Root respiration may account for as much as 60% of total soil respiration. Therefore, factors that regulate the metabolic activity of roots and associated microbes are an important component of terrestrial carbon budgets. Root systems are often sampled by diameter and depth classes to enable researchers to process samples in a systematic and timely fashion. We recently discovered that small, lateral roots at the distal end of the root system have much greater tissue N concentrations than larger roots, and this led to the hypothesis that the smallest roots have significantly higher rates of respiration than larger roots. This study was designed to determine if root respiration is related to root diameter or the location of roots in the soil profile. We examined relationships among root respiration rates and N concentration in four diameter classes from three soil depths in two sugar maple (Acer saccharum Marsh.) forests in Michigan. Root respiration declined as root diameter increased and was lower at deeper soil depths than at the soil surface. Surface roots (0-10 cm depth) respired at rates up to 40% greater than deeper roots, and respiration rates for roots < 0.5 mm in diameter were 2.4 to 3.4 times higher than those for roots in larger diameter classes. Root N concentration explained 70% of the observed variation in respiration across sites and size and depth classes. Differences in respiration among root diameter classes and soil depths appeared to be consistent with hypothesized effects of variation in root function on metabolic activity. Among roots, very fine roots in zones of high nutrient availability had the highest respiration rates. Large roots and roots from depths of low nutrient availability had low respiration rates consistent with structural and transport functions rather than with active nutrient uptake and assimilation. These results suggest that broadly defined root classes, e.g., fine roots are equivalent to all roots < 2.0 mm in diameter, do not accurately reflect the functional categories typically associated with fine roots. Tissue N concentration or N content (mass x concentration N) may be a better indicator of root function than root diameter.  相似文献

Increasing global temperatures could potentially cause large increases in root respiration and associated soil CO2 efflux. However, if root respiration acclimates to higher temperatures, increases in soil CO2 efflux from this source would be much less. Throughout the snow-free season, we measured fine root respiration in the field at ambient soil temperature in a sugar maple (Acer saccharum Marsh.) forest and a red pine (Pinus resinosa Ait.) plantation in Michigan. The objectives were to determine effects of soil temperature, soil water availability and experimental N additions on root respiration rates, and to test for temperature acclimation in response to seasonal changes in soil temperature. Soil temperature and soil water availability were important predictors of root respiration and together explained 76% of the variation in root respiration rates in the red pine plantation and 71% of the variation in the sugar maple forest. Root N concentration explained an additional 6% of the variation in the sugar maple trees. Experimental N additions did not affect root respiration rates at either site. From April to November, root respiration rates measured in the field increased exponentially with increasing soil temperature. For sugar maple, long-term Q10 values calculated from the field data were slightly, but not significantly, less than short-term Q10 values determined for instantaneous temperature series conducted in the laboratory (2.4 versus 2.62.7). For red pine, long-term and short-term Q10 values were similar (3.0 versus 3.0). Sugar maple root respiration rates at constant reference temperatures of 6, 18 and 24 degrees C were measured in the laboratory at various times during the year when field soil temperatures varied from 0.4 to 16.8 degrees C. No relationship existed between ambient soil temperature just before sampling and root respiration rates at 6 and 18 degrees C (P = 0.37 and 0.86, respectively), and only a very weak relationship was found between ambient soil temperature and root respiration at 24 degrees C (P = 0.08, slope = 0.09). We conclude that root respiration in these species undergoes little, if any, acclimation to seasonal changes in soil temperature.  相似文献

The prevalence of endocrine‐disrupting chemicals (EDCs) in the aquatic environment has been associated with the wide detection of alterations in the development and physiology of vertebrates. Zebrafish, as a model species, has been extensively used in toxicological research. In this review, we focus on recent published evidence of the harmful effects of EDCs on reproductive function in zebrafish, including skewed sex ratio, immature gonads, diminished sexual behaviour, decreased sperm count, reduced spawning and fertilization. These impairments mostly result from disruption to sex‐steroid hormones induced by endocrine disruptors. We also discuss other effects of exposure to EDCs. In EDC exposure research, despite incomplete assessments of altered gonad histopathology and sexual behaviour, these present potential effective biomarkers or pathways for evaluating the reproductive function in zebrafish on EDC exposure. To date, the pernicious effects of some EDCs on the reproductive performance in laboratory zebrafish are well understood; however, similar alterations remain for further determination in wild‐type fish and more kinds of EDCs. More studies should be performed under established scientific regulatory criteria to investigate the impact of EDCs on reproduction in zebrafish. Moreover, further research is required to explain the definite mechanism of sexual differentiation, which helps in understanding the shift of sexual phenotype with EDC exposure.  相似文献

