Possibility of using interspecific hybridization to increase genetic diversity of drought tolerance in safflower (Carthamus tinctorius L.) and effectiveness of different selection methods is poorly understood. The main objectives of this study were to evaluate the effectiveness of (a) interspecific hybridization to expand genetic diversity in safflower for agronomic traits and drought tolerance in advance generations and (b) different selection methods (direct vs. indirect) for improving seed yield. Interspecific hybridizations were performed between species of C. tinctorius × C. palaestinus (TP), C. palaestinus × C. oxyacantha (PO) and C. tinctorius × C. oxyacantha (TO). Based on the field screening of genotypes in F3 generation, four groups of selected genotypes (derived from combination of direct and indirect selection under both drought and normal environments along with selection based on drought tolerance index (STI)) were evaluated in F4 and F5 generations. Indirect selection for seed yield was significantly more efficient than direct selection. Selection in stress environment resulted to more efficiency than selection in normal environment. No significant difference was observed between direct selection and selection based on STI. The result showed that TP was the best population to combine high seed yield and drought tolerance in safflower. 相似文献

Wild species of safflower, Carthamus oxyacanthus Bieb., is highly crossable with cultivated species, C. tinctorius L. and could be directly exploited in broadening safflower gene pool and improving the crop for biotic and abiotic stress
environments. In this study, genetic diversity among accessions of C. oxyacanthus and their relationships with cultivated safflower were evaluated using agro-morphological traits and polymorphic inter-simple
sequence repeats (ISSR) markers. Significant variation was observed among accessions particularly for seeds per capitulum,
seed yield per plant, harvest index and capitula per plant. Cluster analysis based on agro-morphological traits classified
the wild accessions in two groups according to their geographical regions, and separated them from the cultivated genotypes.
ISSR marker also revealed a high genetic variation among the accessions, and cluster analysis based on this marker divided
genotypes into four groups, with cultivated ones in a separate clade. Genetic variation observed among the wild safflower
germplasm at the DNA level was higher than the agro-morphological traits, indicating that ISSR is an effective marker system
for detecting diversity among safflower genotypes and their genetic relationships. Accessions of C. oxyacanthus with high genetic relationship to cultivated species could be used for interspecific hybridization in breeding programs of

Microsatellite markers were used to analyse the biodiversity of 57 accessions of different subspecies and varieties of wild
Aegilops tauschii (2n = 2x = 14; D genome) collected across the major areas where it grows in Iran. Levels of diversity were high, with numbers of alleles
averaging 7.3 (ranging up to 12) and polymorphism information contents averaging 0.6591. One accession was notably more similar
to two of the D genome in hexaploid wheats (Triticum aestivum) used as outgroups. Within the Ae. tauschii accessions, no markers were characteristic for taxa or geographical origin, suggesting high gene flow between the subspecies
and varieties, although some groupings, which could be related to geographical origin, were evident. This survey demonstrates
the high diversity present in wild goatgrass in Iran, and indicates that there is value in sampling for useful genes for wheat

Safflower (Carthamus tinctorius L.) is an important oil seed crop with growing importance in many countries around the world. However, safflower fly (Acanthiophilus helianthi Rossi) is one of the main limiting factors to expand the production area of the crop in several countries. Since host plant-resistance is the most efficient method for pest management, we evaluated a germplasm of wild accessions (Carthamus oxyacanthus Bieb.) with mostly brown–black seed coat color along with some white-seeded cultivated safflower genotypes for resistance to safflower fly. The results showed that there was a significant difference between two species for safflower fly damage with all cultivated genotypes being susceptible. However, for the accessions of C. oxyacanthus, a wide range of resistance was observed. In a choice experiment and natural infestation, 35.3–78.9 percent of heads per plant were infested in genotypes of C. tinctorius while for the accessions of C. oxyacanthus this was much smaller and ranged from 1.6 to 13.6%. In addition, percentage of seed yield loss per infested plant was more drastic in cultivated safflower (29.0–72.8%) than the wild accessions (0.0–21.4%). In the no-choice experiment, fly population was dramatically decreased in the wild accessions due to larval mortality and possibility of antibiosis. There was a strong relationship between brown–black seed coat color and resistance to safflower fly indicating the possibility of using this trait in breeding programs of safflower to develop fly-resistant cultivars. 相似文献

