Hardwoods contain a substantial amount of hemicellulose-type oligosaccharides, the chemical structures of which are typically categorized in the xylan saccharide class. The efficient and selective isolation of these hemicelluloses is regarded as one of the most critical obstacles to overcome for their eventual biomaterial and bioenergy utilization. Therefore, the objective of the current work was to perform a fundamental exploration of the function of extraction pH, temperature, and time on the final chemical properties of the extracted hemicelluloses. The extraction chemistries employed varied pH and showed that acidic conditions provided a higher extractive yield versus alkaline conditions. An alkaline environment gave higher yields than an acidic environment and also resulted in much higher lignin removal from wood, especially at high temperature. In general, control of the pH chemistry leads to a change in the carboxylic acid groups present in the extracts. When the extraction pH changed from acidic to alkaline, the total carboxylic acid group content in the hemicelluloses removed significantly increased due to more acetic acid group hydrolysis and increased lignin degradation.  相似文献

Forest soils are pivotal to understanding global carbon (C) cycling and evaluating policies for mitigating global change. However, they are very difficult to monitor because of the heterogeneity of soil characteristics, the difficulty of representative sampling, and the slow time scale of response to environmental change. Here we demonstrate that use of gamma-ray spectroscopy facilitates in situ non-destructive analysis of C and other elements in forest soils. In this approach the element-specific gamma-rays are induced by fast and thermal neutrons interacting with the nuclei of the elements present in the soil. Background gamma-rays emanating from naturally occurring radionuclides in the forest are recorded as well. We applied this approach in a mature northern hardwood forest on glacial till soils at the Bartlett Experimental Forest in New Hampshire, USA. The inelastic neutron scattering (INS) system yielded strong signals in gamma-ray counts/h, from C and other elements present in the soil matrix that included silicon, oxygen, hydrogen, iron, aluminum, manganese and potassium. The INS sensitivity for carbon was 20.656 counts h−1 kg−1 C m−2 based on current net C gamma-ray counts and the data for the O horizon and mineral soil to a depth of 30 cm obtained from a nearby quantitative soil pit (7.35 kg C m−2). We estimate the minimum detectable change to be ∼0.34 kg C m−2, which is ∼5% of the current soil C content, and the minimum detectable limit to be ∼0.23 kg C m−2. Eight % reproducibility from 11 measurements was limited, in part, by the large variability in the system counting geometry due to the uneven forest microtopography. The INS approach has the potential to revolutionize belowground monitoring of C and other elements, because the possibility of detecting a 5% change in forest soils has not been possible with destructive sampling methods.  相似文献

The recently developed protocol for isolating enzymatic mild acidolysis lignins (EMAL) coupled with the novel combination of derivatization followed by reductive cleavage (DFRC) and quantitative (31)P NMR spectroscopy were used to better understand the lignin isolation process from wood. The EMAL protocol is shown to offer access at lignin samples that are more representative of the overall lignin present in milled wood. The combination of DFRC/(31)P NMR provided a detailed picture on the effects of the isolation conditions on the lignin structure. More specifically, we have used vibratory and ball milling as the two methods of wood pulverization and have compared their effects on the lignin structures and molecular weights. Vibratory-milling conditions cause substantial lignin depolymerization. Lignin depolymerization occurs via the cleavage of uncondensed beta-aryl ether linkages, while condensed beta-aryl ethers and dibenzodioxocins were found to be resistant to such mechanical action. Condensation and side chain oxidations were induced mechanochemically under vibratory-milling conditions as evidenced by the increased amounts of condensed phenolic hydroxyl and carboxylic acid groups. Alternatively, the mild mechanical treatment offered by ball milling was found not to affect the isolated lignin macromolecular structure. However, the overall lignin yields were found to be compromised when the mechanical action was less intense, necessitating longer milling times under ball-milling conditions. As compared to other lignin preparations isolated from the same batch of milled wood, the yield of EMAL was about four times greater than the corresponding milled wood lignin (MWL) and about two times greater as compared to cellulolytic enzyme lignin (CEL). Molecular weight distribution analyses also pointed out that the EMAL protocol allows the isolation of lignin fractions that are not accessed by any other lignin isolation procedures.  相似文献

