Oryzalin as an Efficient Agent for Chromosome Doubling of Haploid Apple Shoots in vitro 总被引:4,自引:0,他引:4
In an outbreeding species such as apple, haploid plants may be especially useful in breeding programmes for the production of homozygous material. However, methods must be available to induce chromosome doubling in the haploid plants. Two antimitotic agents, colchicine and oryzalin, were compared as regards their efficiency in inducing chromosome doubling of in vitro haploid apple shoots. Three colchicine levels (0.025, 0.25 and 1.25 mM) and three oryzalin levels (5, 15 and 30 μM) were evaluated. Three techniques were also used and compared. Survival rate and chromosome counts were determined. Differences were observed between the two antimitotic agents and between the three techniques. This study demonstrates that oryzalin could be a better choice than colchicine for chromosome doubling on haploid apple shoots in vitro. 相似文献

Summary The group 1 and 6 inter-varietal chromosome substitution lines of Cappelle-Desprez (Bezostaya 1) were intercrossed along with the donor and recipient varieties, Cappelle-Desprez and Bezostaya 1, to give 36 genetically different families. The analysis of the means of these families showed that variation in SDS-sedimentation volume fitted a predominantly additive model. There were no significant within or between chromosome interactions among the group 1 and 6 chromosomes. Nor was there any evidence for interactions between these chromosomes and those of the background. Significant dominance/within chromosome interactions amongst the background chromosomes were however detected. Some of the positive effects on SDS-sedimentation were associated with increased grain hardness. Chromosome effects on % grain protein were not correlated with SDS-sedimentation. 相似文献

The meiotic behaviour of 13 spontaneous interspecific F1 hybrids of Amaranthus was studied. The hybrids between species with n= 16 chromosomes had 16 bivalents but varied considerably in pollen stainability (0–55%). These results suggest the existence of cryptic structural hybridity. The hybrids involving A. cruentus (n = 17) and species with n = 16 (A. caudatus and A. quitensis) always formed 15II+1 III with very low pollen stainability (5–7%). Further observations indicated that Amaranthus species are allotetraploids with basic numbers of x= 8 and x= 9 but exhibit x= 16 and x= 17 as secondary basic numbers, as demonstrated by (a) the frequent presence of 811 + 171 in the meiosis of the hybrid A. spinosus (n = 17) × A. hybridus (n= 16); and the occurrence of secondary associations between bivalents in MI. Numerous cytogenetic studies have been performed on alfalfa (Medicago sativa L. 2n= 4x= 32), but only a few have examined the somatic chromosomes. Because the major factor limiting the study of somatic chromosomes of M. sativa is their rather uniform morphology, characterization was attempted using the C-banding technique instead of the traditional Feulgen method. Chromosome morphology and position of the heterochromatic bands resulted in satisfactory characterization of the chromosomes and provided further evidence of the autotetraploid origin of the species. It was possible to identify the homologous chromosomes and arrange them into eight groups of four. The ideogram of the basic genome of eight chromosomes was constructed. The impact of these results on cytogenetic studies and breeding research in alfalfa is discussed. 相似文献

Two disomic barley chromosome addition lines and five translocated chromosome addition lines of common wheat cultivar Shinchunaga
were isolated. They were derived from a hybrid plant between Shinchunaga and cultivated barley Nyugoruden (New Golden) by
backcrossing with wheat and self pollination. Barley chromosomes added to chromosome arms involved in the translocated chromosomes
were identified by C-banding method and by crossing these lines with Chinese Spring/Betzes addition lines. Two disomic addition
lines were identified to have chromosome 6 and 7 of barley, respectively. Two of the five translocated chromosome addition
lines were clarified to have same chromosome constitution, 42 wheat chromosomes and a pair of translocated chromosomes constituted
with a long arm of chromosome 5B of wheat and a short arm of chromosome 7 of barley. The other three lines could not be identified
due to chromosome rearrangement. Performances of these seven lines on agronomic characters were examined. Addition of barley
chromosome 7 induced early heading, and chromosome 6 showed lated heading. Almost all of the lines except that of chromosome
6 showed short culm length and all showed reduced number of tillers, spikelets and grains per ear, and low seed fertility.
These lines would be useful for genetic analyses in wheat and barley and for induction of useful genes of barley into wheat.

Summary We have succeeded in producing useful interspecific hybrid using ovule-embryo culture between Gypsophila paniculata L. Red Sea and G. manginii, an incompatible combination by ordinary cross breeding methods. The hybrid plant had double flowers with a color of pale purplish pink. Hybrid characteristics of the plant were firmed by observation of plant form, flower type, chromosome number and peroxidase isozyme patterns. 相似文献

Using monosomic lines of wheat cultivars ‘Palur’ and ‘Compal’ as recipient parents as well as disomic substitution lines of chromosomes 5A and 5D of the wheat cv. ‘Atlas 66’, F3-populations and BC1′- to BC3′-populations with limited and free recombination of the 20 and 21 parental chromosomes, respectively, were realized and tested in field trials in comparison to the corresponding recipient cvs. ‘Palur’ and ‘Compal’. F3- and BC'-populations with the homozygous chromosomes 5A and 5D of the wheat cv. ‘Atlas 66’ expressed higher and more stable grain protein values than the comparable populations with free recombination of the same chromosomes. The grain protein content of populations with limited recombination was significantly increased compared with the recipient cultivars. Summary This study was conducted using the isozymes ACP-1, ADH-1, GOT-2, GOT-3, MDH, 6-PGD-1 and PGI-2 to: a) compare isozyme banding patterns of tall fescue somaclones with parents and b) correlate tissue culture-induced chromosome abnormalities with variant banding patterns. The 174 somaclones were grouped into seven categories based on their meiotic analyses and time of regeneration from culture. Differences in isozyme frequency between categories compared by chi-square tests were greatest for MDH, 6-PGD-1 and PGI-2, and least for ACP-1. The most significant differences in frequency were found between somaclones and parents. In comparisons of somaclone categories, the most different isozyme distributions were between the early vs. late regenerated somaclones. No significant differences in isozyme frequencies were found between all 42-chromosome somaclones vs. aneuploid somaclones and the three somaclone groups (42-normal, 42-abnormal, aneuploid) compared to each other. This study suggests that culture-induced isozyme variation alters the distribution of the isozyme phenotypes, but is not directly correlated with chromosome abnormalities. 相似文献

Summary Meiotic observations in PMCs were made in eight kinds of interspecific hybrids in Lilium. Three hybrids, 6134-S × L. cernuum. L. regale × L. leichtlinii maximowiczii and L. auratum platyphyllum × L. henryi showed respectively the mean chromosome association frequencies of 1.4II, 0.8II and 1.5II per cell at MI which were the lowest values hitherto reported in Lilium hybrids. In L. longiflorum × L. henryi, 33.3° of the cells had 1 or 2 multivalents of three to five chromosomes in addition to uni- and bivalents. 12° bivalents and 12 univalents were invariably observed in the triploid hybrid L. longiflorum × L. cernuum. The hybrids between the species belonging to the different sections of the genus generally showed high pollen sterility, with some exceptions. A more or less remote genomic homology was found between the different sections in Lilium. 相似文献
