The adaptive response of the omnivorous ide,Leuciscus idus melanotus, to drastic metabolic conditions was analyzed on different levels of organisation investigating a variety of parameters: Organism (condition factor, liver-somatic index), organ (liver structure), cellular and subcellular level (hepatocyte structure, glycogen and lipid storage, contents and distribution of nucleic acids, enzyme alterations).During starvation, ide were able to maintain liver integrity in a biphasic process: after an initial phase of disturbance, ide established a new structural and metabolic homeostasis. Recovery from starvation was possible only with a complete diet but not with a sucrose diet. Carbohydrate overload, as evoked by sucrose refeeding, did not result in liver or carcass fattening as known from mammals.To the best of our knowledge, the present study is the first to use enzyme histochemistry in fish nutrition research. In mammals, histochemistry is particularly helpful for understanding processes of hepatic metabolic adaptation. In fish, however, on the basis of our results, enzyme histochemical studies appear to be of limited value, as long as no further data are available on a zonal distribution of enzyme activities in teleost liver parenchyma. For many fish species, dietary fish oil (FO) has been substituted with other oils such as poultry oil (PO) without affecting growth performance. However, in barramundi, the mechanisms by which fatty acid metabolism is regulated are poorly understood, and the effects of FO substitution are unknown. This study defined changes in the expression of genes controlling the metabolism of fatty acids in barramundi over a 24‐h time period after a single meal. From one to 12 h after a single feeding event, the expression of fatty acid synthesis genes in the liver was upregulated, while genes involved in the β‐oxidation showed minimal alteration. However, the expression of β‐oxidation genes was significantly correlated with the expression of genes regulating fatty acid synthesis. In a second experiment, the changes in liver fatty acid composition and gene expression were defined after FO was substituted with PO. Liver fatty acid profile reflected the diet composition, with some subtle exceptions supporting the enrichment of certain long‐chain polyunsaturated fatty acids in the liver. The fish from all experimental groups preferentially retained more docosahexaenoic acid than eicosapentaenoic acid in the liver, suggesting a bioconversion of this fatty acid to intermediate fatty acids. Replacement of FO with PO significantly regulated genes controlling both fatty acid synthesis and catabolism pathways, potentially related to a higher percentage of monounsaturated fatty acids, in the livers of fish fed these diets. The results demonstrated that diet composition significantly altered the lipid metabolism in barramundi and that there was a balance between direct dietary effects and endogenous synthetic capacity.  相似文献

For the first time, pre‐ and post‐hepatic plasma lipid profiles were monitored following a single meal in a free‐swimming, non‐anaesthetized fish. Rainbow trout (Oncorhynchus mykiss; 700–1500 g; 10 °C) were equipped with cannulae in the dorsal aorta (DA) and hepatic portal vein (HPV). Simultaneous blood samples, taken from both cannulae at 0, 3, 6, 12, 24 and 48 h postprandial, revealed the time course of the plasma lipid profiles following a single meal (1% of body mass). Primarily monounsaturated fatty acids with the exception of 18:1n ? 9, increased significantly from baseline by 12 h postprandial without greatly affecting total plasma lipid concentrations. Total plasma lipids then showed a small peak at 24 h postprandial, coinciding with a peak in triacylglycerols. We conclude that assimilation of lipids from the digest into the plasma is slower than reported for proteins and carbohydrates in the same species. Furthermore, as there were no significant differences between the HPV and DA, no measurable effect of hepatic passage on plasma lipid levels was resolved. Therefore, we also conclude that, in contrast to that in higher vertebrates, hepatic passage does not seem to have a major role in rainbow trout for modulating the postprandial plasma profile of lipids.  相似文献

