The phylogenetic relationship between different Cichorium intybus cultivars and cultivar groups was studied. A total of 127 random amplified polymorphic DNA markers have been used to distinguish between 51 Cichorium intybus cultivars belonging to three different cultivar groups. A principal component analysis was performed on the complete data set and a clear grouping of chicory cultivar groups was observed. The three chicory groups tested (root chicory, witloof and pain de sucre) were separated from one another in the principal component plots. The first two axes accounted for 43.9% of the total variation. Although the chicory used in traditional forcing in soil and the red chicory cultivars were related more to each other and to the chicory cultivars from the witloof group, they still formed separate groups in the principal component plot.  相似文献

With the aim of unravelling the role of airborne Fusarium graminearum inoculum in the epidemic of Fusarium head blight (FHB) caused by this species in wheat spikes, a network of Burkard air samplers was set up in five wheat fields distributed in Belgium from 2011 to 2013. Each year from April to July, the daily amounts of F. graminearum inoculum above the wheat canopy were quantified using a newly developed TaqMan qPCR assay. The pattern of spore trapping observed was drastically different per year and per location with a frequency of detection between 9 and 66% and a mean daily concentration between 0.8 and 10.2 conidia-equivalent/m3. In one location, air was sampled for a whole year. Inoculum was frequently detected from the wheat stem elongation stage until the end of the harvesting period, but high inoculum levels were also observed during the fall. Using a window-pane analysis, different periods of time around wheat flowering (varying in length and starting date) were investigated for their importance in the relation between airborne inoculum and FHB parameters (FHB severity, frequency of F. graminearum infection and DON). For almost all the combinations of variables, strong and significant correlations were found for multiple window lengths and starting times. Inoculum quantities trapped around flowering were highly correlated with F. graminearum infection (up to R?=?0.84) and DON (up to R?=?0.9). Frequencies of detection were also well correlated with both of these parameters. DON concentrations at harvest could even be significantly associated with the F. graminearum inoculum trapped during periods finishing before the beginning of the anthesis (R?=?0.77). Overall, these results highlight the key role of the airborne inoculum in F. graminearum epidemics and underline the importance of monitoring it for the development of disease forecasting tools.  相似文献

One‐hundred and twenty‐four amplified fragment length polymorphism (AFLP) and 49 random amplified polymorphic DNA (RAPD) markers have been used to distinguish between 20 and 23 commercial chicory cultivars, respectively. These were all Cichorium intybus var. foliosum F1 hybrids, currently used in hydroponic forcing. Five‐hundred and twenty RAPD primers (OPERON) were tested, of which 156 resulted in reproducible patterns and 26 yielded polymorphisms. Two‐hundred and fifty‐six AFLP primer‐combinations were tested and six combinations were selected for identification purposes. Similarity indices were measured and clustering has been done using pairwise comparison. Both types of marker provide similar conclusions. Two major clusters are formed, representing late and early cultivars. All cultivars were identified using 10 informative RAPD primers or three AFLP primer combinations. A low degree of polymorphism was detected between some early cultivars, suggesting a narrow genetic base in their breeding strategy.  相似文献

The purpose of this study was to gain knowledge about the micromorphology of the porcine common carotid artery (CCA) during the period of growth over the bodyweight range of 10–40 kg. CCA samples from German landrace pigs (DL) aged either 2 or 3 months (DL-2 and DL-3) were compared with samples from Göttingen minipigs (GM) aged either 18 or 40 months (GM-18 and GM-40) using transmitted light (phase-contrast mode) and transmission electron microscopy. The GM-18, GM-40 and the DL-3 groups had typical muscular artery histological characteristics. Contrasting to this, the 2-month-old DL pigs had a transitional artery type being characterized by a significantly higher proportion of elastic fibres and a significantly lower number of smooth muscle cells than did the 1 month older DL-3. During the period of maturation, the tunica media of the CCA in GM animals thickened by 1.3× and in DL animals by 2.5× resulting in an overall increased vessel wall thickness. The cumulated thickness of the tunica interna (endothelium, stratum subendotheliale and internal elastic lamina) and the tunica media (including the external elastic lamina) of DL-3 and GM-40 pigs were similar to each other and comparable to that of humans. With an increasing vessel wall thickness, the luminal diameter decreased in GM by 19% and in DL by 11%. Additionally, in the older age groups, GM-40 and DL-3, the internal elastic lamina principally was continuous, but there were also interrupted large segments of elastic lamina separated by gaps. In addition, the principal internal elastic lamina was duplicated in several places.  相似文献

