Identification of sperm morphometric subpopulations in cooled‐stored canine sperm and its relation with sperm DNA integrity 下载免费PDF全文
The aims of this study were to (i) identify different morphometric subpopulations in cooled‐stored canine sperm and their patterns of distribution during cool‐storage for up to 240 hr and (ii) determine whether or not morphometric sperm subpopulations (sP) are related to sperm DNA integrity. For that purpose, morphometric parameters were analysed by computer‐assisted sperm analysis (CASA) and sperm DNA fragmentation (sDFi) using the sperm Halomax test. Four morphometric sperm heads subpopulations were identified: sP1 (large and rounded), sP2 (large and elongated), sP3 (small and rounded) and sP4 (small and elongated). sP1 was the most predominant subpopulation for up to 72 hr and thereafter sP3 increased progressively. sDFi increased after 48 hr of cool‐storage. Although sP3 showed a positive correlation with sDFi, and both increased over time, it could not be ensured that only the sperm with fragmented DNA are accumulated in sP3. In conclusion, sP3 and DNA fragmentation increased progressively during cool‐storage, becoming possible indicators of sperm damage. Mammalian gametes and embryos are particularly vulnerable to oxidative stress-induced damage, which is mainly caused by reactive oxygen species (ROS) originating from normal metabolism and/or the external environment. Several researchers have implicated the role of oxidative stress in the activation of apoptosis, causing peroxidative damage to sperms/oocytes and inducing embryo fragmentation, arrest, or demise. Melatonin is a tryptophan derivative that is known for its powerful free radical-scavenging activity and broad-spectrum antioxidant property. Numerous studies have shown that melatonin and its metabolic derivatives can sequentially detoxify ROS in an antioxidant cascade, and modulate various antioxidant enzymes via its receptors to prevent radical-mediated damage. The identification of melatonin receptors in cumulus/granulosa cells, oocytes, and epididymal tissues implies that melatonin has protective actions on gametes and embryos. Enriching the semen extender or culture medium with melatonin significantly benefits sperm characteristics, improves oocyte maturation potential and quality, and enhances the developmental competence of preimplantation embryos. Certainly, further comparative studies are needed to show the unique antioxidant role and the advantage of melatonin in this field. 选用518只母鸡、20只公鸡(海赛克斯褐父父母代鸡),公鸡精液用高压静电场处理后人工受精。分析研究高压静电场对鸡精子受精率及其胚胎发育的影响。结果表明用一定剂同压静电场处理鸡精液能提高精子受精率、缩短出雏持续时间,但剂量过大时降低受精率、增加鸡早期胚胎死亡,从而又影响孵化率。本次实验表明600kv/m*15min剂量的高压静电场对鸡精子受精率和胚胎孵化率具有显著的提高作用。即受精率提高6.56个百分点,孵化率提高10.33个百分点。这对养殖业生产有着积极的意义。 相似文献

研究了中国对虾卵水的采集、保存方法、紫外吸收特性和有效期等方面的内容,并应用卵水研究了交尾期虾、雌虾及产卵期雌虾精荚或纳精囊中精子对卵水的应答开始时间、必要的反应时间等响应卵水的时间特性以及精子的存活期等。结果表明,卵水经液氮保存或先经液氮后转入普通冰柜保持冻结状态7个月后诱导精子激活效力无显著差异。精子在自然温度(10℃)普通海水中可在10h内保持对卵水的响应能力。交尾期精子最初响应卵水有5 ̄1 相似文献

将用于体外受精的种公牛精液,分别添加咖啡因、茶碱和己酮可可碱3种黄嘌呤活性化物质,探讨其对精子运动性及受精率的影响。结果表明:精子的运动性,以添加己酮可可碱处理的较高,随着时间的推移,添加茶碱处理的相对较高,但试验组与对照组差异不显著。受精率除添加己酮可可碱处理与对照组有显著差异外,其他组间均无显著差异。不同种公牛的精子运动性存在差别,但受精率无显著差异。 相似文献

Ultrasonographic appearance of the testis and epididymis, and seminal characteristics, with regard to localization of spermatic granuloma were studied. In rams with spermatic granuloma (n = 9), diagnosed by clinical or ultrasonographic examinations before histopathological confirmation, localization of each lesion was recorded. Epididymal granulomas, caput and cauda, were seen as anechoic or hyperechoic areas with a distinct margin with or without a hyperechoic capsule. Granulomas in the testis were microscopic and, therefore, could not be detected by ultrasonography. Enlargement in the mediastinum testis was detected in all rams when there were granulomas in the caput. Heterogeneous testis parenchyma invaded by numerous hyperechoic foci, representing testicular degeneration, was associated with granulomas both in the epididymis and testis. Ultrasonographic appearance of the lesions did not differ with regard to season. Seminal characteristics varied between rams. It was concluded that ultrasonographic evaluation may give valuable information in the diagnosis of sperm granuloma in the ram. 相似文献

探讨了8种稀释液在5℃、10℃、15℃和20℃下保存犬精液的效果。结果表明:15℃为犬精液液态保存的较适温度,3号液为优等液,其中3号液在15℃精子的存活时间为105±10h,与对照液在15℃的保存效果差异极显著(p<0.01)。2、4、5号液是良等液,在15℃精子的存活时间是64±8h,与对照液在15℃的保存效果差异显著(p<0.05)。中等液为6号液,在15℃、20℃保存时与对照液没有统计学上的差异(p<0.05),在其他2种温度保存时不及对照液。7、8号液为差等液,在15℃保存时与对照液差异不显著(p<0.05),在其他3种温度时不及对照液。 相似文献
