AIM To investigate the effect of microRNA-92b-5p (miR-92b-5p) on renal injury and inflammatory response in diabetic nephropathy (DN) rats and its mechanism. METHODS The rats were divided into control group, DN group, lentiviral negative control (LV-NC) group, LV-miR-92b group, LV-high mobility group protein B1 (LV-HMGB1) group and miR-92b+HMGB1 group, with 15 rats in each group. After fasting for 12 h, the model rats were intraperitoneally injected with streptozotocin at dose of 60 mg/kg, and the control rats were intraperitoneally injected with an equal volume of citrate buffer. Three days later, the rats in each treatment group were intravenously injected with 100 μL LV-NC, LV-miR-92b and LV-HMGB1 (1×1011 U/L) twice a week for 8 consecutive weeks. Urinary protein, blood glucose, blood urea nitrogen and serum creatinine were detected by an automatic biochemical analyzer. The expression of miR-92b-5p and HMGB1 mRNA was detected by RT-qPCR. The targeting relationship between miR-92b-5p and HMGB1 was verified by dual-luciferase reporter assay. HMGB1 expression in kidney tissue was detected by Western blot. The kidney damage was observed by HE staining. The apoptosis was detected by flow cytometry. The levels of interleukin-6 (IL-6), IL-1β and tumor necrosis factor-α (TNF-α) in renal tissues were detected by ELISA. RESULTS In DN model rats, miR-92b-5p was down-regulated, while HMGB1 was highly expressed. There was a binding site between miR-92b-5p and HMGB1 3'-untranslated region. High expression of miR-92b-5p inhibited the luciferase activity of the wild-type HMGB1 plasmid (P <0.01), but had no effect on the luciferase activity of the mutant HMGB1 plasmid. Compared with DN group, urinary protein, blood glucose, serum creatinine and blood urea nitrogen in LV-miR-92b group were significantly reduced (P <0.01). The degree of hyperplasia, swelling and inflammatory cell infiltration of glomerular mesangium and basement membrane tubules, the apoptosis rate of renal tissues, and the content of IL-6, IL-1β and TNF-α in renal tissues were significantly decreased (P <0.01). Co-transfection of LV-HMGB1 significantly reversed the effect of miR-92b-5p on DN rats. CONCLUSION miR-92b-5p reduces renal injury and inflammatory response in DN rats by targeting HMGB1 and down-regulating its expression. 相似文献

从鸡脾脏中提取总 RNA,以总RNA为模板,5’-ATC TAG AGG TAC CGG ATC C-(T)15-3’为反转录引物,采用TaKaRa AMV反转录酶合成First-strand cDNA (fcDNA)。根据GenBank中登录的鸡的BF2基因胞外区序列,设计了含EcoRⅠ和 HindⅢ 酶切位点的2对引物,分别扩增鸡BF2的胞外区基因。PCR产物经纯化回收后,插入到含LacZ基因的原核克隆载体pGEM-T Easy中,转化至大肠杆菌JM109感受态细胞,经EcoRⅠ和Hind Ⅲ双酶切及PCR鉴定,筛选出阳性克隆。再把所克隆的片段亚克隆到表达载体pMAL-p2X vector,构建重组质粒p2X/BF2(1-5)。将重组表达质粒转化入大肠杆菌TB1感受态细胞,筛选阳性克隆,经IPTG诱导表达后,进行聚丙烯酰胺凝胶电泳(SDS-PAGE)分析。结果表明,本研究克隆了5类鸡BF2的胞外区基因序列,试验结果证明本研究已成功克隆并可溶性表达了5类BF2胞外区蛋白质分子,全长约807~819 bp,编码约269~273个氨基酸。采用淀粉树脂柱亲和层析和DEAE凝胶过滤方法对表达产物进行了纯化,并对纯化的表达产物进行了western blot鉴定。本研究为进一步利用BF2的胞外区在体外构建MHC Ⅰ类分子结合筛选抗原表位肽平台奠定了基础。 相似文献

基于电磁感应仪的河口地区底聚型盐分剖面特征的解译 总被引:3,自引:1,他引:2
针对长江河口地区存在的盐渍化问题,以该地区分布较广泛的底聚型盐分剖面类型为研究对象,通过电磁感应仪EM38测量与田间采样,建立了磁感式表观电导率和土壤电导率之间的多元回归模型。在分析土壤盐分剖面分布特征的基础上提出了Logistic模型,并运用该模型对盐分剖面进行了参数拟合和验证。结果表明:磁感式表观电导率水平读数和垂直读数呈极显著线性相关,回归模型和Logistic模型均具有很好的预测效果。通过模型精度检验,二者的预测精度差异不显著,但Logistic模型所需参数少,表明该文提出的Logistic模型不仅能降低待估参数数量,同时还具有较高的预测精度。该研究结果为利用电磁感应仪快速、精确地进行土壤盐分预测及土壤次生盐渍化的防控提供了一定的理论参考。 相似文献

