Utility of diagnostic tests used in diagnosis of infection in dogs experimentally inoculated with a North American isolate of Leishmania infantum infantum

Eight female beagles were infected with 1 x 10(7) (low dose, LD) or 2 x 10(8) (high dose, HD) promastigotes of a North American isolate of Leishmania infantum infantum (LIVT-1 strain) isolated from naturally infected Virginia Foxhounds. Two female beagles served as negative controls and 2 male beagles chronically infected (> 3 years) with Leishmania infantum chagasi were positive controls. Bone marrow (BM) and lymph node (LN) aspirates were collected every 6-8 weeks for cytologic evaluation, parasite culture, and polymerase chain reaction (PCR). Serum samples were collected monthly for determination of serologic responses by indirect fluorescent antibody test (IFAT) and diagnostic rK39 antigen. Cultures of BM and LN aspirates and cytology evaluation were consistently positive in positive control dogs during the course of study. Negative control dogs were negative on BM and LN cultures and on cytologic evaluation of aspirates. Amastigotes were present on cytological examination of BM aspirates in 2 experimentally infected dogs. Cultures of LN aspirates were positive on 22 samples, whereas BM cultures were positive on 12 samples for all dogs. IFA titers ranged from 0 to 1 :400 in experimentally infected dogs during the course of the study. Recombinant K39 immunoassay tests were consistently positive in positive control dogs and in the HD dogs by approximately 8 weeks after infection. BM PCR products were identified more consistently in the HD dogs compared with the LD dogs. Kappa statistics indicated PCR correlated better with cultures and cytology than did IFAT or the rK39 immunoassay results in the experimentally infected dogs.  相似文献

An indirect enzyme-linked immunosorbent assay (IELISA) was developed for the detection of equine serum antibodies to lipopolysaccharide of Salmonella enterica subsp. enterica serovar Abortusequi (LPS), a causative organism of Equine Paratyphoid. The data presented demonstrates that horses immunized with S. abortusequi LPS developed antibodies detectable by the IELISA. By comparison, the tube agglutination test (TAT) did not detect antibody to S. abortusequi LPS as consistently as the IELISA. The data suggests that the IELISA may be a more suitable test for the detection of serum antibodies to S. abortusequi than the TAT.  相似文献   

舟曲泥石流经济损失评估(英文)

[目的]对舟曲泥石流造成的直接和间接经济损失进行评估。[方法]在 2010 年 8 月 7 日舟曲发特大泥石流发生的后第一时间收集了相关数据,建立评估模型,采用比例系数法估计潜在的经济损失。[结果]评估结果表明,此次灾害共造成经济损失 16.57 亿元人民币,其中间接经济损失高达2.42 亿,直接经济损失 14.15 亿。[结论]至目前为止,比例系数法仍是一种快速有效的评估间接经济损失的方法。  相似文献   

刚地弓形虫RH株SAG3蛋白抗体检测的ELISA方法建立

[目的]利用纯化的刚地弓形虫RH株表面蛋白SAG3的原核重组蛋白为基础,建立抗体间接ELISA检测方法。[方法]采用棋盘滴定法,确定间接ELISA的最佳条件。采用鼠抗新孢子虫阳性血清、鼠抗安氏隐孢子虫阳性血清、鼠抗柔嫩艾美尔球虫阳性血清、鼠抗蓝氏贾第虫阳性血清以及鼠抗微小隐孢子虫阳性血清和健康小鼠阴性血清作对照,检测血清之间是否有交叉反应。[结果]最佳包被条件为:抗原包被浓度0.25μg/孔,被检血清稀释度1∶400,酶标二抗稀释度1∶3 000,封闭剂5%脱脂奶粉的PBST溶液;血清之间没有交叉反应。该方法的灵敏度为1∶1 600。[结论]该方法灵敏度高,特异性较强,可重复性较好,可用于对弓形虫感染的检测及流行病学调查。  相似文献   