The diploid goatgrass Aegilops
tauschii is considered the D-genome donor of bread wheat and has probably a centre of diversity in north of Iran. In order to measure
the genetic diversity of and the relationships among different populations, varieties and subspecies belonging to Ae. tauschii in Iran, DNA was extracted from 48 accessions of Ae. tauschii collected across the geographic range of the species in the Country and the genetic diversity was assessed using AFLPs based
on eight PstI/MseI +3 primer pairs resulted in 277 bands, 198 of which were polymorphic. High level polymorphism was detected, with an average
of polymorphism rate of 0.715; relatively low genetic similarity (0.455) between accessions and significant difference between
the lowest (0.179) and the highest genetic similarity (0.817). The Iranian Ae. tauschii populations showed high level of genetic diversity. The populations studied were divided into two groups: one group was mainly
representing Northern populations collected from Southern Caspian Sea shore and the other group was mainly representing Northeast
and Northwest populations. Based on the results of this study, it can be suggested that Ae. tauschii possesses two separate gene-pools in Iran: Northern and Northeastern–Northwestern. Considering the needs for introducing
new characteristics and alleles for wheat improvement purposes, Ae. tauschii Iranian gene-pool is assumed to be of high importance for more investigation in the future. 相似文献

BACKGROUND: Phelebotumus papatsi is considered to be the main vector of zoonotic cutaneous leishmaniasis as well as sand fly fever in Iran. There are several measures for vector control, with emphasis on insecticides. The objective of this study was to determine the baseline susceptibility of this vector to the commonly used insecticides in an endemic focus of the disease in central Iran. P. papatasi collected from the field were used for susceptibility status. Its baseline susceptibility to DDT and pyrethroids was assessed on about 6866 specimens collected from Badrood rural district, Esfahan Province, Iran, during the summer of 2010. The LT50 and LT90 values were measured according to the World Health Organisation (WHO) test using probit analysis and regression lines. RESULTS: Results of tests against female P. papatasi revealed LT50 values to DDT 4%, permethrin 0.75%, deltamethrin 0.1%, cyfluthrin 0.15% and lambdacyhalothrin 0.05% of 1104.97, 182.35, 26.79, 15.42 and 1.48 s respectively. The figures for male P. papatasi were 973.51, 59.5, 4.4, 2.65 and 1.5. CONCLUSION: The results of this study provide a guideline for implementation of different vector control measures. Furthermore, guidelines are needed for monitoring and evaluation of insecticide susceptibility tests against sand flies. Copyright © 2012 Society of Chemical Industry 相似文献

The mountain rye Secale strictum is native to the Middle East and is the progenitor of the cultivated rye. Regarding lack of information about the genetic
diversity of this species in Iran, this study was aimed to evaluate its genetic variation and to examine the patterns of diversity
related to the varieties and geography. Fifteen wheat and rye derived microsatellite markers were used to achieve this aim.
High levels of diversity, with an average number of 6.1 alleles per locus (ranging up to 11) and high level polymorphism with
polymorphism rate averaging 0.624 (between populations) and 0.357 (within populations) were observed among 125 individuals
from 19 populations collected from various regions of Iran. The Northwestern populations showed the highest and the Northern
populations showed the lowest polymorphism and diversity. One population of the Northwest of the country was notably closer
in its allele range to the S. cereale accessions used as outgroup. No taxon or geographic specific marker was detected, suggesting high gene flow between the populations,
however some groupings which can be related to the geographic regions and varieties, were evident. The analysis of molecular
variance attributed same portions of genetic diversity to the within and between populations with no significant variation
among different geographic regions. The results of this study indicated that the Iranian genepool of Secale strictum is valuable to search for new useful alleles for crop improvement. 相似文献

Domestication and selection significantly changed phenotypic and behavioural traits in modern domestic animals. In this study, to identify the genomic regions associated with mastitis, genomic data of German Holstein dairy cattle were analysed. The samples were genotyped using the Bovine 50 K SNP chip. For each defined healthy and sick group, 133 samples from 13,276 genotyped dairy cows were selected based on mastitis random residual effects. Grouping was done to infer selection signatures based on XP-EHH statistic. The results revealed that for the top 0.01 percentile of the obtained XP-EHH values, five genomic regions on chromosomes 8, 11, 12, 14 and 26 of the control group, and four regions on chromosomes 3, 4 (two regions) and 22 of the case group, have been under selection. Also, consideration of the top 0.1 percentile of the XP-EHH values, clarified 21 and 15 selective sweeps in the control and case group, respectively. This study identified some genomic regions containing potential candidate genes associated with resistance and susceptibility to mastitis, immune system and inflammation, milk traits, udder morphology and different types of cancers. In addition, these regions overlap with some quantitative trait loci linked to clinical mastitis, immunoglobulin levels, somatic cell score, udder traits, milk fat and protein, milk yield, milking speed and veterinary treatments. It is noteworthy that we found two regions in the healthy group (on chromosomes 12 and 14) with strong signals, which were not described previously. It is likely that future research could link these identified genomic regions to mastitis. The results of the current study contribute to the identification of causal mutations, genomic regions and genes affecting mastitis incidence in dairy cows. 相似文献
