Ashwagandha (Withania somnifera) is an important medicinal plant and its dried roots are used in traditional systems of medicine. The market price of roots is determined by physical (textural) quality. Brittle roots with high starch and low fiber are considered to be superior because of ease in grinding. Genetic diversity studies based on root textural parameters have not been done so far. So the present study was designed to assess genetic diversity for morphometric traits and root textural quality parameters among two morphologically distinct groups: Poshita and Nagore. The PCA separated the morphometric and root texture variables distinctly into two different principal components: PC-1 and PC-2 respectively, indicating that both are negatively associated. All the morphotypes in Poshita group showed high positive loadings in PC-1 indicating that component genotypes are high root yielding. Nagore morphotypes were low yielding but the root texture was good. Clustering of morphotypes grouped Poshita and Nagore separately with high inter-cluster distances indicating that both groups are highly divergent from each other, suggesting that there is sufficient scope for varietal improvement through hybridization.  相似文献

Characterization of microsatellites is extremely important for the development of molecular markers. Here, we present the detection and abundance of microsatellites or simple sequence repeats (SSRs) in UniGene sequences of Citrus sinensis. A total of 427 SSRs were mined in 8786 UniGene sequences downloaded from National Center for Biotechnology Information (NCBI). Depending on the repeat units, the length of SSRs ranged from 14 to 21 for mono-, 14 to 48 for di-, 18 to 48 for tri-, 24 to 40 for tetra- and 42 bp for hexa-nucleotide repeats. Average density of SSRs (1SSR/12.92 kb of 5518.71 kb sequences mined) suggests that only 4.43% of sequences contained SSRs. Di-nucleotide repeats were most frequent repeat type (49.41%) followed by tri-nucleotide repeats (41.45%). An attempt was made to design primer pairs for 427 identified SSRs but these were found only for 216 sequences. The positions of SSRs with respect to open reading frame (ORF) detected and annotation of sequences containing SSRs were also carried out to assign function to each of the sequences.  相似文献

Summary A comparative study of the standard tube agglutination test (SAT), Rose Bengal plate agglutination test and counter immuno-electrophoresis (CIEP) was made on 647 sera from naturally aborting ewes, orchitic, in-contact and apparently healthy sheep with no history of vaccination against brucellosis. No individual test could detect all the 13 known positive reactors (the foetuses of which yieldedBrucella melitensis) but by combination of two tests all 13 were positive. The SAT detected more reactors during the early stage of infection while CIEP performed better in later stages of infection. All these tests may be carried out in a field laboratory at very low cost.
Resumen Se llevó a cabo un estudio donde se comparó la efectividad de la aglutinación en tubo, la prueba Rosa de Bengala y la contrainmunoelectroforesis en 647 sueros de ovejas que habían abortado naturalmente, machos con orquitis, ovejas en contacto y ovejas aparentemente sanas, sin previa historia de vacunación contra brucelosis. Ninguna de las pruebas utilizadas pudo detectar individualmente todos los 13 reactores positivos (de los fetos de los cuales se aislóBrucella melitensis), pero la combinación de dos pruebas detectó todos los reactores positivos. La prueba de la aglutinación en tubo detectó la mayoría de reactores en los estadíos iniciales de la infección, mientras que la contrainmunoelectroforesis se comportó bien en estadíos iniciales de la infección, mientras que la contrainmunoelectroforesis se comportó bien en estadíos tartíos de la infección. Todas estas pruebas pueden realizarse, en laboratorios de campo a un costo bajo.

Résumé Les auteurs ont réalisé une étude comparative entre l'agglutination en tube standard (SAT), le test d'agglutination sur lame au Rose Bengale (RBPT) et la contre immunoélectrophorèse (CIEP), à partir de 647 sérums prélevés sur des brebis ayant avorté naturellement ou sur des moutons atteints d'orchite ou ayant été en contact mais restés apparement sains et sans passé vaccinal contre la brucellose. Aucun test individuel n'a pu reconna?tre comme positif l'un des 13 animaux reconnus réagissant (dont les foetus hébergeaientBrucella melitensis), mais par combinaison des deux tests, tous les 13 se sont révélés positifs. Le SAT a décelé plus de réagissants au cours des premiers stades de l'infection. Par contre, le CIEP a été plus sensible dans les derniers stades. Tous ces tests peuvent être réalisés dans un laboratoire de terrain à, un co?t trés bas.

