Contents: Oxytocin, progesterone and testosterone production of porcine granulosa cell culture in absence - or in presence of 0.01–10 μg/ml LH-RH or its antagonist (D Phe2, D Phe6) LH-RH were analysed. It was found that both LH-RH and its analoque stimulated granulosa oxytocin and progesterone (but not testosterone) secretion in a dose-dependent manner. Present results demonstrate (I) a direct stimulatory influence of LH-RH and its antagonist on porcine ovarian peptide and steroid hormone production, as well as (2) the relative independence of the LH-RH -related peptides actions at the hypophysial and at the ovarian level .

Oxytocin, Progesteron und Testosteronproduktion porciner Granulosazellkulturen wurde in Abwesenheit oder Anwesenheit von 0,01–10 μg / ml LH-RH bzw. des Antagonisten (d-Phe2, d Phe6) LH-RH analysiert. Es wurde festgestellt, daβ sowohl LH-RH als auch ein Analogpräparat die Oxytocin und Progesteronsekretion in dosisabhängiger Form stimulieren, Testosteron aber nicht beeinfluβt wird. Die Ergebnisse Zeigen einen stimulierenden Effekt von LH-RH und auch des Antagonisten auf die Synthese ovarieller Peptide und Steroidhormone. Gleichzeitig stellt sich eine relative Unabhängigkeit von LH-RHgebunderen Peptiden auf hypophysärem und ovariellen Level dar .  相似文献

We examined individual differences in serum oxytocin concentrations (OT) of calves, and assessed whether these differences were correlated with their dams’ milk and serum OT. Eight Holstein (H), nine Japanese Shorthorn (JS), and six Japanese Black (JB) calves were examined. Blood was collected three times during the first month in H calves, while their dams’ blood was collected three times prior to parturition. Milk was collected twice after parturition from H cows. Blood from JS and JB calves were collected at 1 and 4 months old, while of their dams only once before parturition. Serum OT in H calves at 7 days old was significantly correlated with that at 30 days. Serum OT of JS calves at 1 month old was significantly correlated to that at 4 months, while of JB calves was also positively correlated (r = 0.70). Serum OT of calves showed significant individual differences in each breed. Serum OT of calves was not correlated with the milk OT of dams, except for 3‐day‐old calves that fed on their dams’ milk. We concluded that although serum OT differed among individuals, this difference was stable within each individual and not affected by the serum OT of the dams.  相似文献

This study was conducted to investigate the arginine‐vasopressin (AVP)‐ and oxytocin‐induced changes in plasma adrenocorticotropic hormone (ACTH), growth hormone (GH), insulin and glucagon levels and their metabolite concentrations in goats. In this study, five goats were intravenously injected with either AVP (0.3 nmol/kg body weight (BW)) or oxytocin (0.7 IU/kg BW). AVP injection significantly increased ACTH and GH secretions compared to controls, although insulin and glucagon concentrations were not altered. The incremental areas (ICAs) of the ACTH and GH concentrations were higher in the AVP group than in the saline group. Oxytocin injections increased insulin and glucagon secretions, while ACTH level was not altered. GH levels became elevated 30 min after the oxytocin injection. The ICAs of insulin and glucagon after oxytocin was injected were higher than those of the control. Results indicate that AVP is a potent stimulant of ACTH and GH secretions, while oxytocin uses different pathways to regulate insulin and glucagon secretions in goats.  相似文献

Abstract

AIM: To compare the efficacy of oxytocin given once daily, either I/V or I/M, on Days 7–14 post-ovulation, on the expression of oestrus in mares through to 65 days post-ovulation.

METHODS: Eighteen mares of various breeds that were displaying normal oestrous cycles were randomly assigned to one of three treatment groups on the day of ovulation (Day 0), detected using transrectal ultrasonography. Mares in the control group (n = 6) were given 1 mL saline I/V; mares in the I/V and I/M groups (n = 6 per group) were injected with 10 IU oxytocin I/V and I/M, respectively. All treatments were given once daily on Days 7–14. Mares were teased by a stallion three times per week, up to 65 days post-ovulation, to detect oestrous or dioestrous behaviour. Ovarian follicular and luteal activity were monitored using transrectal ultrasonography three times weekly, and daily when a follicle >30 mm diameter was present until ovulation. Blood samples were collected weekly for analysis of concentrations of progesterone in serum. Prolonged dioestrus was defined as a period of >30 days of dioestrous behaviour after Day 0, confirmed by detection of corpora lutea and concentrations of progesterone in serum >4 nmol/L.

RESULTS: Overall, 8/18 (44%) mares showed prolonged dioestrus. These included 2/6 (33%) mares in the control group, compared with 5/6 (83%) and 1/6 (16%) mares in the I/V and I/M groups, respectively (p = 0.11). The median duration of the first dioestrus was longer for the I/V group (64 (min 16, max 66) days) compared with the control group (18 (min 12, max 64) days) (p = 0.05), but was not different between the control group and the I/M group (16 (min 13, max 65) days) (p = 0.57). For all mares there was strong agreement between teasing behaviours, ultrasonographic assessment of ovarian activity, and concentration of progesterone in serum.