The use of aqueous suspension of nanoparticles of titanium dioxide for photocatalytic removal of pollutants is not suitable for industrial applications due to the inconvenient and expensive separation of nanoparticles of titanium dioxide for reuse. The nanosized titanium dioxide needs to be immobilized on the support for improving the efficiency and economics of the photocatalytic process. In the present paper, nanoparticles of titanium dioxide have been immobilized on the surface of the support using three different techniques. The immobilized films of titanium dioxide have been characterized using X-ray diffraction and scanning electron microscopy to notice any change in the phase composition and photocatalytic properties of the titanium dioxide after immobilization on the support. A photocatalytic test has been performed under similar reaction conditions to compare the photocatalytic performance of the films of immobilized titanium dioxide prepared using different techniques.  相似文献

Key message

Historic transfer of larch from Alpine sources to Southern and Eastern Carpathians has been verified by means of nuclear genetic markers. Tyrolean populations can be differentiated into a north-western and south-eastern group, while Romanian populations are separated according to the Southern and Eastern Carpathians. Low-level introgression from Alpine sources is found in autochthonous Carpathian populations.

Context

Large scale human mediated transfer of forest reproductive material may have strongly modified the gene pool of European forests. Particularly in European larch, large quantities of seeds from Central Europe were used for plantations in Southern and Eastern Europe starting in the mid nineteenth century.

Aims

Our main objective was to provide DNA marker based evidence for the anthropogenic transfer of Alpine larch reproductive material to native Carpathian populations.

Methods

We studied and compared 12 populations (N?=?771) of Larix decidua in the Alps (Austria, Italy) and in the Southern and Eastern Carpathians (Romania) using 13 microsatellites.

Results

High genetic diversity (He?=?0.752; RS?=?9.4) and a moderate genetic differentiation (FST?=?0.13; GST?=?0.28) among populations were found; Alpine and Carpathian populations were clearly separated by clustering methods. A Tyrolean origin of plant material was evident for one out of four adult Romanian populations. In the transferred population, a genetic influence from Carpathian sources was found neither in adults nor in juveniles, while the natural regeneration of two Romanian populations was genetically affected by Alpine sources to a minor degree (2.2 and 2.9% allochthonous individuals according to GeneClass and Structure, respectively).

Conclusion

Tracing back of plant transfer by means of genetic tools is straightforward, and we propose further studies to investigate gene flow between natural and transferred populations.

During two measurement campaigns, from August to September 2008 and 2009, we quantified the major ecosystem fluxes in a hemiboreal forest ecosystem in Järvselja, Estonia. The main aim of this study was to separate the ecosystem flux components and gain insight into the performance of a multi-species multi-layered tree stand. Carbon dioxide and water vapor fluxes were measured using the eddy covariance method above and below the canopy in conjunction with the microclimate. Leaf and soil contributions were quantified separately by cuvette and chamber measurements, including fluxes of carbon dioxide, water vapor, nitrogen oxides, nitrous oxide, methane, ozone, sulfur dioxide, and biogenic volatile organic compounds (isoprene and monoterpenes). The latter have been as well characterized for monoterpenes in detail. Based on measured atmospheric trace gas concentrations, the flux tower site can be characterized as remote and rural with low anthropogenic disturbances.Our results presented here encourage future experimental efforts to be directed towards year round integrated biosphere-atmosphere measurements and development of process-oriented models of forest-atmosphere exchange taking the special case of a multi-layered and multi-species tree stand into account. As climate change likely leads to spatial extension of hemiboreal forest ecosystems a deep understanding of the processes and interactions therein is needed to foster management and mitigation strategies.  相似文献
