‘BJ 104’ was the most widely grown pearl millet hybrid in India until it became susceptible to downy mildew (DM) in 1984—85. Residual variability for resistance was found in both parental lines, 5141 B (maintainer of 5141 A) and J 104, and through four generations of pedigree selection under intense disease pressure in the DM nursery, two lines, IC-MA841 (from 5141 B) and ICMP 84814 (from J 104), were selected resulting in a reconstituted DM resistant hybrid (‘ICMH 84814’) which was equal in yield to the original ‘BJ 104’. The reconstituted hybrid, though phenotypically similar, can be distinguished from “BJ 104” being slightly taller, flowers later, has heavier heads, and 1000-seed weight, but tillers less. A similar exercise was attempted on Tifton 23 B, the female parent of the first widely grown hybrid (‘HB 3’— the male parent was also J 104), but no variability for resistance to DM was found.  相似文献

Sunspots, dark magnetic regions occurring at low latitudes on the Sun's surface, are tracers of the magnetic field generated by the dynamo mechanism. Recent solar dynamo models, which use the helioseismically determined solar rotation, indicate that sunspots should form at high latitudes, contrary to observations. We present a dynamo model with the correct latitudinal distribution of sunspots and demonstrate that this requires a meridional flow of material that penetrates deeper than hitherto believed, into the stable layers below the convection zone. Such a deep material flow may have important implications for turbulent convection and elemental abundance in the Sun and similar stars.  相似文献

The reversible blockage of synthetic pores formed by rigid-rod beta barrels, either by substrates or products, was used to sense a variety of enzymatic reactions in high-throughput format with "naked-eye" fluorescent detection. Improvement of sensor sensitivity beyond three orders of magnitude by straightforward internal mutations underscores the functional plasticity of rigid-rod beta barrels. Such detectors of enzyme activity with the aforementioned characteristics are needed in areas as diverse as proteomics and environmentally benign organic synthesis.  相似文献

Current widespread and intensive soil degradation in India has been driven by unprecedented levels of population growth, large-scale industrialization, high-yield agriculture, urban sprawl and the spread of human infrastructure. The damage caused to managed and natural systems by soil degradation threatens livelihoods and local services and leads to national socio-economic disruption. Human-induced soil degradation results from land clearing and deforestation, inappropriate agricultural practices, improper management of industrial effluents and wastes, careless management of forests, surface mining, urban sprawl, and ill-planned commercial and industrial development. Of these, inappropriate agricultural practices, including excessive tillage and use of heavy machinery, over-grazing, excessive and unbalanced use of inorganic fertilizers, poor irrigation and water management techniques, pesticide overuse, inadequate crop residue and/or organic carbon inputs, and poor crop cycle planning, account for nearly 40% (121 Mha) of land degradation across India. Globally, human activities related to agriculture contribute to the transgression of four of the nine Planetary Boundaries proposed by Rockström et al. (2009): Climate Change, Biodiversity Integrity, Land-system Change, and altered Phosphorus and Nitrogen Biogeochemical Flows. This review focuses on how knowledge of soil processes in agriculture has developed in India over the past 10 years, and the potential of soil science to meet the objectives of the United Nations' Sustainable Development Goal 2: Zero Hunger (End hunger, achieve food security, improved nutrition and promote sustainable agriculture), using the context of the four most relevant Planetary Boundaries as a framework. Solutions to mitigate soil degradation and improve soil health in different regions using conservation agricultural approaches have been proposed. Thus, in this review we (1) summarize the outputs of recent innovative research in India that has explored the impacts of soil degradation on four Planetary Boundaries (Climate Change, Biodiversity Loss, Land-system Change, and altered Biogeochemical Flows of Phosphorus and Nitrogen) and vice-versa; and (2) identify the knowledge gaps that require urgent attention to inform developing soil science research agendas in India, to advise policy makers, and to support those whose livelihoods rely on the land.  相似文献

A 60 days feeding trial was conducted to illustrate the effect of graded levels of protein on the growth and metabolic enzymes of grey mullet (Mugil cephalus L.) fingerlings reared in inland saline water (ISW). Six isoenergetic (16 MJ/kg) and isolipidic (60 g/kg) diets containing 240, 260, 280, 300, 320 and 340 g crude protein (CP)/kg diet were formulated and fed to triplicate. Weight gain %, specific growth rate, protein utilizing efficiency, feed efficiency and RNA:DNA ratio were significantly higher (p < .05) in 320 and 300 g CP/kg diets. Fish fed with 240 g CP/kg diet showed significantly higher (p < .05) feed intake, whole‐body lipid content, hepatosomatic index value and liver glycogen content. Transaminase enzymes and malate dehydrogenase activities were elevated in fish fed 340 g CP/kg diet. Protease activity increased with increasing dietary CP level, but amylase activities showed an inverse relationship. No significant (p > .05) variations were observed for lactate dehydrogenase, oxidative stress enzymes, blood parameters and serum osmolality among all the treatment groups, but red blood cell count increases with increasing dietary CP levels. Based on the results, feeding dietary protein level of 300 g CP/kg is economically viable for rearing of grey mullet in ISW.  相似文献

