The objective of this study was to investigate the effect of infusing whole dead semen (WDS) after AI with diluted commercial semen on uterine inflammatory reaction and embryonic survival rate in gilts. Sixty Yorkshire-Landrace gilts were assigned at their second estrus to one of the following AI treatments: 1) commercial semen adjusted to 1 x 10(9) sperm cells (S1) per dose, followed by an infusion of 80 mL of WDS (S1-WDS); 2) S1 followed by an infusion of 80 mL of Beltsville Thawing Solution (S1-BTS); 3) commercial semen adjusted to 3 x 10(9) sperm cells (S3) per dose, followed by an infusion of 80 mL of BTS (S3-BTS); and 4) a negative control group, in which gilts received two infusions of 80 mL of BTS (BTS). Two days after the first AI, eight gilts from Groups 1, 2, and 4 were slaughtered and reproductive tracts were collected. One horn was cut open longitudinally along the antimesometrial aspect and endometrial samples were taken and immediately frozen for analysis of messenger RNA (mRNA) abundance for inflammatory cytokines and growth factors. The other horn was flushed with 20 mL of PBS, and the contents of interferon-gamma (IFN-gamma), tumor necrosis factor-alpha (TNF-alpha) and transforming growth factor-beta1 (TGF-beta1) were determined by ELISA. On d 25 after AI, gilts from Groups 1, 2, and 3 were slaughtered and their reproductive tracts were collected to evaluate the number of fetuses and corpora lutea. On d 2 after the first AI, only TGF-beta1 was detected in the flush of all gilts, and no difference was observed between S1-WDS, S1-BTS, and BTS gilts. Endometrial levels of IFN-gamma and interleukin (IL)-6 mRNA were marked in all gilts, but they were not affected by the AI treatments, whereas the mRNA abundances for IL-1 and IL-2 were negligible. Infusions of WDS or BTS after a fertile AI did not affect IGF-I, IGF-I receptor, or IGF-II mRNA levels compared with gilts infused with BTS only, whereas the mRNA abundance for the IGF-II receptor was decreased (P < 0.05) in WDS-infused gilts. In gilts inseminated with S1 doses, infusion of WDS did not affect the number of live embryos. Although infusions of WDS did not affect the mRNA level and secretion of the cytokines measured and did not improve embryonic survival rates, further studies are needed to better understand the influence of semen composition on the uterine response after mating.  相似文献

The present experiment was conducted to determine the influence of dietary fatty acids C18:2n-6 and C18:3n-3 on the modulation of intrauterine synthesis of prostaglandin E2 (PGE2) and F2alpha (PGF2alpha) during early pregnancy in pigs. Prostaglandin E2 in uterine fluid has been previously reported to be associated with embryo survival and development. Thirty-two Yorkshire-Landrace nulliparous gilts were randomly allocated to four diets containing 5% supplemental fat. The four dietary treatments were: HT, hydrogenated tallow (26.5% C16:0 and 54.8% C18:0); SO, sunflower oil (61.3% C18:2n-6); LO, linseed oil (50.4% C18:3n-3); and SO(CLA), a mixture of sunflower oil and conjugated linoleic acids to provide 20% CLA. Treatments started 2 d after the first pubertal estrus (d -21) and lasted for 36 d (slaughter), which was 15 d after the second estrus (d 0; insemination). Fatty acids and PGE2 were measured in the peripheral blood plasma on d -19, d -7, d 0, and d 14. Fatty acids in endometrial tissues and PGE2 and PGF2alpha in the uterine fluid collected on d 15 were also measured. Concentrations of fatty acids in the plasma reflected the content of fatty acids in the diet as early as d -7. From d -7, PGE2 concentrations in the plasma were higher in gilts fed SO compared with HT (P < 0.05). Plasma PGE2 concentrations were lower (P < 0.01) on d 14 in gilts fed LO compared with HT. Total PGF2alpha contents in the uterine fluid of gilts fed LO were more than 70% lower (P < 0.05) than for the HT group. A similar trend was observed for total PGE2 content and for the ratio PGF2alpha:PGE2, but the effect (LO vs HT) was less marked (P < 0.07 and P < 0.10, respectively). There was no effect of SO or SO(CLA) on total PGE2 contents in the uterine fluid. Dietary enrichment in C18:2n-6 and/or C18:3n-3 for early pregnant gilts can influence fatty acids in plasma and endometrial tissue and can modulate circulatory and intrauterine prostaglandins.  相似文献

