Dynamic elastic modulus (EL) and wave velocity (V) were determined using resonance vibrations from initially green, 100 × 50 mm sample boards of Eucalyptus regnans F. Muell., and after several stages of drying to oven dry. EL and V were determined from impact induced vibrations and spectral analysis. EL and V from green wood were positively related to basic density and normal shrinkage, only V was negatively related to green density, and both EL and V were negatively related to green moisture content and the number of internal checks after drying. The latter relationship has the potential to provide a simple method of segregating highly check prone material. No significant relationships were obtained with collapse. Outside the hygroscopic range, in low shrinkage material, EL increased little or gradually, while in high shrinkage, collapse prone material, it increased more rapidly, but no clear breakpoint was evident. In the hygroscopic range, EL increased rapidly in all samples. V increased curvilinearly throughout the entire moisture range, but no difference between collapse prone and non-collapse prone material was observed. Received 16 February 1998  相似文献

In the Balkan countries, where trichinellosis is a re-emerging zoonosis, it is of great importance to determine Trichinella infection prevalence among the major hosts, including horses. One method for monitoring prevalence is serological surveillance; however, the validity of serological methods in horses is not well understood. The dynamics of anti-Trichinella IgG production and circulating excretory/secretory (ES) antigens were investigated in three horses experimentally-infected with Trichinella spiralis. Horses were slaughtered at 32 week post infection (p.i.). Low worm burdens were found in all three animals. Anti-Trichinella IgG was detected up to 32 weeks p.i. by an indirect immunofluorescence assay (IFA) and by Western blot (Wb), but not by ELISA. The ELISA test detected antibodies for only a short period of time (up to 18 weeks p.i. using ES antigen or up to 20 weeks p.i. using tyvelose-BSA antigen). The presence of circulating muscle larvae ES antigen in sera of infected horses was observed by dot blot from the 4th week p.i. up to the 32nd week p.i.  相似文献

Maedi-Visna (MV) and ovine pulmonary adenocarcinoma (OPA) are two retroviral diseases occurring worldwide that affect adult sheep. Differences in incidence, which may be related to sheep-rearing and housing choices, as well as to genetics, and disease progression have been reported for both diseases. In this work four microsatellites located in immune-relevant regions, the major histocompatibility complex (MHC) region, interferon-γ and interleukin-12p35, were genotyped to determine their association with disease progression. The analysed sample included Latxa sheep with and without OPA and MV-characteristic lesions in their lungs. The microsatellites in the MHC were the most diverse, while the ones located in the cytokines were the less polymorphic. In the case of IFN-γ the results suggested the presence of null alleles. Significant results were detected for several microsatellite alleles in the association analysis carried out by logistic regression. All statistical analyses included a flock effect adjustment to avoid false positives due to genetic structuration. MHC Class I microsatellite alleles OMHC1*205 and OMHC1*193 were associated with disease progression for Maedi and OPA, respectively. Moreover, MHC Class II microsatellite allele DRB2*275 was associated with presence of lesions in Maedi. Furthermore, the MHC microsatellites were combined for a bioinformatic haplotype inference with the PHASE software. In total, 73 haplotypes were detected, 18 of them in more than 6 animals. After standard and weighted logistic regression analysis, two of them were significantly associated with susceptibility: OMHC1*205-DRB2*271 for Maedi and OMHC1*193-DRB2*271 for OPA, both with the Class I microsatellite alleles associated in the marker by marker study. Although more extensive analyses are needed to disentangle the relationship between host genetics and disease, as far as we know this is the first study demonstrating a significant association between sheep MHC Class I microsatellite alleles and susceptibility to Maedi-Visna and OPA viral diseases.  相似文献

Development of optimal ways to predict juvenile wood stiffness, strength, and stability using wood properties that can be measured with relative ease and low cost is a priority for tree breeding and silviculture. Wood static modulus of elasticity (MOE), modulus of rupture (MOR), radial, tangential, and longitudinal shrinkage (RS, TS, LS), wood density (DEN), sound wave velocity (SWV), spiral grain (SLG), and microfibril angle (MFA) were measured on juvenile wood samples from lower stem sections in two radiata pine test plantations. Variation between inner (rings 1–2 from pith) and outer (rings 3–6 from pith) rings was generally larger than that among trees. MOE and MOR were lower (50%) in inner-rings than in outer-rings. RS and TS were higher (30–50%) for outer-rings than inner-rings, but LS decreased rapidly (>200%) from inner-rings to outer-rings. DEN had a higher correlation with MOR than with MOE, while MFA had a higher correlation with dry wood MOE than with MOR. SLG had higher significant correlation with MOE than with MOR. DEN and MOE had a weak, significant linear relationship with RS and TS, while MOE had a strong negative non-linear relationship with LS. Multiple regressions had a good potential as a method for predicting billet stiffness (R 2 > 0.42), but had only a weak potential to predict wood strength and shrinkage (R 2 < 0.22). For wood stiffness acoustic velocity measurements seemed to be the most practical, and for wood strength and stability acoustic velocity plus core density seemed to be the most practical measurements for predicting lower stem average in young trees.  相似文献