Tail and fin rot disease (TFRD) is a big issue in the production of spotted snakehead, Channa punctata Bloch. The aims of the present study were to isolate and identify the bacterial pathogen causing TFRD, to detect histopathological changes in tissues (fin, tail, liver, and kidney), and to ascertain the antibiotic sensitivity pattern of the isolate. Out of six bacterial isolates, only the isolate DJ1990 was found to be the causal candidate of TFRD in C. punctata. Identical histopathological changes were detected in tail, fin, liver, and kidney under light and scanning electron microscope in both collected diseased fish and artificially infected fish. The isolate was identified as Pseudomonas aeruginosa strain DJ1990 (National Center for Biotechnology Information Ace. No. KX709967) based on the biochemical characterization tests and 16S rDNA sequence‐based phylogeny analysis. Artificial challenge test demonstrated that the strain DJ1990 was highly virulent (100% mortality at 48 h of postinjection period at the concentration of 1.5 × 107 CFU/g of body weight) for C. punctata. The isolate exhibited sensitivity to the broad‐spectrum antibiotics but was resistant against aztreonum. To the best of our knowledge, this is the first report of P. aeruginosa as a TFRD‐causing candidate in C. punctata.  相似文献

水稻单片段替换系群体的建立及QTL定位   总被引:3,自引:0,他引:3  
水稻中许多重要的农艺性状属数量性状,由多基因(QTL)控制。研究QTL的遗传特性和遗传效应对于培育高产和稳产的水稻品种具有十分重要的意义。本研究以6个优良的品种为供体亲本,以华粳籼74为轮回亲本,通过微卫星标记辅助回交选择培育了一批单片段替换系,随后利用所培育的单片段替换系进行了QTL分析和基因定位。主要结果有:1、利用258个微卫星标记对6个供体亲本和轮回亲本间的多态性进行了筛选。6个供体亲本与轮回亲本间的多态率在32.98%至60.78%之间,平均47.81%,粳型供体亲本比籼型供体亲本的多态性要高。2、随着回交代数的增加,植株所含的替换片段数逐渐减少。在BC2F1、BC3F1、BC3F2和BC3F3代,平均每个植株携带有12.50、5.98、1.69和1.46个替换片段。替换片段的平均长度也随回交和自交代数的增加而逐渐变短。在BC2F1、BC3F1、BC3F2和BC3F3代,替换片段的平均长度分别为25.43cM、22.38cM、20.78cM和18.15cM.回交世代替换片段变短的速率(11.99%)比自交世代变短速率(7.15%)要快。在BC2F1、BC3Fl、BC3F2和BC3F3代,轮回亲本基因组的恢复率分别为82.24%、92.55%、98.04%j阳98.52%。3、在BC3F2和BC3F3代,共选育出111个单片段替换系,其中独一无二的单片段替换系共42个。BC3F2代替换系中替换片段的估算长度在2.00cM到64.80cM之间,平均为21.75cM,而BC3F3代中替换片段的估算长度在6.05cM到48.90cM之间,平均为20.95cMc,12条染色体中仅第11染色体没有选择到单片段替换系。所选育的单片段替换系中替换片段的总长为2367.50cM,基因组的覆盖长度为704.50cM,覆盖率为39.25%。4、在52个单片段替换系的22个性状中共鉴定出了234个QTL。每个性状鉴定出的QTL数在3到19个之间,平均4.50个。每个替换系鉴定出的QTL在2到15个之间,平均10.64个。QTL加性效应的大小因性状和替换系不同而不同,低至-0.02(0.79%)的加性效应(如谷粒宽度)均能检测到。在RM237.RM322、RM225和RM481标记附近的各种性状中同时检测到了增效和减效的QTL。5、通过染色体替换作图,对7个单片段替换系中16个性状的QTL进行了定位。利用携带有相同替换片段的替换系进行QTL位置的确定。6、通过分析复杂性状的加性效应,对影响植高、每穗粒数和粒型的相关性状进行了分析,分析QTL之间的相关性。7、对控制抽穗期、稃尖颜色、植株高度和粒型的基因进行了定位。抽穗期基因Hd-8表现为单基因显性遗传,定位于第8染色体上,与PSMl55紧密连锁。紫色稃尖基因Pa-6表现为单基因显性遗传,定位于第6染色体上,与RM253紧密连锁。株高基因Ph-1-3定位于第1染色体上,与PSM331紧密连锁。粒型基因Rlw-8-2为首次报道,定位于第8染色体的末端,与RM447紧密连锁,长粒表现为单基因隐性遗传。本研究通过分子标记辅助选择培育了一批单片段替换系,并成功地对这批材料进行了QTL分析和基因定位,从而证实了在水稻中通过微卫星标记辅助回交选育单片段替换系和进行QTL分析的可行性。  相似文献

The present study determined the effective concentrations of clove oil and MS‐222 in juvenile rohu Labeo rohita for quick induction and recovery. The immune‐biochemical responses due to 0, 1 and 24 hr exposure to those anaesthetics were also evaluated. Of four concentrations of the anaesthetics examined, the lowest effective concentration of clove oil and MS‐222 were 50 µl/L and 125 mg/L respectively. Clove oil and MS‐222 significantly increased the myeloperoxidase, total protein and alkaline phosphatase activity at some of the holding durations. However, superoxide anion production (after 0 and 1 hr) and antiprotease activity (after 24 hr) were significantly reduced in fish exposed to clove oil. Serum glucose content was significantly elevated in the MS‐222‐treated group. Furthermore, the clove oil‐treated group showed significantly higher levels of serum Na+ and K+, while the aspartate and alanine aminotransferase activities were significantly enhanced in the MS‐222 group. The use of both clove oil and MS‐222 is advised as an anaesthetic agent for rohu with a bias towards clove oil, considering its economic and operational feasibility.  相似文献
