Around 20,000 samples are analysed each year for the presence of antibacterial residues in Danish finisher pigs, and between zero and five samples are detected positive above the maximum residue level (MRL). The intention was to develop a risk-based surveillance programme involving fewer samples while ensuring equal sensitivity. Therefore, risk indicators were searched for. Data were obtained from the Danish slaughterhouse database covering the period from July 2010 to December 2012. Residues were found or suspected in 17 incidents. In nine of these, the farmer had called in to prevent the pigs from being slaughtered. Hence, eight suspect cases were found through the surveillance programme, and two of these were above MRL. For these eight case herds, the number of pigs slaughtered and the number in which each of the following lesions were found were included in a statistical analysis: chronic pleuritis, tail bite, chronic pericarditis, chronic pneumonia, chronic peritonitis, osteomyelitis, abscess in hindquarters, abscess in leg/toe and abscess in forequarters. Only chronic pleuritis was associated with the presence of residues. Next, data from all herds delivering pigs for slaughter to the same abattoir were included covering a 3-month period prior to the residue finding. The prevalence of chronic pleuritis was on average 1.7 times higher in the eight case herds compared to all other herds. In two herds, the prevalence was significantly higher (P ≤ 0.05), and in one herd substantially higher, but only borderline significant (P = 0.1). In the remaining herds, the prevalence did not differ from the other herds delivering pigs to the abattoir. This indicates that chronic pleuritis might be considered as a risk indicator for use in surveillance. Other risk indicators/factors – reported in the cases where the farmers called in – were inadequate marking of treated animals and incorrect use of medication dispensers. These factors are not suited for use in surveillance and should be dealt with otherwise.  相似文献

The analgesic effect of wound infiltration with bupivacaine was evaluated in cats undergoing bilateral mastectomy. Twenty-one female cats with mammary gland tumors were anesthetized with propofol and oxygen-isoflurane anesthesia following premedication with atropine. In the trial group (Group I; n=11), 30 ml of saline containing 2 mg/kg of bupivacaine was infiltrated topically into the surgical wound right after removal of the mammary glands, whereas only saline solution was infiltrated in the control group (Group II; n=10). At the same time, carprofen (4 mg/kg) was also administered subcutaneously in both groups. Behavioral signs of pain were monitored during the recovery period after general anesthesia. In order to examine the behavioral changes associated with acute pain, a questionnaire was prepared and given to the owners to be completed 4 hr and then 10 hr after the operation. According to the owners’ anwers to the questionnaire, a pain score was specified using a “numerical rating scale” for each cat. Although some cats showed mild to moderate pain, the pain score recorded at 4 hr after the operation was significantly lower in Group I (P<0.001). No significant difference was found at 10 hr after the operation between the groups. The incidence of vocalization, aggression and convulsion within 2 hr after the operation was also lower in Group I. In conclusion, wound infiltration with bupivacaine before incisional closure provided reliable analgesia at least 4 hr after bilateral radical mastectomy in cats.  相似文献

AIM:To investigate the expression of stem cell factor (SCF) in tryptase -positive mast cells (MCs) in different types of human periapical diseases for determining the role of SCF and MCs in the pathogenesis of periapical diseases. METHODS:A total 50 cases of specimens were involved in this study, including healthy control (n=20), periapical cyst (n=15) and periapical granuloma (n=15). The tissue material was fixed in 10% formalin for at least 48 h, stained with hematoxylin and eosin for the observation of histopathology, stained with immunohistochemistry for identifying MCs and MCs degranulation, and stained with double immunofluorescence for identification of tryptase-SCF double positive MCs. RESULTS:Compared with healthy control, significantly higher densities of both total and degranulated MCs in human periapical lesions were observed. The densities of both total and degranulated MCs in the periapical cyst were significantly higher than that in the periapical granuloma. The density of tryptase-SCF double positive MCs in the periapical lesions was significantly higher than that in the healthy controls. The density oftryptase-SCF double positive MCs in the periapical cyst was significantly higher than that in periapical granuloma. No significant difference in the density of MCs between immunohistochemistry staining and double immunofluorescence staining was observed. CONCLUSION: The tryptase-SCF double positive MCs play an active role in the pathogenesis of the periapical inflammatory lesions, particularly in the formation of fibrous tissue in periapical cyst. The potential role of the tryptase-SCF double positive MCs relates with the initiation, development, and persistence of the periapical inflammatory process.  相似文献

AIM:To examine the expression of T-cell immunoglobulin mucin 1 (TIM-1) on tryptase-positive mast cells (MCs) in different severities of human chronic periodontitis. METHODS:Human gingival specimens (n=92) were involved in this study, including healthy control (n=27), mild chronic periodontitis (n=34) and severe chronic periodontitis (n=31). The gingival specimens were fixed in 4% formaldehyde. Paraffin embedding and serial sectioning with hematoxylin and eosin staining were performed for histopathological examination, and double-immunofluorescence staining was conducted for identification of tryptase-TIM-1 double-positive MCs in the gingival tissues. RESULTS:Compared with the healthy controls, the densities (cells/mm2) of tryptase-TIM-1 double-positive MCs were significantly increased in both mild chronic periodontitis group (P<0.05) and severe chronic periodontitis group (P<0.01). However, compared with mild chronic periodontitis group, both the score of gingival tissue inflammation and the density of tryptase-TIM-1 double-positive MCs in the gingival tissues were significantly increased in severe periodontitis group (P<0.05). CONCLUSION:Significantly increased number of tryptase-TIM-1 double-positive MCs has the similar tendency as the severity of periodontitis inflammation in human chronic periodontitis, suggesting that tryptase-TIM-1 double-positive MCs may play an important role in human chronic periodotitis.  相似文献

