BACKGROUND: Left ventricular (LV) torsional deformation plays an important role in myocardial function. However, it has never been assessed in the awake dog, because magnetic resonance imaging and sonomicrometry have been the only methods available so far. HYPOTHESIS: Two dimensional speckle tracking echocardiography (STE), a new ultrasound imaging technique, provides a repeatable and reproducible noninvasive assessment of systolic LV wringing motion in the awake dog. ANIMALS: Six healthy dogs were used to determine the repeatability and reproducibility of STE variables (study 1). These variables also were prospectively assessed in a population of 35 healthy dogs (study 2). METHODS: Peak LV basal and apical systolic rotations were measured by STE from right parasternal short-axis views using automatic frame-to-frame tracking of gray-scale speckle patterns. Systolic LV torsion (LVtor, degrees ) was defined as apical rotation relative to the base. RESULTS: All within-day and between-day coefficients of variation were <20% (6.8-18.0%). Amplitude of apical systolic rotation was significantly higher (P < .001) than the basal value (5.4 +/- 3.2 degrees and -3.1 +/- 1.3 degrees , respectively). Global LVtor was significantly correlated with systolic longitudinal LV myocardial velocity gradient assessed by tissue Doppler (P < .05), but not with either systolic radial LV myocardial velocity gradient or the ratio of early mitral inflow velocity to early mitral annular velocity (Em/Ea). CONCLUSIONS AND CLINICAL IMPORTANCE: Speckle tracking echocardiography is a repeatable and reproducible method for assessing systolic LV torsional deformation. The combination of these new STE indices with tissue Doppler variables could provide a new approach for quantifying canine LV systolic function.  相似文献

BACKGROUND: Atrial fibrillation (AF) in dogs can be managed by electrical cardioversion to sinus rhythm, but early recurrence of AF occurs in some dogs. In humans, the commonly evaluated clinical variables for prediction of early relapse of AF are left atrial size and duration of AF. It is unclear whether the duration of AF affects maintenance of sinus rhythm after cardioversion in dogs with spontaneous AF. HYPOTHESIS: That duration of sinus rhythm after cardioversion is related to the chronicity of AF. ANIMALS: Forty-one consecutive dogs that had undergone successful transthoracic cardioversion for spontaneous AF were evaluated. METHODS: The relationship between the duration of documented AF and the duration of sinus rhythm after cardioversion was statistically evaluated using data obtained retrospectively. The effects of structural heart disease and pretreatment with amiodarone were also evaluated. RESULTS: The presence of structural heart disease and the duration of documented AF had significant effects on maintenance of sinus rhythm after cardioversion (P <.001 and P=.022, respectively). The duration of documented AF was inversely related to the duration of sinus rhythm (P=.022) in dogs with and without structural heart disease. CONCLUSIONS AND CLINICAL IMPORTANCE: Estimates of duration of sinus rhythm based on the duration of documented AF are provided for dogs with and without heart disease allowing prediction of risk for early AF relapse.  相似文献

Heart failure (HF) is the end stage of various cardiovascular diseases. The prognosis of patients with HF is poor. The precise pathophysiological mechanism of HF is still not fully elucidated. Recent studies have shown that mitochondrial dysfunction is closely associated with HF. Mitophagy plays a crucial role in mitochondrial quality control. In this article, we review the present evidence implicating the role of mitophagy in HF, and provide the potential ther?apeutic strategies targeting mitophagy to improve outcome in the patients with HF.  相似文献

