Bambara groundnut (Vigna subterranea) was fermented to produce a dawadawa-type product using a starter culture of Bacillus licheniformis isolated from naturally fermenting bambara groundnut beans. Fermentation was carried out at 30 and 37 °C for four days and at 45 °C for two days. The pH of the substrate decreased after 24 hours and then rose at 30 and 37 °C but remained constant at 45 °C after the initial drop. Total titratable acidity of the fermenting beans mimicked the pH values. Proximate analyses for moisture, protein and fat of the cotyledons showed an increase in all three constituent at each of the three fermentation temperatures. At the end of fermentation, total available carbohydrate was 55%, 59% and 62% of the original value at 30, 37 and 45 °C, respectively. Fermentation of bambara groundnut at 45 °C for two days is recommended as the ideal fermentation temperature and time.  相似文献

Cloacal swabs from 62 green iguanas (Iguana iguana), including 47 wild and 15 domestic ones from five parishes of Grenada, were sampled during a 4‐month period of January to April 2013 and examined by enrichment and selective culture for the presence of Salmonella spp. Fifty‐five per cent of the animals were positive, and eight serovars of Salmonella were isolated. The most common serovar was Rubislaw (58.8%), a serovar found recently in many cane toads in Grenada, followed by Oranienburg (14.7%), a serovar that has been causing serious human disease outbreaks in Japan. Serovar IV:48:g,z51:‐ (formerly, S. Marina) highly invasive and known for serious infections in children in the United States, constituted 11.8% of the isolates, all of them being from domestic green iguanas. Salmonella Newport, a serovar recently found in a blue land crab in Grenada, comprised 11.8% of the isolates from the green iguanas. The remaining four less frequent serovars included S. Javiana and S. Glostrup. Antimicrobial susceptibility tests conducted by a disc diffusion method against amoxicillin–clavulanic acid, ampicillin, cefotaxime, ceftazidime, ciprofloxacin, enrofloxacin, gentamicin, nalidixic acid, streptomycin, tetracycline and trimethoprim–sulfamethoxazole showed that drug resistance is minimal, with intermediate susceptibility, mainly to streptomycin, tetracycline and cefotaxime. This is the first report of isolation and antimicrobial susceptibilities of various Salmonella serovars from wild and domestic green iguanas in Grenada, West Indies.  相似文献

Soil and microbiological properties of a tropical rain forest soil were evaluated 17 years after oil spillage to access the chronic effects of, and interrelationship between population of petroleum hydrocarbon utilizing and nitrifying microorganisms. The spatial distribution of petroleum hydrocarbons (oil), the nutrient status and the abundance of heterotrophic microbes along soil transect lines in the contaminated zones served as the index for corroborating the results. The pH status of soil in the contaminated (Heavy impact — HI, and moderate inpact — MI) zones varied from acidic, that is 4.0 to near neutral PH, that is 6.0. The C content of soils decreased from 3.6% at the HI zones to 2.84% at the MI zones. Total N in the HI and MI zones differed by a factor of 0.10%. Available P was higher at the MI than HI zone, while CEC decreased from a combined mean of 6.48 at the HI zones to 4.46 at the MI zones. Although residual oil content was higher in the HI zones than MI zone, the soil nutrient status within these two zones did not vary significantly (P=0.05). However, soil microbes responded differently. For instance, petroleum hydrocarbon utilizers correlated positively with the distribution of oil in the environment. But, not the nitrifying microorganisms. Aerobic nitrifiers were abundant at the HI than MI zones, while anaerobic nitrifiers were higher at the MI than HI zones. With the presence of petroleum hydrocarbon utilizers anf nitrifying microbes. It is possible to enhance the degradation of oil in the 17 yr old spillage by adopting bioremediation.  相似文献

