This study was undertaken to determine the influence of dietary protein levels on growth, feed utilization and liver histology in common sole. Four isoenergetic diets were formulated to contain four different crude protein levels: 39 (P39), 45 (P45), 51 (P51) and 57 (P57) % dry weight. Fifty animals per tank (initial weight 10.2 ± 0.4 g) were randomly distributed into twelve 500 litre square tanks (bottom surface: 5600 cm2) connected to a closed recirculation system. The diets were tested in triplicate for 84 days. At the end of the experiment, the final weight ranged from 19.6 (P39) to 25.4 g (P57). The specific growth rate showed statistical differences between groups, with the best results in the group fed diet P57 (1.07% day?1). Significant differences between groups were also recorded for the feed conversion ratio, with values of 1.31, 1.28, 1.12 and 0.94 in P39, P45, P51 and P57 respectively. Gross lipid efficiency was also significantly affected by the dietary treatment, with the highest value (42.07%) found in P57. Ammonia excretion, expressed as g 100 g?1 protein intake, was significantly lower for group P39 (2.46) than groups P51 (4.70) and P57 (4.75). Increased incidence of lipid droplets in hepatocytes was observed when the dietary protein levels increased and/or dietary lipid decreased. 相似文献

为研究香菇多糖对力竭后小鼠肝组织的影响,通过建立灌服香菇多糖的小鼠力竭游泳实验模型,采用试剂盒法测定了小鼠安静时肝糖元及安静与力竭后谷胱甘肽过氧化物酶(GSH PX)、丙二醛(MDA)、血浆谷丙转氨酶(GPT)活性。结果显示:肝糖元,安静对照组(RC)(36 93±9 81)mg g,安静服药组(RM)(73 44±21 71)mg g,RM组明显升高(P<0 001);高强度游泳时间,运动对照组(EC)(42 52±7 82)min,运动服药组(EM)(49 90±5 31)min,EM组明显延长(P<0 05);GSH PX,RC组(60 25±5 31)U mg,RM组(68 37±7 92)U mg,RM组明显升高(P<0 05)。EC组(56 77±7 30)U mg,EM组(63 28±4 23)U mg,EM组明显升高(P<0 05);MDA水平,RC组(5 76±0 87)mmol L,RM组(4 59±0 57)mmol L,RM组明显降低(P<0 05)。EC组(14 1±1 28)mmol L,EM组(12 53±4 93)mmol L,EM组明显降低(P<0 01);GPT活性,RC组(6 14±1 21)U L,RM组(6 25±1 02)U L,差异不显著(P>0 05)。EC组(12 53±4 93)U L,EM组(7 42±2 90)U L,EM组明显降低(P<0 05)。表明香菇多糖具有明显的抗疲劳、抗氧化、抗损伤功效,可减轻大强度运动后自由基造成的损伤。 相似文献

Fifteen pesticides, representatives of different chemical groups, were tested for their inhibitory effect on the glucuronidation of 4-nitrophenol (4-NP) and phenolphthalein (PPh) by rat liver microsomes. Three herbicides (simazine, chlorsulfuron, tribenuron-methyl), two insecticides (dioxacarb, carbaryl) and one fungicide (zineb) significantly decreased the UDP-glucuronosyltransferase (UDPGT) activity. The carbamate insecticide dioxacarb was found to be the most potent inhibitor, at 1 mM concentration suppressing 4-NP-UDPGT activity completely, and reducing by 55% the activity associated with the conjugation of PPh. One millimole simazine and carbaryl affected only 4-NP glucuronidation, while chlorsulfuron and zineb exerted a marked inhibition of both 4-NP and PPh conversion. Concentrations of 0·1 mM carbaryl, dioxacarb and zineb were still inhibitory against 4-NP-UDPGT, with zineb producing 40% inhibition of PPh glucuronidation. As a whole, UDPGT isoforms conjugating PPh were less sensitive to the agrochemicals tested. Kinetic studies with dioxacarb, chlorsulfuron and carbaryl revealed a mixed type of inhibition with respect to the acceptor substrate 4-NP, with apparentKi values of 70 μM , 120 μM and 160 μM , respectively. 相似文献

