Cryopreservation has become anessential tool for operational application offorest tree embryogenic cultures, due to thelong evaluation periods needed for treesregenerated from these cultures. Fiveyellow-poplar (Liriodendron tulipifera)and seven sweetgum (Liquidambar spp.)embryogenic culture lines werestored in liquid nitrogen for 48 hours, afterwhich they were thawed and tested for regrowthand ability to produce somatic seedlings.Combinations of two sorbitol pretreatments andthree dimethylsulfoxide (DMSO) cryoprotectantlevels were evaluated for their impact onrecovery following cryogenic storage. The bestresults were obtained with 0.4 M sorbitol and5% DMSO, which provided 100% recovery.Somatic seedlings were regenerated from allculture lines and treatments, except for atransgenic sweetgum line.  相似文献

Near-infrared (NIR) spectroscopy is a rapid, non-destructive and accurate technique for analyzing a wide variety of samples, thus, the growing interest of using this technique in soil science. The objective of this study was to evaluate the potential of NIR spectroscopy to predict organic carbon (OC), total nitrogen (TN), available phosphorus (P) and available potassium (K) in the soil. NIR spectra from 20 cm3 of soil samples were acquired on the range of 750 to 2500 nm in diffuse reflectance mode, resolution of 16 cm?1 and 64 scans. Eight models of calibration/validation were constructed. Calibration and validation models showed that the predictive potential of NIR varied with the specific soil property (OC, TN, P and K) under evaluation and according to the methodology employed in the model construction (cross-validation or test set). Good prediction models were obtained for OC and TN content based on the statistical parameters. Test set methodology was able to predict soil OC, TN, P, and K better than cross-validation methodology.  相似文献

Real-time PCR with fluorogenic hydrolysis probes (5' nuclease assay) is increasingly used for the detection of pathogens for diagnostic purposes. Nevertheless, the size of the probes, usually 25–40 nucleotides, might limit their use to detect pathogens with high genome variability between isolates, where an identical sequence cannot be found without multiple mismatches. In this report, we describe a 5' nuclease assay, to detect Apple stem pitting virus, based on the use of a shorter probe which is chemically modified with a minor groove binder in order to increase duplex stability and raise the melting temperature to a value suitable for real-time analysis. The short size of the probe, which is critical to target a conserved cluster sequence of 14 nucleotides in the RNA polymerase gene, circumvents the genome variability of the virus. The assay correlates at 96 percent with gel analysis and is more reliable than biological indexing to detect Apple stem pitting virus field isolates. It is fast and fully compatible with automation, and therefore particularly suitable for plant certification.  相似文献

Daylily (Hemerocallis × hybrida Hort.) is a popular ornamental plant and hybrid cultivars are frequently selected for medium size, large number of flowers per stem, and flower color diversity. However, daylily selection is limited due to the lack of information on genetic parameters and their correlation to phenotypic characteristics. This study aimed at estimating genetic parameters and genetic correlations among full sib families of 13 daylily cultivars using REML/BLUP (restricted maximum likelihood/best linear unbiased prediction) and to select families with high genotypic values for plant height and number of floral buds per stem. Crosses were performed among those cultivars and 24 families were obtained. In the third year, plant height, leaf length and width, stem number, number of floral buds per stem, and number of floral buds per plant were evaluated. The estimates of average heritability of families were superior to the estimates of restrict individual heritability for all characters evaluated. High heritability estimates were obtained for plant height, leaf length, leaf width, stem number, and number of floral buds per plant. Selected families showed genetic gain of 19.80% for plant height, 32.48% for stem number, and 38.64% for number of floral buds per plant.  相似文献

The aim of the present study was to evaluate the effect of phloroglucinol in the recovery and survival of cryopreserved Dendrobium nobile protocorms. The exposure of protocorms to 2 M glycerol osmoprotective solution for 20 min followed by 10 min in PVS2 vitrification solution with 1% phloroglucinol resulted in the highest protocorm recovery and survival (68%). A positive effect of phloroglucinol was observed when combined with glycerol and PVS2. Phloroglucinol added at 1% provided an increase of over 100% in protocorm recovery and survival as compared to the same treatment without phloroglucinol. However, when sucrose was added to treatments, a negative effect was observed with a reduction in survival by 90%. Protocorms that survived cryopreservation were successfully regenerated into plants and acclimatized with 100% survival in greenhouse. Survival of cryopreserved D. nobile protocorms was a determining factor for seedling survival and growth into normal and fully functional plants. This study demonstrated an efficient procedure for cryopreservation and subsequent recovery and survival of cryopreserved D. nobile protocorms using phloroglucinol as a cryoprotectant additive.  相似文献