The present study investigated genetic diversity, structure and hybridization in a collection of the endangered wild pear species Pyrus pyraster (L.) Burgd. A total of 278 putative ‘true type’ P. pyraster trees originating from seven populations in the federal state of Saxony in Germany were analyzed along with 35 pear cultivars commonly cultivated in Saxony. The genetic analysis was performed using nine nuclear microsatellite markers (ncSSR) and two paternally inherited chloroplast marker (cpDNA) amplifying in the intergenic spacer region trnQrps16 and the intron region rps16. On basis of the ncSSR dataset after STRUCTURE analysis 80 % of the wild pear individuals were assigned as ‘true type’ P. pyraster genotypes. The cpDNA analysis showed shared haplotypes in P. pyraster and P. communis but with an unequal frequency in both species. The analysis of molecular variance resulted in a moderate (ncSSR) and great (cpDNA) variation among ‘true type’ P. pyraster and the pear cultivars. The genetic diversity in the ‘true type’ P. pyraster populations was still high and the genetic structure between the populations low (ncSSR and cpDNA) indicating a genetic exchange between the populations by pollen and seeds. The clear discrimination between the P. pyraster and P. communis confirms our expectation of the existence of ‘true type’ P. pyraster individuals in the study area. The existing genetic integrity and the high genetic diversity argue for the implementation of preservation measures in P. pyraster.  相似文献

Three ornamental bromeliads, i.e. Aechmea ‘Maya’ (CAM), Aechmea fasciata ‘Primera’ (CAM) and Guzmania ‘Hilda’ (C3) were grown under greenhouse conditions at ambient (380 ppm) and elevated (750 ppm) CO2. Malus sylvestris (L.) Mill. is the only indigenous wild apple species in Europe and recognized as rare and endangered species. An important prerequisite for the preservation of M. sylvestris is the identification of ‘true type’ M. sylvestris individuals because hybrids with cultivated apple could be also occurred. In this study variations among 625 putative M. sylvestris trees originated in the East Ore Mountains were evaluated with 20 selected morphological traits. The importance of these traits concerning the differentiation of ‘true type’ and hybrids of M. sylvestris were determined by statistical analysis. The results demonstrated a strong correlation of individual traits and those contributing mostly to variation of M. sylvestris were detected by the Principal Component Analysis. The relationship of 284 M. sylvestris, 18 M.?×?domestica, M. floribunda 821 and M. robusta 5 individuals were calculated based on the morphological data and presented in a dendrogram. The individuals grouped in two main clusters and 68 hybrids of M. sylvestris were identified. The main result of the morphological data of these hybrids indicates that the grouping is mainly based on the flower and leaf pubescence and less on the fruit size.  相似文献

Bromeliaceae are tropical ornamentals with a variety of colors and inflorescence shapes. Leaves with spined margins are characteristic for the genus Aechmea. Crosses between spineless and spined Aechmea cultivars were attempted in order to obtain a spineless offspring. Crossing spined and spineless genotypes resulted in a progeny with on average 50% spineless plants. The hybrids of two spineless plants had on average 75% spineless leaves. This indicated that the spineless character was dominant. Homozygous plants were selected; these plants are interesting for further breeding and propagation by seed.
