Spleen cells from Toxoplasma lysate antigen (TLA)-sensitized BALB/c mice showed the strong cytotoxic activity against both natural killer (NK)-sensitive cells (YAC-1 and RL male-1) and NK-insensitive cells (P-815), when incubated with TLA or recombinant human IL-2 (rhIL-2). The increment of TLA concentration in culture medium increased the cytotoxic activity. Treatment of effector cells; spleen cells from TLA-sensitized mice incubated with TLA, with anti-asialo GM1 or anti-Thy-1 plus complement inhibited the cytotoxic activity of effector cells, whereas treatment with anti-mouse Lyt-2.2 serum plus complement had no effect on the cytotoxic activity. Treatment of spleen cells from TLA-sensitized mice with anti-asialo GM1 and/or anti-Thy-1 plus complement inhibited cytotoxic activities of effector cells. These results suggested that spleen cells sensitized with TLA both in vivo and in vitro were asialo GM1 positive and Thy-1 positive, and the majority of cytotoxic cells induced by TLA were similar to lymphokine-activated killer (LAK) cells induced by IL-2.  相似文献

We investigated the effect of mixed rearing of barrows and gilts on the backfat thickness and the serum metabolite profiles of Kagoshima-Kurobuta (Berkshire) pigs. A total of 149 pigs with an average body weight of 35 kg were divided into the following groups: 100%, 90%, 70%, 50%, 30%, 10%, and 0% groups consisting of 10 barrows (1 pen), 9 barrows + 1 gilt (3 pens), 7 barrows + 3 gilts (2 pens), 5 barrows + 5 gilts (3 pens), 3 barrows + 7 gilts (2 pens), 1 barrow + 9 gilts (3 pens), and 9 gilts (1 pen), respectively. All pigs were raised to a shipping weight of 120 kg. Mixed rearing significantly reduced (p < 0.001) backfat thickness, and the optimum mixing ratio of barrows and gilts was 7:3 (the 70% group). Four types of circulating sex steroids were found in both the barrows and gilts in the 50% group but were not detected in barrows from the 100% group. These results indicated that mixed rearing of barrows and gilts was effective for reducing the backfat thickness of barrows, and induced sex steroid hormones may influence the backfat thickness of barrows in mixed-reared groups.  相似文献

DNA from 111 ticks collected by flagging in Tokachi district, Eastern Hokkaido, Japan were examined for infection with Rickettsia and Ehrlichia, by PCR and sequencing methodology. For Rickettsia, analysis of the partial sequence of the citrate synthase gene was successfully performed on 11 DNA samples from I. persulcatus, and 7 of them showed 99.8% identical with Rickettsia helvetica while the other 4 showed 99.8% identical with ;Candidatus Rickettsia tarasevichiae'. For Ehrlichia, a partial sequence of the 16S rRNA gene detected from I. persulcatus was 100% identical with that from Ehrlichia muris, and another DNA sample from I. ovatus showed 99.8% identical with Ehrlichia species detected from I. ovatus. The results suggest that the pathogens detected here might be distributed in this area.  相似文献

The usefulness of magnetic resonance (MR) is already established, but it has a disadvantage of requiring a long scanning time. A short-time examination is more or less needed so as to be more practical in veterinary clinics. A protocol of the short-time MR examination was devised based on parameters determined, and validity of the protocol was assessed through the diagnosis of clinical cases with intervertebral disc diseases. With this protocol, it was possible to complete an MR examination for the spine within 15 min. The MR images and myelographic findings were correlated well in this study, suggesting the short-time protocol of MR examination can be used in the clinical diagnosis of spinal diseases.  相似文献

The process of decline of an endangered tree species,Pinus armandii var.amamiana, was monitored on the southern slope of Mt. Hasa-dake in Yaku-shima Island from 1994 to 1998. There are 163 trees ofP. armandii var.amamiana over 1.3 m in height. They are distributed on steep ridges and rocks with a thin soil layer mostly consisting of friable granite. During the monitoring period, 21 trees ofP. armandii var.amamiana died and the mortality rate was 12.9%. Dead trees were categorized into three types: standing, uprooted and landslide. The uprooted- and landslide-typed dead trees were found only after a severe typhoon struck Yaku-shima Island. This suggests that the combination of fragile site conditions and severe typhoons play an important role in the process of decline ofP. armandii var.amamiana. The standing-typed dead trees were presumed to have been killed by pine wilt disease, accounting for 71.4% of the dead trees. However,Bursaphelenchus xylophilus, the cause of pine wilt disease, was not detected from any of the wood chips or branch samples from the standing-typed dead trees ofP. armandii var.amamiana. This might indicate that some factor(s) other than pine wilt disease could be responsible for the standing-typed death ofP. armandii var.amamiana in natural habitats. Environmental factors, such as global solar radiation, wind speed, air temperature, humidity, and CO2 concentration, were measured above and within the canopy of a tropical rainforest in Lambir Hills National Park, Sarawak, Malaysia. Few data concerning the environment of this forest have been reported. Intensive observations were carried out in 1998, 1999, and 2000 with the following results: (1) The fraction of global solar radiation reaching the upper layer of the canopy varied with global solar radiation above the canopy. Even though the global solar radiation above the canopy fluctuated, the fraction of that reaching the lower canopy and the ground was constantly approximately 5%. (2) The fraction of wind speed reaching each layer of the canopy increased with wind speed above the canopy. Little wind was usually present at the lower canopy. (3) The daytime air temperature at the canopy top was higher than that near the ground. The maximum difference between the air temperature at the canopy top and that at the ground was about 5°C, and the diurnal temperature ranges at the canopy top and those at the ground were about 8°C and about 5°C, respectively. The highest daytime water vapor pressure occurred within the canopy and particularly near the ground. Vertical gradients of water vapor pressure during the day were steep, probably because of high transpiration. (4) In the 1998 observation the minimum and the maximum CO2 concentrations were 360 ppm in the day and 450 ppm at night, while in the 2000 observation the minimum and the maximum CO2 concentrations were 350 ppm in the day and 540 ppm at night. The higher CO2 concentration in the daytime and the lower concentration at night observed during the 1998 observation period were probably due to reduced photosynthesis and soil respiration caused by exceptional dry conditions during the observation period.  相似文献

