茶叶对人体健康有众多益处是因为它富含儿茶素类化合物。茶叶中的儿茶素类化合物有10种以上,其中含量最高的是表没食子儿茶素没食子酸酯(EGCG)。许多研究表明,以EGCG为代表的茶叶儿茶素类化合物对乳腺癌具有抑制作用,其作用包括:抑制致癌物诱导的活性氧自由基(ROS)升高及其引起的DNA损伤、降低细胞对肿瘤坏死因子的响应、阻碍雌激素与雌激素受体的结合、抑制DNA甲基化、保护线粒体免受氧化损伤、抑制肿瘤血管生成、减少癌细胞的转移和增殖、改善代谢综合征以减少抗癌药物副作用、与抗癌药物产生协同效应等。本文综述了该研究领域的进展,并讨论导致研究结果不一致的原因和未来的研究方向。 相似文献

Estrogen plays an important role in regulating testicular Sertoli cell number. Furthermore, S-phase kinase-associated protein 2 (SKP2) plays a central role in mammalian cell cycle progression. The objective of this study was to determine whether 17β-estradiol can regulate the expression of SKP2, and the Sertoli cell cycle, via estrogen receptor β (ERβ), the cyclic adenosine monophosphate (cAMP)-protein kinase A (PKA) and extracellular signal-regulated kinase (ERK1/2) pathway. When cultured immature boar Sertoli cells were treated with 17β-estradiol, a time-dependent increase in SKP2 mRNA and protein level was observed by real-time PCR and Western blot, and 17β-estradiol activity peaked at 30 min. Treatment with ICI182780 and ERβ antagonist reduced 17β-estradiol-induced expression of SKP2 and proliferating cell nuclear antigen (PCNA), while increasing the protein concentration of p27kip1. However, the effect of ERa antagonist on these parameters was lower than that of ICI 182780 and ERβ. Forskolin had a similar effect as 17β-estradiol on the expression of SKP2, PCNA and p27kip1, Rp-cAMP, H-89 and U0126 treatment reduced 17β-estradiol-induced changes, while H-89 also inhibited ERK1/2 activation. Therefore, 17β-estradiol mainly regulates SKP2 mRNA and protein expression via ERβ-cAMP-PKA and ERK1/2 activation. SKP2 and PCNA expression were positively correlated, while increased SKP2 expression likely resulted in p27kip1 degradation. 相似文献

The female sex hormone estrogen exerts anti‐inflammatory effects. The G‐protein‐coupled estrogen receptor (GPER) has been recently identified as a novel membrane‐type estrogen receptor that can mediate non‐genomic estrogenic effects on many cell types. We previously demonstrated that GPER inhibits tumor necrosis factor alpha‐induced expression of interleukin 6 (IL‐6) through repression of nuclear factor‐kappa B (NF‐κB) promoter activity using human breast cancer cells. Although several reports have indicated that GPER suppresses Toll‐like receptor‐induced inflammatory cytokine expression in macrophages, the molecular mechanisms of the inhibition of cytokine production via GPER remain poorly understood. In the present study, we examined GPER‐mediated inhibition of IL‐6 expression induced by lipopolysaccharide (LPS) stimulation in a mouse macrophage cell line. We found that the GPER agonist G‐1 inhibited LPS‐induced IL‐6 expression in macrophage cells, and this inhibition was due to the repression of NF‐κB promoter activity by GPER. G‐1 treatment also decreased the phosphorylation of inhibitor of κB kinases. Among the mitogen‐activated protein kinases, the phosphorylation of c‐jun N‐terminal kinase (JNK) was increased by G‐1. These findings delineate the novel mechanism of the inhibition of LPS‐induced IL‐6 through GPER‐activated JNK‐mediated negative regulation of the NF‐κB pathway in murine macrophage cells, which links anti‐inflammatory effects to estrogen. 相似文献

为探讨雌激素对子宫内膜容受性的影响,实现主动干预子宫内膜容受性、提高体外受精-胚胎移植(IVF-ET)的成功率,本研究利用免疫组化法检测了雌激素受体在小鼠发情周期子宫内膜中的表达规律.结果显示,自然发情受孕组、自然发情假孕组(对照组)和超数排卵组小鼠在见栓后第2天,子宫内膜上雌激素受体(ER)表达量3组之间差异不显著(P>0.05);之后的第4、6、8天,超数排卵组雌激素受体表达量显著高于其他2组(P<0.05);自然发情受孕组小鼠子宫内膜中ER的表达在第4、6天时显著高于自然发情假孕组(P<0.05),在第8天时,自然发情受孕组小鼠子宫内膜中的ER阳性率与自然发情假孕组差异不显著(P>0.05).与自然发情受孕组相比,自然发情假孕组和超数排卵组小鼠子宫内膜容受性下降,窗口期开放延迟,而自然发情假孕组和超数排卵组差异不明显(P>0.05).以上结果表明,雌激素通过ER对小鼠胚胎着床及窗口期子宫内膜容受性变化具有调节作用. 相似文献

