目的 探讨小儿牛黄清心散对热性惊厥后脑损伤的保护作用。方法 将74例复杂性热性惊厥患儿随机分为治疗组40例和对照组34例,均予以常规治疗,治疗组加用小儿牛黄清心散口服,疗程14 d。比较两组治疗前后血清神经元特异性烯醇化酶(NSE)水平及脑电图异常率。结果 治疗后治疗组血清NSE水平恢复情况优于对照组(P<0.05);脑电图异常率也显著低于对照组(P<0.05)。结论 小儿牛黄清心散治疗热性惊厥后脑损伤有明显疗效,且安全性良好。  相似文献

The enolase [EC 4.2.1.11] is an essential enzyme in the glycolytic pathway catalyzing the conversion of 2-phosphoglycerate (2-PGE) to phosphoenolpyruvate (PEP). In this study, a full-length cDNA encoding α-enolase was cloned from rice brown planthopper (Nilaparvata lugens) and is provisionally designated as NlEno1. The cDNA sequence of NlEno1 was 1,851 bp with an open reading frame (ORF) of 1,305 bp and encoding 434 amino acids. The deduced protein shares high identity of 80–87% with ENO1-like protein from Hemiptera, Diptera, and Lepidoptera speices. The NlEno1 showed the highest mRNA expression level in hemolymph, followed by fat body, salivary gland, ovaries and egg, and showed trace mRNA levels in testis. The mRNA of NlEno1 showed up-regulated level in virulent N. lugens population Mudgo, IR56 and IR42 when compared with TN1 population. Injection of double-stranded RNA (dsRNA) of NlEno1 into the adults significantly down-regulated the NlEno1 mRNA level along with decreased eggs and offspring. Moreover, injection of NlEno1-dsRNA decreased mRNA level of Vitellogenin (Vg) gene. These results showed that the NlEno1, as a key glycolytic enzyme, may play roles in regulation of fecundity and adaptation of N. lugens to resistant rice varieties.  相似文献

目的:观察氦氖激光血管内照射(ILIB)治疗脑梗死患者对血液流交学、血清神经元特异性稀醇化酶(NSK)的影响。方法:38例脑梗死随机分为两组:ILIB组21例,对照组17例,两组均在基础治疗下,ILIB组加用ILIB治疗,对照组不用ILIB。治疗后第3天和第14天检测血液流变学、血清NSE浓度,并行神经功能缺损的评定。结果:两组同步治疗后血液流变学指标除红细胞体积分数外,其余4项指标均有下降,但两组治疗后比较,ILIB组较对照组下降更显著,差异有显著性;血清NSK浓度与神经功能缺损评定,治疗后两组均有下降,但ILIB组下降也更显著,差异有显著性。结论:ILIB可改善脑梗死患者血液流变学情况,且可降低血清NSK浓度,有利于促进神经功能恢复。  相似文献

表面烯醇化酶(surface enolase,SEN)和3-磷酸甘油醛脱氢酶(surface glyceraldehydes -3 - phosphate dehydro - genase,sGAPDH)都是A群链球菌(group A Streptococcus,GAS)没有踌膜结构的膜表面蛋白,它们可能在GAS感染中起到重要的作用.在本研究中,对SEN和sGAPDH在M6型GAS不同生长阶段(即在对数期和稳定期)的表达情况进行检测分析.取对数期(OD600 =0.5 -0.6)和稳定期(OD600 =1.5- 1.6)的GAS,分别测定SEN和sGAPDH活性.同时用变溶菌素(mutanolysin)提取这2个生长阶段细胞壁结合蛋白,用Western blot检测该提取物中的SEN和sGAPDH.结果表明在相同菌细胞浊度的条件下,SEN在M6型GAS对数期时的表达量及活性比其稳定期时的高45%左右.虽然sGAPDH的表达量较高但活性很低(<0.07mM NADH/min),并且在这2个生长阶段没有明显的差异.  相似文献

AIM: To investigate the effect of astragaloside IV on neurological function and the protein expression of neuron-specific enolase (NSE), Notch1 and NF-κB in acute cerebral hemorrhage (ACH) rats.METHODS: ACH model in rats was established via injection of autologous blood, and the rats were divided into 4 groups:sham, ACH, ACH+astragaloside IV (100 mg/kg) and ACH+astragaloside IV (200 mg/kg) groups. The impairment of neurological function in each group was graded, and the brain coefficient and water content were calculated. The serum level of NSE was measured by ELISA. Additionally, the expression of Notch1 and NF-κB was determined by Western blot.RESULTS: Astragaloside IV improved the neurological function and decreased the brain coefficient and water content in ACH rats. Moreover, the ACH-induced increase in NSE was inhibited after astragaloside IV treatment. Similarly, astragaloside IV also significantly attenuated the expression of Notch1 and NF-κB in ACH rats.CONCLUSION: Astragaloside IV attenuates the impairment of neurological function in ACH rats, which may be through decreasing the NSE level and down-regulating the expression of Notch1 and NF-κB.  相似文献

