Purification of a lipoxygenase enzyme from the cultivar Tresor of durum wheat semolina (Triticum turgidum var. durum Desf) was reinvestigated furnishing a new procedure. The 895-fold purified homogeneous enzyme showed a monomeric structure with a molecular mass of 95 +/- 5 kDa. Among the substrates tested, linoleic acid showed the highest k(cat)/K(m) value; a beta-carotene bleaching activity was also detected. The enzyme optimal activity was at pH 6. 8 on linoleic acid as substrate and at pH 5.2 for the bleaching activity on beta-carotene, both assayed at 25 degrees C. The dependence of lipoxygenase activity on temperature showed a maximum at 40 degrees C for linoleic acid and at 60 degrees C for bleaching activity on beta-carotene. The amino acid composition showed the presence of only one tryptophan residue per monomer. Far-UV circular dichroism studies carried out at 25 degrees C in acidic, neutral, and basic regions revealed that the protein possesses a secondary structure content with a high percentage of alpha- and beta-structures. Near-UV circular dichroism, at 25 degrees C and at the same pH values, pointed out a strong perturbation of the tertiary structure in the acidic and basic regions compared to the neutral pH condition. Moreover, far-UV CD spectra studying the effects of the temperature on alpha-helix content revealed that the melting point of the alpha-helix is at 60 degrees C at pH 5.0, whereas it was at 50 degrees C at pH 6.8 and 9.0. The NH(2)-terminal sequence allowed a homology comparison with other lipoxygenase sequences from mammalian and vegetable sources.  相似文献

In this study, for the first time, the impact of the genetic factor on the contents of oleuropein in olive leaves was not only evaluated but the influence exerted by the color/age of leaves (green, green-yellowish, and yellow) and the collecting period (spring or autumn) was also evaluated. A repetitive high-resolution gas chromatographic quantitation method and an accurate high-performance liquid chromatographic method were developed. These analytical methods gave results showing a highly linear relationship. Samples of olive leaves were taken from seven major Italian olive cultivars, such as Dritta, Leccino, Caroleo, Coratina, Castiglionese, Nebbio, and Grossa di Cassano. Such a vegetal raw material could actually be exploited for recovering oleuropein, considered to be a high-added value molecule. This could be converted into hydrxytyrosol, a compound known to possess strong bioactive properties. Olive leaves showed considerable contents of oleuropein, which with some cultivars were even higher with respect to those present in the corresponding olive fruits (reported in the literature). The amounts of oleuropein in the collected leaves were markedly modified by the color/age and genetic factors, whereas meaningless variations were ascribable to the quantitation method and the collecting period factors. Various chemometrics, applied to the obtained analytical data, appeared to be effective in discriminating the samples on the basis of the above-examined experimental factors, thus confirming how these should be taken into account in future industrial recovery of oleuropein from olive leaves.  相似文献

Mapping quantitative trait loci (QTLs) responsible for malting quality traits in barley populations has been the main genetic approach to malting quality breeding. A ‘winter × spring’ doubled haploid barley population ‘Nure’ × ‘Tremois’, where such traits were segregating, has been recently developed. Our objective was to map QTLs for malting quality from 2 years of trials in two contrasting locations. QTLs were found on six chromosomes, with a main cluster on chromosome 1H. For wort viscosity and malt extract, favourable alleles at two loci on chromosome 5H were carried by the winter feeding parent ‘Nure’. Doubled‐haploids with higher quality than the spring malting cultivar ‘Tremois’ showed either a facultative or a winter growth habit and a level of frost tolerance comparable with that of the winter tolerant parent ‘Nure’. Markers and QTLs of quality traits were further validated on a separate set of DH lines, coming from the same cross, by means of marker‐assisted selection. This showed that, at least in the present cross combination, positive contributions to malting quality can be found in winter feed barley.  相似文献

