Reason for performing study: Current therapy protocols to treat persistent post mating endometritis and retained fetal membranes in mares typically include the administration of ecbolic drugs. Evaluation of the pharmacokinetics and tolerability of carbetocin, a long‐acting oxytocin analogue, after i.v. administration is required. Objectives: To determine the pharmacokinetic parameters (principally half‐life) of carbetocin in horses. Methods: Five mature mares and one gelding received 0.175 mg carbetocin i.v. All animals were monitored periodically throughout the study for elevation in rectal temperature, heart rate, respiratory rate and signs of pain or discomfort. Plasma samples were collected for determination of carbetocin concentrations by radioimmunoassay. Results: Administration of carbetocin was well tolerated by all horses and its half‐life was 17.2 min. Conclusions: The half‐life of carbetocin is greater than that previously reported for oxytocin (6.8 min). Potential relevance: Carbetocin is an attractive alternative to oxytocin therapy in broodmare management.  相似文献

通过对泥鳅进行生殖激素的注射,研究分别注射25、30、35IU/g剂量的hCG、分别注射10、12、14μg/尾剂量的LHRH-A2、分别注射16IU/g+10μg/尾、18IU/g+ 10μg/尾、20IU/g+ 10μg/尾剂量的hCG+LHRH-A2对泥鳅催产效果的影响.结果表明:单独使用hCG注射量以30IU/g效果最好.单独使用LHRH-A2其注射量以12μg/尾效果最好.复合激素hCG+LHRH-A2其注射量以18IU/g+10μg/尾效果最好.综合考虑抗药性以及价格因素后,实际生产中建议使用12μg/尾LHRH-A2为宜.  相似文献

试验旨在评价39.1℃恒温下绵羊催产素在渗透泵(ALZET®)中的稳定性。试验分为3个组,分别为渗透泵组、微量离心管组和对照组。除对照组始终冻存于-80℃以外,另外两组置于39.1℃培养箱中进行孵育,分不同时间点采集样品。所有样品经超高效液相色谱串联四级杆/飞行时间质谱(ultra-performance liquid chromatography tandem quadrupole/flight time mass spectrometry,UPLC-Q-TOF MS)方法获得氧化产物提取离子流色谱图,通过比较各产物的峰面积,分析催产素在39.1℃缓释泵中的氧化过程。结果表明,催产素在39.1℃缓释泵及微量离心管中均表现出氧化程度随时间逐渐增加的趋势,两试验组之间差异不显著(P>0.05)。渗透泵组和微量离心管组不同时间点采集样品中催产素原型占对照组原型的百分比分别为83.6%~117.8%和83.2%~116.2%,说明在渗透泵植入绵羊体内试验中需要考虑因催产素氧化引起的含量变化对试验结果的影响。  相似文献

为了揭示不同生理期催产素(Oxytocin,OT)在下丘脑-垂体-性腺轴中的分布特点,采用链霉素抗生物素蛋白-过氧化物酶(Streptaridin-peroxidase,SP)法研究了OT免疫反应产物在乏情期奶山羊下丘脑中的分布。结果显示:OT样神经元存在于视上核、室旁核弓状核、室周核、穹隆周核、下丘脑前核、下丘脑后核、腹外侧核、背内侧核、下丘脑外侧区以及乳头体各核团等27个核区,分布区域呈前端尖、中部宽、后钝圆的梭形带。此外,在室旁核观察到大量的强阳性神经纤维,第三脑室附近的纤维伸向脑室内。说明OT在下丘脑广泛存在,且下丘脑OT经神经垂体、第三脑室、血管3条途径释放,经血液循环最终作用于靶细胞而发挥生理作用。  相似文献

Overdose and death of both target and non-target species are not uncommon when α–chloralose is used for animal control. Alpha–chloralose appears to depress the central nervous system by having an affect on the GABA-A receptor in a manner similar to ethanol. Recently central administration of oxytocin was shown to block ethanol-induced impairment in rodents. Our study investigates whether IM administration of oxytocin speeds avian recovery from α–chloralose and reduces mortality under simulated field conditions. We found that when injected twice with oxytocin (30 uL/kg) chickens entered α–chloralose narcosis more quickly and showed greater behavioral impairment. When fed the LD50 dose of α–chloralose, repeated injection with oxytocin produced no significant difference in mortality rate or time to death in comparison to the control group. The effects of peripheral oxytocin on female chicks were different than results found previously in cockerels given oxytocin centrally. Oxytocin is not a suitable antidote for α–chloralose overdose during animal capture in the field but instead accentuates α–chloralose narcosis, making capture more likely and efficient.  相似文献

采用免疫组化链霉素抗生物素蛋白-过氧化物酶法(SP法),检测羔羊期和性成熟期雌性奶山羊下丘脑中催产素表达的变化规律.结果显示,催产素免疫反应阳性神经元在羔羊和性成熟羊的视上核、室旁核、乳头体内侧核等核团均有分布,在羔羊的室周核、视前内侧核等核团广泛分布,但在性成熟羊的这些核团却未见分布.在视上核、室旁核、乳头体内侧核等核团的催产素免疫阳性神经元数量,性成熟羊显著高于羔羊(P<0.01);这些核团中催产素免疫阳性产物的平均灰度值,羔羊显著低于性成熟羊(P<0.01),在其他核团则羔羊高于性成熟羊,且大部分差异明显(P<0.01).结果提示,性成熟羊下丘脑中除室周核、视前内侧核等核团外,室旁核、视上核、乳头体内侧核、乳头体后核等核团,催产素的表达高于羔羊,说明这些核团对生殖系统的发育成熟可能有重要作用.  相似文献

