Harvesting, handling and transporting quails to the slaughterhouses, other farms and laboratories might covertly reduce their welfare. The aim of this study was to evaluate the effects of two major sources of energy in poultry nutrition on reducing transportation stress in Japanese quail (Coturnix Coturnix japonica). Male quails (n = 60) were divided into two groups. The first group was fed corn‐based diet, and the second was fed wheat‐based diet supplemented with xylanase and phytase. At the end of the experiment (day 35), quails were subjected to 80 km of transportation. Immediately on arrival and after 24 h, heterophil counts, lymphocyte counts and H:L ratios were measured. On arrival, H counts were lower, L counts were higher, and H:L ratios were lower for corn‐fed group. After 24 h, wheat‐fed group showed lower increment of H counts, greater increment of L counts and also decrement of H:L ratios rather than corn‐fed group which showed increment of H:L ratios. However, these ratios were still lower in corn‐fed group. Results indicate that corn‐based diets can help Japanese quail to better resist transportation stress, although it seems that feeding wheat‐based diets supplemented with xylanase and phytase could have positive effects for coping better with stress after journeys.  相似文献

Accurate diagnosis of foetal sex in pregnant mare is helpful for many breeders, both for private or commercial purposes. In this study, in order to pre-natal foetal sexing in equine, we used TaqMan duplex real-time PCR to detect the specific regions of SRY and TSPY genes on extracted cell-free foetal DNA from maternal blood. Peripheral blood samples from 50 pregnant Arabian mares with singleton foetuses were collected. Cell-free foetal DNA was extracted from maternal plasma, and duplex real-time PCR assays were performed with TaqMan probes and primers. Amplification of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene was used as control of DNA extraction procedure. From the 50 sampled mares, 28 cases had female and 22 mares had male foetuses. The final results for 46 samples were conclusive, and from them, 43 cases were predicted correctly. Sensitivity, specificity and accuracy of the test were 90.48%, 96% and 93.48%, respectively. In conclusion, a TaqMan duplex real-time PCR was set up to pre-natal detection of foetal sex in equine. The method was fast and decreased the false-positive and false-negative results. The technique can be used as a routine procedure in farms by collecting only a blood sample.  相似文献

Summary A simple laboratory method was developed to measure changes in saturated hydraulic conductivity and bulk density as affected by intermittent saturating and draining of soil columns. Significant changes in soil properties were measured after intermittent wetting and draining. Laboratory experiments showed significant changes in bulk density and saturated hydraulic conductivity of soil samples during drainage period following saturation of initially dried samples. The rate of changes in soil properties during the drainage period was a function of soil type and degree of desaturation. The effect of intermittent application of water in reducing the soil hydraulic conductivity and subsequently the infiltration rate was verified in the field by measuring the changes in soil intake rate during intermittent (Surge-flow) irrigation.  相似文献

Extraction using ethylenediaminetetraacetic acid (EDTA), and other chelates has been demonstrated to be an effective method of removal of Pb from many contaminated soils. However, column leaching of Pb from alkaline soils with EDTA has been problematic due to extremely low soil permeability. The first purpose of this study was to develop batch extraction procedures and methods of analysis of batch extraction data to provide Pb solubility information which can be used to model the column extraction of Pb from soils. The second purpose was to determine the effect of the addition of KOH and CaCl2 to K2H2EDTA extract solution on both hydraulic conductivity and Pb removal. A Pb-contaminated soil sample was collected from an abandoned battery recycling facility. Both batch shaker extractions and column leaching experiments were completed using 5 different EDTA extract solutions. When only CaCl2 was added to EDTA no change in the amount of Pb removed by batch extraction was observed. As expected, lead solubility was observed to decrease as pH was increased by the addition of KOH. However, Pb solubility was only slightly decreased by the addition of both CaCl2 and KOH. The amount of time required to leach 6.0 L of extraction solution through the soil columns varied from 2 to 33 days. The addition of CaCl2 and/or KOH resulted in increased soil hydraulic conductivity relative to the EDTA-only solution. The hydraulic conductivity was related to residual calcium carbonate content, suggesting that dissolution of CaCO3 and subsequent production of CO2 gas in the soil pores was partially responsible for the observed reductions in soil permeability. However, Pb removal was diminished with the addition of CaCl2 and KOH because of the decreased Pb solubility and also kinetic limitations associated with the shorter residence time of the extract solution in the column.  相似文献

