本试验采用乳品分析仪对槟榔江水牛、摩拉水牛和荷斯坦牛乳蛋白、乳脂和乳糖等含量进行测定。同时采用SDS-PAGE技术对乳蛋白各组分进行分离,并利用凝胶成像系统进行扫描定量。结果表明,槟榔江水牛和摩拉水牛乳蛋白、乳脂含量差异不显著(P>0.05),槟榔江水牛乳糖含量显著低于摩拉水牛(P<0.05);而水牛乳蛋白、乳脂和乳糖含量均极显著高于荷斯坦牛乳(P<0.01)。酪蛋白百分含量在三种牛乳蛋白中占优势,并且水牛乳CN和β-LG百分含量与荷斯坦牛乳差异不显著(P>0.05);而水牛乳中其他蛋白组分含量则显著高于荷斯坦牛乳(P<0.05)。  相似文献

[目的]建立一种高效的水牛子宫内膜腺上皮细胞和基质细胞分离培养方法,为胚胎着床和子宫内膜相关疾病的分子生物学机制研究奠定基础。[方法]采用酶消化、刮取、系列过滤和差速贴壁相结合的方法分离水牛子宫内膜腺上皮细胞和基质细胞,用免疫细胞化学法和台盼兰染色法检测分离细胞的纯度和活率。[结果]成功分离培养出子宫内膜腺上皮细胞和基质细胞;免疫荧光染色和细胞计数表明上皮细胞和基质细胞的纯度均达90%以上;台盼兰染色结果显示,上皮细胞的活率为91%,基质细胞的活率为78%。[结论]采用酶消化、刮取、过滤和差速贴壁相结合的方法可分离出高纯度的永牛子宫内膜腺上皮细胞和基质细胞。  相似文献

11头5~8月龄公水牛犊和2头7日龄黑白花奶牛犊,分别人工感染80~2000万个水牛源枯氏住肉孢子虫孢子囊,2头同龄公水牛犊不感染作对照。感染后12、22和32天在大、中、小血管和毛细血管内出现第一代裂殖体,32.86×16.43(19.08~46.64×12.72~21.2)μm,含100~450个裂殖子;32~46天在毛细血管和小血管内发现大量的第二代裂殖体,18.21×9.87(10.60~29.68×6.36~13.78)μm,含30~110个裂殖子;28~40天,第二代裂殖子出现于血液中或单核细胞内;46天初级幼包囊形成,78天部分包囊成熟,98天(黄牛)包囊成熟。  相似文献

Reproductive efficiency is the primary factor affecting productivity in female buffalo, but is greatly hampered by late attainment of puberty, seasonality of calving, long postpartum anoestrus and subsequent calving interval. Moreover, artificial insemination (AI) is seldom performed because of the weakness of oestrus symptoms and the variability of oestrus length, which makes oestrus detection very difficult.Researchers have tried to cope with each of the critical physiological aspects of buffalo reproductive performance mentioned above. In the present review, the main factors affecting the low reproductive efficiency in buffalo are examined and proposals made for its improvement, taking into account both our research work and literature results. Previous research on the changes in hormone profile during the oestrus cycle and in follicle development were used to develop models for improving reproductive efficiency. Hormonal treatments can be used: (1) to anticipate puberty in order to reduce the age at first calving; (2) to improve fertility in the low breeding season; and (3) to increase the effectiveness of AI programmes by overcoming the difficulty in oestrus detection.  相似文献

The effect of the digestion process in the gastro-intestinal tract (GIT) of animal models on the fate and integrity of plant DNA has been widely evaluated since DNA availability and integrity is a key factor for hypothetical horizontal gene transfer of recombinant DNA from GM crop-derived feeds to animal and human gut microflora. In this study, plant DNA sequences from high and low copy number genes were monitored in GIT and tissues of buffaloes and rabbits. Using a real-time PCR approach to track plant DNA in animal samples, we demonstrated the persistence of fragmented plant DNA blood and tissues of buffaloes and rabbits raised with conventional feeding.  相似文献

Identification of genes of importance regarding production traits in buffalo is impaired by a paucity of genomic resources. Choice to fill this gap is to exploit data available for cow. The cross‐species application of comparative genomics tools is potential gear to investigate the buffalo genome. However, this is dependent on nucleotide sequences similarity. In this study, gene diversity between buffalo and cattle was determined using 86 gene orthologues. There was approximately 3% difference in all genes in terms of nucleotide diversity and 0.267 ± 0.134 in amino acids, indicating the possibility for successfully using cross‐species strategies for genomic studies. There were significantly higher non‐synonymous substitutions both in cattle and buffalo; however, there was similar difference in terms of dN– dS (4.414 versus 4.745) in buffalo and cattle, respectively. Higher rate of non‐synonymous substitutions at similar level in buffalo and cattle indicated a similar positive selection pressure. Results for relative rate test were assessed with the chi‐squared test. There was no significance difference on unique mutations between cattle and buffalo lineages at synonymous sites. However, there was a significance difference on unique mutations for non‐synonymous sites, indicating ongoing mutagenic process that generates substitutional mutation at approximately the same rate at silent sites. Moreover, despite of common ancestry, our results indicate a different divergent time among genes of cattle and buffalo. This is the first demonstration that variable rates of molecular evolution may be present within the family Bovidae.  相似文献