A 1H NMR‐based metabolomics approach was to explore the effect of ultrafiltered fish protein hydrolysate (UF) levels on the liver and muscle metabolic profile of juvenile turbot (Scophthalmus maximus L.). Fish protein hydrolysate (FPH) was produced from by‐products by enzymatic treatment, and UF was obtained by diluting FPH followed filtration. Fish were fed diets containing fish meal protein, which was, respectively, replaced by UF protein 0, 50, 100, 150 and 200 g kg?1 of dietary total protein (UF‐0, UF‐5, UF‐10, UF‐15 and UF‐20) for 68 days. OPLS‐DA of liver and muscle showed that high levels of UF in diets may lead to corresponding tissue metabolites changes. In liver tissue, changing metabolites included dimethylamine, N,N‐dimethylglycine, valine, isoleucine, leucine, arginine and alanine and were mainly involved in choline metabolism and amino acid metabolism. For muscle tissue, changing metabolites included lactate, alanine, proline, fumarate, tyrosine, histidine, cystathionine and taurine. And the metabolites were mainly involved in glycolysis and gluconeogenesis, proline metabolism and taurine metabolism. 1H NMR‐based metabolomics is a useful approach to investigate different levels of UF on metabolic profile in liver and muscle tissues combined with the growth. 相似文献

To explore the mechanism of fatty liver formation induced by high non‐protein energy diets in grass carp (Ctenopharyngodon idella), basal diet and high‐energy diets were fed to juvenile grass carp for 9 weeks. The experimental groups fed on high‐energy diets which included a high‐lipid diet (H‐LIP), a high‐carbohydrate diet (H‐CHO) and a high‐lipid and carbohydrate diet (H‐CL). The control group fed on basal diet. Growth performance, liver fat accumulation, serum biochemical indexes and the expression levels of lipid metabolism‐related genes (SREBP‐1, PPARγ, FAS, ACC1, and LPL) and miRNAs (miR‐33, miR‐122, and miR‐370) were examined at the end of the feeding trial. There were no significant differences in growth rate and feed efficiency among the four groups. However, significant increase in mesenteric and liver fat contents, and lipid droplets in the liver was induced by high‐lipid and high‐carbohydrate diets. There were significant differences in serum biochemical indicators such as AST/ALT, GLB, TG and TP, and liver fatty acid composition between the control and experimental groups. The expression levels of SREBP‐1, PPARγ, FAS, ACC1 and LPL were upregulated, while CPT‐1 was downregulated with the high‐energy treatments. Additionally, the expression levels of miR‐33, miR‐122 and miR‐370 in the liver were higher in the three high‐energy treatments than those in the control (P < 0.05). The results suggest that modifications of lipid metabolism‐related genes and miRNAs may be involved in fatty liver formation induced by high non‐protein energy diets in grass carp. 相似文献

采用CloneMinerTM cDNA文库构建试剂盒构建了患败血症的鲢(Hypophthalmichthys molitrix)肝和肾的cDNA文库.经检验,文库的滴度为1.34×107 cfu/mL,总库容为2.68×107 cfu,阳性克隆率为97.5%,平均插入片段大于1.2 kb.对80个随机挑选的克隆进行两端测序,结果显示有74个测序成功.经在GenBank上BLAST比对,其中57个为已知功能基因,属于45个不同基因,另外17个没有明显的同源序列.在已知功能的57个克隆中,含有全长编码区序列的克隆共有49个,全长cDNA比率为61.25%.采用PCR法对文库进行筛选,获得一个含MHC classⅠ完整编码区的序列,该序列具有典型的脊椎动物MHC class Ⅰ的序列特征.本研究完成鲢肝和肾较高质量的cDNA文库的构建,旨在为今后开展鲢功能基因研究奠定基础. 相似文献