AIM:To investigate the relationship between galectin-3(Gal-3) and myocardial fibrosis,and to clarify the role of Gal-3 in ventricular remodeling in rabbits with ischemic cardiac insufficiency.METHODS:A rabbit model of ischemic cardiac insufficiency was established by ligation of the anterior descending branch of the coronary artery.The 20 rabbits were randomly divided into sham operation group and cardiac insufficiency group by random number table method.After 4 weeks of coronary artery ligation,the cardiac function was measured by cardiac echocardiogram.Real-time PCR and Western blot were used to detect the expression of Gal-3,type I collagen and type Ⅲ collagen at mRNA and protein levels in the myocardium.The serum Gal-3 contents were measured by ELISA.HE staining and Masson staining were used to observe the degree of fibrosis development in myocardial tissues after infarction.RESULTS:Compared with sham operation group,the mRNA expression of Gal-3 in cardiac insufficiency group was significantly increased.At the same time,type I collagen,type Ⅲ collagen and collagen type I/Ⅲ ratio were also increased significantly.The protein contents of Gal-3,type I collagen and type Ⅲ collagen were increased significantly.The serum Gal-3 levels were significantly increased.The pathological changes were observed in cardiac insufficiency group as the myocardial cell morphological disorder and marked hyperplasia of fibrous tissue were seen.CONCLUSION:Gal-3 aggravates myocardial fibrosis in rabbits with ischemic cardiac insufficiency,and promotes the ventricular remodeling and the occurrence of heart failure.  相似文献

为研究海水酸化和重金属污染物对滩涂贝类的联合影响,采用静态急性毒性实验方法,以自然海水(pH 8.2)为对照,研究了不同酸化条件下(pH 7.7和7.3),菲律宾蛤仔(Ruditapes philippinarum)耗氧率(OR)、滤水率(FR)和心率对重金属Cu (0、0.06和0.60 mg/L)和Cd (0、0.03和0.30 mg/L)胁迫的生理响应。结果显示,Cu胁迫显著降低了蛤仔的OR和FR (P<0.05),随胁迫时间的延长,蛤仔心率显著下降(P<0.05)。Cd胁迫对蛤仔OR和FR无显著影响(P>0.05),但在0.30 mg/L Cd胁迫下,随着时间的增加,蛤仔心率显著升高(P<0.05)。pH 7.7和7.3两种酸化胁迫对蛤仔的OR和FR无显著影响(P>0.05),但在pH 7.3的海水中,蛤仔心率出现短暂下降并快速恢复的现象。研究表明,蛤仔短期暴露于Cu、Cd或酸化海水中均会影响其生理功能,影响程度排序为Cu>Cd>pH。实验中未发现短期海水酸化与Cu或Cd对菲律宾蛤仔存在联合毒性效应。本研究分析了菲律宾蛤仔响应海水酸化和重金属胁迫的机制,同时探讨了把贝类心率作为环境胁迫指标的可行性。  相似文献

AIM: To observe the role of exogenous and endogenous basic fibroblst growth factor (bFGF) on myocardial ischemia/reperfusion(I/R) injury of rats.METHODS:bFGF and bFGF antiserum were applied to rat isolated I/R heart. Myocardial function, coronary effluent volume,protein and myoglobin content as well as LDH activity in coronary effluent fluid, myocardial calcium, MDA and ATP concentration as well as PKC, MAPK activity were measured. RESULTS:Compared with control, myocardial function in I/R group significantly decreased. Protein, myoglobin content and LDH activity in coronary effluent liquid as well as myocardial MDA and calcium content increased, while myocardial ATP concentration decreased(all P<0.01). Compared with I/R group, ±LV dp/dtmax in bFGF group increased by 43% and 26%, respectively. LVEDP decreased by 40%. HRr/HRi and B/A augmented by 42% and 20%, respectively. Protein and myoglobin content as well as LDH activity lowered by 29%,30% (all P<0.01) and 33% (P<0.05) respectively. Myocardial MDA and calcium content decreased by 44% and 35%, respectively, while myocardial ATP level as well as PKC and MAPK activity increased by 34%,41% and 10% (all P<0.01), respectively. In bFGF antiserum group, ±LV dp/dtmax were 35% and 38% lower than those in I/R group. LVEDP increased by 93%. HRr/HRi and B/A decreased by 36% and 45%, respectively. Protein and myoglobin content as well as LDH activity augmented by 54%,96% (all P<0.01) and 34% (P<0.05) respectively. Myocardial MDA and calcium content increased by 24% and 50%, respectively, while myocardial ATP level as well as PKC and MAPK activity lowered by 28%,21% and 8% (all P<0.01), respectively. CONCLUSION:Endogenous bFGF is a protective factor against myocardial ischemia/reperfusion injury of rats.  相似文献

