大肠杆菌耐热性肠毒素ST1—K99／LacZ基因重组的研究

将构建的ｐＢＳＴ２～６工程菌质粒用限制性内切酶ＢａｍＨＩ／ＢｇｌⅡ酶切，经琼脂糖凝胶电泳和电洗脱，回收１４７ｂｐ的目的ＳＴ１基因。随之将该基因分别重组到能有效表达Ｋ９９菌毛抗原和ＬａｃＺ酶的ｐＧＫ９９之Ｋ９９基因ＢｇｌⅡ位点和ｐＵＣ１８的ＢａｍＨＩ位点中。通过ＳＴ１基因探针菌落原位杂交、特定酶切分析及ＤＮＡ序列分析，筛选并鉴定出了理想重组子，从而构建出了能分别表达ＳＴ１融合基因产物的工程菌株ｐＳＫ２１９和ｐＸＳＴ１。  相似文献   

Bronchoalveolar lavage in horses: effect of exercise and repeated sampling on cytology

SUMMARY Bronchoalveolar lavage (BAL) was performed at weekly intervals in 10 Thoroughbred horses in race training (group 1) and in 4 rested horses (group 2) for 10 weeks. Lavages were continued on a weekly basis in 4 group 1 horses for an additional 9 weeks (group 3). Cytological analysis of samples included leukocyte counts, erythrocyte counts, differential leukocyte counts, and haemosiderophage score. The mean leukocyte concentration was significantly lower in group 1(92.1 ± 4.6 cells/μL) when compared with group 2 (133.5 ± 8.2 cells/μL), P = 0.037. The differential leukocyte data were not significantly different between groups. There was a large amount of variability in the percent-age of macrophages and lymphocytes in the differential counts over time with no obvious trends. The proportion of neutrophils demonstrated a tendency to decrease over time for both groups 1 and 2. Erythrocyte counts and haemosiderin scores were significantly higher in the exercising group than the rested horses. Neither exercise nor the technique itself evoked an inflammatory response in the BAL fluid.  相似文献

Pig breeding is generally conducted among many herds, so EBV comparisons across populations are necessary. Genetic connectedness is required for reliable between-farmanimal EBV comparisons.Five quantitative overall connectedness measures among populations have been proposed so far,coefficient of connectedness(γ*), coefficient of determination (CD) and overall indices of precision, connectedness rating, number of direct genetic links between subpopulations due to common sires and dams (GLt), and average genetic covariance (AGC) are reviewed and theirproperties are discussed in this paper. It is recommended to use AGC at present for measuring genetic connectedness between herds.  相似文献

应用PCR及Nested PCR技术检测柑桔木虱及其寄主九里香的结果表明:PCR只可检测最低2头带菌木虱,Nested PCR可检测到单个带菌木虱。100头带菌木虱中,单虫检出率为96％。检测田间重、中等、轻病的柑桔园内的木虱,其带菌率依次为87％、53％和21％。在病芦柑上饲菌不同天数的木虱均能检测出带菌,其饲菌时间最短为1d。城市九里香叶片及在其叶上取食的木虱单虫,均能用Nested PCR检测出病原。饲菌木虱接种九里香及芦柑健苗,在植株尚未表现症状时,常规PCR难检测出病原,但用Nested PCR则能检测到病原,说明九里香不仅是木虱的寄主,而且是黄龙病病原的隐症寄主。  相似文献   

出血性大肠杆菌O157eae-stx1/2B融合基因的构建和表达

构建表达eae和stx1／2B的融合基因，克隆eae基因的C端280个氨基酸残基(Int280)基因部分，以正确地阅读框定向插入到含有stx1／2B融合基因的质粒，构建重组质粒，将其转化于BL21(DE3)，用IPTG进行诱导表达，经SDS—PAGE电泳检测，该融合蛋白获得了高效表达。薄层扫描分析表明：目的蛋白表达量占菌体总蛋白含量的50．67％。由于该融合蛋白由eae、stx1B、stx2B等三部分抗原组成，可刺激机体产生针对紧密素和StxB的抗体，在EHEC O157亚单位疫苗设计或单克隆抗体抗制备中具有重要价值。  相似文献

日粮中添加牛至油对河西绒山羊育肥性能的影响研究

牛至精油对家畜生产性能和免疫力的提高有重要的影响。通过在日粮中添加牛至精油育肥河西绒山羊,研究其对河西绒山羊生长性能、屠宰性能及胴体品质的影响。对照组饲喂基础日粮,试验组在饲喂的基础日粮中分别添加4和7 g宜生饲宝(含牛至精油的添加剂),4和7 g宜生饲宝中牛至精油的含量分别为52和91 mg。对照组和试验组均为同月龄、同体重的河西绒山羊羯羊,试验组和对照组均设3个重复,每个重复5只羊。育肥期为90 d,每30 d为1个阶段。试验结果表明:育肥全期,对照组、4 g组和7 g组河西绒山羊平均日增重分别为96.30、172.22和169.82 g·d^-1,4 g组和7 g组分别显著高于对照组(P<0.05),4 g组和7 g组无显著差异(P>0.05);屠宰性状,胴体重对照组、4 g组和7 g组分别为20.24、24.18和23.90 kg,无显著差异(P>0.05),屠宰率对照组、4 g组和7 g组分别为48.86%、51.21%和51.56%,4 g组和7 g组分别显著高于对照组(P<0.05),4 g组和7 g组无显著差异(P>0.05),胴体净肉重对照组、4 g组和7 g组分别为12.90、16.27和15.99 kg,4 g组和7 g组分别显著高于对照组(P<0.05),4 g组和7 g组无显著差异(P>0.05);肉质性状,大理石纹4 g组和7 g组分别显著高于对照组(P<0.05),4 g组和7 g组无显著差异(P>0.05),pH、熟肉率、失水率、嫩度、蒸煮损失对照组、4 g组、7 g组间均无显著差异(P>0.05)。日粮中添加牛至油可以显著提高河西绒山羊的育肥性能,本试验日粮中添加4 g宜生饲宝(含牛至精油52 mg)效果最佳。  相似文献

木薯优良新品种桂垦09-26选育初报

2008年以华南5号为母本,华南205为父本,进行木薯品种间杂交,以选育高产、高淀粉、高抗逆性木薯新品种。当年获得杂交种子936颗,经多代系统选育,于2013年获得综合性状优良的新品系桂垦09-26,鲜薯产量达48.64 t/hm2,比对照华南205增产90.43%;淀粉含量达30.0%,与华南205相近。综合各种农艺性状表现,桂垦09-26可以进一步参加区域性试验和生产性试验。  相似文献   

The evolutionary patterns of intercity co-invention networks in the Greater Pearl River Delta,China: A comparative analysis based on the technological intensity of industry

A substantial amount of research has examined the collaborations within technology-based industries, but how technological intensity relates to the evolution of interregional research collaboration is less well understood. This paper investigates the dynamic changes of spatial structure and determinants of intercity research collaborations in the sectoral innovation networks of high-tech industries contrasting to low and medium technology (LMT) industries in the Greater Pearl River Delta (GPRD), China. Using the sectoral patent application data over the period 2008–2016, the social network analysis results confirm a higher level of triangles in the co-invention network of high-tech industries than within LMT industries. Moreover, the empirical results through Exponential Random Graph Model (ERGM) show that, compared to LMT industries, collaboration in the co-invention network of high-tech industries is less sensitive to geographical and linguistic distance, while more subject to economic distance, regional educational level and government support with time. In addition, cognitive proximity and regional R&D intensity are both important prerequisite in the formation of these two collaborative networks.  相似文献