Lead contamination of soils is a common problem throughout the world. Laboratory batch test and bench scale experiments have shown that EDTA can be used to remove lead from contaminated soils. However, due to the high cost and laborious task associated with actual environmental remediation of a lead contaminated soil, there is a need to be able to predict the outcome of a remediation process in advance in order to optimize the process and minimize the cost. This paper describes the development and validation of a computer model which can be used to simulate the removal of lead from a contaminated soil column using EDTA as the chelating agent. The model is able to simulate the lead removal from soil based on equilibrium as well as kinetic dissolution of Pb. The comparison of the simulated results with actual lead concentrations both in effluent and soil shows that the model can predict the lead removal process with reasonable accuracy.  相似文献

A simple irrigation scheduling approach for pecans   总被引:1,自引:0,他引:1  
Pecans are a major crop in New Mexico's Lower Rio Grande Valley (LRGV). It is estimated that New Mexico is responsible for about 21% of the world's pecan production (Lillywhite et al., 2007). Currently, approximately 12,000 ha of pecan orchards at various stages of growth consume 45% of the area's irrigation water. Pecan evapotranspiration (ET) varies with age, canopy cover, soil type, crop density and method of water management. Intense competition for the LRGV's limited water supply has created a serious need for better water management through improved irrigation scheduling. Annual pecan ET ranges from as low as 500 mm to as high as 1400 mm. Diversity of the pecan crop coefficient (Kc) and ET makes the task of irrigation scheduling for this crop very complicated. Using remote sensing technology and field ET measurements, a simple relationship was developed to relate crop coefficient and ET to canopy cover. This relationship is then used in combination with climate data to calculate daily and weekly water requirements for each orchard. The difference between annual ET values estimated from canopy cover and values measured with an eddy covariance flux tower ranged from 2 to 5%. The average ratio of estimated monthly ET values over measured ET values was 1.03 with the standard error of the estimate ranging from 10 to 20 mm/month. This methodology provides a simple tool that farmers can use to schedule irrigation of pecan orchards. Even though the methodology was developed for irrigation scheduling in the LRGV, it can be used in other locations by transferring the reference crop coefficients using Kc-GDD relationships.  相似文献

A wild strain of Eimeria tenella was isolated and utilized for immunization studies. Its optimal sporulation was attained at room temperature 24-25 degrees C after 24-48 h. Two groups of chicks were immunized by dosing a graded dose of five oocysts/chick/day for 6 days followed by 50 oocysts/chick/day for 7 days. A third group was not immunized and served as a negative control. Immunized chicks gained mass at the same rate as unimmunized ones, but when challenged with 200,000 oocysts/chick, mass gains declined in the unimmunized group. The growth rate of immunized chicks was not affected by challenge (P > 0.05). Upon challenge, unimmunized chicks produced significantly more oocysts than immunized chicks (P < 0.005). Immunized chicks withstood a challenged with 200,000 oocysts/chick without developing any clinical signs whereas the unimmunized chicks developed typical clinical signs of coccidiosis. Unimmunized chicks had significantly more severe lesions in the caecum than any other group (P > 0.005) and also produced significantly more oocysts than any other group (P > 0.005).  相似文献

Pecan is a major crop in the lower Rio Grande Valley (LRGV), New Mexico. Currently, about 11,000 ha of pecan orchards at various stages of growth are consuming about 40% of irrigation water in the area. Pecan evapotranspiration (ET) varies with age, canopy cover, soil type and method of water management. There is a need for better quantification of pecan ET for the purpose of water rights adjudication, watershed management and agronomical practices. This paper describes a process where remote sensing information from Landsat-5 and Landsat-7 were combined with ground level measurements to estimate pecan ET and field scale actual crop coefficient (K c) for the LRGV. The results showed that annual pecan water use for 279 fields ranged from 498 to 1,259 mm with an average water use of 1,054 mm. For fields with NDVI > 0.6 (normalized difference vegetation index), which represented mature orchards (total of 232 fields), the annual water use ranged from 771 to 1,259 mm with an average water use of 1,077 mm. The results from remote sensing model compared reasonably well with ground level ET values determined by an eddy covariance system in a mature pecan orchard with an average error of 4% and the standard error of estimate (SEE) ranging from 0.91 to 1.06 mm/day. A small fraction (5%) of the pecan fields were within the range of maximum ET and K c.  相似文献
