Brucellosis is a zoonotic disease worldwide distributed and having the economic as well as public health importance. The prevalence of brucellosis among sheep flock having history of abortions was studied. A total of 229 samples comprising of 157 blood and 72 clinical samples (vaginal swabs) were collected from 157 animals. Clinical samples were processed for the isolation of Brucella melitensis. Serum samples (n = 157) were tested by Rose Bengal plate test (RBPT) and i-ELISA. A total of 68 (43.31%) and 104 (66.24%) samples were positive by RBPT and ELISA, respectively. Brucella isolates (n = 2) were recovered from clinical samples. Both isolates demonstrated amplification for bcsp 31 and IS711 genes. On AMOS PCR, both the isolates amplified at 731 bp, i.e., belongs to B. melitensis species. The incidence of B. melitensis in a migratory flock warns the thorough testing and culling of Brucella-infected sheep from the flock on a continuous basis; otherwise, such incidence will be routine and poor farmers will be at a loss.  相似文献

Two wide hybrids, Diplotaxis erucoides (2n = 14) × Brassica rapa (2n = 20) and B. maurorum (2n = 20) × B. rapa, were developed using the sequential ovary–ovule culture. Reciprocal crosses failed, possibly as a consequence of strong unilateral incompatibility. The F 1 hybrids in each combination were completely male sterile and morphologically intermediate to the respective parents. DNA marker polymorphism and chromosome counts confirmed their hybrid nature. High frequency of bivalents in the F 1 and the presence of trivalents/quadrivalents in the derived amphiploids suggested genomic duplications and homoeology of the parental genomes. Up to three homoeologous pairs between the D. erucoides (DeDe) and B. rapa (AA) genomes, and one between B. maurorum (BmBm) and B. rapa genomes were observed. Successful synthesis of the F 1 hybrids and amphiploids of B. rapa with D. erucoides and B. maurorum, and allosyndetic chromosome pairing are expected to permit introgressions of desirable loci into the cultivated Brassica germplasm, especially for resistance to Alternaria brassicae and Albugo candida.  相似文献

A diverse array of wild and weedy crucifers was screened under laboratory conditions for their resistance to Lipaphis erysimi (Kaltenbach) (Homoptera: Aphididae). Among these, Brassica fruticulosa and Brassica montana were found to be the most promising. The availability of a synthetic amphiploid, AD-4(B. fruticulosa × Brassica rapa var. brown sarson) as well as a set of Brassica juncea lines carrying genomic introgressions from B. fruticulosa allowed us to investigate B. fruticulosa resistance in greater detail. This assessment was carried out along with susceptible check B. rapa ssp. brown sarson cv. BSH-1 in a series of choice and no choice experiments. The mustard aphid showed maximum preference for feeding on BSH-1 while least preference was recorded for B. fruticulosa followed by AD-4 as evidenced by the number of aphids that settled on circular leaf bits of these genotypes 24 and 48 h after release in a choice experiment. Brassica fruticulosa exhibited strong antibiosis against mustard aphid in no choice experiment and all the released aphids died within 5–8 days of their release, while the maximum survival (76.7%) was recorded on BSH-1. The survival on AD-4 (40%) was significantly lower than that on BSH-1. Almost similar trend was observed with respect to other demographic parameters of L. erysimi viz. development time, fecundity and longevity. In the screen house studies, there was no seedling mortality in B. fruticulosa and AD-4 after 30 days of aphid release while 80% mortality was observed on BSH-1. Excellent variation for aphid resistance was recorded in B. juncea introgression lines, emphasizing heritable nature of fruticulosa resistance. The biochemical analysis suggested the possibility of high concentration of lectins to be associated with low aphid infestation in B. fruticulosa.  相似文献