高密度养殖彭泽鲫造成雄鱼较多的分子机制及成因分析 总被引:1,自引:0,他引:1
前期将同一批经雌核发育产生的F1彭泽鲫仔鱼(Pcc)分别置于实验室和池塘进行养殖,发现实验室养殖缸中出现了高比例的雄鱼。本研究为验证雌雄比例是否与密度有关,采用F1彭泽鲫雌鱼经人工雌核发育技术繁育出PccF 2,分高、低密度在实验室进行养殖,并比较了高、低养殖密度下雌雄鱼中性腺分化相关基因的表达。结果表明在PccF 2高、低密度养殖组中,雌鱼中dmrt1c、抗缪勒氏管激素基因amh、芳香化酶基因cyp19a1a的表达显著高于雄鱼中对应基因的表达;雌鱼中dmrt1a、dmrt1b、3β羟基类固醇脱氢酶(3bhsd)、11β羟基类固醇脱氢酶2(11bhsd2)、17α羟化酶/17,20碳链裂解酶a1(cyp17a1)和类固醇激素急性调节蛋白(star)的表达显著低于雄鱼中对应基因的表达。实验室不同密度养殖、不同性别PccF 2彭泽鲫中差异表达基因主要为类固醇合成酶类基因,这些基因的表达差异可能造成雄鱼过多。 相似文献

本文建立了鲜牛乳及奶粉中4种雌激素(17β-雌二醇、雌三醇、雌酮、己烯雌酚)的HPLC分析方法。奶粉样品溶解于水,QuEChERS方法萃取,用乙腈沉淀蛋白质并提取雌激素,正己烷除脂,经氨基柱净化,C18色谱柱分离,采用紫外检测器,检测波长为230 nm。该方法对鲜牛乳及奶粉中17β-雌二醇、雌三醇、雌酮、己烯雌酚的检出限(S/N=3计)分别为0.03 mg/kg、0.026 mg/kg、0.03 mg/kg、0.012 mg/kg和0.024 mg/kg、0.021 mg/kg、0.025 mg/kg、0.01 mg/kg。定量限(以S/N=10计)分别为0.1 mg/kg、0.09 mg/kg、0.1 mg/kg、0.04 mg/kg和0.07 mg/kg、0.07 mg/kg、0.08 mg/kg、0.03 mg/kg在0.1、1.0、2.0 mg/kg 三个浓度水平添加的平均回收率在81%~97%的范围内,RSD值在3.2%~8.7%之间。 相似文献

ABSTRACT: Because blood vitellogenin (Vg) has been considered a biomarker for environmental estrogens, the basal levels of Vg and 17β-estradiol (E2 ) were determined in male Japanese whiting reared under natural conditions. Serum levels of Vg and E2 were measured and gonadal development was assessed by gonadosomatic index (GSI) and histological observation in 8–10 male fish at monthly intervals throughout the annual reproductive cycle. Serum E2 was <60 pg/mL throughout the study period. In contrast, serum Vg exhibited seasonal changes: serum levels of Vg gradually increased from April to May (mean 63 ± 13 ng/mL and 124 ± 48 ng/mL in April and May, respectively), and then reached a peak value (mean 352 ± 68 ng/mL) in June. Thereafter, serum Vg gradually decreased, reaching undetectable levels (<50 ng/mL) in October. Serum levels of Vg tended to increase in the male fish in which the GSI was >1%. Histological observation revealed that testes in such male fish were in active spermatogenesis and then all of the testes of male fish in which serum Vg decreased to ND levels were regressed. These results suggest that Vg productive potency (sensitivity to estrogens) may increase in the spermatogenic stage, resulting in production of Vg in response to very low levels of natural or xenobiotic estrogens. 相似文献

为检测成年雌兔背根神经节中是否存在雌激素受体(ER)与M2型乙酰胆碱受体(M2mAchR)的分布与共存,采用免疫荧光双标记染色法对ER与M2mAchR在背根神经节中的分布情况进行了研究。结果显示,在成年雌兔背根神经节中:(1) ER分布主要以神经元为主。其中,ERα主要表达于中小型神经元,而ERβ则多分布于大中型神经元。(2)相较ER而言,M2mAchR分布更为广泛,除神经元外,神经纤维等组织上也有弱阳性表达。(3)两种ER与M2mAchR均有共存现象,但共存情况存在差异,ERα/M2mAchR主要共分布于中小型细胞,ERβ/M2mAchR主要共分布于大中型神经元。结果表明,首先,雌激素具有通过影响背根神经节部分神经元的活动而实现对躯体内脏感觉活动间接调节的条件。其次,在此影响的机制中,雌激素除可以直接作用于神经元上的受体之外,还具有与共表达的M2mAchR交互作用而实现的条件。为研究雌激素影响神经系统调控活动的机制以及背根神经节的神经内分泌调节机制提供了重要的形态学依据。 相似文献
