A study was conducted to determine the extent of genetic diversity among African cassava (Manihot esculenta Crantz) accessions resistant to the cassava mosaic virus disease (CMD), using simple sequence repeat (SSR) markers. The accessions
included a breeding stock (clone 58308), five improved lines, 62 CMD resistant and 10 CMD susceptible landraces. Genetic diversity
was assessed among accessions in five cluster groups derived from UPGMA analysis on data from 18 SSR primer pairs. Average
gene diversity, He, was high in all cluster groups, with an average heterozygosity of 0.591 ± 0.061. The estimator of inbreeding
Fis revealed a low level of inbreeding within groups and averaged −0.262 ± 0.142. Gene diversity among all accessions was
51.4% and gene diversity within cluster groups was 46.6%, while 4.8% was due to diversity between the different cluster groups.
The amount of genetic differentiation measured by Gst and Fst were 9.6% and 12.1% respectively, indicating a weak genetic

Common bean is one of the widely consumed food security crop in Africa, Asia and South America. It is a rich source of protein, minerals and micronutrients. High genotype by environment interaction is one of the main challenge in breeding for high grain micronutrient concentration. The objective of this study was to estimate SNP markers associated with grain Fe and Zn concentration using 289 common bean genotypes and 11,480 SNP markers. The study revealed that 43 quantitative trait loci (QTLs) were associated with grain Fe and Zn concentration. Five quantitative trait nucleotides (QTNs), that is, QTN Fe_1.1, QTN Fe_6.3, QTN Fe_6.5, QTN Fe_10.3 and QTN Fe_11.6 were detected both at Haramaya and Melkassa locations. Two of the markers, that is, QTN Fe_6.3 and QTN Fe_6.5, were located on chromosome 06 while QTN Fe_1.1, QTN Fe_10.3 and QTN Fe_ 11.6 were residing on chromosomes 01, 10 and 11, respectively. Among these, QTN Fe_11.6 had a large and positive consistent effect across locations. The five stable QTNs along with the potential candidate genes could be used for Fe biofortification through marker assisted selection. 相似文献

We evaluated the agronomic performance and diversity for morphological traits among a collection of local and exotic cassava genotypes in the Guinea Savannah agroecology of Ghana. One hundred and fifty cassava genotypes were assessed using 28 agro-morphological (18 qualitative and 10 quantitative) traits recorded from three months after planting until harvesting at 12 months after planting. There was significant genetic variability among the genotypes for most of the traits evaluated. High broad sense heritability was observed for most of the traits. Principal component analyses of the traits indicated that the first three principal component axes which accounted for more than 55% of the variation for 10 quantitative traits included storage root yield, number of leaves, harvest index, height at first branching, and plant height as the most important traits. A greater proportion of the phenotypic variances of plant height, height at branching, and harvest index were accounted for by genetic variances indicating that progress could made with selection. Pearson correlation analysis indicated that a number of traits were related to growth and yield of cassava in the Guinea Savanna ecology, making indirect selection for root yield possible. The study revealed ample genotypic variation in these morphological and agronomic traits to support selection for improved varieties. 相似文献

Striga hermonthica can cause as high as 100% yield loss in maize depending on soil fertility level, type of genotype, severity of infestation and climatic conditions. Understanding the mode of inheritance of Striga resistance in maize is crucial for introgression of resistance genes into tropical germplasm and deployment of resistant varieties. This study examined the mode of inheritance of resistance to Striga in early‐maturing inbred line, TZdEI 352 containing resistance genes from Zea diploperennis. Six generations, P1, P2, F1, F2, BC1P1 and BC1P2 derived from a cross between resistant line, TZdEI 352 and susceptible line, TZdEI 425 were screened under artificial Striga infestation at Mokwa and Abuja, Nigeria, 2015. Additive‐dominance model was adequate in describing observed variations in the number of emerged Striga plants among the population; hence, digenic epistatic model was adopted for Striga damage. Dominance effects were higher than the additive effects for the number of emerged Striga plants at both locations signifying that non‐additive gene action conditioned inheritance of Striga resistance. Inbred TZdEI 352 could serve as invaluable parent for hybrid development in Striga endemic agro‐ecologies of sub‐Saharan Africa. 相似文献