Biomass and carbon sequestration rate of a young (four year old) mixed plantation of Dalbergia sissoo Roxb., Acacia catechu Willd., and Albizia lebbeck Benth. growing in Terai region (a level area of superabundant water) of central Himalaya was estimated. The plantation is seed sown in the rainy season of year 2004 and spread over an area of 44 ha. Allometric equations for both above and below ground components were developed for three tree species. The density of trees in the plantation was 1322 trees ha−1 The diameters of trees were below 10 cm. Five diameter classes were defined for D. sissoo and A. catechu and 3 for A. lebbeck. 5 trees were harvested in each diameter class. Individual tree allometry was exercised for developing the allometric equations relating tree component (low and above ground) biomass to d.b.h. Post analysis equations were highly significant (P > 0.001) for each component of all species. In the plantation Holoptelia integrifolia Roxb. (Family Ulmaceae) has been reduced to shrub form because of frost. Only the aboveground biomass of H. integrifolia and other shrubs were estimated by destructive harvesting method. Herbaceous forest floor biomass and leaf litter fall were also estimated. The total forest vegetation biomass was 10.86 Mg ha−1 in 2008 which increased to 19.49 Mg ha−1 in 2009. The forest is sequestering carbon at the rate of 4.32 Mg ha−1 yr−1.  相似文献

Calcium uptake in experimentally induced scorbutic snake heads, Channa (= Ophicephalus) punctatus, fed vitamin C-deficient diet for 210 days is reported, using 45Ca as tracer. When compared with a parallel control on completely synthetic diet, the results show decreased absorption and utilization of calcium by gill, skin, muscle and bone of scorbutic fish from surrounding water. The physiological significance of these results is discussed, and its practical utility in aquaculture is indicated.  相似文献

This study was undertaken to ascertain the antipsychotic properties of Rauwolfia tetraphylla L. leaves and to isolate and characterize the antipsychotic constituents. Among the MeOH extract and some alkaloidal fractions at different pHs, the alkaloidal CHCl(3) fraction at pH-9 (2C) showed the highest antipsychotic activity against dopaminergic (DA-D(2)) and serotonergic (5-HT(2A)) receptors in-vitro and amphetamine induced hyperactive mouse model in-vivo. The activity guided isolation of CHCl(3) fraction (2C) afforded six indole alkaloids: 10-methoxytetrahydroalstonine (1), isoreserpiline (2), an isomeric mixture of 11-demethoxyreserpiline (3) and 10-demethoxyreserpiline (4), α-yohimbine (5) and reserpiline (6). Given orally, alkaloids 3-6 showed significant antipsychotic activity in a dose dependent manner. None of the extract, alkaloidal fractions or alkaloids showed any extra pyramidal symptoms at the tested doses. It was also observed that MeOH extract was behaving similar to other clinically used novel atypical antipsychotics in having 5-HT(2A) occupancy greater than the DA-D(2) receptor at the tested doses. Further toxicity and safety evaluation studies of MeOH extracts of R. tetraphylla leaves at different doses (10, 100, 300 and 2000 mg/kg) on female Swiss albino mice showed that MeOH extract is non toxic. The isolated alkaloids, 3-6 could serve as a promising lead structure for drug development of treating psychotic conditions in human.  相似文献

Climate is a critical factor affecting forest ecosystems and their capacity to produce goods and services. Effects of climate change on forests depend on ecosystem-specific factors including dimensions of climate (temperature, precipitation, drought, wind etc.). Available information is not sufficient to support a quantitative assessment of the ecological, social and economic consequences. The present study assessed shifts in forest cover types of Western Himalayan Eco-region (700?4500 m). 100 randomly selected samples (75 for training and 25 for testing the model), genetic algorithm of rule set parameters and climatic envelopes were used to assess the distribution of five prominent forest cover types (Temperate evergreen, Tropical semi-evergreen, Temperate conifer, Subtropical conifer, and Tropical moist deciduous forests). Modelling was conducted for four different scenarios, current scenario, changed precipitation (8% increase), changed temperature (1.07°C increase), and both changed temperature and precipitation. On increasing precipitation a downward shift in the temperate evergreen and tropical semi-evergreen was observed, while sub-tropical conifer and tropical moist-deciduous forests showed a slight upward shift and temperate conifer showed no shift. On increasing temperature, an upward shift in all forest types was observed except sub-tropical conifer forests without significant changes. When both temperature and precipitation were changed, the actual distribution was maintained and slight upward shift was observed in all the forest types except sub-tropical conifer. It is important to understand the likely impacts of the projected climate change on the forest ecosystems, so that better management and conservation strategies can be adopted for the biodiversity and forest dependent community. Knowledge of impact mechanisms also enables identification and mitigation of some of the conditions that increase vulnerability to climate change in the forest sector.  相似文献