AIM:To investigate the cardioprotective effects of monophosphoryl lipid A(MLA) and its molecular mechanisms.METHODS: In the ischemia/reperfusion(I/R) model of porcine hearts, the effect of MLA on myocardial infarct size was observed. The protein levels of extracellular signal regulatory kinases (ERKs) and heat shock protein (HSP) 86 were detected by using western blotting method. RESULTS: MLA pretreatment significantly limited the infarct size in the porcine hearts subjected to I/R. It is also found that MLA pretreatment upregulated the protein levels of HSP86 and ERKs. CONCLUSION: MLA pretreatment can attenuate I/R injury in porcine hearts. The protective mechanism might be associated with ERKs-induced upregulation of HSP86 synthesis.  相似文献

AIM:To investigate whether calcium activated potassium channel (KCa) and mitochondrial permeability transition pore (mPTP) contribute to cardioprotective effect elicited by anoxic postconditioning.METHODS:The isolated perfused hearts of male Sprague-Dawley rats were subjected to 30 min global anoxic followed by 120 min reoxygenation. Formazan content of myocardium was measured at 490 nm spectrophotometrically,and the level of lactate dehydrogenase (LDH) in the coronary effluent was detected. The absorbance of isolated heart mitochondria at 520 nm was determined. RESULTS:Anoxic postconditioning increased formazan content,reduced LDH release,improved the hemodynamic parameters of the left ventricular developed pressure,maximal rise/fall rate of left ventricular pressure,left ventricular end-diastolic pressure and rate pressure product and attenuated the decrease of coronary flow during reperfusion. Pretreatment with paxilline (1 μmol/L) inhibited the effect of anoxic postconditioning. The opening of mPTP was suppressed in the mitochondria isolated from A/R hearts treated with anoxic postconditioning.CONCLUSION:The findings indicate that in the isolated rat heart,anoxic postconditioning protects myocardium against anoxic/reoxygenation injury via inhibiting KCa and the mPTP opening.  相似文献

AIM: To study the function of autonomic nervous system by the method of heart rate variability (HRV) and arterial blood pressure variability (BPV).METHODS: The rectal temperature of rats decreased gradually with their body surfaces contacted with ice,the range of the rectal temperature varied from 19 ℃ to 37 ℃.Both the dynamic electrocardiogram (ECG) signals and dynamic blood pressure signals were recorded,respectively.RESULTS: Using the method of HRV,there was an increase in RR interval (RRI),which implied a corresponding decrease in heart rate with the rectal temperature varying from 19 ℃ to 29 ℃ (P<0.01).There was a decrease in the value of normalized low frequency power (LFn) (P<0.05) and an increase in the value of normalized high frequency power (HFn) (P<0.01) with the rectal temperatures varying from 19 ℃ to 21 ℃.Therefore,the balance control of cardiac autonomic nervous was transferred to increase of vagal nerve drive (P<0.05).Using the method of BPV,there was an increase (P<0.01) in the value of HFn related to respiratory with the body temperatures falling to 31 ℃.While the body temperature varied from 19 ℃ to 29 ℃ (except 27 ℃),there was an increase in the value of HFn related to respiratory rates (P<0.05 or P<0.01) and the balance control of autonomic nervous was transferred to increase of vagal nerve drive (P<0.05).CONCLUSION: As the body temperature reduces,the vagal nerve activity in cardiovascular system increases,and the effect of hypothermia on HRV is sensitive to BPV.  相似文献
