Water absorption in individual white-core and non-white-core grains of rice varieties used in sake brewing was observed during soaking by magnetic resonance imaging (MRI). Water was absorbed faster in white-core grains than in non-white-core grains and was pooled in the core. Moisture distributions were compared using the line profiles of nuclear magnetic resonance signal intensities (SI profiles) generated from magnetic resonance images of grains soaked for 2 h. SI profiles of white-core and non-white-core grains overlapped despite slight differences in the central and intermediate regions of the grains. A white-core-like structure with loosely packed starch granules was found to exist in the endosperm of non-white-core grains. SI profiles of grains polished to 70% of the yield weight of brown rice were similar in shape to those of grains polished to 90% of the yield, but their overall moisture content was higher after the removal of the hydrophobic grain periphery. A varietal difference in the thickness of the intermediate region, which may affect fissuring resistance in the polishing process, was detected in SI profiles. Differences in water absorbability between parents and their offspring were also apparent from their SI profiles. MRI enabled the detection of variety-specific water-absorption properties in grains.  相似文献

Coxiella burnetii is the causative agent of Q fever, and the main route of infection in humans is inhalation of contaminated aerosols. Although oral transmission by contaminated raw milk or dairy products is also a possible route of human infection, there have been few studies investigating the presence of C. burnetii in dairy products. We developed a new method of extracting DNA from cheese and detecting C. burnetii DNA in cheese samples with a nested PCR assay. The limit of detection was 6.0 × 10(2) C. burnetii particles per gram. We subsequently used this method to examine the presence of C. burnetii in cheese at commercial markets in Tokyo from June 2005 to December 2008. Twenty-eight of 147 cheese samples were found to be positive for C. burnetii DNA. However, when we assessed the viability of C. burnetii by inoculating mice with DNA-positive samples, all of the samples were found to be negative. Thus, the viability of C. burnetii appears to have been lost in these cheese samples.  相似文献

Salmonella enterica serovar Choleraesuis (S. Choleraesuis) isolates derived from diseased pigs in Japan during 2001 and 2005 were analyzed for biotype, based on H(2)S production and dulcitol fermentation, pulsed-field gel electrophoresis (PFGE) profile, and antimicrobial resistance profile. S. Choleraesuis biotype Choleraesuis (biotype Choleraesuis) was classified into one genotype, while varietas Kunzendorf (var. Kunzendorf) was classified into two genotypes. The isolates of var. Kunzendorf belonging to one genotype were isolated in a limited area of Japan. Variation in the antimicrobial resistance pattern was observed in isolates of both biotypes Choleraesuis and var. Kunzendorf. We have also shown that the PFGE profile was associated with the biotype and isolation region of each isolate.  相似文献

The water penetration into a rice grain during soaking was monitored by three-dimensional gradient echo magnetic resonance imaging, which has the advantage of a short measurement time and a sufficient sensitivity for low-moisture samples. The water penetration route was compared between the milled and brown rice grains of two japonica cultivars. In normal milled rice grains of Koshihikari, water first penetrated the ventral side surface and the embryo attachment site of the endosperm, then migrated along the central line and transverse cracks, and finally diffused to all parts of the endosperm. In milled rice grains of Yamadanishiki with a white core, water quickly infiltrated into the cracks or chalky parts on the dorsiventral line and then diffused to the lateral side. In brown rice grains of both cultivars water penetrated extremely slowly and did not infiltrate directly into transverse cracks or the white core due to inhibition by the pericarp and seed coat. The central part of every grain allowed more water to penetrate. The route, pattern and speed of water penetration were determined by the morphological structure, crack formation and hardness distribution associated with the packing of the starch granules in the grain.  相似文献

We performed biosafety assessments of transgenic poplars prior to field trials. Constitutive expression of the Aspergillus aculeatus xyloglucanase in Populus alba increased the cellulose content and specific gravity of its stem, the leaves of which were visibly greener, thicker, and smaller than those of the wild-type plant. Although the young transgenic poplars grew faster than the wild type in a growth chamber, there was no distinguishable difference in growth between the poplars when they were placed in a special screened greenhouse. Allelopathic tests showed that the transgenic poplars do not produce harmful substances. Based on all the biosafety assessments and the scientific literature on poplar species, we came to the conclusion that transgenic poplars probably do not disturb the biological diversity of the surrounding environment, even when they are submitted to field trials.  相似文献

The long-term changes of the area of Sasa nipponica grassland, bark stripping damage to trees by sika deer, Cervus nippon, and sika deer densities on Mt. Ohdaigahara were assessed with aerial photographs, information from previous studies, and field experiments. The grasslands expanded year after year. The expansion rate was highest from 1982 to 1992, and lowest from 1992 to 1997. Bark-stripping damage on coniferous trees was heavier than that on broadleaf trees in both the early 1980s and 1998. The deer density in the eastern part of the area had been high (14.4–64.3 km−2) from 1984 until 1998. We presume that rich growth of S. nipponica and heavy feeding pressure by sika deer over the long term caused expansion of S. nipponica grasslands by killing canopy trees by bark stripping and preventing forest regeneration.  相似文献

A total of 518 fecal samples collected from 183 apparently healthy cattle, 180 pigs and 155 broilers throughout Japan in 1999 were examined to determine the prevalence and antimicrobial susceptibility of Salmonella. The isolation rates were 36.1% in broilers, 2.8% in pigs and 0.5% in cattle. S. enterica Infantis was the most frequent isolate, found in 22.6% of broiler fecal samples. Higher resistance rates were observed against oxytetracycline (82.0%), dihydrostreptomycin (77.9%), kanamycin (41.0%) and trimethoprim (35.2%). Resistance rates to ampicillin, ceftiofur, bicozamycin, chloramphenicol and nalidixic acid were <10%. CTX-M-2 β-lactamase producing S. enterica Senftenberg was found in the isolates obtained from one broiler fecal sample. This is the first report of cephalosporin-resistant Salmonella directly isolated from food animal in Japan.  相似文献

The flagellin genes (fliC) of Clostridium chauvoei, Clostridium haemolyticum, Clostridium novyi types A and B, and Clostridium septicum were analysed by PCR amplification and DNA sequencing. The five Clostridium species have at least two copies of the flagellin gene (fliC) arranged in tandem on the chromosome. The deduced N- and C-terminal aminoacid sequences of the flagellin proteins (FliCs) of these clostridia are well conserved but their central region aminoacid sequences are not. Phylogenic analysis based on the N-terminal aminoacid sequence of the FliC protein revealed that these clostridia, which belong to Clostridium 16S rDNA phylogenic cluster I (), are more closely related to Bacillus subtilis than to Clostridium difficile, which belongs to the cluster XI. Moreover, a multiplex polymerase reaction (PCR) system based on the fliC sequence was developed to rapidly identify C. chauvoei, C. haemolyticum, C. novyi types A and B, and C. septicum. PCR of each Clostridium amplified a species-specific band. The multiplex PCR system may be useful for rapid identification of pathogenic clostridia.  相似文献