用免疫组化ABC法,对发情周期中奶山羊下丘脑-垂体-卵巢轴催产素(OT)分布进行了观察研究.结果表明,下丘脑中分泌OT的神经元主要分布在室旁核和视上核,在穹窿周核、腹内侧核、腹外侧核、交叉上核、背内侧核、乳头体、下丘脑外侧区、下丘脑前核等核团也有一定数量的阳性神经元;阳性神经纤维仅见于室旁核、下丘脑前核、视上核等少数核团,在正中隆起和第3脑室室周可见到一定数量的阳性神经纤维.在垂体前叶未见到OT免疫反应阳性产物,自垂体柄和正中隆起的一侧可见到平行排列的OT阳性神经纤维断续地延伸至神经部.卵巢的卵泡及间质未见OT免疫阳性反应,,在黄体组织中存在数量较多的免疫反应阳性细胞,阳性细胞主要呈圆形、卵圆形,小梁两侧及黄体中央近腔区域的阳性细胞呈长梭形,有相当数量的阳性细胞具有突起.连续切片HE染色对照观察显示,黄体中OT主要由大黄体细胞产生,但小黄体细胞也存在OT免疫阳性反应.  相似文献

Factor XI (F XI) deficiency is an autosomal recessive coagulopathy found in Holstein cattle. Affected animals have a 50% greater prevalence of repeat breeding. Therefore, several parameters describing ovarian function were studied. Daily blood sampling revealed that progesterone concentrations were slower to decline from a peak at day 16 (p<0.01) to values less than 3 nmol/L in F XI-deficient cows (5.14±0.69 days (mean ± SD) versus 4.05±0.63 days in control animals), resulting in an oestrous cycle length of 24.7±2.1 days compared to 22.9±3.0 days, respectively. This was not due to an alteration in the availability of prostaglandin F2 (PGF2) or oxytocin (OT) involved in luteolysis. No significant differences (p>0.05) were seen between normal (n=7) and F XI-deficient (n=7) cows in the peak values or the area under the curve for the pulse in 13,14-dihydro-15-keto PGF2 in response to OT challenge or in the parameters describing the pulse of ovarian OT secretion after PGF2 injection (n=7 for each) between days 12 and 14. Ovulatory follicular development was assessed by ultrasound monitoring and plasma 17-oestradiol values at 8-h intervals after a luteolytic injection of cloprostenol (n=6 for each). Follicular diameter was smaller (p<0.05) and accompanied by lower peak oestradiol values near the time of ovulation in F XI-deficient cows. This study examined the feasibility of transcervical embryo recovery after the hormonal treatment to induce cervical dilation, following the 7‐day oestrous synchronization protocol in multiparous Santa Inês ewes. A total of 23 cyclic ewes received two doses of 37.5 μg of d‐cloprostenol by latero‐vulvar route 7 days apart. After the second injection of d‐cloprostenol, the ewes were checked for oestrus (every 12 hr) and then mated by fertile rams throughout the oestrous period. All ewes received 37.5 μg of d‐cloprostenol (latero‐vulvar) and 1 mg of oestradiol benzoate by either intramuscular (EBim group; n = 12) or intravaginal (EBivg group; n = 11) route 16 hr before embryo flushing. Twenty minutes before the flushing, 50 IU of oxytocin were administered intravenously. The oestrous response (i.e., the percentage of ewes that showed signs of oestrous behaviour after the second d‐cloprostenol injection) was 91.3% (21/23). The proportion of successfully penetrated ewes (81.8% compared with 80.0%), the mean duration of embryo flushing (24.7 ± 2.0 min compared 26.2 ± 1.9 min), the flushing fluid recovery rate (94.8 ± 1.3% compared with 91.0 ± 2.9%) and the average number of structures recovered per ewe (0.5 ± 0.4 compared with 0.8 ± 0.4) did not vary (p > 0.05) between the EBim and EBivg groups. Viable embryos were recovered from 41.2% (7/17) of successfully penetrated ewes. It can be concluded that nonsurgical (i.e., transcervical) embryo collection can be performed in oestrous‐synchronized Santa Inês ewes pretreated with d‐cloprostenol, oxytocin and oestradiol benzoate, with the latter hormone administered by either the intramuscular or intravaginal route.  相似文献

Oxytocin receptor (OTR) is a member of G-protein coupled receptor, which has been found in many tumors and cancer cell lines. Many studies revealed oxytocin (OT) may inhibit the tumor and cancer cell growth and proliferation, but the mechanism of this inhibition is not wel known. Some experiments indicated cAMP-PKA system participates in the signal transduction that oxytocin inhibits the cancer cell proliferation. However, other experiments showed the signal transduction for oxytocin in the Hs578T carcinosacoma cel is the same as that in the normal cells.