Intestinal samples from 156 small Indian mongooses (Herpestes auropunctatus) collected island-wide in Grenada from April 2011 to March 2013 were examined for the presence of Salmonella enterica spp. Nineteen (12%) mongooses were culture-positive for S. enterica spp. of which five serotypes were identified. Salmonella javiana and S. Montevideo were the most commonly isolated serotypes. The other serotypes isolated were S. Rubislaw, S. Panama and S. Arechavaleta. All isolates were susceptible to amoxicillin–clavulanic acid, ampicillin, cefotaxime, ceftazidime, ciprofloxacin, enrofloxacin, gentamicin, nalidixic acid, imipenem and trimethoprim–sulfamethoxazole. One isolate (S. Montevideo) showed resistance to tetracycline and intermediate resistance to streptomycin. The five isolated Salmonella serotypes are potential human pathogens suggesting that the mongoose may play a role in the epidemiology of human salmonellosis in Grenada.  相似文献

Percentage germination, plant height, leaf area, relative dry matter yield, plant nutrient concentration, soil chemical characteristics and source/sink relationship were assessed for maize (Zea mays L TZ-SR-Y) planted in a soil polluted with 3% (v/w) crude oil or 0% (untreated soil), after remediation of soil with different organic (Poultry manure, peptone water, sawdust and yeast extract) and inorganic (NPK, KNO3, NH4H2PO4 and (NH4)2SO4) nutrient supplements applied 7 days after oil treatment. Germination of maize in oil polluted soil was not significantly affected by nutrient supplementation (P=0.05). Maize performance in terms of other studied parameters was highest in the oil polluted soil supplemented with poultry manure, and least in oil polluted soil supplemented with sawdust. Oil treatment correlated significantly with organic C, N, Na, Mg and ferrous in the soil (P=0.05) and with nutrient composition in maize plant tissues (P=0.05). For nutrient uptake, significant correlation was only established between soil N and plant N. Population of petroleum hydrocarbon tolerant microbes increased initially but decreased with time. There is every indication that nutrient supplementation of oil polluted soil especially with organic nutrient sources is beneficial for maize growth, because the C/N ratio is narrowed while the rate of biodegradation of oil and soil recovery is also enhanced. Poultry manure is recommended, however sawdust is not recommended since it tends to impose adverse effect by widening the C/N ratio in soil.  相似文献

Study was carried out to determine the spoilage association of traditionally fermented ugba, product of African oil bean seed. Samples were collected from three markets within Enugu metropolis in Eastern Nigeria. Microbial population of ugba was isolated, characterized and identified. Effect of time on product quality, total viable cell count and individual growth pattern of isolates was evaluated. Microorganisms isolated were Proteus sp., Klebsiella sp., Staphylococcus epidermidis, E. coli, Bacillus licheniformis and Bacillus subtilis. Initial viable cell count increased from 4.6 x 10(8) -6.2 x 10(8), 6.0 x 10(9) -6.9 x 10(9) and 3.9 x 10(8) -5 x 10(8) cfu g(-1) by the 4th day for samples from Mayor, Kenyatta and Garki markets, respectively. Organoleptic changes in texture and colour were seen to be a factor of time of storage. A decline in growth of Proteus sp., E. coli, Klebsiella sp. and S. epidermidis by the 4th day of storage was observed for all samples. Viable cells of B. licheniformis increased from 6.1 x 10(7) -17.7 x 10(7) cfu g(-1), 13.9 x 10(8) -20.0 x 10(8) and 8.7 x 10(7) -15.5 x 10(7) by the 4th day of storage for samples from Mayor, Kenyatta and Garki markets respectively. Growth of B. subtilis increased from 9.2 x 10(7) -19.9 x 10(7), 14.9 x 10(8) -21.2 x 10(8) and 11.5 x 10(7) -17.2 x 10(7) cfu g(-1) for samples from Mayor, Kenyatta and Garki markets respectively. Our results indicate that ugba spoilage is primarily a result of the continued activity of African oil bean seed fermentative organisms B. subtilis and B. licheniformis.  相似文献

