Chronically cannulated rainbow trout were exposed in acid water (pH 4.0) for 72h. The gill potential was strongly dependent on water pH, being blood side negative in neutral water, but positive in acid water. Catecholamine levels increased irregularly during acid exposure, and the Bohr and Root effects were not completely erased by the effect of catecholamines during acid exposure. Long term exposure to low water pH, although causing an acidosis in the fish, did not suppress resting oxygen consumption. Prolonged exposure to acid conditions, however, resulted in an increase in ammonia excretion. Changes in plasma sodium and chloride were similar to that reported previously for trout exposed to low calcium, acid water. We conclude that exposure of trout to pH 4 soft water, although impairing oxygen transport, does not limit resting oxygen consumption but does reduce the scope for activity. More extreme acid conditions do impair resting oxygen uptake. 相似文献

The present study investigated the effects of sequential collection of milt, time of post-mortem storage and anesthesia on rainbow trout (Oncorhynchus mykiss) sperm motility parameters (using computer-assisted sperm analysis – CASA) as well as seminal plasma osmolality and sperm
concentration. The post-mortem storage and time of anesthesia altered motility characteristics of rainbow trout sperm to different extents. The moderate
impact of time of anesthesia was manifested in a shortened duration of sperm motility after 10 min exposure of fish to anesthetic.
The prolonged post-mortem storage (≥40–60 min), in addition to lowering sperm motility duration, also significantly influenced sperm motility parameters,
such as sperm velocities, percentage of motile sperm and sperm trajectory parameters. These results clearly demonstrate that
when milt from sacrificed fish is used for sperm motility studies, the time of post-mortem storage significantly alters sperm motility characteristics. Since sperm motility rate and swimming velocity could predict
fertilizing ability, detrimental effects of prolonged post-mortem storage may lead to reduced fertilization success. Sperm concentration and seminal plasma osmolality were lower in the first
fractions and increased with successive collections of milt. It suggests the presence of urine contamination of the first
milt fractions which were collected by stripping. Therefore, testing of sperm concentration and/or seminal plasma osmolality

Two trials were designed to quantify the effect of feeding ratio and fish size on the cohabitation transmission of Loma salmonae, the causative agent of microsporidial gill disease (MGD) in salmonids, Oncorhynchus spp. To evaluate the effect of feeding ratio on disease onset, groups of 45 rainbow trout, O. mykiss (Walbaum) (RBT), were fed daily at 1% (low), 2% (medium) or 4% (high) of the fish biomass in the tank. There were three tanks at each feeding level: two tanks were exposed to the pathogen and one was a control. For the second objective, 300 RBT were separated into seven tanks so that the weight classifications were small (17-23 g), medium (32-38 g) and large (57-63 g). Each size class was done in duplicate with one control tank containing medium-sized fish. Separately for each trial, on day 0 post-exposure (PE) five highly infectious RBT were added to each tank (not including the control tanks) to begin the cohabitation exposure period. Beginning on day 21 PE and continuing biweekly until days 70 and 77 PE for the feeding and fish size trials, respectively, each fish was evaluated for visible branchial xenomas to determine disease onset time. Using survival analysis, the survival curves for the low, medium and high feedings were not significantly different from each other. However, there were significant differences amongst the small, medium and large weight classes in the size trial. The median numbers of days to the development of branchial xenomas was 31, 38 and 42 for small, medium and large size fish, respectively. On any given day, a medium or large sized fish had a hazard ratio for developing branchial xenomas of 0.66 and 0.63, respectively, compared with a small fish. Four diets differing in crude protein/crude lipid concentrations (CP/CL), 570/200, 510/220, 460/240, 430/260 (g kg?1 dry diet) were fed to near‐satiety to rainbow trout (initial body weight, IBW = 268 g, domestic strain) and Atlantic salmon (IBW = 456 g, wild strain) for 308 days to determine the effect of diets, species and fish size on efficiency of feed, nitrogen (N) and energy utilization. Within species, weight gain, feed efficiency (FE), and energy retention efficiency (ERE, E gain/E intake) were not affected by diet (P < 0.05). N retention efficiency (NRE, N gain/N intake) increased linearly (P < 0.05) with decreasing CP/CL for both species. NRE and FE were higher for salmon compared with trout. Whole body CP of salmon was significantly (P < 0.05) higher, and lipid significantly (P < 0.05) lower, than those of trout. There was a significant (P < 0.05) linear decrease in FE as fish grew, regardless of diet and species. NRE linearly decreased (P < 0.0001) and lipid to protein deposition ratio (LD/PD) increased (P < 0.05) as trout grew. NRE linearly increased (P < 0.05) while ERE and LD/PD linearly decreased (P < 0.05) as salmon grew. Data on reproductive traits in a cultured population of brown trout (Salmo trutta L.) in southern Chile were collected over three consecutive reproductive seasons: 1999, 2000 and 2001, corresponding to the first (at 3 years), second and third spawnings in females. Data were collected from individual females (n=238, 273 and 169 respectively). The reproductive season lasted mainly from June to August. The peak (July) tended to increase with each season (55.46%, 62.27% and 80.81% of total spawning fish respectively). Female body weight (470.8±102.5, 735.0±150 and 1263.9±263.4 g), total fecundity (1182±344, 1904±595 and 2744±605) and egg diameter (4.64±0.11, 4.67±0.27 and 5.24±0.12 mm) increased significantly (P<0.01) over successive reproductive seasons, particularly between the second and third spawnings. Relative fecundity, on the other hand, decreased significantly with each season (3577±471, 2591±900 and 2181±360). Following analysis of the variables over the three seasons, the correlation pattern of female body weight with total fecundity (r=0.91, n=458; P<0.001) and relative fecundity (r=?034, n=451; P<0.001) proved similar to that described in other salmonid females. Fertilization rate and survival to the eyed egg stage were also positively correlated (r=0.73, n=453; P<0.001). The systematically high values obtained for these latter variables over the seasons evaluated (consistently above 90%) are clearly greater than those we registered in other species of salmonids bred in Chile under similar conditions and suggest highly efficient biological variables that determine the gamete fertilization of this species. The brown trout is, therefore, an interesting potential aquaculture resource in Chile. 相似文献