Human activity has increased the amount of N entering terrestrial ecosystems from atmospheric NO3 deposition. High levels of inorganic N are known to suppress the expression of phenol oxidase, an important lignin-degrading enzyme produced by white-rot fungi. We hypothesized that chronic NO3 additions would decrease the flow of C through the heterotrophic soil food web by inhibiting phenol oxidase and the depolymerization of lignocellulose. This would likely reduce the availability of C from lignocellulose for metabolism by the microbial community. We tested this hypothesis in a mature northern hardwood forest in northern Michigan, which has received experimental atmospheric N deposition (30 kg NO3-N ha−1 y−1) for nine years. In a laboratory study, we amended soils with 13C-labeled vanillin, a monophenolic product of lignin depolymerization, and 13C-labeled cellobiose, a disaccharide product of cellulose degradation. We then traced the flow of 13C through the microbial community and into soil organic carbon (SOC), dissolved organic carbon (DOC), and microbial respiration. We simultaneously measured the activity of enzymes responsible for lignin (phenol oxidase and peroxidase) and cellobiose (β-glucosidase) degradation. Nitrogen deposition reduced phenol oxidase activity by 83% and peroxidase activity by 74% when compared to control soils. In addition, soil C increased by 76%, whereas microbial biomass decreased by 68% in NO3 amended soils. 13C cellobiose in bacterial or fungal PLFAs was unaffected by NO3 deposition; however, the incorporation of 13C vanillin in fungal PLFAs extracted from NO3 amended soil was 82% higher than in the control treatment. The recovery of 13C vanillin and 13C cellobiose in SOC, DOC, microbial biomass, and respiration was not different between control and NO3 amended treatments. Chronic NO3 deposition has stemmed the flow of C through the heterotrophic soil food web by inhibiting the activity of ligninolytic enzymes, but it increased the assimilation of vanillin into fungal PLFAs.  相似文献

Abstract

AIM: To investigate and characterise an inborn error of metabolism in a dog with skeletal and ocular abnormalities.

METHODS: A 2.5-year-old small male Miniature Poodle-like dog was presented with gross joint laxity and bilateral corneal opacities. Clinical examination was augmented by routine haematology, serum chemistry, radiographs, pathology, enzymology and molecular genetic studies. Euthanasia was requested when the dog was 3 years of age because of progressively decreasing quality of life.

RESULTS: Radiology revealed generalised epiphyseal dysplasia, malformed vertebral bodies, luxation/subluxation of appendicular and lumbosacral joints with hypoplasia of the odontoid process and hyoid apparatus. These clinical and radiographic findings, together with a positive urinary Berry spot test for mucopolysaccharides, and metachromatic granules in leucocytes, were indicative of a mucopolysaccharidosis (MPS), a lysosomal storage disease. Histological lesions included vacuolation of stromal cells of the cornea, fibroblasts, chondrocytes, macrophages and renal cells. The brain was essentially normal except for moderate secondary Wallerian-type degeneration in motor and sensory tracts of the hind brain. Dermatan sulphate-uria was present and enzymology revealed negligible activity of N-acetylgalactosamine-4-sulphatase, also known as arylsulphatase B, in cultured fibroblasts and liver tissue. A novel homozygous 22 base pair (bp) deletion in exon 1 of this enzyme's gene was identified (c.103_124del), which caused aframe-shift and subsequent premature stop codon. The “Wisdom pure breed-mixed breed” test reported the dog as a cross between a Miniature and Toy Poodle.

CONCLUSIONS: The clinicopathological features are similar to those of MPS type VI as previously described in dogs, cats and other species, and this clinical diagnosis was confirmed by enzymology and molecular genetic studies. This is an autosomal recessively inherited lysosomal storage disease.

CLINICAL RELEVANCE: The prevalence of MPS VI in Miniature or Toy Poodles in New Zealand and elsewhere is currently unknown. Due to the congenital nature of the disorder, malformed pups may be subject to euthanasia without investigation and the potential genetic problem in the breed may not be fully recognised. The establishment of a molecular genetic test now permits screening for this mutation as a basis to an informed breeding policy.  相似文献

A high-intensity laser was used to shock-compress liquid deuterium to pressures from 22 to 340 gigapascals. In this regime deuterium is predicted to transform from an insulating molecular fluid to an atomic metallic fluid. Shock densities and pressures, determined by radiography, revealed an increase in compressibility near 100 gigapascals indicative of such a transition. Velocity interferometry measurements, obtained by reflecting a laser probe directly off the shock front in flight, demonstrated that deuterium shocked above 55 gigapascals has an electrical conductivity characteristic of a liquid metal and independently confirmed the radiography.  相似文献

The spatially dependent de-excitation of a beam of metastable argon atoms, traveling through an optical standing wave, produced a periodic array of localized metastable atoms with position and momentum spreads approaching the limit stated by the Heisenberg uncertainty principle. Silicon and silicon dioxide substrates placed in the path of the atom beam were patterned by the metastable atoms. The de-excitation of metastable atoms upon collision with the surface promoted the deposition of a carbonaceous film from a vapor-phase hydrocarbon precursor. The resulting patterns were imaged both directly and after chemical etching. Thus, quantum-mechanical steady-state atom distributions can be used for sub-0.1-micrometer lithography.  相似文献