CONCLUSIONS AND CLINICAL RELEVANCE: This study found that low doses of oxytocin did not increase the proportion of mares with prolonged dioestrus, compared with controls, although I/V oxytocin did increase the median duration of dioestrus. The results must be interpreted with some caution as group numbers were small, and a variety of breeds were used. Further investigation of oxytocin given I/V may be warranted as a potential method of oestrus suppression in mares exhibiting oestrous cycles that is low cost, safe and well-tolerated, and potentially reversible with prostaglandin.  相似文献

The present study was conducted to determine the difference in plasma prostaglandin F2α metabolite concentrations following oxytocin (OT) challenge between pregnant and non‐pregnant cows. Experiment 1: cows were subjected to the OT challenge test on days 12, 14 or 16 (day of estrus = day 0) with or without prior insemination and plasma 13,14‐dihydro‐15‐keto prostaglandin F2α (PGFM) concentrations were measured from ?30 to 180 min after OT injection. On day 16, the increment of plasma PGFM concentrations in response to OT injection was significantly smaller in pregnant than that in cyclic cows. On days 12 and 14, there was little OT‐induced PGFM secretion and no difference in PGFM increase between the pregnant and cyclic cows. Experiment 2: cows were inseminated on day 0 and subjected to the OT challenge test on day 16. Cows were classified into non‐pregnant/early embryonic death (NP/EED), late embryonic death (LED) and pregnant (PREG) groups. The increment of PGFM concentrations in response to OT injection was less in both PREG and LED groups than that in the NP/EED group. In conclusion, plasma PGFM secretion induced by OT is suggested as the base of pregnancy diagnosis prior to returning estrus in cows.  相似文献

The objectives of this study were to (1) compare the effect of twice versus once daily administration of oxytocin on days 7-14 after ovulation on the duration of corpus luteum (CL) function and (2) determine the effect of oxytocin treatment on endometrial oxytocin receptor concentration in mares. In experiment 1, mares were randomly assigned to three groups on day 7: (1) untreated control group (n = 7), (2) twice daily oxytocin treatment group (n = 7), and (3) once daily oxytocin treatment group (n = 8). Oxytocin-treated mares received 60 U of oxytocin intramuscularly (IM) the respective number of times each day on days 7 through 14. One of seven control mares (14%), five of seven (71%) twice daily oxytocin-treated mares, and five of eight (63%) once daily oxytocin-treated mares had prolonged CL function. There was no significant difference in the proportion of mares with prolonged CL function between the two oxytocin-treated groups, and collectively, oxytocin treatment increased (P < .05) the proportion of mares with prolonged CL function compared with no treatment. In experiment 2, mares were randomly assigned to two groups (n = 5/group): (1) saline-treated control mares, and (2) oxytocin-treated mares. Beginning on day 7, control mares received 3 mL of sterile saline IM twice daily, and oxytocin-treated mares received 60 U of oxytocin IM twice daily through day 14. On day 15, endometrial oxytocin-binding capacity was determined (as a measure of oxytocin receptor concentration), and there was no difference (P > .1) between control and oxytocin-treated mares (1,465.7 ± 108 and 1,382.8 ± 108 fmol/mg protein [mean ± standard error of mean], respectively).  相似文献

A major challenge in Clariid catfish seed production is our inability to induce voluntary captive spawning of the brooders. Since voluntary captive spawning had been previously elicited in a Clarias species by simulating natural breeding environment, we speculated that the major obstacle in voluntary spawning might be the absence of an inducing agent that triggers mating response in the brooders. To address the issue, oxytocin was injected to the brooders along with Ovatide (a commercial inducing agent comprising salmon gonadotropin‐releasing hormone analogue and domperidone). Administration of 40 mIU/kg body weight of oxytocin after 12 hr of Ovatide injection resulted in voluntary spawning of the fish (81.5%, i.e. 31 out of 38 trials), with more than 70% hatching rate in simple and low‐cost spawning facilities. Histology of male gonads indicated sustained gamete maturation and the gonads were observed to be visibly turgid in the fish that received both the hormones. In contrast, gamete maturation slackened after 10 hr in the control fish, injected with Ovatide only. Administration of oxytocin along with Ovatide was also observed to induce aggressive sexual behaviour among the males. Neither of the hormones, acting alone, could induce voluntary spawning in the fish or influence sexual behaviour. Our results clearly show that oxytocin plays a significant role in sustaining the male gamete maturation induced by Ovatide and trigger the behavioural mating response, leading to voluntary spawning in C. magur. The results hold potential to make a paradigm shift in the commercial catfish seed production technology.  相似文献

We examined the associations between natural individual variations in basal oxytocin (OXT) in postnatal cattle and social behavioral traits. At 1, 2 and 6 weeks of age, the basal OXT exhibited individual variability in 20 Holstein heifer calves. Cluster analysis of mean OXT for these time periods obtained two subgroups: high OXT (HOXT; n = 9) and low OXT (LOXT; n = 11). Social behaviors were observed for 2 days at week 6 after introduction into a four‐peer group, and at 10–14 months of age (10 months) immediately and 1 week, 1 month and 5 months after introduction into 11–15 heifers. At week 6, the main effect of the OXT groups was not significant for all social behaviors. At 10 months, there tended to be interactions between the OXT groups and time periods with respect to the frequency of escape behaviors. LOXT heifers exhibited more escape behaviors than HOXT heifers on the first day of the second sociality tests. At 10 months, HOXT heifers exhibited both attacking and affiliative behavior for peers more than LOXT heifers during 5 months after the second social introduction. This suggests that postnatal OXT concentrations may have long‐lasting effects on individual differences among social behavioral traits in cattle.  相似文献