Mitochondrial NAD(P)-dependent malic enzyme [EC 1.1.1.39, L-malate: NAD+ oxidoreductase (decarboxylating)] was purified from herring testicular tissue to a specific activity of 26.4 μmol NADH/min/mg protein. Herring testicular tissue is one of the most abundant sources of this enzyme. The purification procedure involved differential centrifugation of mitochondria and then chromatography on DEAE-Sephacel, Red Agarose and Sephacryl S-300. This enzyme catalyzes the oxidative decarboxylation of malate in the presence of Mn2+ and either NAD or NADP. Under Vmax conditions the ratios for the rate of NAD/NADP reduction was 1.8. A study of the reductive carboxulation reaction indicated that this enzyme reaction is reversible; at pH 7.0 the reverse reaction exhibited 22% of the activity of forward reaction. Some kinetic characteristics of the enzyme were determined. ATP was found to be a competitive inhibitor with respect to malate. Fumarate reversed ATP inhibition. Regulation of NAD(P)-dependent malic enzyme from herring testicular tissue mitochondria could respond to changing levels of mitochondrial ATP and fumarate in vivo.  相似文献

测定了海捕银鲳亲鱼肌肉,肝脏和性腺组织的氨基酸,并分析了不同性腺发育阶段的氨基酸变化。结果表明,雄性V期亲鱼精巢中的牛磺酸含量最高(27.8mg/g),显著高于肌肉和肝脏组织,而雄性V期亲鱼氨基酸总量在肌肉中最高(631.4 mg/g),其次是精巢(544.8 mg/g)和肝脏(468.0 mg/g)组织。雌性亲鱼肝脏中的牛磺酸含量(13.7 mg/g)显著高于卵巢和肌肉组织。在卵巢从III期发育到V期过程中,雌性亲鱼肌肉和肝脏中的氨基酸总量在V期下降显著,而卵巢氨基酸总量维持稳定。在总氨基酸的组成上,肌肉中含量高的氨基酸为Glu,Lys, Asp,Leu和Arg;肝脏中含量高的为Glu,Lys, Asp,Val和Leu;精巢中含量高为Glu,Asp, Lys, Arg和Val;卵巢含量高中为Glu,Lys,Leu,Val和Asp。性别和性腺发育阶段对亲鱼肌肉总氨基酸组成无影响。游离氨基酸含量在III 、IV和 V期卵巢中的含量分别为17.7 mg/g ,41.5 mg/g和29.6 mg/g,且其组成随性腺发育阶段的不同而有显著差异。  相似文献

A feeding trial was conducted to investigate the influence of dietary protein levels on growth performance, carcass proximate composition and liver lipid classes of juvenile Spinibarbus hollandi (Oshima), a cyprinid fish. White fish meal was the primary protein source in the study. Eight experimental diets containing 13–55% crude protein were fed to three replicate groups of six fish weighing nearly 8.5 g each for 10 weeks. Both percentage weight gain and feed efficiency ratio increased significantly with increasing dietary protein levels up to 31%, but there was no further increase for protein levels from 31% to 55%. Application of broken‐line regression analysis to the percentage weight gain provided an estimate of 32.7±1.5% dietary protein for maximum growth. The protein efficiency ratio and productive lipid value were inversely correlated with dietary protein level. The hepatosomatic index and the viscerosomatic index were also inversely related to dietary protein level. The carcass protein of fish fed lower protein diets was significantly lower than that of the fish fed higher protein diets. Carcass lipid content decreased with increasing dietary protein levels, whereas moisture was inversely related to lipid content. Both liver glycogen and liver lipid contents decreased with increasing dietary protein levels. Triglyceride was the major component in the liver lipid, and the amount of triglyceride deposited in the liver also decreased with dietary protein levels. The results indicated that both carcass proximate composition and liver lipid class of juvenile Spinibarbus hollandi were affected by dietary treatments.  相似文献

