Piscirickettsia salmonisis the causative bacterial pathogen of piscirickettsiosis, a salmonid disease that causes notable mortalities in the worldwide aquaculture industry. Published research describes the phenotypic traits, virulence factors, pathogenicity and antibiotic‐resistance potential for various P. salmonisstrains. However, evolutionary and genetic information is scarce for P. salmonis. The present study used multilocus sequence typing (MLST) to gain insight into the population structure and evolution of P. salmonis. Forty‐two Chilean P. salmonisisolates, as well as the type strain LF‐89T, were recovered from diseased Salmo salar, Oncorhynchus kisutchand Oncorhynchus mykissfrom two Chilean Regions. MLST assessed the loci sequences of dnaK, efp, fumC, glyA, murG, rpoD and trpB. Bioinformatics analyses established the genetic diversity among P. salmonis isolates (H = 0.5810). A total of 23 sequence types (ST) were identified, 53.48% of which were represented by ST1, ST5 and ST2. Population structure analysis through polymorphism patterns showed few polymorphic sites (218 nucleotides from 4,010 bp), while dN/dS ratio analysis indicated purifying selection for dnaK, epf, fumC, murG, and rpoD but neutral selection for the trpB loci. The standardized index of association indicated strong linkage disequilibrium, suggesting clonal population structure. However, recombination events were detected in a group of seven isolates. Findings included genogroups homologous to the LF‐89T and EM‐90 strains, as well as a seven‐isolate hybrid genogroup recovered from both assessed regions (three O. mykiss and four S. salar isolates). The presented MLST scheme has comparative potential, with promising applications in studying distinct P. salmonis isolates (e.g., from different hosts, farms, geographical areas) and in understanding the epidemiology of this pathogen.  相似文献

Nutrient content and pH of rain samples collected at nine sites during 1988-1996 were studied to determine the amounts of N, P and S added by precipitation over Maracaibo Lake basin. The basin is a wide depression occupied by Lake Maracaibo and extensive alluvial plains and delimited by a mountainous zone of variable physiolgraphic characteristics with different cattle ranching and industrial activities. Rain pHs averaged ≈ 4 to 6, individual samples occasionally reached as low as pH 3 in the south basin. NH4 + levels were significantly higher (up to 2.6 mg/l NH4-N) than NO3 ? plus NO2 ?. Phosphorus levels were low, ranging from undetectable to 1.5 mg/l. The amounts of inorganic N/ha/yr added ranged from ≈ 5.4 Kg in the north to ≈ 8 Kg in the south basin; around the lakeshore, it was estimated that ≈ 9.8–16.8 Kg TN/ha (≈ 55 % as organic-N) are annually deposited. The annual amounts of SO4-S/ha added range from ≈ 10 to 13 Kg. The contribution of precipitation to P in the ecosystem is very small; ≈ 0.02 – 0.8 Kg of PO4-P/ha/yr was added in basin. The results of this study show that precipitation represents a significant factor for the total nitrogen and sulfur loading to Maracaibo Lake Basin.  相似文献

In order to reduce nutrient wastes to the environment the supply should be in accordance to the demand for these. Two experiments were conducted to study and quantify the effect of temperature, irradiance, and plant age on the uptake of nitrate (NO3?), ammonium (NH4+), dihydrogen phosphate ion (H2PO4?), potassium (K+), calcium (Ca2+), magnesium (Mg2+), and sulfate (SO42). In the first experiment, various levels of temperature and irradiance were applied to plants in a growth chamber, while in the second experiment the uptake was studied along the crop season under greenhouse conditions. The uptake rates were calculated at 2-hour intervals through sampling the nutrient solution and analyzing it by inductively coupled plasma atomic emission spectrometry (ICP-AES). Increasing light and temperature enhance the uptake rates, while the rates decrease with plant age. Nitrogen absorption was similar during the day as during the night. No differences were found in the absorption of H2PO4?, K+, Ca2+, Mg2+, and SO42? between day and night. Nitrate absorption was found to have a positive correlation with the absorption of all the ions except for NH4+.  相似文献

In order to make an estimation of thewet deposition levels of the major nitrogen compoundsin Lake Maracaibo system, the precipitation wascollected by events at five sites located on shorearound the Lake from 1991 to 1994. Also the phosphorusconcentrations were determined in rain samples. RainpHs averaged about 5 to 6. NH4 + levels weresignificantly higher (up to 1.44 mg L-1NH4-N) than NO3 -plus NO2 -ions which resulted in the highest pHs values in thestrait Maracaibo. The strait is the nearest zone toAmmonia Plant located upwind at the TablazoPetrochemical Complex. At the strait, theinorganic-nitrogen ((NH4 + NO3 +NO2)-N) concentrations were greater than organicnitrogen levels, but at the other sites were lower.Phosphorus levels were low, ranging from undetectableto 1.5 mg L-1; organic-P was estimated to accountfor about 40% of total-P. Total mean nitrogen wetdepositions were 6.93 kg ha-1 yr-1 (≈59% asNH4-N) at the strait and 9.85–16.84 kg ha-1yr-1 (≈55% as organic-N) around the lakeshore.Inorganic and organic nitrogen account for about equalportions of the total nitrogen loading. These amountsrepresents ≈24% of the annual total-N inputsby the tributary rivers to the lake and aresubstantially higher than those reported in the shoreof Lake Valencia (north-central part of Venezuela) andother tropical areas. The contribution ofprecipitation to P in lake is very small; ≈0.6 kg ha-1 of total-P are added annually. The N and Pinputs were greater than the sewage contributions. Themean molar ratio N/P is ≈22 at the straitsite whilst at lake sites the N/P ratio isconsistently >22 (about 35~1 to 100~1). The results of this preliminary study show that the atmosphere represents a significant factor for the total nitrogen loading to Lake Maracaibo system.  相似文献