Bambara groundnut, an indigenous African legume, was subjected to fermentation by three strains of Rhizopus. One strain B. arrhizus could not ferment the substrate. Mycelial penetration and binding was good when strains NRRL 2710 ( R. oligosporus) and NRRL 1477 ( R. stolonifer) were used. Fermentation by both strains resulted in increases of pH, moisture, protein and fat while total carbohydrate decreased by 50%. Sensory evaluation showed that bambara groundnut tempes rated similar (p>0.5) in taste and texture and higher (p<0.05) in color and flavor than soybean tempe. Bambara groundnut would be an acceptable food product in the diet as a good protein supplement.  相似文献

Soil moisture is known to be a major control of greenhouse gas (GHG) emissions from agricultural soils. However, there is little data regarding GHG exchange from the organic matter-rich soils characteristic of shelterbelts—especially under elevated soil moisture conditions. In the present study, we quantified CO2, CH4 and N2O fluxes from shelterbelts under elevated soil moisture (irrigated) and semi-arid (rainfed) conditions. Studies were carried out at the Canada-Saskatchewan Irrigation Diversification Centre (CSIDC) near Outlook, Saskatchewan. Non-steady state vented chambers were used to monitor soil GHG fluxes from three shelterbelts in 2013 and 2014. The shelterbelts consisted of a single row of caragana with a north–south orientation and a single row of Scots pine with either a north–south or east–west orientation. Each shelterbelt was divided into two areas based on whether or not it received irrigation. During the 2-year study period, N2O emissions from the irrigated shelterbelts (IR-SB) (0.93 kg N2O-N ha?1) were significantly greater than those from the rainfed shelterbelts (RF-SB) (0.49 kg N2O-N ha?1). Soil CH4 oxidation was significantly lower in the IR-SB compared to the RF-SB (?0.85 and ?1.20 kg CH4-C ha?1, respectively). Irrigation activities stimulated CO2 production/emission in 2014, but had no effect on CO2 emissions during the much drier 2013 season. Correlation analyses indicate a strong dependence of CO2 and CH4 fluxes on soil moisture in both IR-SB and RF-SB sites. There was a significant relationship between N2O emissions and soil moisture for the IR-SB sites in 2013; however, no such relationship was observed in either the IR-SB or RF-SB sites in 2014. Our study suggests that changes in precipitation patterns and soil moisture regime due to climate change could affect soil-atmosphere exchange of GHGs in shelterbelts; however, elevated soil moisture effect on GHG emissions will depend on the availability of N and C in the shelterbelts.  相似文献

Cloacal swabs and caecal contents sampled from 58 cane toads (Bufo marinus) in St George’s parish, Grenada, during a 7‐month period in 2011 were examined by an enrichment and selective culture method for presence of Salmonella spp. Twenty‐four (41%) toads were positive for Salmonella spp. of which eight were Salmonella enterica serovar Javiana, and eight were S. enterica serovar Rubislaw. The other serovars were as follows: Montevideo, 6; Arechavaleta, 1; and serovar: IV:43:‐:‐, 1. The high frequency of isolation of serovar Javiana, an emerging human pathogen associated with several outbreaks in the recent years in the eastern United States, suggests a possible role for cane toads in transmission of this serovar. Although S. Rubislaw has been isolated from lizards, bats and cases of some human infections, there is no report of its carriage by cane toads, and in such high frequency. The rate of carriage of S. Montevideo, a cause for human foodborne outbreaks around the world was also over 10% in the 58 toads sampled in this study. The antimicrobial drug susceptibility tests against amoxicillin‐clavulanic acid, ampicillin, cefotaxime, ceftazidime, ciprofloxacin, enrofloxacin, gentamicin, imipenem, nalidixic acid, streptomycin, tetracycline and trimethoprim‐sulfamethoxazole showed that drug resistance is minimal and is of little concern. Antimicrobial resistance was limited to ampicillin and amoxicillin‐clavulanic acid in one isolate of S. Javiana and one isolate of S. Rubislaw. This is the first report of isolation and antimicrobial susceptibilities of various Salmonella serovars not identified previously in cane toads in Grenada, West Indies.  相似文献