Root respiration may account for as much as 60% of total soil respiration. Therefore, factors that regulate the metabolic activity of roots and associated microbes are an important component of terrestrial carbon budgets. Root systems are often sampled by diameter and depth classes to enable researchers to process samples in a systematic and timely fashion. We recently discovered that small, lateral roots at the distal end of the root system have much greater tissue N concentrations than larger roots, and this led to the hypothesis that the smallest roots have significantly higher rates of respiration than larger roots. This study was designed to determine if root respiration is related to root diameter or the location of roots in the soil profile. We examined relationships among root respiration rates and N concentration in four diameter classes from three soil depths in two sugar maple (Acer saccharum Marsh.) forests in Michigan. Root respiration declined as root diameter increased and was lower at deeper soil depths than at the soil surface. Surface roots (0-10 cm depth) respired at rates up to 40% greater than deeper roots, and respiration rates for roots < 0.5 mm in diameter were 2.4 to 3.4 times higher than those for roots in larger diameter classes. Root N concentration explained 70% of the observed variation in respiration across sites and size and depth classes. Differences in respiration among root diameter classes and soil depths appeared to be consistent with hypothesized effects of variation in root function on metabolic activity. Among roots, very fine roots in zones of high nutrient availability had the highest respiration rates. Large roots and roots from depths of low nutrient availability had low respiration rates consistent with structural and transport functions rather than with active nutrient uptake and assimilation. These results suggest that broadly defined root classes, e.g., fine roots are equivalent to all roots < 2.0 mm in diameter, do not accurately reflect the functional categories typically associated with fine roots. Tissue N concentration or N content (mass x concentration N) may be a better indicator of root function than root diameter.  相似文献

A bovine leukemia virus (BLV) control program was instituted at a high-prevalence, purebred Holstein dairy. The specific control program required single use of needles and obstetrical sleeves, disinfection of tattoo equipment before use, dehorning by use of electrical burning devices, replacement of whole milk feeding with high-quality milk replacer, and heat treatment of colostrum before feeding. The effect of the program was estimated by comparing the pre- (1987) and postintervention (1989) prevalences of BLV-agar gel immunodiffusion titers within particular heifer age cohorts (0 to 90, 91 to 180, 181 to 360 days, and 361 days to parturition). Random serologic testing was conducted quarterly within each cohort, but was analyzed on a combined yearly basis. Seroprevalence of BLV decreased in all age cohorts greater than 180 days old, with significant decreases in the 361 days to parturition (P = 0.003) and the combined 181 days to parturition cohorts (P = 0.0004). Prevalence within the latter cohort decreased from 0.44 in 1987 to 0.17 in 1989. These results were obtained without either culling or segregation of BLV-agar gel immunodiffusion-positive cattle.  相似文献

The purpose of the study, carried out in a beef processing plant, was to evaluate the effectiveness of a new prototype for steam pasteurization treatment in controlling microbiological hazards. Samples were taken by swabbing randomly selected sites before and after pasteurization and again after chilling to obtain total aerobic counts (TAC), total coliform counts (TCC), and generic Escherichia coli counts (ECC) on Petrifilm plates and to determine the prevalence of Salmonella spp., Listeria monocytogenes, and E. coli O157:H7 using standard enrichment techniques. Escherichia coli and L. monocytogenes strains were tested for various factors associated with their virulence by using colony hybridization and polymerase chain reaction (PCR), respectively. Antimicrobial susceptibility was determined for each isolate that was potentially pathogenic to humans by using the disk-diffusion method. Mean values for TAC, TCC, and ECC were 2.18, 0.16, and 0.06 log10 CFU/cm2, respectively, before pasteurization; 1.17, 0.03, and 0.01 log10 CFU/cm2 after pasteurization; and 0.89, 0.02, and 0.01 log10 CFU/cm2 after chilling. Prevalence of L. monocytogenes, Salmonella spp., and E. coli O157:H7 on carcasses was 0.8%, 0.0%, and 0.0%, respectively, before pasteurization; 2.6%, 0.0%, and 0.0% after pasteurization; and 3.1%, 0.1%, and 0.0% after chilling. The prevalence of E. coli containing > or = 1 virulence gene was 14.7%. More specifically, 11.88% of the isolates obtained before pasteurization, 22.2% obtained after pasteurization, and 31.2% obtained after chilling had virulence genes. All L. monocytogenes isolates tested positive for the presence of 3 major virulence factors (hlyA, inlB, and plcB). Antibiograms showed that certain isolates were susceptible to all antibiotics, some showed an intermediate sensitivity, and others were multiresistant. Overall, these results suggest that steam pasteurization is an effective means of improving safety quality of beef carcasses. However, pasteurization may indirectly contribute to the growth of some pathogenic microorganisms, such as L. monocytogenes.  相似文献

OBJECTIVE: To evaluate healing of pinch-grafted wounds on the distal aspect of the limbs of ponies bandaged with equine amnion or a standard nonadherent wound dressing material. ANIMALS: 6 ponies. PROCEDURE: A 2.5x2.5-cm full-thickness section of skin was removed from the dorsal aspect of each limb at the midpoint of the metacarpus or metatarsus. Six days later, wounds were grafted with partial-thickness pinch grafts. Half the wounds were bandaged with amnion, and the other half were bandaged with a nonadherent dressing. Bandages were changed every 3 days until wound healing was complete. At each bandage change, numbers of grafts lost were recorded, and wounds were measured. RESULTS: Percentage of grafts lost from wounds bandaged with amnion was not significantly different from percentage lost from wounds bandaged with the nonadherent dressing. Median healing time for wounds bandaged with amnion (30 days) was significantly less than median healing time for wounds bandaged with the nonadherent dressing (39 days). All wounds were healed by day 45. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that amnion can be used for bandaging pinch-grafted wounds on the distal aspect of the limbs of ponies.  相似文献
