A protocol for somatic embryogenesis and plant regeneration of Ilexparaguariensis St. Hil. from embryos cultures was developed. Heart stage zygotic embryos were removed from seeds of immature, light green fruit and treated with antimicrotubule agents (0.1; 0.2, and 0.5% colchicine for 24 and 48 h; 1; 10, and 20 M of either trifluralin, - trifluoro- 2,6-dinitro-N,N- dipropyl-p-toluidine, or oryzalin, 3,5-dinitro-N4, N-dipropylsulphate during 48 h). The embryos were cultured aseptically on quarter-strength Murashige and Skoog medium containing 3% sucrose, 0.65% agar (1/4MS), and 0.46 M zeatin. Cultures were incubated in darkness at 27 ± 2 °C. All thetreatments provoked a diminution of the number of germinated embryos and in some of the treated embryos somatic embryogenesis was induced. Somatic embryo maturation and conversion into whole plants could be achieved by culturing the embryos on 1/4MS lacking hormones and incubated at 27 ± 2 °C, 14 h photoperiod (116 mol m-2s-1). Mostof the plants regenerated from somatic embryos appeared morphologically normaland grew under greenhouse conditions. Only 2 plants out of 152 studied contained the tetraploid number of the chromosomes (2n = 4x = 80), meanwhile the rest of the plants had the normal diploid number of chromosomes (2n =2x = 40). Somatic embryos with abnormal morphology were also observed.  相似文献

Highly varible organogenic regeneration has been obtained in sunflower, depending upon culture conditions, genotype and their interactions and now the effect of cytoplasm upon regeneration ability has been analysed. Six sunflower inbred lines in their male-sterile and malefertile forms were evaluated in vitro : HA89, GP762, HA300, RK416, RK456, and KLM280. The basal medium was agar solidified Murashige and Skoog (MS) medium supplemented with plant hormones: KIN 2mg/1 + IAA 1 mg/1 (Ml); BA 0.5mg/1 4- IAA 0.5mg/1 (M2). Cotyledons of mature seeds were used as explants. Regeneration ability (R) was evaluated on day 30 after initiation of culture. Significant differences were observed between male-sterile and male-fertile forms of some inbred lines for R on M2 medium. The presence of interaction among the factors line-cytoplasm-regeneration on M2 was demonstrated by using a G-test. This nucleus-cytoplasm interaction affects the regeneration ability of this species and should be taken into account when using tissue culture for crop improvement.  相似文献

A cryopreservation process usingencapsulation/dehydration technique was setup for apical meristem-tips of invitro plantlets of `paradise tree' (Melia azedarach L. var. gigantea,clone `El dorado'). Apical meristem-tipswere cultured for one day on MS basalmedium with 2 M BA and 0.5 M IBAand encapsulated with 3% sodium alginate.The highest shoot proliferation rate aftercryopreservation was obtained whenencapsulated apical meristem-tips werepregrown for 3 days in liquid medium with0.5, 0.75 and 1 M of sucrose for 24 hoursprogressively, desiccated for 5 hours withsilicagel followed by rapid or slowcooling. Survival after freezing in liquidnitrogen ranged between 67–83% andshoot proliferation ranged between 43–60%. This cryopreservation treatmentpreserved genetic stability, when it wasevaluated using the electrophoreticpatterns of nine isozyme systems and RAPDprofiles.  相似文献

Mesquite (Prosopis alba Grisebach), an important species in arid and semiarid regions, is currently commercially propagated by seeds and so there is great interest in developing techniques for species propagation, domestication and improvement. Cloning through mini-cuttings is a new and promising technique for the production of clonal seedlings of forest species. The main objective of this work was to evaluate vegetative propagation using the mini-cutting technique and indolebutyric acid (IBA) at different concentrations (0, 3,000, 4,500, 6,000 and 7,500 mg L?1) on rooting of P. alba clones. Rooting was achieved in 98–100 % of the mini-cuttings at all concentrations tested. Increasing IBA concentration resulted in an increase in the number of leaves and leaflets, as well as in fresh matter weight and number and length of roots of clone seedlings, until an optimum point (between 3480 and 4800 mg L?1) was reached. Plants were also vegetatively propagated in the field via conventional clonal garden cuttings under the same rooting conditions to compare the efficiency of the two propagation techniques. Mini-cuttings showed higher rooting and survival percentages than cuttings. A correlation analysis conducted between characteristics of stock plants (height, diameter, number of shoots) and shoots (height, diameter) showed a positive correlation between rooting and height of shoots with total root length, leaflet number, and height of clonal seedlings. To identify plant growth promotion ability of phosphorus-solubilizing native bacteria, we have examined a collection of isolates representing the diversity of culturable phosphate-solubilizing bacteria from acid soils of the northeast of Argentina. Assays in growth medium supplemented with tricalcium phosphate revealed different phosphorus solubilization activity and temporal patterns of solubilization. Acidification of the broth medium coincided with phosphorus solubilization. The isolates were grouped according to their Rep fingerprinting profiles and phylogenetically classified by 16S rDNA and biochemical analyses. These isolates were assigned to the genera Enterobacter, Pantoea, Pseudomonas, Acinetobacter, Burkholderia, and Exiguobacterium. Four isolates showing high phosphorus solubilizing activity in in vitro assays were inoculated on common beans (Phaseolus vulgaris); some of them promoted plant growth and increased photosynthesis and the P and N content of leaves. The results indicated that the ability to in vitro solubilize P is not necessarily associated to the promotion of plant growth.  相似文献

In vitro regeneration of complete plants from nodal single-bud segments of 2-year-old Australian Cedar (Toona ciliata) trees were obtained under defined nutritional and environmental conditions. Explants were dissected from plants obtained by germination of seeds and growth in pots in a greenhouse. The best medium for shoot regeneration was that of Murashige and Skoog at 1/4 strength with 3% sucrose (1/4 MS), supplemented with 0.1 mg/l IBA and 0.5 mg/l BAP. Rooting of regenerated shoots was observed in MS medium with 0.1 mg/l IBA. Using mature tree material was more difficult. Forced flushing was used to induce shoot development on branches of a 10-year-old tree. Nodal segments of these epicormic shoots formed shoots in vitro on 1/4 MS + 0.01 mg/l IBA + 5 mg/l BAP, but rooting was never observed.  相似文献

Cotyledons from 20 inbred lines of sunflower were evaluated in vitro for their regeneration ability on two culture media supplemented with growth regulators. Plant regeneration by direct organogenesis was observed after 4 weeks. Significant differences among inbred lines were found for the in vitro traits ‘percentage of organogenic explants’,‘percentage of shooting explants’,‘percentage of hypertrophing explants’ and ‘percentage of callusing explants’. The medium effect was significant for all variables. Some in vitro traits displayed significant genotype‐culture medium interaction. Regeneration ability was influenced by culture medium, genotype and their interaction and genetic parameters were estimated taking this interaction into account. Heritability values ranged from 0.74 to 0.45. Strong positive correlations were observed among the organogenesis‐related traits ‘percentage of organogenic explants’, ‘percentage of shooting explants’ and ‘proliferation rate’.  相似文献
