为研究、防控广东地区猪流行性腹泻,首先通过RT-PCR法确定某猪场粪便样品中存在猪流行性腹泻病毒,随后利用Vero细胞进行盲传,最后经核酸类型鉴定、RT-PCR、病毒TCID50的测定、目的基因测序和序列分析等方法,确认分离到2株猪流行性腹泻病毒毒株,命名为PEDV GDKP-2013和PEDV GDKP2-2013,通过ORF3全基因核苷酸序列分析可知,GDKP-2013株与GDKP2-2013株与经典参考毒株的氨基酸序列一致性为 JX261936-CHGD-01,98.2%,98%;JQ039903-CH-GD-2011,98.6%,98.4%;AF353511-CV777,95.4%,95.1%;KC344845-STPED0810,95.1%,94.8%;EU054929-DR13,95.5%,95.2%;KF272920-USA-Colorado-2013,95.1%,94.8%,表明此2毒株与国内分离株的氨基酸同源性高,与韩国、泰国和美国分离株的氨基酸同源性要低一些,同时,与疫苗参考株的氨基酸同源性也相对较低;基因系统进化分析可知,GDKP-2013株与GDKP2-2013株主要处在Ⅰa分支,均与弱毒疫苗株(Ⅰb分支)相互之间遗传进化关系较远。  相似文献

为了探讨归参汤(GST)对免疫抑制小鼠血清中IL-2、TNF-α和脾细胞Th1/Th2细胞因子的影响。本试验采用环磷酰胺制造免疫抑制模型,利用双抗夹心ELISA法,观察归参汤对小鼠血清中IL-2、TNF-α的影响;RT-PCR法观察归参汤对小鼠脾细胞中INF-γ、IL-4、T-bet和GATA-3的mRNA表达的影响。结果显示:归参汤高剂量组可以显著增加免疫抑制小鼠血清中IL-2、TNF-α的含量(P<0.01),提高INF-γ的表达(P<0.05),降低IL-4的含量(P<0.05),升高T-bet(P<0.05),降低GATA-3的表达(P<0.05)。归参汤的免疫增强作用与升高血清中IL-2、TNF-α有关,且升高INF-γ/IL-4纠正免疫抑制小鼠Thl/Th2细胞因子的比例紊乱,其机制可能部分归因于对T-bet和GATA-3的mRNA表达的影响。  相似文献

为了减少普通地膜和氮肥的投入,实现该地区农业生态良性循环和土壤的可持续利用,在甘肃武威试验站采用完全随机裂区设计,研究了不同种植模式(玉米单作、玉米//毛苕子和玉米//箭筈豌豆)和覆盖方式(普通地膜覆盖、生物可降解地膜覆盖、秸秆覆盖和无覆盖)对玉米和绿肥作物氮素吸收以及土壤硝态氮累积的影响。结果表明:覆盖和间作对作物的氮素吸收利用具有显著促进作用。玉米收获期,与无覆盖相比,普通地膜覆盖、生物可降解地膜覆盖和秸秆覆盖的玉米植株吸氮量分别增加21.46%,11.22%和34.63%,与玉米单作相比,玉米//毛苕子和玉米//箭筈豌豆的玉米吸氮量分别增加6.94%和6.26%。玉米收获期,秸秆覆盖的牧草植株吸氮量较普通地膜覆盖、生物可降解地膜覆盖和无覆盖分别增加46.75%,72.31%和21.76%,间作毛苕子的吸氮量较间作箭筈豌豆增加25.26%。玉米收获期,各处理玉米带、牧草(裸露)带0~140cm土壤硝态氮累积量均低于牧草收获期,无论牧草收获期还是玉米收获期,单作玉米的玉米带、裸露带土壤硝态氮累积量均低于间作玉米。玉米收获期,普通地膜覆盖和生物可降解地膜覆盖的玉米带0~140cm土壤硝态氮累积量较无覆盖分别显著增加32.87%和27.82%,普通地膜覆盖的牧草(裸露)带0~140cm土壤硝态氮累积量较生物可降解地膜覆盖、秸秆覆盖和无覆盖分别增加9.17%、6.07%和35.45%。  相似文献