In vitro germination and seedling development from Dendrobium Swartz. hybrid ‘Sena Red’, ‘Mini WRL’, ‘Jaquelyn Thomas’, and ‘BFC Pink’ seeds cryopreserved through vitrification (PVS2) were evaluated. Germination percentages after cryopreservation (LN) were variable among different controls and treatments, despite the initial high seed viability for all hybrids. Seeds exposed to PVS2 at ice temperature from 1 to 3 h prior to LN exhibited significantly higher germination than seeds exposed to PVS2 at room temperature for the same time periods. No significant differences in germination percentages were observed for time exposure to PVS2 at 1, 2 or 3 h for ‘Sena Red’, ‘Mini WRL’, and ‘BFC Pink’. Seeds of ‘Jaquelyn Thomas’ exposed to 1 h prior to LN showed higher germination percentage than for exposure to 2 or 3 h. The combination of a pre-cooling treatment (ice) with a dehydration treatment (PVS2) for a period of time of 1–3 h was essential to allow proper germination of cryopreserved seeds. Although variability in seed germination among different hybrids and treatments was observed, germination was above 50% of the controls and all germinated seeds developed into normal seedlings with healthy shoot and root formation. No abnormalities, nutritional deficiencies, or diseases were observed in developed seedlings and no significant differences were observed for seedling growth and development from germinated seeds among the different hybrids. Seedlings transplanted to pots acclimatized well and developed into normal plants within 6–8 months in greenhouse. Transplanted seedlings exhibited 100% survival for all hybrids.  相似文献

Liners of angelonia ‘Pink’ (Angelonia angustifolia Benth.), shooting star (Pseuderanthemum laxiflorum), coreopsis ‘Early Sunrise’ (Coreopsis grandiflorum Hogg ex Sweet.), and scutellaria ‘Purple Fountains’ (Scutellaria costaricana H. Wendl.) were transplanted into containers filled with, by volume, 1) 100% compost; 2) 60% compost, 25% vermiculite, 15% perlite; 3) 30% compost, 30% sphagnum peat, 25% vermiculite, 15% perlite; 4) 0% compost, 60% sphagnum peat, 25% vermiculite, 15% perlite. Two compost products were evaluated: SW, a 1:1 by volume mixture of partially composted seaweed and partially composted yard trimmings and SYT, a 1:1 by weight mixture of biosolids compost and yard trimmings compost. There was no difference in angelonia shoot dry mass among the different percentages of SW compost but angelonia plants grown in 0 and 30% SYT had greater shoot dry mass than plants grown in 60 or 100% SYT. Shoot dry mass of shooting star plants increased as the percentage of SYT compost increased from 0 to 30 % and then decreased while shoot dry mass of plants grown in SW compost decreased as the percentage of compost increased. There was no difference in coreopsis shoot dry mass or scutellaria shoot dry mass between the two compost products and there also was no difference in shoot dry mass among the different percentages of either compost product.  相似文献

Wild blueberries are a rich source of polyphenols and other compounds that are highly metabolized by the intestinal microbiota and may, at the same time, affect the intestinal environment itself. A repeated-measure, crossover dietary intervention on human volunteers was designed to study the effect of six week consumption of a wild blueberry ( Vaccinium angustifolium ) drink, versus a placebo drink, in modulating the intestinal microbiota. Relative to total eubacteria, Bifidobacterium spp. significantly increased following blueberry treatment (P ≤ 0.05), while Lactobacillus acidophilus increased after both treatments (P ≤ 0.05). No significant differences were observed for Bacteroides spp., Prevotella spp., Enterococcus spp., and Clostridium coccoides . Bifidobacteria, which have been largely proposed to be of benefit for the host, appeared to be selectively favored suggesting an important role for the polyphenols and fiber present in wild blueberries. Results obtained suggest that regular consumption of a wild blueberry drink can positively modulate the composition of the intestinal microbiota.  相似文献