Freshly isolated rainbow trout hepatocytes were exposed to tert-butyl hydroperoxide (BuOOH), a substrate for glutathione peroxidase.
BuOOH at a concentration approximately equimolar (1 mM) with intracellular reduced glutathione (GSH) caused a reversible increase
in intracellular glutathione disulphide (GSSG) but did not compromise cell viability or damage membrane lipids. BuOOH at 10
mM caused a large irreversible increase in intracellular GSSG followed by efflux into the medium. Considerable leakage of
lactate dehydrogenase and loss of highly unsaturated fatty acids, particularly docosahexaenoic acid also occurred. Dependence

Serum and mucosal antibody responses of juvenile rainbow trout, Oncorhynchus mykiss, were characterized by enzyme‐linked immunosorbent assay (ELISA) following immunization with various preparations of formalin‐killed Flavobacterium psychrophilum cells. The protective nature of these preparations was then determined by immunizing rainbow trout fry and challenging with the bacterium. Juvenile rainbow trout immunized intraperitoneally (i.p.) with formalin‐killed F. psychrophilum emulsified with Freund's complete adjuvant (FCA), and i.p. with formalin‐killed F. psychrophilum either with or without culture supernatant generated significant serum antibody responses by 6 and 9 weeks, respectively. Significant mucosal antibody responses were detected by 9 weeks only in fish immunized i.p. with killed F. psychrophilum/FCA. Following immunization and bacterial challenge of rainbow trout fry, protective immunity was conferred in F. psychrophilum/FCA and saline/FCA groups with relative per cent survival values of up to 83 and 51, respectively. Significant protection was not observed in treatment groups immunized by immersion or i.p. without adjuvant at the challenge doses tested. Results suggest that stimulation of non‐specific immune factors enhances the ability of fish to mount a protective immune response, but specific antibody appears necessary to provide near complete protection. In this study, an ELISA was developed to monitor anti‐F. psychrophilum antibody production in trout. The relationship of such responses to protective immunity suggests that future vaccination strategies against coldwater disease may require stimulation of both the innate and adaptive arms of the immune response. 相似文献