安徽省部分地区牛衣原体的血清学调查

牛衣原体病是由鹦鹉热衣原体引起的一种接触性人兽共患传染病。本调查采用间接血凝法(IHA)对安徽省部分地区的535份牛血清进行了衣原体抗体检测,结果显示,血清抗体阳性率为22.43%(120/535);4个奶牛场的群阳性率为75%,个体阳性率为23.21%(68/293);9个肉牛场的群阳性率为88.9%,个体阳性率为21.49%(52/242)。对不同用途、不同年龄段、不同购入来源以及不同送检地区的牛衣原体抗体阳性率进行统计分析。结果表明安徽省的牛群中存在不同程度的衣原体感染。IHA法检测衣原体抗体简便、快速,适合在基层推广。  相似文献

运用3种ELISA法对提纯的Bt杀虫蛋白进行检测,结果表明:直接法ELISA和夹心法ELISA的灵敏度相同,都为0.94 ng/孔,但直接法ELISA中Bt蛋白量与OD值间呈极显著的线性关系,夹心法ELISA中Bt蛋白量与OD值间呈显著的线性关系;间接法ELISA的灵敏度较前两者低,使用HRP-IgG的灵敏度为15 ng/孔,Bt蛋白量与OD值间呈显著的线性关系,使用AKP-IgG的灵敏度为3.75ng/孔,Bt蛋白量与OD值间呈极显著的线性关系。  相似文献

用毛细管离子分析(CIA)法测定了茶叶中的锌、锰、铜、铅和镉,在10mmol/L咪唑,10mmol/Lα-羟基乙酸,pH=4.0的电泳介质中,五种离子在10min内得到了分离,用间接紫外法检测,以外标法定量,五元素的线性关系良好,相关系数为0.9973~0.9997,平均回收率在96.4%~104.2%之间,相对平均偏差为2.1%~3.3%,检测限为0.02~0.2μg/ml。此方法快速简便、准确度高、测定成本低。  相似文献

选用自然感染严重的绵羊肉孢子虫病病羊肌肉和从食道挑出的虫体制备诊断抗原,用微量间接血凝法对326只绵羊血清进行诊断。结果表明,34只(份)带虫绵羊血清检出阳性反应32只(份),阳性符合率达94%,血凝效价达1∶64—1∶1024。9只(份)健康羊血清均为阴性反应,血凝效价仅达1∶4—1∶16。240只(份)被检0.5岁羔羊血清中有166只(份)血清呈阳性反应,阳性率达69.2%。被检成年羊血清43只(份),41只(份)血清呈阳性反应,阳性率达95.33%。试验表明,用绵羊肉孢子虫滋养体制作的可溶性蛋白抗原(蛋白含量为200—300μg/ml),致敏健康绵羊红细胞作为诊断液,敏感性高,重复性稳定,特异性强,不与弓形虫和棘球蚴发生交叉反应。是一种较为理想的诊断方法。  相似文献   

Correlation analysis of the total IgY level in hen serum,egg yolk and offspring serum

The correlation between IgY levels of the serum and the yolk has been well documented in wild and domestic birds. The levels of total yolk IgY can be an index of the general health status of birds and may contribute to breeding programs when fitness of the offspring is a concern. We measured the levels of total serum IgY and yolk IgY in three different breeds （White Leghorn, Silkie and Dongxiang blue-shell） using indirect ELISA, and found that there was a significantly positive correlation between the levels of total serum IgY and total yolk IgY in all three breeds （White Leghorn： r = 0.404, P 〈 0.001, n = 100; Silkie： r = 0.561, P 〈 0.001, n = 70; Dongxiang blue-shell： r = 0.619, P 〈 0.001, n = 30）. We also measured the total serum IgY levels in the 3-day-old offspring hatched from the Silkie hens and results were significantly correlated for serum IgY levels （r = 0.535, P 〈 0.001, n = 70） and the yolk IgY levels （r = 0.481, P 〈 0.001, n = 70）. The regression analysis showed simple linear regression between IgY levels in hen serum, yolk and offspring serum. Our results suggest that total IgY level could be used as an index for chicken fitness.  相似文献