We examined the distribution of ground-flora species (herbaceous and woody species <1 m tall) across riparian areas of northeastern Wisconsin in an effort to understand how hierarchical landscape properties, such as the physiographic system (ground moraine and outwash plain), valley system (constrained and unconstrained), and valley floor landforms influence distribution patterns of ground-flora species and functional plant guilds across riparian areas of small streams and rivers in a glacial landscape. A total of 162 species were recorded on 417 (1 m2) plots stratified by four different valley types that reflect the dominant physiographic system and valley system (constrained ground moraine, constrained outwash plain, unconstrained ground moraine, unconstrained outwash plain) and transverse geomorphic structure (valley floor landforms including floodplains, terraces, slopes, and adjacent uplands). Although distribution patterns of individual ground-flora species are highly variable among the four valley types, canonical correspondence analyses (CCA) of individual valley types indicate that ground-flora vegetation is related strongly to hierarchical landscape properties, including valley type and the transverse geomorphic structure of the stream valley. Vegetation ultimately reflects the influence of hydrogeomorphic processes that shape valley floor landforms. Constrained valley types tend to be characterized by diverse floodplain ground-flora communities dominated by graminoids and pteridophytes, while the terraces and slope plant communities are comprised of facultative upland and obligate upland perennial forb and woody species whose distribution appears to be a function of topographic features, such as aspect. However, ground-flora communities of unconstrained valley types appear to be responding not only to changes associated with the transverse geomorphic structure of the riparian ecotone, but also to more localized changes in environmental conditions associated with flooding and hydrologic regime. As a result, floodplain, terrace, and slope ground-flora communities tend to be diverse, dominated by a mixture of obligate wetland, facultative wetland, and facultative species. Thus, variation in riparian plant community characteristics can be explained using a nested, hierarchical landscape framework to organize and group different riparian settings based on the underlying geomorphic processes shaping stream valleys. Based on these results, we suggest that riparian management zones (RMZs) designed to maintain riparian function should be variable in width rather than fixed, encompassing variation in valley floor landforms and valley walls, regardless of the physiographic system.  相似文献

We examined fine-root (< 2.0 mm diameter) respiration throughout one growing season in four northern hardwood stands dominated by sugar maple (Acer saccharum Marsh.), located along soil temperature and nitrogen (N) availability gradients. In each stand, we fertilized three 50 x 50 m plots with 30 kg NO(3) (-)-N ha(-1) year(-1) and an additional three plots received no N and served as controls. We predicted that root respiration rates would increase with increasing soil temperature and N availability. We reasoned that respiration would be greater for trees using NO(3) (-) as an N source than for trees using NH(4) (+) as an N source because of the greater carbon (C) costs associated with NO(3) (-) versus NH(4) (+) uptake and assimilation. Within stands, seasonal patterns of fine-root respiration rates followed temporal changes in soil temperature, ranging from a low of 2.1 micro mol O(2) kg(-1) s(-1) at 6 degrees C to a high of 7.0 micro mol O(2) kg(-1) s(-1) at 18 degrees C. Differences in respiration rates among stands at a given soil temperature were related to variability in total net N mineralized (48-90 micro g N g(-1)) throughout the growing season and associated changes in mean root tissue N concentration (1.18-1.36 mol N kg(-1)). The hypothesized increases in respiration in response to NO(3) (-) fertilization were not observed. The best-fit model describing patterns within and among stands had root respiration rates increasing exponentially with soil temperature and increasing linearly with increasing tissue N concentration: R = 1.347Ne(0.072T) (r(2) = 0.63, P < 0.01), where R is root respiration rate ( micro mol O(2) kg(-1) s(-1)), N is root tissue N concentration (mol N kg(-1)), and T is soil temperature ( degrees C). We conclude that, in northern hardwood forests dominated by sugar maple, root respiration is responsive to changes in both soil temperature and N availability, and that both factors should be considered in models of forest C dynamics.  相似文献

Pheromone-responsive neurons of insects not only require specific receptors but in addition several auxiliary components, including the “sensory neuron membrane protein,” SNMP. Accordingly, SNMP is considered as a marker for neurons responding to pheromones. For the desert locust Schistocerca gregaria, it is known that the behavior, including aggregation behavior and courtship inhibition, is largely controlled by pheromones. However, little is known about pheromones, their receptors, and the pheromone-responsive cells in locusts. In this study, we have identified two SNMP subtypes, SNMP1 and SNMP2, and compared their phylogenetic relationship and primary structure motifs with SNMPs from other species. Both SNMPs were found in chemosensory tissues, especially the antennae. Employing double in situ hybridization, we identified and localized the SNMP-expressing cells in the antennae. Cells expressing SNMP1 were localized to sensilla trichodea but also to sensilla basiconica, which in locust respond to pheromones. One or a few cells express SNMP1 within the multineuron clusters from sensilla basiconica, whereas the SNMP2 subtype was expressed in cells surrounding the neuron clusters, possibly supporting cells. Based on the finding that SNMP1 is expressed in distinct neurons under chemosensory sensilla, it is conceivable that these cells may represent pheromone-responsive neurons of the desert locust.  相似文献