Recent studies have shown that factors from adipose tissue influence and regulate the reproductive system. Hormones such as leptin and resistin are now known to regulate several reproductive processes. Adiponectin is the most abundant protein secreted by adipose tissue, and its circulating concentration is inversely related to adiposity and body mass index. Little is known about the involvement of adiponectin in reproduction. In the present study, the effect of recombinant adiponectin on the meiotic maturation and early embryo development in vitro was investigated, using porcine oocytes. Adiponectin receptors, AdipoR1 and AdipoR2, were found to be expressed in porcine oocytes and cumulus cells of both small and large follicles. Both AdipoR1 and AdipoR2 were immunolocalized to cumulus-oocyte complexes (COCs), oocytes, and early developing embryos. When included in oocyte maturation medium for 46 h, adiponectin significantly decreased the frequency of meiotic immature oocytes derived from large follicles (3-6 mm) but not from small follicles (<3mm). From studies of oocytes matured in the presence of adiponectin and mitogen-activated protein kinase (MAPK) pathway inhibitors MEK1 (PD98059), MEK1/2 (U0126), and p38MAPK (SB203580) it was concluded that adiponectin enhances oocyte maturation thought the p38MAPK pathway. Finally, a superior rate of embryo development to the blastocyst stage was achieved by embryos cultured in the presence of adiponectin. These results indicate that adiponectin has a positive effect on the meiotic maturation and in vitro embryo development of porcine oocytes and suggests a physiological role for this adipokine in early development in mammals.  相似文献

A greenhouse experiment was conducted to study and quantify the daily uptake rate of nitrate, ammonium, phosphoric acid, potassium, calcium, magnesium, and sulfate (NO3?, NH4+, H2PO4?, K+, Ca2+, Mg2+ and SO42?) and to characterize the uptake daily pattern at different plant ages in a lettuce crop. The uptake rates per gram of plant fresh weight were calculated at 2-hour intervals through sampling the nutrient solution and analyzing it by NO3?/NH4+ conductivity and inductively coupled plasma atomic emission spectrometry (ICP-AES). The uptake rate of nitrogen, phosphorus and potassium (N, P and K) per unit mass of plant decreased with plant age following a reduction in plant relative growth rate. No significant differences were found in the absorption of Ca, Mg and sulfur (S) between the different weeks of growth. The daily absorption patterns showed no preference for the absorption of any of the ions during the daytime. A significant reduction in the absorption peaks of all the ions with increasing plant age was observed.  相似文献

This study investigated the effect of porcine follicular fluid (PFF) and dibutyryl cyclic adenosine monophosphate (dbcAMP) during in vitro maturation (IVM) of porcine oocytes on meiotic maturation, fertilization and embryo development, and compared the effect of supplementing the embryo culture media with PFF or foetal bovine serum (FBS) on embryo development. Oocytes from pre‐pubertal gilts were IVM for 44 h, and parthenogenetically activated or in vitro‐fertilized. Embryos were cultured in porcine zygote medium (PZM3) for 7 days. Cleavage and blastocyst rates were evaluated at 48 h and 7 days of culture. The supplementation of the IVM medium with 25% PFF and 1 mm dbcAMP for the first 22 h resulted in more (p < 0.05) embryos developing to the blastocyst stage as compared with the inclusion of dbcAMP alone. The dbcAMP + PFF combination increased (p < 0.05) the average number of nuclei per blastocyst as compared with either of these components alone or in its absence. A synergistic effect of dbcAMP + PFF during IVM was also reflected in the capacity of oocytes to regulate sperm penetration and prevent polyspermy, as twice as many oocytes from the control group were penetrated by more than one sperm as compared with those matured in the presence of both dbcAMP and PFF. The supplementation of PZM3 with 10% FBS from days 5 to 7 of culture significantly improved the total cell quantity in embryos derived either from control or dbcAMP + PFF matured oocytes. There was no effect on the total cell quantity when FBS was replaced by the same concentration of PFF. These studies showed that dbcAMP, PFF and FBS can improve both the quantity (57.3% vs 41.5%) and quality (74.8 vs 33.3 nuclei) of porcine blastocysts derived from oocytes recovered of pre‐pubertal gilts.  相似文献

Under conditions of limited nutrient supply, plant nutrient uptake is controlled by the external concentration of the ions. Limited information exists about the whole-plant regulation of nutrient uptake when the supply is adequate. To study the relationship between growth rate and carbon dioxide (CO2) assimilation with nutrient uptake, growth chamber experiments were conducted with temperatures ranging from 10 to 35°C at medium (600 µmol m?2 s?1) and high (1200 µmol m?2 s?1) light intensities. Nutrient solution samples were collected every 24 hours and the concentration of ions was analyzed by Inductively coupled plasma -atomic emission spectroscopy (ICP-AES) and nitrate and ammonium (NO3?/NH4+) conductivity. Leaf photo-synthesis was measured using a closed gas exchange system and the total amount of CO2 assimilated was calculated from dry weight increases. The daily absorption of NO3?, Total nitrogen (N), dihydrogen phosphate (H2PO4?) and potassium (K+) responded linearly to plant growth, while ammonium (NH4+) and sulfate (SO42?) uptake showed a curvilinear response. All the ions studied showed a curvilinear relation with CO2 assimilation.  相似文献
