Resistance to pre-harvest sprouting is an important breeding objective for cereal crops like barley and wheat. Seed dormancy, which determines the resistance or susceptibility to pre-harvest sprouting (PHS), is a complex trait. It is largely controlled by the antagonistic action of the plant hormones abscisic acid and gibberellic acid, but also has a large component of genotype?×?environment interaction. Recent studies have revealed a role for epigenetic changes through histone modification in controlling seed dormancy. However, the role of DNA methylation in seed development and dormancy is not known. In this study, we explored the role of ARGONAUTE4_9 class genes of the DNA methylation pathway in seed development and dormancy in barley. Our results show that the two AGO4_9 class genes in barley, i.e. AGO1002 and AGO1003, are preferentially expressed in ovaries at meiosis and in embryos 25?days after pollination (DAP). The expression of AGO1003 is two to fivefold higher than that of AGO1002 in these tissues, demonstrating differential expression of these genes. We also analysed the expression of AGO1003 in embryos of PHS-resistant and -susceptible varieties at 25?DAP and found a significant variation in the expression of this gene in seeds of dormant and non-dormant lines. The observed expression pattern of AGO1002 and AGO1003 suggests a possible role in sporogenesis and post-fertilization seed development. Indirectly these results imply a potential role of DNA methylation in seed development and seed dormancy.  相似文献

This study assessed the net above-ground carbon stock in six community forests in the Dolakha district, Nepal. A survey was conducted of above-ground timber species, using random sampling. A tree-ring chronology for Pinus roxburghii was created to construct a growth model representative of the various mainly-pine species. The allometric model combined with tree ring analysis was used to estimate carbon stock and annual growth in the above-ground tree biomass. The out-take of forest biomass for construction material and fuelwood was estimated on the basis of interviews and official records of community forest user groups. The average annual carbon increment of the community forests was 2.19 ton/ha, and the average annual carbon out-take of timber and fuelwood was 0.25 ton/ha. The net average carbon balance of 1.94 ton/ha was equivalent to 117.44 tons of carbon per community forest annually. All the community forests were actively managed leading to a sustainable forest institution, which acts as a carbon sink. It is concluded that community forests have the potential to reduce emissions by avoiding deforestation and forest degradation, enhance forest carbon sink and improve livelihoods for local communities.  相似文献

Transition proteins (TNPs) are essential in chromatin condensation during spermiogenesis, and hence, they are the candidate genes for identifying sperm motility markers. Coding and in silico predicted promoter regions of these genes were investigated in crossbred and purebred cattle, and also, their mRNA quantification was done to explore its use as a diagnostic tool of infertility. PCR‐SSCP analysis revealed two band patterns in fragment III of TNP1 and fragment II of TNP2 gene. Sequence analysis revealed a deletion of “G” nucleotide in 3′UTR region of TNP1 and C>T SNP in intronic region of TNP2 gene. Least square analysis of variance did not reveal any significant influence of nucleotide deletion on any sperm motility parameters in both crossbred and purebred cattle. However, C>T SNP had a significant effect on initial progressive motility (p < 0.05) in purebred cattle and post‐thaw motility in overall cattle population. RT‐qPCR analysis did not reveal any significant variation in TNP1 and TNP2 gene expression among poorly motile and good quality spermatozoa of Vrindavani bulls.  相似文献

Progesterone (P4) plays a key role in the establishment and maintenance of pregnancy in most mammals. Unravelling the expression of progesterone‐regulated genes can expand the understanding of the embryonic mortality. Accordingly, we studied the relative mRNA expression of the P4‐regulated genes in the buffalo. Uteri were collected from the abattoir and categorized into nonpregnant late luteal phase, stage I (28–38th days of gestation) and stage II (48–56th days of gestation) of pregnancy (n = 6/group). After extraction of total RNA from the endometrial tissues, we carried out qRT‐PCR for determining the relative mRNA expression of the P4‐regulated genes using nonpregnant late luteal phase as calibrator group. The expression of LGALS3BP (essential for maternal recognition of pregnancy) gene was found to be significantly upregulated (p < 0.05), while MUC1 (important for embryo attachment) gene was downregulated in stage I and II of pregnancy. We observed no significant change in the expression of LGALS1, LGALS9 and CTSL genes. The SLC5A11 and SLC2A1 genes (involved in the transport of glucose to endometrium) in early pregnancy were upregulated in the pregnancy stage I (p < 0.05) relative to nonpregnant late luteal phase. The CST3 gene was significantly upregulated in pregnancy stage II (p < 0.01). These results provide molecular insights into the specific pathways involved in foeto‐maternal communication during early pregnancy in buffaloes.  相似文献

A decrease in net photosynthesis (Pn) in leaves of main shoot with age ( 7 -28 days after opening) at two stages of peanut ( Arachis hypogaea L. var. M-13 ) reproductive growth (70 and 90 DAS) is reported. Pn rate decreased with advance in age of leaves, the decline was rapid during peak period of pod filling (90 DAS). Foliar application with aliphatic alcohols, maintained the chlorophyll content and enhanced PEP carboxylase activity. The present studies suggest that aliphatic alcohols enhance functional capabilities of leaf for a larger duration and hence maintain the supply of photoassimilates to the developing kernels of peanut leading to increased yield.  相似文献

