Three groups of eight Friesian calves, reared parasite-free, were experimentally infected with 1000 infective larvae of Dictyocaulus viviparus. Two groups were injected subcutaneously with 1% doramectin at 0.2 mg/kg body weight, one group 5 days after infection and the other 25 days after infection. A third group served as untreated controls. Faecal samples were examined for lungworm larvae on days 28, 32, 33, 34 and 35 after infection; the calves were killed and necropsied 39 or 40 days after infection and any lungworms present recovered and counted. Doramectin proved 100% effective against both 5-day-old and mature D. viviparus infections. 相似文献
1. The perfusion of livers with the vital dye trypan blue was performed to test for evidence of tissue hypoxia in 3 groups of young broiler chickens, namely, ascitic, hypoxia‐induced and controls.
2. Hepatocytes that stained with trypan blue were considered to be dead or dying before fixation and represented damaged cells.
3. The proportion of trypan blue‐stained hepatocytes in the livers of ascitic birds was slightly less than half that observed in the hypoxia‐induced birds but significandy more than the proportion of stained cells observed in control birds.
4. Liver damage in the ascitic birds was also assessed biochemically by an altered enzyme profile.
5. The study demonstrated that increased trypan blue uptake in the livers of ascitic birds reared at sea‐level may be the consequence of hypoxia stress caused by reduced oxygen utilisation. 相似文献
To produce large quantities of primary cell suspensions on efficient technique is described which is named "Biomix-Method". By this technique first tissue mince is performed automatically using tissue amounts between 300 g and 500 g. Trypsinization takes place in a three-litre trypsinization vessel using a stirring mechanism according to a defined trypsinization mode. In regard to optimal cell seeding concentration and a justifiable productivity one kidney of newborn or suckling calf yields 6--9 1 cell suspension and that of piglet 1,8-2,3 1, respectively. omnipresence of multiple viroid species among citrus and other Middle Eastern and Mediterranean fruit trees and grapevines.
However, neither means could explain viroid infections of wild trees — scattered on terrains inaccessible to humans — nor
the finding of similar viroids among graft-incompatible plants. Northern hybridization of RNA extracts made of scrapings from
the surfaces of goat (Capra hircus) horns that were rubbed against etrog (Citrus medica) stems infected with a citrus viroids complex, revealed accumulation of considerable amounts ofCitrus exocortis viroids (CEVd) andHop stunt viroids (HSVd). Experimental transmission of both CEVd and HSVd was obtained by rubbing healthy citrus plants with goat horns that
had been rubbed 24 h earlier on infected etrog stems. These results implicate goats as possible vectors of viroids. Transmissionvia goats could have facilitated the long-range spread of viroids among cultivated and wild plants andvice versa and also among graft-incompatible plants. 相似文献
Viruses in the genus Mastrevirus (family Geminiviridae), including those infecting sugarcane, have natural geographical ranges almost exclusively restricted to Africa and the Indian Ocean islands off the African coast. Only sugarcane white streak virus (SWSV) in Barbados and sugarcane striate virus (SStrV) in Florida and Guadeloupe are known to infect a few sugarcane varieties in the Western Hemisphere. In this study, PCR assays were developed to detect these two viruses in sugarcane. Five hundred and seventy-one DNA samples from Saccharum species and interspecific hybrids from the Miami World Collection of sugarcane and related grasses were tested for the presence of SStrV and SWSV by PCR. No variety was found infected by SWSV but SStrV was detected in 19 varieties. PCR data were confirmed by sequencing amplified fragments (248 bp). These fragments shared 93%–100% nucleotide identity with SStrV sequences from the GenBank database. SStrV isolates were distributed in six phylogenetic groups, including the four strains of the virus. Most varieties infected by SStrV originated from Asia, thus confirming a previous hypothesis stating that this virus originated from this continent. Absence of SStrV in commercial sugarcane in Florida also suggested that this virus has not been spread in this location, while infected plants have been present for several decades. 相似文献
Precipitation has generally increased in Norway during the last century, and climate projections indicate a further increase. The growing season has also become longer with higher temperatures, particularly in autumn. Previous studies have shown negative effects of high temperatures and, depending upon temperature conditions, contrasting effects of waterlogging on hardening capacity of timothy. We studied effects of waterlogging on seedlings of timothy (Phleum pratense, cv. Noreng) under three pre-acclimation temperatures: 3°C, 7°C, 12°C, and in autumn natural light in a phytotron at Holt, Tromsø (69°N). After temperature treatments, all plants were cold acclimated at 2°C for three weeks under continued waterlogging treatments. Freezing tolerance was determined by intact plants being frozen in pots at incremental temperature decreases in a programmable freezer. Waterlogging resulted in a higher probability of death after freezing, and a significantly reduced regrowth after three weeks at 18°C, 24 hrs light in a greenhouse. Increasing pre-acclimation temperatures also had a clear negative effect on freezing tolerance, but there was no interaction between temperatures and waterlogging. The results indicate that waterlogging may have negative implications for hardening of timothy and may contribute to reduced winter survival under the projected increase in autumn temperatures and precipitation. 相似文献
