SNP切实可行,并有望将EST-SNP用于陆地棉遗传图谱构建、重要性状的基因定位以及分子标记辅助育种。 相似文献

钠猪毛菜在我国仅分布在新疆,具有很强的抗干旱和抗盐碱能力。研究钠猪毛菜不同类型种子形态及其萌发特性,结果表明,钠猪毛菜产生3种类型的种子,其形态特征、萌发特性均有显著差异。A型种子绿色,圆形,着生方式为横生,直径为(1.294±0.089) mm,百粒重(90.71±0.42) mg,花被背部有翅状附属物;B型种子红色,圆形,着生方式为直立,直径为(1.166±0.069) mm,百粒重(85.52±0.49) mg,花被背部有短/无翅状附属物;C型种子黄色,圆形,着生方式为直立,直径为(1.044±0.062) mm,百粒重(83.84±0.31) mg,花被背部无翅状附属物。A型种子在5 ℃/15 ℃、10 ℃/20 ℃、 15 ℃/25 ℃、 20 ℃/30 ℃、 25 ℃/35 ℃(光/暗=12 h/12 h)变温条件下萌发率均>92%,B型种子在5种变温条件下萌发率<60%,为非深度生理休眠,低温层积2周后可打破休眠;C型种子在5种变温条件下萌发率<5.3%,为深度生理休眠,划破种皮可打破休眠。浓度低于0.2 mol·L-1的NaCl溶液对A型种子萌发的影响不大,但从0.8 mol·L-1起,萌发率随着浓度增高而降低,直至为零,将在0.05~4.00 mol·L-1NaCl溶液中处理的A型种子转移至蒸馏水后,仍有一定的恢复萌发率。苞片延缓A型种子吸胀,影响种子的最终萌发率,去除苞片可显著提高种子的萌发率。通过研究钠猪毛菜种子异型性及其萌发行为,为揭示其生态适应机制提供科学依据,并为新疆荒漠地区生态建设提供基础资料。 相似文献

Resource polymorphism may play an important role in the process of speciation. The Arctic char (Salvelinus alpinus) exhibits great phenotypic and genetic diversity across its range, making it an ideal species for studies of resource polymorphism and divergence. Here, we investigated genetic variation at 11 microsatellite loci among 287 Arctic char from five isolated yet proximate postglacial lakes in south‐western Alaska that were previously examined for resource polymorphism. Significant differences in pairwise FST were detected among all lakes (range from 0.05 to 0.28, all P < 0.02). In one lake (Lower Tazimina Lake), we found evidence for two genetic groups of char and for significant differences in the distribution of microsatellite variability among at least two of the three previously described body size morphotypes (‘large’‐, ‘medium’‐, and ‘small’‐bodied char; maximum FST = 0.09; differences in admixture proportions). We also found a significant association between genetic admixture proportions and gill raker counts among body size morphs (r = ?0.73, P < 0.001). Our data represent the first record of genetically distinct sympatric morphs of Arctic char in Alaska and provide further evidence that differences in morphology associated with feeding (gill rakers) and growth trajectories reflect niche diversification and promote genetic divergence in Holarctic populations of Arctic char. 相似文献