A sterile white fungus was isolated from the healthy looking roots of buffalo grass (Stenotaphrum secundatum) grown on cleared bush land in Perth, Western Australia. The fungal strain was pathogenic on 12 plant species screened under the greenhouse conditions. The clamp connections and dolipore septa indicated that the isolate was a Basidiomycete. Mycelial features, growth rate at different temperatures, as well as pathogenicity patterns of this sterile white basidiomycete (SWB) were distinctly different from those of a strain with a similar morphology, ATCC 28344, previously described as a pathogen in Florida and Georgia (USA). All attempts to induce sporulation failed. The isolates were also compared using the nucleotide sequence analysis of the ribosomal DNA array. Approximately 1 kbp of the 5 end of the large subunit ribosomal RNA gene, complete sequences of the small subunit ribosomal RNA gene and the entire ITS region (including ITS1, ITS2 and 5.8S gene) were sequenced for the purpose. The obtained sequences were compared with the homologous regions of other genera of Agaricales available in GenBank. Relatively low sequence similarities between the American and Australian strains, as well as the phylogenetic analysis of the studied regions has suggested that these two fungi belong to different genera. Interesting results were achieved in the case of the large subunit ribosomal DNA since this region has been widely studied for taxonomy of Basidiomycetes. The Australian strain 3034 appeared to be closely related to the genus Campanella and the American SWB was identified as belonging to the genus Marasmius, possibly to M. graminum. Our data suggest that the Australian strain is a novel pathogen, and is different from the American SWB isolates described to date.  相似文献

The isolation of leptospires from buffaloes worldwide is still limited to a few strains. Thus, the aim of this study was to describe the first Leptospira isolate from buffalo urine, assigned to the Sejroe serogroup, which does not belong to the Wolffi subgroup, traditionally isolated in Brazil. A total of 244 urine samples of water buffaloes (Bubalus bubalis) raised in the Brazilian Amazon were subjected to bacteriological culturing and polymerase chain reaction (PCR) for the detection of leptospires. The obtained isolate was characterized by serogrouping using polyclonal antibodies, partial DNA sequencing, Hardjo-Bovis-specific PCR, multiple-locus variable-number tandem repeat analysis (MLVA/VNTR) and experimental infection in hamsters. PCR was performed on the urine samples; 11/244 were positive (4.5 %) for Leptospira, and only one isolate was recovered (0.4 %). Regarding characterization, the isolate was assigned to the Sejroe serogroup with high titers (12,800) for the Saxkoebing and Sejroe serovar antisera. The isolate was negative for Hardjo-Bovis-specific PCR, and the species Leptospira borgpetersenii was identified by DNA sequencing. The MLVA results showed that the VNTR profile of the isolate was 1−2-5, compatible with that of serovars Sejroe/Istrica. In the experimental infection in hamsters, the animals did not develop clinical signs, and no macroscopic lesions were observed on the organs at necropsy; however, the strain was detected in the kidneys, uterus, and testicles of the animals. The isolate described herein highlights infection by Sejroe strains that may be overlooked in buffaloes and that may be different from those normally isolated and used in serological studies.  相似文献

Progressive weight gain, faecal egg counts, packed cell volume, percent eosinophils in blood, serum antibody and serum levels of glutamate dehydrogenase and gamma-glutamyl transpeptidase were recorded in seven swamp buffalo (Bubalis bubalis), 7 Ongole (Bos indicus) and four Bali calves (Bos sundiacus) which were infected orally with 15 metacercariae of Fasciola gigantica twice weekly for 32 weeks. Similar observations were made on four buffalo, 4 Ongole calves and 3 Bali calves maintained fluke-free as controls. Flukes were counted at slaughter 36 weeks after initial infection. Mean daily weight gains of infected Bali (228 ± 100 (SD) g/day) and infected Ongole calves (328 ± 57 (SD) g/day) were lower (p = 0.026 and 0.067, respectively) than those of control calves (405 ± 107 (SD) g/day), but infected buffalo calves (379 ± 78 (SD) g/day) had similar weight gains to those of the controls (p = 0.57). Throughout the trial, faecal Fasciola egg counts in buffaloes were about one-fifth of counts of Ongole calves, and counts in Bali calves were intermediate. Ongole calves had three times the number of flukes at slaughter in their liver compared to buffalo and Bali calves, which had similar numbers. However, there was evidence that Bali calves had acquired a degree of resistance about 24 weeks after infection commenced and may have lost adult flukes as a consequence.  相似文献