No-tillage (NT) is becoming increasingly attractive to farmers because it clearly reduces production costs relative to conventional tillage (CT). However, many producers in southern Italy are reluctant to adopt this practice because NT can have contrasting consequences on grain yield depending on weather conditions. The effect of NT and CT on continuous durum wheat (Triticum durum Desf.) under rainfed Mediterranean conditions was studied, over a 3-year period (2000–2002) at two locations (Foggia and Vasto) in southern Italy. Yield, grain quality [thousand kernel weight (TKW), test weight (TW) and protein content (PC)] and soil water content were assessed.Higher yield was obtained with NT than CT in the first 2 years at Foggia. In contrast, mean yield and quality parameters at Vasto were similar for the two treatments, except in the third year in which CT produced more than NT (4.6 Mg ha−1 versus 2.9 Mg ha−1). At Foggia, TW and TKW were higher in NT than CT in all years. Highest PC was obtained under CT (19.6% and 15.5% for CT versus 14.7% and 11.4% for NT, respectively, in the growing season 2000–2001 and 2001–2002) indicating that grain was shriveled with low starch accumulation.At Foggia, where this study was part of a long-term experiment started in 1995, a strong correlation was observed between yield and rainfall during the wheat growing season. The coefficient of determination (R2) values for CT and NT were 0.69* and 0.31 ns, respectively, and the regression straight line crossed around 300 mm of rainfall. These results indicate that NT was superior below this rainfall value, whereas more rainfall enhanced yield in CT. We conclude that NT performed better at Foggia with limited rainfall during the durum wheat growing season. The superior effect of NT in comparison to CT, was due to lower water evaporation from soil combined with enhanced soil water availability.  相似文献

The aim of this study was to evaluate the performance of three species of unhulled-wheat (Triticum monococcum L., Triticum dicoccum Schubler and Triticum spelta L.) with respect to durum wheat (Triticum durum Desf. cv. Trinakria) in southern Italy's flat environment.
As far as yield is concerned, the mean yield was significantly different among species. Durum wheat yielded 16.1 %, 37.6% and 69.5 % more than dicoccum, spelta and monococcum species, respectively.
Among unhulled-wheat species, only Triticum dicoccum showed some similarity with durum wheat, such as one-thousand kernels weight, plant height and number of spikes per m2.

Test weight or bulk density in durum wheat is a physical quality characteristic considered by semolina millers. High test weight values are desirable because they positively influence market grade and price. This study reports data on kernel size features, determined on a sample size of only 25 kernels, replicated three-times, of 16 commercial durum wheat cultivars grown in two locations in southern Italy, to ascertain whether some kernel traits could be related to test weight. For each year and cultivar, the analysis of variance for all of characteristics showed that sample size effect was not significant, enabling the use such a small sample for further investigations. The kernel trait with the highest variation for year was kernel width. The absolute variation of 1994 with respect to 1993 growing season was 30.9%. While kernel weight or volume did not correlate with test weight, a negative association with kernel length (r = -0.61, P = 0.05) and perimeter (r = -0.57, P = 0.05) was found. The kernel shape factors, rectangular aspect ratio (RAR) and circularity shape factor (CSF), showed a positive correlation with test weight (r = 0.51, P = 0.05 and r = 0.59, P = 0.05, respectively). The shape factors were negatively correlated both with kernel length and perimeter and positively with kernel width. A predictive model for test weight (TW = 38.7 + 5.1·ETW, where the estimated test weight [ETW] was computed as individual kernel weight/estimated kernel volume [EKV] ratio), was highly correlated with actual test weight values (r = 0.82, P = 0.001). The effectiveness of the linear model was confirmed when a set of 10 advanced lines of durum wheat were considered, although a slightly lower correlation (r = 0.73, P = 0.05) between actual and predicted test weight values was found. Because the extent of predictability of this approach might be more effective in early-generation lines, the application of the findings in a durum wheat breeding program would be advisable.  相似文献