AIM:To investigate the effect of c-Myc inhibitor 10058-F4 on human chronic myeloid leukemia (CML) K562 cells and imatinib-resistant K562/G cells. METHODS:The protein expression of c-Myc was detected by Western blotting. Cell proliferation was evaluated by MTT assay and colony formation assay. PI staining was used to determine the cell cycle distribution. Annexin V-PI staining was applied for apoptosis detection. RESULTS:Imatinib-resistant K562/G cells displayed lower sensitivity to imatinib than K562 cells with high expression of c-Myc. Treatment with specific c-Myc inhibitor 10058-F4 inhibited the cell proliferation in a dose- and time-dependent manner, and K562/G displayed more sensitivity to 10058-F4 than K562 cells. 10058-F4 also induced cell cycle arrest in G0/G1 phase and induced apoptotic cell death in the 2 cells. Importantly, 10058-F4 suppressed the colony formation ability in K562 and K562/G cells. CONCLUSION:c-Myc is a novel target to overcome imatinib-induced drug resistance, and c-Myc inhibitor provides a new approach in CML therapy.  相似文献

ObjectiveTo evaluate the analgesic and physiological effects of epidural morphine administered at the sixth and seventh lumbar or the fifth and sixth thoracic vertebrae in dogs undergoing thoracotomy.Study designProspective, randomized, blinded trial.AnimalsFourteen mixed-breed dogs, weighing 8.6 ± 1.4 kg.MethodsThe animals received acepromazine (0.1 mg kg?1) IM and anesthesia was induced with propofol (4 mg kg?1) IV. The lumbosacral space was punctured and an epidural catheter was inserted up to the region between the sixth and seventh lumbar vertebrae (L, n = 6) or up to the fifth or sixth intercostal space (T, n = 8). The dogs were allowed to recover and after radiographic confirmation of correct catheter position, anesthesia was reinduced with propofol IV and maintained with 1.7% isoflurane. Following stabilization of monitored parameters, animals received morphine (0.1 mg kg?1) diluted in 0.9% NaCl to a final volume of 0.25 mL kg?1 via the epidural catheter, and after 40 minutes, thoracotomy was initiated. Heart rate and rhythm, systolic, mean and diastolic arterial pressures, respiratory rate, arterial hemoglobin oxygen saturation, partial pressure of expired CO2 and body temperature were measured immediately before the epidural administration of morphine (0 minute) and every 10 minutes during the anesthetic period. The Melbourne pain scale and the visual analog scale were used to assess post-operative pain. The evaluation began 3 hours after the epidural administration of morphine and occurred each hour until rescue analgesia.ResultsThere were no important variations in the physiological parameters during the anesthetic period. The post-operative analgesic period differed between the groups, being longer in T (9.9 ± 1.6 hours) compared with L (5.8 ± 0.8 hours).ConclusionsThe use of morphine, at a volume of 0.25 mL kg?1, administered epidurally over the thoracic vertebrae provided longer lasting analgesia than when deposited over the lumbar vertebrae.Clinical relevanceThe deposition of epidural morphine provided longer lasting analgesia when administered near to the innervation of the injured tissue without increasing side effects.  相似文献

ObjectiveTo investigate a topical local anesthesia technique as a means to prevent and/or diminish pain in mice in a laboratory setting associated with tail vein injections performed by personnel in training.Study designProspective, randomized experimental trial.AnimalsThirty six adult female, 23–28 g CD-1 mice from an in-house training colony. They were acclimated to routine training and handling classes.MethodsEutectic mixture of local anesthetics (EMLA) cream (2.5% lidocaine/2.5% prilocaine) or a bland ointment control (n = 18) was applied on the tail prior to intravenous injection. The injections were performed by novices, who had never attempted the procedure, and experienced personnel. All participants were blinded to treatment groups. Three injection attempts were allowed per animal. The mice were observed and scored by blinded evaluators for behavioral and physiological changes, including respiratory rate, vocalization, tail flick, and escape behaviors, during and after the injection.ResultsThis study demonstrates that aversive behaviors induced by lateral tail vein injection were not changed by the preemptive application of EMLA cream. The aversive behaviors associated with lateral tail vein injection were significantly affected by the number of injection attempts and the individual's experience level.Conclusions and clinical relevanceTopical EMLA cream did not reduce signs of aversive reaction to tail vein injection and thus we did not find support for its use in mouse training programs for tail vein injections.  相似文献

Indiscriminate use of pesticides has elevated the risk of contamination of environment and aquatic habitat. Considering the above fact, the present study has been under taken to investigate the alteration of some blood parameters of freshwater teleost Mystus vittatus after chronic exposure to sublethal concentrations of Metasystox (4 ppm.) and Sevin (7 ppm.) individually. The main alterations observed in certain haematological parameters were clotting time (CT), erythrocyte sedimentation rate (ESR), haemoglobin percentage (Hb%), red blood corpuscles (RBCs), white blood corpuscles (WBCs), pack cell volume (PCV), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC). Alterations in organic constituents of blood, such as glucose, blood urea, total plasma protein and cholesterol, and in inorganic constituents, such as calcium, iron, magnesium, and phosphorus, were also studied. Alteration of enzymes such as glucose-6-phosphatase (G-6-pase), fructose-1-6-diphosphatase (F-1-6-dipase), serum glutamic oxaloacetic transaminase (SGOT) and serum glutamic pyruvic transaminase (SGPT) were also studied, so that the changes in the blood biochemistry due to the pesticidal stress could be understood. Results indicated that CT, WBCs, MCH, MCHC, glucose, blood urea, cholesterol, magnesium and SGOT were increased, whereas other parameters were found to be decreased in both cases. Possible reasons for the above elevation are discussed in the light of available literature.  相似文献
