Due to their antioxidant activity, anthocyanins are of increasing interest for nutritionists, food scientists and plant breeders. Anthocyanins in wheat grains are expressed in either the pericarp or aleurone layer. Previous studies revealed that different anthocyanins are present in wheat varieties carrying genes for either the purple pericarp or the blue aleurone trait. Progeny from crosses between red‐, purple‐ and blue‐grained wheat varieties were selected over several cycles for grain colour by visual scoring. Bulked F5 grains were evaluated for their total anthocyanin content by UV‐VIS spectrophotometry and HPLC‐MS. The results demonstrate that it is possible to increase the anthocyanin content by the combination of the different genetic backgrounds for purple pericarp and blue aleurone, even though the majority of progeny were within the range of the purple‐ and blue‐grained check varieties. Visual scoring for grain colour is efficient, reliable and fast for selection in early breeding generations. Advanced breeding lines with high anthocyanin content can be identified by simple extraction methods and spectrophotometric measurements.  相似文献

Bisphenol A (BPA) and reactive black 5 (RB5) dye are among the most persistent and non-biodegradable contaminants in water which require an urgent need for the development of effective removal method. The ubiquitous existence of both contaminants could interfere with the human health and aquatic environmental balance. Photocatalytic process as one of advanced oxidation processes (AOPs) has shown high performance for degradation of organic compounds to the harmless materials under sensible condition. Therefore, this study aims to develop a visible-light-driven photocatalyst that can efficiently degrade BPA and RB5 present in household water. N-doped TiO2 were successfully synthesized via simple and direct sol–gel method. The prepared TiO2 nanoparticles were characterized by field emission scanning microscope (FE-SEM), X-ray diffraction (XRD), Fourier transform infrared (FTIR), and Brunauere Emmette Teller (BET) analysis. The incorporation of nitrogen in TiO2 lattice exhibited excellent optical responses to visible region as revealed by UV–Vis–NIR spectroscopy absorption capability at 400–600 nm. The photocatalytic activity of the N-doped TiO2 nanoparticles was measured by photocatalytic degradation of BPA and RB5 in an aqueous solution under visible-light irradiations. Degradation of BPA and RB5 was 91.3% and 89.1%, respectively after 360 min illumination. The degradation of BPA and RB5 by N-doped TiO2 was increased up to 89.8% and 88.4%, respectively under visible-light irradiation as compared to commercial TiO2 P25. This finding clearly shows that N-doped TiO2 exhibits excellent photocatalytic degradation of BPA and RB5 under visible irradiation, hence have a promising potential in removing various recalcitrant contaminants for water treatment to fulfill the public need to consume clean water.

Chlamydia psittaci was detected by PCR in the lung and equine foetal membranes of two aborted equine foetuses and one weak foal from two different studs in Victoria, Australia. The abortions occurred in September 2019 in two mares sharing a paddock northeast of Melbourne. The weak foal was born in October 2019 in a similar geographical region and died soon after birth despite receiving veterinary care. The detection of C. psittaci DNA in the lung and equine foetal membranes of the aborted or weak foals and the absence of any other factors that are commonly associated with abortion or neonatal death suggest that this pathogen may be the cause of the reproductive loss. The detection of C. psittaci in these cases is consistent with the recent detection of C. psittaci in association with equine abortion in New South Wales. These cases in Victoria show that C. psittaci, and the zoonotic risk it poses, should be considered in association with equine reproductive loss in other areas of Australia.  相似文献

Early and accurate diagnosis is a critical first step in mitigating losses caused by plant diseases. An incorrect diagnosis can lead to improper management decisions, such as selection of the wrong chemical application that could potentially result in further reduced crop health and yield. In tomato, initial disease symptoms may be similar even if caused by different pathogens, for example early lesions of target spot (TS) caused by the fungus Corynespora cassicola and bacterial spot (BS) caused by Xanthomonas perforans. In this study, hyperspectral imaging (380–1020 nm) was utilized in laboratory and field (collected by an unmanned aerial vehicle; UAV) settings to detect both diseases. Tomato leaves were classified into four categories: healthy, asymptomatic, early and late disease development stages. Thirty-five spectral vegetation indices (VIs) were calculated to select an optimum set of indices for disease detection and identification. Two classification methods were utilized: (i) multilayer perceptron neural network (MLP), and (ii) stepwise discriminant analysis (STDA). Best wavebands selection was considered in blue (408–420 nm), red (630–650 nm) and red edge (730–750 nm). The most significant VIs that could distinguish between healthy leaves and diseased leaves were the photochemical reflectance index (PRI) for both diseases, the normalized difference vegetation index (NDVI850) for BS in all stages, and the triangular vegetation index (TVI), NDVI850 and chlorophyll index green (Chl green) for TS asymptomatic, TS early and TS late disease stage respectively. The MLP classification method had an accuracy of 99%, for both BS and TS, under field (UAV-based) and laboratory conditions.