AIM:To analyze the difference of long-chain acyl-CoA synthetase 3 (ACSL3) expression between normal prostate epithelial cells and prostate cancer cells.METHODS:ACLS3 mRNA expression between normal prostate epithelial cells and prostate cancer cells was compared using RT-PCR. Meanwhile, ACSL3 gene was amplified from prostate cancer cells, and the eukaryotic expression plasmid pCDNA3.1(+)-Flag-ACLS3 and lentivirus Lenti-ACLS3 were constructed. After transfection of ACSL3-plasmid and lentivirus into the prostate cancer cells, ACSL3 expressive was detected by RT-PCR and Western blotting, and then Matrigel invasion assay was performed to investigate the alteration of the invasive ability of the prostate cancer cells with over-expression of ACSL3. RESULTS:A significant difference of ACSL3 mRNA level between normal prostate epithelial cells and prostate cancer cells was observed. ACSL3 was highly expressed in localized prostate cancer cells compared to metastatic prostate cancer cells, while ACSL3 expression was higher in androgen-dependent prostate cancer cells than that in androgen-independent prostate cancer cells. Furthermore, the eukaryotic expression plasmid and lentivirus containing ACLS3 gene were successfully constructed. The prostate cancer cell line which stably over-expressed ACLS3 was established. Up-regulation of ACSL3 inhibited the invasive ability of prostate cancer cells. CONCLUSION:ACSL3 plays an antagonistic role in invasiveness of prostate cancer.  相似文献

AIM:To investigate the effects of siRNA targeting integrin-linked kinase (ILK) on the expression of glycogen synthase kinase 3β (GSK-3β) and β-catenin during epithelial-mesenchymal transition (EMT) in human kidney proximal tubular epithelial cell line HKC induced by high glucose. METHODS:HKC cells were divided into 4 groups:normal glucose (NG) group, high glucose (HG) group, HG+HK (a vector containing the non-specific siRNA designed as negative control) group and HG+ILK siRNA group. The inverted fluorescence microscope was used to examine the expression of green fluorescent protein (GFP). The expression of ILK at mRNA and protein levels was detected by RT-PCR and Western blotting. The expression of p-GSK-3β and β-catenin was observed by immunocytochemical staining. The protein expression of total GSK-3β, p-GSK-3β, nuclear β-catenin, total β-catenin, E-cadherin and α-smooth muscle actin (α-SMA) was measured by Western blotting. RESULTS:GFP was observed in HKC cells, indicating that the transfection was successful. Both the protein and mRNA of ILK were down-regulated in HG+ILK siRNA group compared with HG group and HG+HK group, but still higher than those in NG group. Silencing of ILK down-regulated the expression of p-GSK-3β and nuclear β-catenin. No difference of total GSK-3β or total β-catenin was observed among the 4 groups. CONCLUSION:These data support a functional role of ILK, GSK-3β and β-catenin in tubular EMT induced by high glucose. ILK may promote tubular EMT by regulating the activity of GSK-3β and β-catenin, the downstream effectors of the Wnt/β-catenin pathway.  相似文献

AIM:To explore the effect of L-carnitine on nuclear factor of activated T-cells,cytoplasmic 3 (NFATc3) in cardiomyocytes under H2O2 stimulation. METHODS:Primary cultured neonatal rat myocardial cells were stimulated by H2O2 at concentration of 200 μmol/L for 12 h to induce oxidative stress injury. In treatment group, L-carnitine and cyclosporin A (CsA), a specific inhibitor of calcineurin (CaN), were administered 30 min prior to H2O2 stimulation. After treatment, total, cytoplasmic and nuclear NFATc3 protein levels were determined by Western blotting. The method of immunofluoresence was used to evaluate the distribution of NFATc3. RESULTS:H2O2 treatment produced no effect on the expression of total NFATc3, but caused its translocation from the cytosolic to nuclear compartment, which was greatly blunted by L-carnitine pretreatment. CONCLUSION: L-carnitine antagonized oxidative stress injury via alleviating NFATc3 nuclear translocation.  相似文献

AIM:To investigate the relationship of microRNA-7 (miRNA-7) over-expression and epidermal growth factor receptor (EGFR)/phosphatidylinositol kinase-3 (PI3K)/protein kinase B (PKB, also called Akt) pathway in human nasopharyngeal carcinoma 5-8F cells. METHODS:The 5-8F cells were transfected with miRNA-7 mimics (carrying by Lipofectamine 2000). The expression of miRNA-7 was detected by real-time PCR. The cell proliferation was analyzed by CCK-8 assay. The cell colony-forming capability was determined by cell colony formation test. The expression of EGFR/PI3K/Akt at mRNA and protein levels was examined by real-time PCR and Western blotting. RESULTS:The expression level of miRNA-7 was significantly increased in 5-8F cells compared with negative control (NC) group and control group (P<0.01). The proliferation of NPC 5-8F cells was decreased extremely after tansfected with the miRNA-7 mimics (P<0.01), so did the result of the cell colony-formation test. The expression of EGFR/PI3K/Akt at mRNA and protein levels was significantly down-regulated compared with NC group and control group (P<0.01). CONCLUSION: Over-expression of miRNA-7 significantly inhibits the proliferation and colony-forming ability of nasopharyngeal carcinoma 5-8F cells by down-regulation of EGFR/PI3K/Akt pathway.  相似文献