Fifty cassava clones were studied using RAPD technique. They included landraces from the Wenchi, Nkoranza, Dormaa Ahenkoro
and Asonafo districts of the Brong Ahafo region of Ghana and three improved varieties. Genetic diversity of these genotypes
was studied using four primers, OPK-01, OPR-02, OPR-09 and OPJ-14. A total of 41 different bands were detected. Levels of
polymorphic fragments detected by the four primers ranged from 90% to 100%. By pooling bands from individual accessions together,
mean number of fragments per accession per primer ranged from 5.50±1.04 for the Improved cultivars to 7.00±0.71 for populations
of landraces from Dormaa. Mean frequencies of fragments not detected by the primers for the accessions were 0.524±0.12, 0.460±0.12,
0.561±0.12 and 0.523±0.12 for landraces from Wenchi, Nkoranza, Dormaa Ahenkro, Asonafo and the Improved varieties, respectively.
The grand mean frequency of individuals showing fragments not present in populations was 0.522±0.10. Genetic diversity estimates
ranged from 0.290 to 0.425 (mean 0.352±0.05) for primer OPK-01, 0.001 to 0.381 (mean 0.309±0.06) for primer OPR-02, 0.335
to 0.344 (mean 0.283±0.04) for primer OPR-09 and 0.152 to 0.352 (mean 0.261±0.07) for primer OPJ-14. Within the accessions
mean gene diversity estimates were 0.316±0.03, 0.293±0.09, 0.331±0.02, 0.322±0.07 and 0.247±0.03 for accessions from Wenchi,
Nkoranza, Dormaa Ahenkro, Asonafo districts and the Improved varieties, respectively. Interpopulational genetic divergence
ranged from 0.069 to 0.203 (mean 0.119±0.04). Rate of nucleotide substitution among the landraces was 9.8 per cent per site
per year, while that for the Improved varieties was 15 per cent.

Complete indehiscence of anthers accompanied with exerted stigmas outside the hull could be useful in hybrid rice breeding if the genetics and possible environmental effects are well understood. Studies were undertaken to identify the sources, confirm the mode of inheritance, and examine the possible environmental effects and potential application of anther indehiscence with exerted stigmas in hybrid rice breeding. Direct and reciprocal crosses were made between Jasmine 85, selected interspecific and intersubspecific lines and two wide compatibility varieties. The resulting F1 progenies were evaluated for their expression of anther indehiscence and exerted stigmas. Expression of anther indehiscence with exerted stigmas by both direct and reciprocal crosses suggested no effect of cytoplasmic factors on the expression of the trait. Segregation analysis supported the 7:1 (indehiscence:dehiscence) mode of inheritance. Temperature and relative humidity could not modulate the fertility/sterility status of anther indehiscent plants. Utilization of anther indehiscence with exerted stigmas in hybrid rice breeding was hindered by trait fixation and propagation. 相似文献

Citrus Black Spot (CBS), caused by Guignardia citricarpa, was detected for the first time in Ghana and in West Africa. The disease was first observed in the Eastern Region in 1999 with typical disease symptoms including hard spot, virulent spot and false melanose were observed on several citrus species. A survey revealed that the disease has reached epidemic levels in the citrus-producing areas of the Eastern and Ashanti regions and is spreading rapidly within these areas and to other regions of the country. Currently, CBS is the most important fruit disease of citrus in Ghana, causing about 22% crop loss. Although the disease does not cause postharvest decay and the internal quality of the fruit is not affected, significant amounts of blemished fruit are discarded at the markets. Disease incidence and severity was found to be higher on mature than on young citrus trees. Pycnidia were found on fruit with hard spot symptoms, and pycnidia and pseudothecia typical of Guignardia spp. were found on decomposing leaves. Two species, G. citricarpa and G. mangiferae, were isolated from 15% of the samples collected and identified using the Oatmeal Agar test and by PCR with species-specific DNA primers. Isolates of G. citricarpa produced CBS symptoms after 80 to 233?days on 75% of the artificially inoculated young fruit of Valencia Late sweet orange. The fungus was re-isolated from symptomatic, inoculated fruit completing Koch’s postulates. Isolates of the endophyte G. mangiferae did not induce symptoms in the pathogenicity tests. In epidemiological studies, infections were detected from November to February for the minor cropping season and from May to November for the major season. Fruit of Valencia Late sweet orange were susceptible to G. citricarpa infection for up to 7?months after petal fall. Knowledge of the disease cycle in Ghana will improve methods for disease control. 相似文献

This study was carried out to assess genotypic variability in abscisic acid content, carbon isotope ratio, and their relationship to storage root yield and yield components in cassava under irrigation and moisture stress. The study involved 20 cassava genotypes arranged in randomized complete block design with three replications. Irrigation water was applied using a drip irrigation system with a discharge rate of approximately 5.33 L m-2 hr-1. Significant (P < 0.05) genotypic variability was observed for all physiological, growth, and yield traits assessed. Abscisic acid content was higher under stress than irrigation and negatively correlated with root yield (r = -0.45), harvest index (r = -0.43), and above-ground biomass yield (r = -0.20) indicating that it can be used as indirect selection criteria against unproductive genotypes. Carbon isotope ratio was significantly and positively correlated with above-ground biomass yield (r = 0.20) but not root yield (r = 0.09). Estimates of genotypic variability indicated high values for most of the growth and yield components but low heritability values for abscisic acid content, carbon isotope ratio, stomatal conductance, and root yield under stress conditions. However, higher estimates were recorded under irrigation and in the combined analysis. It was also found from this study that carbon isotope ratio influences above-ground biomass but not storage root yield under stress conditions. The results from this study provide useful information on the relationship between abscisic acid content, carbon isotope discrimination, and storage root yield in field-grown cassava. 相似文献