Current in vitro embryo production protocols in the Iberian red deer (Cervus elaphus hispanicus) need to be optimized; oocyte harvesting in situ followed by overnight holding could reduce the human effort and shipping costs. In our work, post‐mortem ovaries were retrieved, and the oocytes were harvested and allocated to G1 group (good quality) or G2 + G3 group (low quality). The oocytes were separately subjected to immediate in vitro maturation (IVM) or held overnight in a holding medium composed of 40% of TCM 199 with Earle's salts, 40% TCM 199 with Hanks' salts and 20% fetal bovine serum (FBS), at room temperature (16 hr). In vitro maturation was carried out in a basal medium supplemented or not with 50 ng/ml of epidermal growth factor (EGF). Our data showed that addition of EGF to the maturation medium increases the percentage of G1 oocytes reaching metaphase II (3.9% vs. 50%, basal vs. EGF; p < .001) and decreased their degeneration rate (69.9% vs. 22.2%, basal vs. EGF; p < .01) when oocytes were immediately matured. Overnight holding increased the meiotic competence of G1 oocytes (37.5% matured in basal medium) and EGF increased prophase arrest in G2 + G3 oocytes (16.1% vs. 38.8% in germinal vesicle [GV] stage in basal medium vs. EGF added medium; p < .05). Our data demonstrate that oocyte holding can be used in Iberian red deer oocytes. Interestingly, EGF addition increases the oocytes' meiotic competence in immediately matured oocytes but not after oocyte holding depending upon initial oocyte quality.  相似文献

To expand our understanding of the roles of thyroid hormones on female reproduction, we induced hypo‐ and hyper‐T rat models to investigate the roles of thyroid hormones on estrous cyclicity, as well as the antioxidative status in the ovaries of rats. In the current study, our data show that hypothyroidism (hypo‐T) and hyperthyroidism (hyper‐T) led to significantly reduced body weights and ovarain weights and delayed vaginal opening day. For hyper‐T, thyroxine (T4), tri‐iodothyronine (T3), progesterone (P4) and follicle‐stimulating hormone (FSH) were significantly increased, while estradiol (E2) and luteinizing hormone (LH) were significantly decreased. For hypo‐T rats, serum levels of total T4 and T3, E2, P4, FSH and LH were significantly increased, while concentrations of E2 and LH were significantly decreased. For ovary morphology, the numbers of secondary and antral follicles were significantly decreased with more atretic antral follicles and less corpora lutea in both hyper‐ and hypo‐T groups. Both hyper‐T and hypo‐T treatment significantly decreased the expressions of thyroid hormone receptor α1 in the ovary. Hypo‐T significantly reduced nitric oxide (NO), total NO synthase (tNOS), inducible NOS and constitutive NOS activities, but hyper‐T increased them. For antioxidative parameters, hypo‐T and hyper‐T treatment significantly increased malondialdehyde (MDA) contents. The activities of both glutathione peroxidase (GSH‐Px) and catalase (CAT) significantly decreased in the hypo‐T group but increased in the hyper‐T group. Total superoxide dismutase (T‐SOD) activity was significantly increased in the hyper‐T group. In summary, thyroid hormones alter estrous cyclicity and antioxidative status in the ovary of the rat may act through the NOS signaling pathway.  相似文献

A new flavanone derivative, malaysianone A (1), four prenylated flavanones, 6-prenyl-3'-methoxyeriodictyol (2), nymphaeol B (3), nymphaeol C (4) and 6-farnesyl-3',4',5,7-tetrahydroxyflavanone (5), and two coumarins, 5,7-dihydroxycoumarin (6) and scopoletin (7), were isolated from the dichloromethane extract of the inflorescences of Macaranga triloba. The structures of these compounds were elucidated based on spectroscopic methods including nuclear magnetic resonance (NMR-1D and 2D), UV, IR and mass spectrometry. The cytotoxic activity of the compounds was tested against several cell lines, with 5 inhibiting very strongly the growth of HeLa and HL-60 cells (IC(50): 1.3 μg/ml and 3.3 μg/ml, respectively). Compound 5 also showed strong antiplasmodial activity (IC(50): 0.06 μM).  相似文献