Summary Assessments of internal checking and the physical properties of 124 trees of Eucalyptus regnans F. Muell. have shown that for material dried under relatively mild predryer conditions (30 °C, 65% RH) internal checking was highly positively correlated with each of collapse, moisture content and normal shrinkage, and weakly negatively correlated with total external shrinkage. Collapse alone explained 47% of the variation in internal checking. Incidence of internal checking in sample boards could be estimated with moderate success by each of the following properties measured on board ends: collapse, the number of internal checks and initial moisture content. Material with high mean basic density above 530 kg/m3 was associated with low levels of internal checking and collapse. However, the maximum naturally occurring density of E. regnans was not high enough to obviate collapse and internal checking. It was observed that growth rings in 100 × 50 mm backsawn boards in which the earlywood air-dry density was below 450 kg/m3 showed internal checking. The size and number of internal checks increased with a decrease in earlywood density. It was shown that drying E. regnans below temperatures of 24–30 °C does not eliminate collapse, thus raising doubt about the validity of a temperature threshold concept in that range. The influence of host genetics on the susceptibility to primary Trichinella spiralis infection has been extensively studied in a mouse model, but has not been clarified for rats. Analyses of interstrain and intrastrain genetic variation in response to infectious agents could be beneficial not only for elucidating the genetic basis of host resistance/susceptibility, but for revealing immune response mechanisms as well. The aim of this study was to analyse interstrain differences in worm burdens and cytokine production between Albino Oxford (AO) and Dark Agouti (DA) rats in muscle phase of T. spiralis infection. Clear strain-dependent variation was observed in the number of T. spiralis larvae per gram (lpg) of muscle tissue where values for DA rats (626.7 ± 171 lpg) vastly exceeded those found in AO rats (49.8 ± 25.9 lpg, p < 0.001). Differences between the strains were also noticed in key cytokine levels. In infected AO rats, the cytokine production remained in favor of Th1 type response, while infected DA rats showed a shift towards a Th2 type response. The level of regulatory IL-10 was significantly increased only in T. spiralis infected DA rats. Our results provide evidence that DA rats express higher susceptibility to T. spiralis infection in comparison to AO rats with respect to muscle larvae burden. The infection in DA rats was accompanied by the production of anti-inflammatory cytokines, while the response of AO rats was characterized by a proinflammatory type of immune response.  相似文献

Summary Shrinkage and moisture content measurements for thin sections of Eucalyptus regnans have shown that although total volumetric shrinkage was positively correlated with specific gravity, the relationship did not conform to the general equation s = f (Stamm 1935, 1952) in that the straight regression line was significantly different from a line passing through the origin. This contrasted with the relationship between shrinkage from 17% EMC to 0% moisture content and specific gravity which, in principle, complied with the Stamm formula. The discrepancy arose because shrinkage from the green condition to 17% EMC was negatively related to specific gravity owing to the high variability of the measured fibre saturation point and its negative correlation with specific gravity. These observations were affirmed in a comparison of unit shrinkage-specific gravity regressions where relationships were positively significant but the regression constants were not.Shrinkage behaviour in blocks of E. regnans did not comply with the Stamm formula although the average fibre saturation point (41.8%) was not significantly different from that for sections (38.2%). The near similarity derived from a significantly greater unit shrinkage in blocks (0.49) than that in sections (0.30). Parasitic nematode Trichinella spiralis exert immunomodulatory effect on the host immune response through excretory–secretory products (ES L1) released from the encysted muscle larvae. Rat bone-marrow derived dendritic cells (DCs) stimulated with ES L1 antigens acquire semi-matured status and induce Th2 and regulatory responses in vitro and in vivo. Priming naïve T cells in vitro with ES L1 pulsed DCs caused strong Th2 polarization, accompanied by elevated production of regulatory cytokines IL-10 and TGF-β and no increase in the proportion of CD4+CD25+Foxp3+ among the effector T cell population. In vivo T cell priming resulted in mixed Th1/Th2 cytokine response, with the dominance of the Th2 type and elevated levels of regulatory cytokines. Significant increase in the proportion of CD4+CD25+Foxp3+ cells was found among recipient's spleen cells. We have achieved to create immune status characteristic for the live infection by in vivo application of DCs educated with ES L1 antigens.  相似文献

Trophoblast adhesion to the uterine wall is the requisite first step of implantation and, subsequently, placentation. At the maternal-fetal interface, we investigated the expression of selectin adhesion systems that enable leukocyte capture from the bloodstream. On the maternal side, human uterine epithelial cells up-regulated selectin oligosaccharide-based ligands during the window of receptivity. On the fetal side, human trophoblasts expressed L-selectin. This ligand-receptor system was functional, because beads coated with the selectin ligand 6-sulfo sLe(x) bound to trophoblasts, and trophoblasts bound to ligand-expressing uterine luminal epithelium in tissue sections. These results suggest that trophoblast L-selectin mediates interactions with the uterus and that this adhesion mechanism may be critical to establishing human pregnancy.  相似文献

Summary Total collapse values from the green condition to various equilibrium moisture contents (EMC) were obtained by comparing shrinkage before reconditioning in matched blocks and sections of the wood of Eucalyptus regnans. It was shown that while collapse to 17% EMC comprised the largest portion of total collapse, a significant component, apparently caused by drying stress, developed below 17% EMC. Between 5% EMC and 0% moisture content a collapse recovery of some 1% occurred, seemingly because of changes in drying temperature. After reconditioning, collapse recovery was only partial and a high level of residual collapse remained. It was shown that collapse recovery was highest near the sapwood-heartwood boundary, while residual collapse was highest near the heartwood centre. While residual collapse was weakly negatively correlated with specific gravity, this relationship was not significant after adjustment was made for change in shrinkage after reconditioning in sections. All types of collapse were positively correlated with R-ratios calculated for blocks. However, relationships were not as well defined when R-ratios for sections were employed, the exception being for collapse below 17% EMC which was highly negatively correlated with R. The difference between shrinkage before reconditioning and shrinkage after reconditioning in sections, while totalling near zero, was positively correlated with specific gravity. It was demonstrated that this quantity could not constitute collapse in the traditional sense of collapse of the cell lumens. A possible association with moisture content of the material was discussed.  相似文献