A total of 518 Gerres sp. were collected around Okinawa Island, Japan, from November 2002 to July 2005, with monthly sampling where the standard length of females (n=218) were 56.2–147.1 mm, and males (n=149) were 62.2–139.4 mm. The maximum ages observed for females were 5+ years and males were 4+ years, estimated by transverse sectioned sagittal otoliths. Mean marginal increment indicated that opaque rings were formed once a year during April to July. The standard length (SL; mm) — body wet weight (BW; g) relationships were described as BW=(3.26×10−5) SL2.97 and BW=(3.13×10−5) SL 2.98 for females and males, respectively, and the standard length at age described by von Bertalanffy growth function for females, L t=137.1(1−e−0.80[t+0.80]) and males, L t=127.3(1−e−0.82[t+0.93]).  相似文献

ABSTRACT:   The diet shift of larval and juvenile Ryukyu-ayu Plecoglossus altivelis ryukyuensis was investigated for the landlocked population stocked in the Fukuji Reservoir, Okinawa Island. Gut contents of 274 specimens (5.1–31.4 mm in body length) including 31 yolk-sac larvae (5.7–8.0 mm) were examined. The diet of larval and juvenile landlocked P. a. ryukyuensis was mainly composed of zooplankton such as dinoflagellates, rotifers, copepods, and cladocerans. Feeding incidence was 48% in total and it gradually increased with growth. Diet analysis clearly demonstrated a diet shift during the larval and juvenile periods. For yolk-sac larvae and 5.0–19.9 mm fish, the diet was dominated by dinoflagellates Gymnodinium sp., and Peridinium sp., both numerically and by frequency, followed by rotifers that mainly consisted of Polyarthra spp. Diet breadth (Levin's standardized index BA) diversified with growth, and feeding on copepods and cladocerans gradually increased in 10.0–14.9 mm fish. Diet overlap (Morisita's index: C) showed that diet drastically changed in 20.0–24.9 mm fish with a specialization on cladocerans Bosminopsis deitersi and Diaphanosoma brachyurum . This diet shift probably corresponded with an increase of feeding ability resulting from an enlargement of the mouth, increased swimming ability, and the development of sense organs.  相似文献

When 8-week-old BALB/c mice were sensitized with two intramuscular injections of Toxoplasma lysate antigen (TLA) at 2 week interval, the numbers of sIg(+), Thy-1,2(+), Lyt-1,2(+) Lyt-2,2(+), and Asialo GM1(ASGM1)(+) cells in the spleen, liver and peripheral blood increased by 2 to 4 times over those found in unsensitized mice of the same age. When TLA-sensitized and unsensitized mice were infected with Babesia, 4 of 10 (40%) of the TLA-sensitized mice survived infection, while none of the unsensitized control mice lived longer than 14 days after Babesia infection. By contrast, sensitization of nude mice with TLA had no effect on survival, and mice did not live more than 12 days. The number of thymic Thy-1,2(+) cells decreased in TLA-sensitized and unsensitized BALB/c mice by almost 80% within 10 days after infection (AI). During the same time, the numbers of B cells, T cells, and NK cells increased in the spleen, liver and peripheral blood of both sensitized and unsensitized mice. Especially notable were increases in numbers of Lyt-2,2(+) cells in the spleen and blood and increases in numbers of NK cells in the spleen, liver and blood in both TLA-sensitized and unsensitized mice. When spleen cells from TLA-sensitized and unsensitized mice were cultured in the presence or absence of TLA for 6 days, assays for cytotoxicity using NK-insensitive P-815 target cells and NK-sensitive YAC-1 target cells demonstrated higher rates of cytotoxicity in cultures of TLA-sensitized spleen cells.  相似文献

The cyclobutadiene (CBD) molecule C(4)H(4) deviates from a high-symmetry square geometry to compensate for its antiaromatic electronic structure. Here, we report a CBD silicon analog, Si(4)(EMind)(4) (1), stabilized by the bulky 1,1,7,7-tetraethyl-3,3,5,5-tetramethyl-s-hydrindacen-4-yl (EMind) groups, obtained as air- and moisture-sensitive orange crystals by the reduction of (EMind)SiBr(3) with three equivalents of lithium naphthalenide. X-ray crystallography reveals a planar and rhombic structure of the Si(4) four-membered ring, with alternating pyramidal and planar configurations at the silicon atoms. The large (29)Si chemical shift differences (Δδ > 350 parts per million) in the solid-state nuclear magnetic resonance spectra suggest a contribution of an alternately charge-separated structure. The rhombic-shaped charge-separated singlet state of compound 1 thus stabilizes its cyclic 4π-electron antiaromaticity in a manner that contrasts sharply with the bond-length alternation, characterizing the rectangular distortion of carbon-based CBD.  相似文献