Sclerotinia rot caused by the fungus Sclerotinia sclerotiorum is one of the most serious and damaging diseases of oilseed rape and there is keen worldwide interest to identify Brassica genotypes with resistance to this pathogen. Complete resistance against this pathogen has not been reported in the field, with only partial resistance being observed in some Brassica genotypes. Introgression lines were developed following hybridization of three wild crucifers (viz. Erucastrum cardaminoides, Diplotaxis tenuisiliqua and E. abyssinicum) with B. napus or B. juncea. Their resistance responses were characterized by using a stem inoculation test. Seed of 54 lines of B. napus and B. juncea obtained from Australia, India and China through an Australian Centre for International Agricultural Research (ACIAR) collaboration programme were used as susceptible check comparisons. Introgression lines derived from E. cardaminoides, D. tenuisiliqua and E. abyssinicum had much higher levels (P < 0.001) of resistance compared with the ACIAR germplasm. Median values of stem lesion length of introgression lines derived from the wild species were 1.2, 1.7 and 2.0 cm, respectively, as compared with the ACIAR germplasm where the median value for stem lesion length was 8.7 cm. This is the first report of high levels of resistance against S. sclerotiorum in introgression lines derived from E. cardaminoides, D. tenuisiliqua and E. abyssinicum. The novel sources of resistance identified in this study are a highly valuable resource that can be used in oilseed Brassica breeding programmes to enhance resistance in future B. napus and B. juncea cultivars against Sclerotinia stem rot.  相似文献

Forest dieback caused by climate-change associated stresses and insect outbreaks has emerged as a global concern, and the biogeochemical consequences of this phenomenon need to be elucidated. We measured biological and chemical traits of soil beneath live trees or trees recently killed by a mountain-pine-beetle outbreak in a subalpine coniferous forest in the Front Range of Colorado. We focused on the top 5 cm of mineral soil just beneath the O horizon and measured microbial biomass, soil invertebrate abundance and composition, and soil chemical characteristics. With the termination of inputs from rhizodeposition, mycorrhizal fungal turnover and fine root turnover, soil total carbon (C) and total nitrogen (N) in the mineral soil at three sites decreased by 38–49% and 26–45%, respectively. Tree mortality was associated with reduced soil microbial biomass but soil nematode and microarthropod densities were unchanged. Nematode trophic structure was altered with an increased proportion of bacterial feeders. Soil inorganic N concentrations were inversely correlated to microbial C:N ratios. Tree death was associated with increased soil pH, a possible loss of calcium (Ca2+), but an accumulation of soil inorganic N, largely as NH4+. Our results suggest that forest dieback results in rapid C and N loss from surface mineral soils and that the accumulation of soil inorganic N, the reduction in microbial biomass, and the more bacterial-based soil food web increase the potential of enhanced N loss from affected ecosystems.  相似文献

A pot-culture experiment was conducted to assess the bioavailability of iron from spinach cultivated in soil fortified with graded levels of iron and zinc (FeSO4 . 7H20 and ZnSO4 . 7H20, respectively). Applications of varying levels of iron to soil increased the total iron and phosphorus contents and decreased the zinc content (P<0.05). The effect of applying varying levels of zinc was the opposite of on the minerals in spinach. The ascorbic acid content was remarkably reduced with varying levels of iron and zinc. Higher levels of zinc and lower levels of iron in the soil increased the bioavailability of iron from spinach (P<0.05). In conclusion, the interactions of 15 ppm zinc with 30 ppm iron significantly enhanced the bioavailability of iron, total iron and zinc contents.  相似文献

The differential proteins associated with plasma membrane of spermatozoa are less known, identification of which shall help overcome limitations of currently used methods of sperm sexing, considered as a high priority for livestock sector of many countries. This study has reported plasma membrane proteomics of unsorted spermatozoa and differential expression of plasma membrane-associated proteins between X- and Y-chromosome bearing spermatozoa of indicus cattle (Bos indicus). Isolation of plasma membrane fraction using percoll gradient, relatively a rapid method, from bovine spermatozoa has been reported to enrich isolation of plasma membrane proteins. Significant enrichment for plasma membrane-associated proteins was observed in plasma membrane fraction (p < .05) as compared to the total cell lysate using LC-MS/MS. Furthermore, these experiments were conducted in flow cytometry sorted, sexed-semen samples. Thirteen proteins were identified as differentially abundant between X- and Y-sorted spermatozoa. Among these, two proteins were downregulated in Y-sorted spermatozoa compared to the X-sorted spermatozoa (p < .05), while four and seven proteins could be noted in X- and Y-sorted spermatozoa, respectively. Proteins that are presumed to support sperm capacitation and sperm migration velocity were found to be abundant in Y-sorted spermatozoa while those associated with structural molecule activity were identified as abundant in X-sorted spermatozoa in the present study. Our study provides better insight into the plasma membrane proteomics of spermatozoa of indicus cattle and furnishes data that might aid in design and development of alternate and open technology for sex-sorting of semen.  相似文献