The aim of this study was to evaluate the agronomical performance of old unhuUed wheat species (farro) in comparison with an old cultivar of durum wheat, selected for morphological and rusticity characteristics similar to farro, in a drought-flat land environment of southern Italy. The trials were carried out during the 1992/93 and 1993/94 growing seasons. Agronomical responses concerned the behaviour of the different species to weeding treatment (diclofop-methyl, 568 g a.i, ha−1) and to three (0%, 50%, 100%) artificial weed levels with Avenafatua and Phalaris arundinacea with respect to grain yield and its components, plant morphological aspects and two qualitative characters. In all of the species the weeding treatment increased the mean values of almost all the parameters analysed except for days to head and grain protein content which remained constant. The effect of weed treatment on the characters considered was a general decrease of mean values when weed level was increased. The grain protein content showed an opposite trend. As far as the grain yield alone is concerned, yielding differential in medium and large farro decreased by 3% whereas in small farro it increased by 4.3% vs, durum wheat when no weeding and weeding treatments were compared. Small and large farro resulted more susceptible than medium farro and durum wheat to weed treatment. In fact, these latter showed, at the maximum weed level, the same grain yield loss as small and large farro at the 50% weed level. Small farro was the most susceptible species, while Triticum dicoccum showed similar affinity and behaviour to durum wheat.  相似文献

The unpredictability of the Mediterranean climate causes a large fluctuation in wheat yield and quality but offers the opportunity for the production of high quality wheats which are lacking in the European Union.

This paper describes the effects of nitrogen fertilization rate and timing on five Triticum aestivum L. cultivars differing in bread-making quality, cultivated in six representative Italian sites (years/locations). Nitrogen was applied at each location at two rates, the first corresponding to the amount estimated to maximize grain yield, the second 30% higher. Three timings of nitrogen applications were tested including a late application at the boot stage. Bread-making quality was evaluated with the Chopin alveograph and each sample was assigned to the following qualitative classes of the Italian market: Class 1, improver wheat; Class 2, for direct bread-making; Class 3, ordinary wheat.

Cultivars differed significantly in all agronomic and qualitative traits. Grain yield was highest in the northern location (7.1 t ha−1) and lowest in the south of the Italian peninsula (2.61 ha−1), while in Sicily abundant rains in both years enabled a yield of 6.41 ha−1 to be obtained. As far as bread-making quality is concerned, the greatest proportion of grain samples belonging to Class 1 was obtained from the northern location. The year of cultivation strongly affected quality, particularly with regard to gluten tenacity, in all locations, though the quality ranking of the cultivars remained substantially stable across locations. Nitrogen applications greater than those considered optimal to maximize yield, combined with a better distribution during the life cycle, significantly improved bread-making quality.

It was concluded that high quality wheats can be obtained in a wide range of growing conditions in the Mediterranean climate. However the likelihood of genotype × environment interactions, statistically significant for both yield and quality, call for a more precise management of nitrogen fertilization in relation to the cultivars chosen and the climatic features of each site.  相似文献

In the last few years, the renewed interest for emmer wheat (Triticum dicoccon Schrank) in Italy has stimulated breeding programs for this crop releasing improved genotypes obtained not only by selection from landraces, but even by crosses with durum wheat (Triticum durum Desf.) varieties. The purpose of this work has been to uncover the genetic make-up of some emmer × durum derivatives, specifically by comparing the differences from their parents. Genetic diversity of advanced breeding lines and varieties derived from a durum × emmer cross has been evaluated on the basis of AFLP and SSR markers in comparison with the corresponding emmer and durum wheat parent for addressing the seminal question of how much ‘wild’ variation remains after selection for agronomic type.  相似文献

Proton high‐resolution magic angle spinning nuclear magnetic resonance (1H HR‐MAS NMR) has been applied for the analysis of two cultivars of durum wheat produced in different Italian geographical areas. Furthermore, on the same samples, isotopic ratios were measured by mass spectrometry (IRMS). The application of chemometrics to these results permitted the discrimination of semolina by cultivar and geographical origin. A similar approach has been applied to the results obtained from chemical analyses. The comparison showed that NMR spectroscopy can provide a faster method for the detection of differences among the durum wheat semolina according to geographical and varietal origin. Furthermore, IRMS data are suitable to provide information about the geographical origin of samples. This present investigation is part of an extensive research project to find a scientific method capable of classifying wheat foods for the assignment of a “denomination of origin” trademark.  相似文献