Storage of recalcitrant seeds remains an unsolved problem. The study of gaseous atmospheres could provide insights in understanding the metabolism of recalcitrant seeds and their potential in being used as means for storage. This study investigated the metabolism of the recalcitrant Quercus alba seeds when subjected under nitrous oxide (N2O) and air atmospheres. Seeds were placed in sealed atmospheric flasks of 98/2% N2O/O2. Three experiments were conducted to determine N2O uptake in relation to oxygen (O2) and carbon dioxide (CO2) levels. For the first experiment, “non-renewed” closed atmospheric systems with 98/2% N2O/O2 and air were used. Two more experiments were also conducted for the 98/2% N2O/O2 atmosphere to determine how rate of gas renewal (two and seven days) affects the N2O, O2, and CO2 gases quantitatively. Cumulative results on non-renewed flasks with 98/2% N2O/O2 showed that as time progressed, N2O uptake increased with variation in the magnitude of N2O utilization throughout the 37 experimental days, while seeds ceased O2 utilization by the 11th day. However, CO2 levels did not decrease, suggesting a shift toward fermentation processes. Finally, the two experiments for the 98/2% N2O/O2 indicated that the O2 levels did not deplete under a two-day gas renewal, suggesting a frequent gas renewal under low O2 storage conditions.  相似文献

Natural forces and anthropogenic activities greatly alter land cover,deteriorate or alleviate forest frag-mentation and affect biodiversity.Thus land cover and forest fragmentation dynamics have become a focus of concern for natural resource management agencies and biodiversity con-servation communities.However,there are few land cover datasets and forest fragmentation information available for the Dhorpatan Hunting Reserve (DHR) of Nepal to develop targeted biodiversity conservation plans.In this study,these gaps were filled by characterizing land cover and forest frag-mentation trends in the DHR.Using five Landsat images between 1993 and 2018,a support vector machine algorithm was applied to classify six land cover classes:forest,grass-lands,barren lands,agricultural and built-up areas,water bodies,and snow and glaciers.Subsequently,two landscape process models and four landscape metrics were used to depict the forest fragmentation situations.Results showed that forest cover increased from 39.4% in 1993 to 39.8% in 2018.Conversely,grasslands decreased from 38.2% in 1993 to 36.9% in 2018.The forest shrinkage was responsible for forest loss during the period,suggesting that the loss of for-est cover reduced the connectivity between forest and non-forested areas.Expansion was the dominant component of the forest restoration process,implying that it avoided the occurrence of isolated forests.The maximum value of edge density and perimeter area fractal dimension metrics and the minimum value of aggregation index were observed in 2011,revealing that forests in this year were most fragmented.These specific observations from the current analysis can help local authorities and local communities,who are highly dependent on forest resources,to better develop local forest management and biodiversity conservation plans.  相似文献

The status of forest conditions before and after intervention of the forestry projects in community forest in three districts of Nepal is examined. Benefits are observed from the adoption of adaptive collaborative management and collective learning and action research in three sampled districts. The adoption of regular silvicultural treatments has increased the availability of forest products to local users. Moreover, improved forest condition and smallholder livelihoods have improved, as has environmental sustainability. However, the community forestry program has several limitations and shortcomings. Elite capture, social disparity, inequitable benefit-sharing and exclusion of poor and marginalized groups from the community forestry program are notable challenges to be solved in coming years. Special attention is needed to make community forestry inclusive with equitable benefit-sharing and a pro-poor focus.  相似文献

In Indian mustard (Brassica juncea L. Czern & Coss), a major oilseed brassica crop, oil yield is the ultimate trait of economic value, which is determined by seed yield (SY) and oil content (OC). SY, the end product of many complex biological processes, is influenced by many traits, such as plant height (PH), branch number, siliqua number, seed weight, seed size and pod length. It is difficult to accurately select for high-yielding genotypes based exclusively on SY, neglecting other yield-determining traits in commercial breeding programs. Therefore, it is desirable for breeders to simultaneously improve several traits of economic importance, most of which are quantitative in nature. The purpose of the present study was to develop selection indices for simultaneous selection of multiple quantitative traits to enhance selection efficiency of the mustard breeding program of Punjab Agricultural University (PAU) located in Abohar, Punjab. A set of 487 diverse mustard genotypes, comprising germplasm accessions of Indian and European origin, advanced breeding lines, landraces, and cultivars, was evaluated using an alpha lattice design for 2 years (2015–16 and 2016–17) at the Regional Research Station of PAU, Abohar, District Fazilka (the major mustard-growing region of the Punjab state). Data on PH, main shoot length (MSL), number of primary branches (NPB), number of secondary branches (NSB), number of siliquae on main shoot (NSMS), siliqua length (SL), number of seeds per pod (NSP), 1000-seed weight (TSW), SY on a plot basis and OC were recorded. To compute selection indices, economic weights, as determined via path coefficient analysis, were assigned to major yield-determining traits. Out of eight recorded agronomic traits, four traits, viz., NSMS, TSW, NSB and NPB, had larger direct effects on SY than other traits, and hence were assigned relative economic weights of 0.60, 0.20, 0.12 and 0.08, respectively. Selection indices were computed using different combinations of traits via Kang_SASPath computer program. When seven traits were used in a selection index (PH, NPB, NSB, NSMS, SY, TSW and MSL; or PH, NPB, NSB, NSMS, SY, TSW and OC), expected genetic advance (GA)at 5% selection intensity was about the same (9.55; selection efficiency or SE = 163% compared with selection based on SY alone).The GA was 9.46 (SE = 161%)for five traits, viz., NPB, NSB, NSMS, SY and TSW; and 9.30 (SE = 159%)for four traits, viz., NPB, NSB, NSMS, and TSW. These GAs were compared with the GA for SY only, which was 5.90 (benchmark; SE = 100%). We concluded that simultaneous selection based on multiple traits was always more efficient than selection based on SY alone. As there was only a small difference between the GA based on four traits (SE = 161%) and GA based on seven traits (SE = 163%), we suggest that mustard breeders use the parsimonious combination of four traits, viz., NPB, NSB, NSMS, and SY, to enhance SY and, consequently, oil yield in mustard.  相似文献