在干旱半干旱地区,土壤盐渍化是常见的土地退化问题之一。本研究选取于田县克里雅河上游边缘典型盐渍化区域作为研究靶区,通过EM38大地电导率仪实测土壤表观电导率,提取不同系数下的土壤调节植被指数(SAVI),分析了SAVI指数与土壤电导率间的相关性,并利用同时期WorldView-2影像的敏感波段建立了基于高分辨率影像数据的土壤盐渍化偏最小二乘回归(PLSR)模型并进行了精度验证。结果表明:①从遥感影像提取SAVI指数时,在系数(L)调节范围内选取固定系数值,系数值(间隔为0.1)从0.1变化到1.0的过程中,相应提取的SAVI指数与土壤电导率的相关性明显提升,相关性系数(r)从0.30提高到0.50,并通过显著性检验(P0.01)。②选取的SAVI1.0、B6、B7、B8四种变量中,以SAVI1.0+B6+B8为变量组合所建立的PLSR模型为最优,该模型较其他变量组合建模的决定系数(R2p)提高了0.11,因此,在研究区该模型具有更好的预测能力,模型精度为RMSEC=0.77dS/m、RC2=0.68、RMSEP=0.79 dS/m、RP2=0.66、RPD=2.2。 相似文献

Over the past five decades, increased pressure caused by the rapidly growing population has resulted in a reclamation of agricultural and urban buffer zones along China's coastline. However, information about the spatio–temporal variation of soil salinity in these reclaimed regions is limited. As such, obtaining this information is crucial for mapping the variation in saline areas and to identify suitable salinity management strategies. In this study, we employed EM38 data to conduct digital soil mapping of spatio–temporal variation and map these variations of different site-specific zones. The results indicated that the distribution of soil salinity was heterogeneous in the middle of, and that the leaching of salts was significant at the edges of, the study field. Afterwards, fuzzy-k means algorithm was used to divide the site-specific management zones within the time series apparent soil electrical conductivity(ECa) data and the spatial correlations of variation. We concluded that two management zones are optimal to guide precision management. Zone A had an average salinity level of about 165 mS m–1, in which salt-tolerant crops, such as cotton and barley can grow normally, while crops such as soybean and cowpeas may be planted using leaching and increasing the mulching film methods to reduce the accumulation of salt in surface soil. In Zone B, there was a low salinity level with a mean of 89 mS m–1 for ECa, which allows for rice, wheat, and a wide range of vegetables to be grown normally. In such situations, measures such as an optimized combination of irrigation and drainage, as well as soil amendment can be taken to adjust and control the salt content. Particularly, flattening the land with a large-scale machine was used to improve the ability of micro-topography to influence salt migration; rice and other dry, land crops were planted in rotation in combination with utilizing salt-leaching multiple times to speed up desalinization. 相似文献

以鲫鱼(Carassius auratus)为试验材料,研究了经不同浓度的2,2’,4,4’-四溴联苯醚(PBDE-47)和十溴联苯醚(PBDE-209)暴露后,鲫鱼体内DNA损伤产物8-羟基脱氧鸟苷(8-OHdG)的含量及p53蛋白的水平。结果表明,采用浓度为0.10~5.00mg·L-1的PBDE-47和10.0~50.0mg·L-1的PBDE-209处理鲫鱼20d,除了0.10mg·L-1PBDE-47和10.0mg·L-1PBDE-209浓度组的8-OHdG和p53蛋白含量与对照组相比无显著差异(P>0.05),其余各浓度组的8-OHdG和p53蛋白含量均随PBDE-47和PBDE-209浓度增加而逐渐上升,呈显著的相关关系(P<0.01),且8-OHdG和p53蛋白之间也呈显著的正相关。说明PBDE-47和PBDE-209对鲫鱼组织的DNA产生了损伤,具有遗传毒性影响。 相似文献

为明确青海春小麦品种‘青春38’成株期抗条锈性的遗传基础,以‘青春38’为父本与感病春小麦品种‘Taichung 29’(T29)杂交构建F2∶3代分离群体。在青海西宁和互助两地田间病圃进行了抗条锈性鉴定,应用植物数量性状主基因+多基因混合遗传模型单个分离世代分析方法,解析‘青春38’的抗条锈性遗传特点。结果表明,‘青春38’/‘T29’F2∶3群体单株的病害严重度和反应型在两个试验点均未呈现连续性分布,也不符合正态分布,初步推测‘青春38’对小麦条锈病的成株期抗性具有质量性状特征;以严重度或反应型数据进行遗传分析,‘青春38’在两个试验点对小麦条锈病的成株期抗性表现的最优遗传模型均属2对主基因遗传,只是主基因的作用方式(C-1:2MG-ADI加性-显性-上位性,C-4:2MG-EA等加性,C-6:2MG-EEAD等显性)有所不同。 相似文献