为进一步了解氨氮对团头鲂幼鱼的毒性毒理影响,以体质量为(14.27±0.01)g的团头鲂幼鱼为研究对象,研究了氨氮胁迫对其鳃、肝、肾组织结构的影响。实验首先进行96h的氨氮胁迫,得出96h LC50,在此基础上,设置对照组(0.472 mg/L)和实验组(25 mg/L)两个氨氮浓度处理组,进行0、6、12、24、48h的氨氮胁迫,取样后剩余团头鲂幼鱼移入曝气自来水进行96h的毒后恢复实验。结果表明:96h LC50为56.492 mg/L;三种组织观察表明,氨氮胁迫6h,鳃丝毛细血管扩张,上皮组织增生;肝细胞肿胀,细胞核肿大,肝细胞空泡化;肾小球萎缩,肾小囊腔膨大,肾小管管腔缩小;胁迫12h,泌氯细胞增生,呼吸上皮细胞出现部分脱落;肝细胞水样变性、血窦扩张、细胞轮廓模糊,形成点状病灶;肾小管上皮细胞肿大、水样变性、浊肿;胁迫24h,鳃小片融合、变短,呼吸上皮细胞大面积脱落;肝细胞水样变性、血窦扩张严重,形成局部病灶;肾组织淋巴细胞浸润严重,充血,肾小球坏死,肾小管坏死;胁迫48h,鳃小片卷曲,上皮细胞部分脱落;肝细胞部分溶解、血窦扩张,形成点状病灶;肾小管上皮细胞坏死,肾小球坏死;96h恢复后,泌氯细胞和上皮组织增生严重;肝组织大面积细胞核肿大,血窦扩张;肾组织淋巴细胞浸润严重,肾小管坏死,肾小球坏死。实验表明,不同的器官之间病症的损伤程度是不同的,肝组织的损伤最严重,然后依次是鳃和肾。随着胁迫时间延长,鳃、肝和肾组织受到的损害增加,同时鱼体也产生防御反应,但96h的恢复期不足以让团头鲂幼鱼在胁迫中完全恢复,而恢复能力最差的是肾组织。  相似文献

采用HE、Mallory氏三色法、Heidenhain氏Azan染色,PAS反应,组织块银浸染方法,对组织切片染色处理,应用光学显微镜和透射电镜对驼背鲈肝脏的显微和超微结构进行了观察,结果表明驼背鲈肝组织中肝细胞界限不明显,不存在双核现象,有暗细胞和亮细胞之分,胞质中细胞器丰富,有大量的线粒体、内质网和溶酶体,高尔基体很难观察到,仅在胆小管和狄氏间隙的附近发现。此外,在肝血窦的附近观察到成纤维细胞、枯否氏细胞和贮脂细胞。肝组织中胆管数量众多包括肝叶中央胆管、支胆管、小叶间胆管和小叶内胆管,并且在中央静脉附近偶见辐射状排列的肝细胞索。  相似文献

在室外水泥池(4m×1.5m×1m)进行石斑鱼(62.3±0.6g)的饥饿实验,每池放养35尾,设3个重复,实验时间分别为0、1、2、3和4周。随着饥饿时间的延长,体重减轻,肥满度和肝体比显著降低;肌肉水分显著增加而蛋白质含量显著下降(P<0.05),脂肪含量在第1周下降后基本保持稳定。肝脏饱和脂肪酸在饥饿前3周基本保持不变,第4周显著下降。单不饱和脂肪酸在饥饿第1周保持不变,然后呈下降趋势。多不饱和脂肪酸和高不饱和脂肪酸的总和随着饥饿时间的延长,含量显著增加。与肝脏相比,肌肉中脂肪酸变化程度较小。随着饥饿时间的延长,肌肉中140减少而205增加。饥饿使肌肉中DHA/EPA比率呈下降趋势,但肝脏DHA/EPA比率显著上升;肌肉和肝脏中DHA EPA的百分比都随饥饿时间的延长而显著上升。  相似文献

SUMMARY: The aim of this study was to investigate the cause of green liver in red sea bream fed substitute protein diets. Red sea bream Pagrus major was given either of the following diets for 28 weeks: (1) control diet (50% fish meal), (2) low level fish meal diet (15% fish meal), and (3) non-fish meal diet (0% fish meal). The green liver was observed in all groups tested, but the incidence was much higher in the experimental diet groups. The feeding of substitute protein diets reduced plasma triglyceride and cholesterol levels. However, there was no significant difference in plasma hepatic enzyme activities and plasma bile salts concentration among the treatments. Fish fed the substitute protein diets showed low hepatic taurine levels with an appearance of a biliverdin in the liver. Moreover, the proportion of ditaurobilirubin to total biliary bile pigments was significantly lower in fish fed the substitute protein diets. These data indicate that feeding of substitute protein diets did not induce any cholestatic hepatobiliary obstructions and that the low hepatic taurine level was one of the probable factors responsible for the occurrence of green liver in red sea bream fed substitute protein diets.  相似文献