Diseases which cause skeletal muscle myopathy are some of the most economically damaging diseases in Atlantic salmon, Salmo salar L., aquaculture. Despite this, there are limited means of assessing fish health non‐destructively. Previous investigation of the serum proteome of Atlantic salmon, Salmo salar L., during pancreas disease (PD) has identified proteins in serum that have potential as biomarkers of the disease. Amongst these proteins, the enzyme enolase was selected as the most viable for use as a biomarker of muscle myopathy associated with PD. Western blot and immunoassay (ELISA) validated enolase as a biomarker for PD, whilst immunohistochemistry identified white muscle as the source of enolase. Enolase was shown to be a specific marker for white muscle myopathy in salmon, rising in serum concentration significantly correlating with pathological damage to the tissue.  相似文献

制备M6型GAS表面烯醇化酶抗体。皮下多点注射GAS表面烯醇化酶重组蛋白(rSEN)免疫家兔,采用亲和层析纯化免疫血清,通过SDS-PAGE、ELISA、Western-Blot等方法检测抗体的特异性,并初步用制备的抗体检测了M6血清型GAS表面烯醇化酶的表达。制备的抗体蛋白纯度较高,检测的特异性较强,用制备的抗体可以检测到活体菌表面的烯醇化酶。成功制备并纯化得到了GAS表面烯醇化酶多克隆抗体。  相似文献

AIM: To explore the character of CD8 after traumatic brain injury (TBI) in peripheral blood. METHODS: Improved Feeney's free-fall method was used to set up traumatic brain injury model. The levels of neuron-specific enolase (NSE) and CD8 in the serum of the rats were detected by ELISA. The correlation of both NSE and CD8 in the serum was analyzed by Pearson product-moment correlation. The FasL expression in the lymphocytes of peripheral blood was determined by Western blot. RESULTS: The elevated level of CD8 lagged behind the level of NSE in the serum after TBI. The serum level of NSE was significantly increased at the 1st day after TBI and reached a peak at the 3rd day, subsequently gradually decreased to a lower level; the serum level of CD8 was increased at the 3rd day after TBI, and reached a peak at the 7th day, then gradually decreased. The serum levels of NSE at 1st, 3rd and 7th days were positively correlated with the serum level of CD8 at 3rd, 7th and 14th days. However, the FasL expression in the CD8+ lymphocytes of peripheral blood showed no variation at different time points after TBI. CONCLUSION: NSE in the serum released from neural tissues after TBI stimulates immune response and induces the augmentation of CD8 in peripheral blood, which may be a cause of secondary injuries in the brain.  相似文献

从茶树‘黄旦’和‘金观音’抑制差减文库中筛选出1条烯醇酶(Enolase,ENO)cDNA片段序列,以‘铁观音’芽叶为材料克隆并验证该序列的全长编码c DNA序列。该序列全长1 753 bp,含有1个1 332 bp完整的开放阅读框,命名为CsENO,登录号KX962311。序列基因编码444个氨基酸,氨基酸序列分析发现,该c DNA与其他植物ENO高度保守,包含ENO特异结构域,与苹果(Malus×domestica)、白梨(Pyrus bretschneideri)的亲缘关系较近,相似性达84%。生物信息学预测结果显示,CsENO属于稳定的亲水蛋白,不存在跨膜结构,无信号肽,可能定位在过氧化体、溶酶体或核糖体等细胞质中,且具有多个磷酸化位点;二级结构主要由α–螺旋构成。实时荧光定量PCR结果表明,CsENO在低温、ABA、高盐、干旱逆境胁迫处理下均有表达,且表达受到不同程度的诱导。  相似文献

【目的】 OsENO2-2OsENO2通过可变剪切产生的短转录本,其cDNA序列从粳稻中花11中分离获得。本研究的主要目的是初步分析OsENO2-2在水稻抽穗期调控中的作用。【方法】 构建了OsENO2-2的超量表达载体,获得转基因植株,通过对表型的观察和统计,分析目的基因在过量表达条件下的功能,并利用反向遗传学方法验证该基因的功能。【结果】 OsENO2-2过量表达导致水稻在长日照条件下的抽穗期推迟,而短日照条件下抽穗期无明显变化。通过qRT-PCR方法对水稻开花关键基因的检测发现,在长日照条件下,RFT1的表达量在超表达材料中显著下调,其他开花重要基因表达量在野生型和超表达材料中没有显著变化;而在短日照条件下,所有检测基因的表达量在野生型和超表达材料中均没有显著变化。【结论】 在长日照条件下,OsENO2-2主要通过调控RFT1基因的表达来调控水稻抽穗期。  相似文献
